Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats (무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. Determination of Cytoplasmic Male Sterile Factors in Onion Plants (Allium cepa L.) Using PCR-RFLP and SNP Markers

  • We have developed a polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) marker that can distinguish male-fertile (N) and male-sterile (S) cytoplasm in onions. The PCR-RFLP marker was located in a chloroplast psbA gene amplicon. Digesting the amplicons from different cytoplasm-containing varieties with the restriction enzyme MspI revealed that N-cytoplasm plants have a functional MspI site (CCGG), whereas the S-cytoplasm plants has a substitution in that site (CTGG), and thus no MspI target. The results obtained using this PCR-RFLP marker to distinguish between cytoplasmic male sterile factors in 35 onion varieties corresponded with those using a CMS-specific sequence-characterized amplified region (SCAR) marker. Moreover, the PCR-RFLP marker can identify N- ot S-cytoplasms in DNA sample mixtures in which they are in up to a 10-fold minority, indicating that use of the marker has high diagnostic precision. We also demonstrated the usefulness of the SNP detected in the psbA gene for high-throughput discrimination of CMS factors using Real-time PCR and a TaqMan probe assay.

Effect of Priming Conditions on Enhancing Germination of Onion (Allium cepa L.) Seeds (Priming 조건이 양파 종자의 발아율 향상에 미치는 영향)

  • Priming of seeds in 150 mM KCI at $10^{\circ}C$ for 6 days was most effective in increasing germination rate and shortening germination time. Germination rate of primed seeds was increased to 96% which is about 14% higher than that of non-primed seeds. Germination temperature for better priming effects from primed onion seeds was 10 to $15^{\circ}C$ which is relatively lower than that of normal germination temperature of $20^{\circ}C$. Drying primed seeds at $20^{\circ}C$ for 5 hours resulted in best priming effects.

Major constituents and antioxidant activities of domestic onion (Allium cepa L.) cultivars (국내 육종 양파 품종별 주요성분의 함량 및 항산화활성 비교)

  • The content of major constituents and antioxidant activities of nine onion cultivars, namely yellow domestic cultivars Meapsihwang, Sinsunhwang, Yeonsinhwang, Yeongbohwang, and Yeongpoonghwang, red domestic cultivar Eumjinara, green domestic cultivar Sweetgreen, yellow-introduced cultivar Sunpower, and red-introduced cultivar Cheonjujeok, were compared. Yeongbohwang and Cheonjujeok had the highest total phenolic and total flavonoid contents among the six yellow and two red cultivars, respectively. The red cultivars displayed the highest DPPH and ABTS+ radical scavenging activities (DRSAs and ARSAs, respectively) and reducing power (RP). Among all cultivars, Cheonjujeok and Eumjinara had the highest DRSA, while both red cultivars had similar ARSAs. Among the yellow cultivars, Sinsunhwang and Yeongbohwang showed excellent DRSAs. The RP of the domestic cultivars was slightly higher than that of the introduced cultivars. Thus, the domestic cultivar Yeongbohwang exhibited excellent characteristics among the yellow cultivars, while the red cultivars Eumjinara and Cheonjujeok were similar.

Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion (흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정)

  • The antioxidant substance in black onion was identified. The assays that used 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazo line-6-sulphonic acid) ($ABTS^+$) radicals showed that the ethyl acetate fraction of black onion methanol extract had a higher level of radical-scavenging activity than the other fractions. Two antioxidative compounds were purified and isolated from the ethyl acetate fraction via column chromatographies of silica gel and Sephadex LH-20 using the guided DPPH radical-scavenging assay. The isolated compounds were identified as 3,4-dihydroxybenzoic acid (1) and quercetin (2) based on mass spectrometry and nuclear magnetic resonance. The isolated compounds showed a high level of DPPH and ABTS+ radical-scavenging activity. Compound 2 had a higher level of radical-scavenging activity than 1.

The Effect of The Planting Time of Mother bulb and Mulching Materials for The Seed Production of Onion (Allium cepa L.) (양파 채종을 위한 모구의 재식시기와 멀칭재료가 채종량에 미치는 영향)

  • This study examines the effects of the planting time of mother bulb and mulching materials for the seed production of onion on the amount of seed production, and the following results are obtained : Growth, bolting rate, flowering rate, floret numbers per scape and fertilization rate are best in the case of the seed production under transparent vinyl mulching between Sept. 10 and Sept. 25, and seed yield per plant and seed yield per 10a are increased in the case of the seed production under transparent vinyl mulching between Sept. 10 and Sept. 25, so it is found that it is appropriate to plant the onion mother bulb under the transparent vinyl mulching between Sept. 10 and Sept. 25 in the southern areas of Korea.

