Development of Olfactory Biosensor Using Olfactory Receptor Proteins Expressed in E. coli

  • Olfactory receptor protein ODR10 was expressed in E.coli as fusion protein with GST and His6 Tag. Crude membrane extract of the expressed protein was coated on the surface of quartz crystal microbalance, and the interaction of the ODR10 with several odorants was examined. Although the expression level was very low, quartz crystal microbalance showed that the expressed protein interacted most strongly with diacetyl (butanedione), which is known to bind to the ODR10 protein selectively. The interaction between ODR10 and diacetyl was $5{\sim}10$ times stronger than the interaction between ODR10 and other odorants. Thus, E. coli cells expressing the olfactory receptor protein could be used as an olfactory biosensor. Also, such system could be used to test which olfactory receptor reacts specifically with which odorant molecules, since there has been no cheap and convenient way to test the interaction of olfactory receptors and odorant molecules yet.

Removal Efficiency of Some Odorants against Water as Sorptive Media: A Case Study on Trimethylamine and Reduced Sulfur Compounds (물을 흡수매체로 이용한 트리메틸아민과 환원황화합물의 제거효율 비교 연구)

  • In this study, the removal efficiency of trimethylamine (TMA) and four reduced sulfur compounds (RSC) of $H_2S$, $CH_3SH$, DMS, and DMDS was investigated using deionized water as absorbent. To this end, two types of experiments were conducted which include: (1) by passing both RSC and TMA standard gases through water contained in an impinger system and (2) by passing TMA gases through a water spray system. In the former method, TMA standard gases were effectively removed (below detection limit) in all experiments. Likewise, minor fractions of some RSC ($CH_3SH$, DMS, and DMDS) were removed by water in line with theoretical expectations. In addition, the first type of our experiment was extended further to test some commercially available odor treatment products. The results of this test showed that removal efficiencies of RSC $(8.2\pm13.7\sim43.7\pm2.48%)$ were different significantly among 4 kinds of absorbent, while the removal of TMA was consistently good as water. In the second phase of experiment, removal efficiency of TMA was tested by passing its standard gas through a water spray system. The overall results of our study showed that the actual removal efficiency of odorants by water (as sorptive media) approached the values that can be predicted theoretically (by Henry's law).

A Study on the Thermal Adsorption and Desorption Characteristics of Industrial Odorants using Activated Carbon (활성탄을 이용한 산업체 악취물질의 열적 흡탈착 연구)

  • Desorption reaction characteristics of the commercial activated carbons which were used for the removal of industrial odorants were investigated. BET specific surface area was analyzed to investigate the chemicophysical property of activated carbon. Adsorptivity of activated carbon was estimated by iodine number. Thermogravimetric analyzer (TGA) was used to investigate the desorption characteristics. Activation energies and reaction orders for reaction characteristics according to adsorption and desorption of activated carbons were estimated by employing the Friedman method and Freeman-Carroll method. Adsorptivity of reprocessed activated carbons were significantly lower than that of fresh activated carbons. In this study, it was found that the activation energies were 15.9~23.4 kJ/mol in Friedman method and 22.7~33.8 kJ/mol in Freeman-Carroll method.

Chemical Composition of the Essential Oils from Ligularia fischeri and Ligularia fischeri var. spiciformis (곰취와 한대리곰취 정유의 화학적 성분)

  • This study investigated the volatile flavor composition of essential oils from Ligularia fischeri and Ligularia fischeri var. spiciformis. The essential oils obtained from the plants were analyzed by gas chromatography (GC) and GC-mass spectrometry (GC-MS). 99.63% volatile flavor compounds were identified in the essential oil from the L. fischeri. The major compounds were (E)-3-hexenol (30.73%), longiverbenone (13.23%), viridiflorol (12.39%), ${\gamma}$-muurolene (7.32%), limonene (6.12%), and caryophyllene (${\beta}-4.24%$). 99.76% volatile flavor compounds were identified in the essential oil from the L. fischeri var. spiciformis. The major compounds were ledol (42.81%), (E)-15-heptadecenoic acid (33.91%), ${\beta}$-bisabolol (3.23%), viridiflorol (3.08%), and cis-${\alpha}$-farnesene (2.60%). Although the two plants are very similar, the chemical composition of the essential oils was significantly different in quality and quantity. In the case of L. fischeri., it has high contents of monoterpene and sesquiterpene. (E)-3-hexenol, longiverbenone, ${\alpha}$-phellandrene, and ${\alpha}$-myrcene were regarded as the characteristic odorants of L. fischeri, but they were not identified in L. fischeri var. spiciformis. Ledo, (E)-15-heptadecenoic acid, and ${\beta}$-bisabolol were regarded as the characteristic odorants of L. fischeri var. spiciformis, but they were not identified in L. fischeri. The ratio of limonene, ${\gamma}$-muurolene and viridiflorol can be used as an indicator to distinguish between these two plants.

