Selectivity of the optical isomers of KR30031 on MDR reversal activity and cardiotoxicity

  • The present study was performed to compare the cardiovascular adverse effects of verapamil. KR30031 and their each optical isomers, and also to measure their ability to overcome multidrug resistance (MDR). R-isomer of KR30031 (R-KR30031) was equipotent with S-isomer of KR30031 (S-KR30031) and 25 fold less potent than R-isomer of verapamil (R-verapamil) in relaxing the aorta isolated from rat (EC50: 11.8, 10.2 and 0.46 ${\mu}$M, respectively).   • The stereoselectivity on the electron-transfer reaction between optical active ${\bigwedge}$-[CO(EDDS)]- and conformationally restricted complex $[Co({\pm}chxn)_3]^{2+}$ has been examined in aqueous solution. The products are four conformational isomers $(lel_3,\;lel_2ob,\;lelob_2,\;and\;ob_3)$ of ${\bigwedge}$-[Co(chxn)$_3]^{3+}$ with optical purities of 22% e.e, 25% e.e, 11% e.e, and 10% e.e, respectively. The reaction between ${\bigwedge}$-[CO(EDDS)]- and $[Co({\pm}chxn)_3]^{2+}$ in DMSO produced lel3-${\Delta}$ and lel2ob-${\Delta}$-[Co(chxn)$_3]^{3+}$ whose optical purities are 100% e.e, and 75% e.e, respectively.

  • Using the plasma that we developed to generate a low-temperature plasma at atmospheric pressure, we have investigated the etching possibility of an air-exposed zinc oxide(ZnO) thin films. Hydrogen and methane radicals generated from the plasma were observed and their intensity was found to be dependent on the isomer of butyl acetate by an analysis with optical emission spectrosxopy. The etching ability of this plasma was evaluated by an emission intensity, etching time, rf power.

  • After establishment of methodology for the separation and quantitation of optical isomer existed in food or biomaterial, the relationship between isomer and nutrient or biological sample was investigated. The optical isomers of standard amino acids and free amino acids were quantitized and the protein was assayed from the Korean bean, pasted bean, soy sauce, gochujang, powderd milk and cataract followed by hydrolysis and dervatization with TFA-IPA for GLC analysis with chirasil val column. Amino acids showing the racemization were alanine, aspartic acid, glutamic acid and phenyl alanine. The most convenient amino acid deducing age and biological activity was aspartic acid. Glutamic acid and phenyl alanine have shown poor resolution with less racemization. The ratio of d-form amino acids was 3~6% for home made pasted bean, about 3% for commerical pasted bean, 2~4% for soy sauce, about 1% for bean, 1~2% for cataract, 1.0~1.5% for powdered milk. The racemization during fermentation process was significant.

  • (-)-(1R, 2R) and (+)-(1S, 2S)-menthyl-trans-2-phenylcyclopropanecarboxylat e have been synthesized with the aid of chiral Cu(II) complex catalyst by the addition reaction of l-menthyldiazoacetate to styrene. The yield was 75%, with the purity of trans isomer over 95% and the optical purity of 95%.

  • The effects of molecular environments on photoisomerization of an azobenzene group were investigated using In-situ UV/Vis spectroscopy and optical anisotropy measurement technique. The reversible and repeatable photoisomeritation reactions of azobenzene were observed by irradiating the film containing 4-hydroxyazobenzene and by measuring absorption intensities of the characteristic bands of trans and cis isomers simultaneously. When the self-assembled monolayer with azobenzene groups was used as an alignment layer for a liquid crystal cell, the homeotropic alignment was induced due to their compact packing structures of azobenfene groups along the vertical direction of the substrate. By irradiating UV light on this cell, the trans-azobenzene groups change to cis-isomers through the photoisonlerieation and then resulting in the planar alignment of liquid crystal molecules.

Development of Wide-Band Compensation Film to Improve Viewing Angle of Vertical Alignment Liquid Crystal Display (수직배향 액정디스플레이의 시야각 향상을 위한 광대역 보상필름 개발)

  • To expand the viewing angle of vertical alignment liquid crystal display (VA-LCD), a wide-band compensation film has been newly developed. VA-LCD needs a retardation film with improved oblique viewing property. The retardation film for VA-LCD has been fabricated by solvent casting and subsequent drawing triacetylcellulose (TAC) containing optical additives. Here, optical birefringence and wavelength dispersion of the retardation film strongly depend on the chemical structures of optical additives. Utilizing quantum mechanical computation, the polarizing anisotropy of retardation film was calculated with respect to additives. It is also realized that the wavelength dispersion of polarizing anisotropy depends on the type and location of substituent to the selected main isomer. When two propionates are substituted to the meta position of the selected main isomer, the wavelength dispersion of polarizing anisotropy shows the most gentle slop, which is well matched with the experimental results. The most gentle slop of wavelength dispersion means that the viewing angle characteristics of LCD are less influenced according to the wavelength of light source of LCD, and it's possible to make better image quality than the present level.

Synthesis of Stereoisomeric Trifluoroethylmandelates and Their Stereospecificity for the Uses as the Substrate of Lipases in Organic Solvent (유기용매내에서 리파제의 입체특이성 반응기질로서 이용하기 위한 Trifluoroethylmandelate의 이성질체 합성 및 입체특이성)

  • Stereoisomers of trifluoroethylmandelate(mandelic acid trifluoroethylester) were synthesized from each isomer of mandelic acid and trifluoroethanol with p-toluene sulfonic acid in order to study the enantioselectivity of lipase in organic solvent. The products were identified by $^1H$ NMR and elemental analysis and their physical properties such as melting point, densities and specific optical rotations($[{\alpha}]_{25}{^D}$) were also characterized. $[{\alpha}]_{25}{^D}$ of (+)- and (-)-trifluoroethylmandelate were +74° and -75.4°, respectively. The trifluoroethylmandelate was found out to be as a good substrate for the transesterfication stereoselectivity of lipases in organic solvent. Any significant difference of the lipase catalyzed transesterification activity between (+)- and (-)-methylchloropropionate was not found, and even lipase activity of transesterfication was not found with high optical polar (+)-and (-)-methylmandelate.

Enantioselective Membranes Based on Chitosan for The Separation of D- And L-Tryptophan

  • Chitosan membranes crosslinked with glutaraldehayde that contained chiral environment were prepared. The chitosan membranes were characterized using FTIR and swelling index measurements. Their swelling index in water ranged from 100 to 70%, depending on the crosslinking time. The separation of D- and L-isomers of tryptophan was achieved through a pressure driven membrane separation process, using the self-supporting crosslinked chitosan membranes. The chiral separation performance of the membranes depended strongly on the swelling index of the membranes and the separation conditions such as concentration of feed solutions and different operating pressures. Especially when a chitosan membrane with a swelling index of 70% was used, almost complete optical resolution of D- and L-tryptophan was obtained ; enantiomeric excess (ee %) of 97.92% and flux of 2.26 g/㎡$.$h.