Effects of top-dressing period on Yield and Storage Quality of Onion(Allium Cepa L.) (추비시기가 양파의 수량 및 저장성에 미치는 영향)

  • This study was carried out to investigate the yield and storability of onions on top-dressing period. Top-dressing period was applied in February, March and March, April with liquid fertilizer and solid fertilizer of farmhouse practice during the onion growing season. The yields of onion in experimental station field and farmhouse field treated with conventional fertilization were 53.2 and 56.6MT, respectively. However, they were 58.2 and 60.8MT with application of liquid fertilizer in February and March, and 53.6 and 59.1MT with application of liquid fertilizer in March and April. Rotting rates until the end of August were lower with application of liquid fertilizer on February and March(15.8%, 28.9%), compared with application of solid fertilizer by farmhouse practice(23.6%, 41.0%), and were 20.7% and 31.7% by treatment of liquid fertilizer on March and April. In conclusion, treatment of liquid fertilizer on February and March decreased the rotting rate.

Effects of Ventilation Condition and Ventilating Hole Sizes to Improve Quality Onion(Allium cepu. L) under Room Temperature (양파 간이저장시 통풍조건 및 통풍구 재료의 크기가 저장에 미치는 영향)

  • This study was conducted to improve the storability of onion bulbs by assembly simple house storage and the reduce the rotteness caused by field open storage. Allium cepa cv. Changnyungdeago, late strain was used for the test at the storage condition of natural ventilation of levels 2, forced ventilation of levels 2, field open storage and 75mm, 100mm, and 125mm ventilating holes. Mean tamperature and relative humidity were not significantly different by ventilation conditions. Mean temperature was lower in forced ventilation than that of in natural ventilation and non-ventilation, and relative humidity was a little higher in ventilation treatment than those of the others. Weight loss of onion bulbs were 2.5%, 2.9%, 3%, 4.3% in field open storage, non ventilation, natural ventilation of levels 2 and forced ventilation of levels 2 respectively. Rotting rate in natural ventilation of levels 2 and farced ventilation of levels 2 were 27.7% and 25.4% respectively but 34.6% and 37.8% in non ventilation and field open storage. Therefore, the treatment of ventilation reduced the rotteness of storage onion bulbs. The smaller the size of a ventilating hole, the lower mean temperature was maintained. The relative humidity was some high in July, but didn’t showed significantly difference in August and September. With small size of a ventilating hole, the strong wind velocity was obtained, and wind velocity by position was weaker in the middle part than both ends. Rutting rates in 75㎜, 100㎜, and 125㎜ ventilating holes were 17.9%, 15.3% and 14.1% respectively.

Changes in Onion (Allium cepa L.) Bulb Quality during Storage at Room Temperature and Greenhouse (비닐하우스를 이용한 양파의 상온 저장 중 품질 변화)

  • This study was conducted to investigate the possibility of changes in onion (cv. 'Turbo' and 'Changnyongdaego') bulb quality during storage at room and cold temperature. Increased weight loss was found in the onion bulb stored at both cold and room temperature conditions. 4% weight loss was found in bulbs stored at room temperature and 2% at cold storage as compared to fresh cultivated onion bulb; after three months. Initially, there was no significant difference in the firmness of bulbs between cultivars and storage conditions. Hunter a value of bulb stored at room temperature was higher than the value of bulb stored at cold temperature. 27% and 7.9% decay was found after three months in 'Changnyongdaego' and 'Turbo' cultivars stored under room temperature condition respectively. In contrast, only 2% decay was noticed in both cultivars stored at cold storage condition. Hunter L value of onion bulb gradually decreased in both room and cold temperature storage conditions. In conclusion, quality of onion bulb can be maintained until three months. For long term storage and quality maintenance, under room temperature conditions a resistance cultivar should be selected.

Pyruvic Acid and Sugar Contents during Storage Duration in Onion (Allium cepa L.) (양파 저장 중의 Pyruvic Acid와 당 함량의 변화)

  • This study was carried out to access the stauts of pyruvic acid and sugars in onion bulbs during storage at room temperature. Pyruvic acid content remained in a steady state until 3 months of storage. However the pyruvic acid content gradually increased as the onions started to root, and rapidly increased after sprouting. There was no difference in dry matter contents during storage.e. Sucrose content increased as storage duration extended, and then gradually decreased right after the onions started to sprout. Fructose content gradually decreased after 45 days of storage, and remained in a steady state after sprouting. Glucose content had a tendency to increase as storage duration extended. Pyruvic acid/total sugar (PA/TS) ratio decreased after 1 month of storage, and then increased after 135 days, and was 83% higher at 150 days of storage than at harvest. Sweetness gradually decreased until 1 month of storage, but thereafter remained steady. Therefore it is desirable that pyruvic acid analysis for sweet onion selection should be conducted before breaking the dormancy.