  • We investigated the responses of six types of lipid-coated AT-cut quartz crystal resonators to chemical vapour sensors. The responses of quartz crystal at 9 MHz coated with phosphatidylglycerol (PG), phosphatidylinositol(PI), phosphatidylethanolamine(PE), phosphatidylserine(PS), and lipid A(LA) were obtained for amyl acetate, acetoin, menthane and other organic gases which showed different affinities for each lipid. The identification of odorants depending on the species of lipid used for coating is discussed in terms of the normalized resonant frequency shift pattern.

  • 본 연구는 향의 감성 구조를 밝히고, 향 선호도를 예측하는 감성요인을 규명하며, 향에 대한 자율 신경계 반응을 밝히는 것이 목적이다. 향에 대한 주관적인 감성을 측정하기 위하여, 25개의 감성 형용사로 구성된 향 감성 측정 척도를 개발하였다. 개발된 이 척도를 이용하여 lavender, lemon, rose, jasmin, peppermint의 다섯 가지 향에 대해 대학생 50명을 대상으로 향 감성 평가를 실시하였다 향에 대한 감성 구조는 심미성, 강도, 자연성, 여성성-남성성, 개성, 중량감의 여섯 요인으로 구성되었다. 향에 대한 자율 신경계 반응 측정은 대학생 20명을 대상으로 lavender, lemon, rose, jasmin, peppermint 향에 대해 실시하였다. 향에 대한 자율 신경계 반응 측정 결과, PPG 반응은 남녀 모두 lavender, lemon 향에서 유의미하게 감소하였다. 남자의 경우, PPG 반응은 rose, peppermint 향에서도 유의미하게 감소하였고, SKT 반응은 lavender, jasmin 항에 대해서 유의미하게 증가하였다.

Utilization of Masking Techniques to Ameliorate Agricultural Odorants

  • Different masking materials were evaluated for their ability to ameliorate odor of model poultry manure solution by assessing their effects on sensory pleasantness and odor intensity. Results indicated extracts from Eastern red cedar leaves, Loblolly pine needles', and commercial masking agents such as pine extract and odor neutralizer were effective (p<0.05) for masking odor of model poultry manure solution by increasing (p<0.05) pleasantness (82 and 86% increases in pleasantness using red cedar and pine needle extracts, respectively) and decreasing (p<0.05) odor intensity (odor intensity reduction by 66 and 76% using red cedar pine needle extract). The most odor-active compound in Loblolly pine needle extract was ${\alpha}$-terpineol (1,573.8 ug/g) which is responsible for aroma of pine trees (piney) and effective for ameliorating agricultural odors.

  • The impressions for odors are subjective and have individual differences. In this study, the Impressions of odors were investigated by covariance structure analysis. 46 subjects (men in their twenty) recorded their reactions to ten odorants by grading them on a seven-point scale in terms of twelve adjective pairs. Their reactions were quantified by using factor analysis and covariance structure analysis. The factors were extracted as "preference", "arousal" and "persistency". The subjects were classified into three groups according to the most suitable causal models (structural equation models). Each group had different causal relationship and different impression structure for odors. It was suggested that there is a possibility to evaluate the subjective impression of odor using covariance structure analysis.

Removal Effect of malodorous Substances of Piggery Wastes by Air Injection Method (공기주입에 따른 양돈분뇨중 악취제거 특성 연구)

  • This study was conducted to investigate the removal effect of malodorous substances of piggery wastes by air injection. With 7% of TS weight of artificial piggery wastes, this study focused on the changes of the concentration of VFAs, H2S during treatment period of 14 days. As a results of this study, air injection volume for removal of malodorants in this study was 150$m\ell$ / min$.$l$.$Under this condition, we found the concentrations of VFAs, H2S, NH3 have very high relationship with the pH, ORP, BOD, {{{{ { NH}`_{4 } ^{+ } }}-N. Especially, the concentration of Volatile Fatty Acids, one of most offensive odorants and one of BOD compounds were depended on ORP which can be controled by air injection in artificial piggery wastes.
