Molecular Biomarkers of Octachlorostyrene Exposure in Medaka, Oryzias latipes, using Microarray Technique (Microarray를 이용한 Octachlorostyrene-노출 송사리(Oryzias latipes)에서의 분자생물학적 지표연구)

  • Octachlorostyrene (OCS) is a primarily concerning chemical in many countries because of its persistent and bioaccumulative properties in the environment. OCS is not commercially manufactured or used but it may be produced during incineration or chemical synthetic processes involving chlorinated compounds. There are several reports that OCS was found in the waters, sediments, fish, mussels, and also in human tissues. However, systematic studies on the OCS toxicities are scarce in literature. In this study, we tried to get the gene expression data using medaka DNA chip to identify biomarkers of OCS exposure. Medaka (Oryzias latipes.) was exposed to OCS 1 ppm for 2 days and 10 days, respectively. Total RNA was extracted and purified by guanidine thiocyanate method and the Cy3- and Cy5-labelled cDNAs produced by reverse trancription of the RNA were hybridized to medaka microarray. As results, eighty five genes were found to be down-or up regulated by OCS. Some of the genes were listed and confirmed by real-time PCR.

Acute Toxicity Studies of Octachlorostyrenes for Environment Organisms (Octachlorostyrenes의 환경생물에 대한 급성독성 연구)

  • Octachlorostyrene (OCS) is a persistant and bioaccumulative toxic subtance (PBTs). In this study, acute toxicity tests on algae, daphnia and fish for octachlorostyrene and its isomers were done to determine effective concentration ($EC_{50}$), Lethal concentration ($LC_{50}$), no observed effect concentration (NOEC) or lowest observed effect concentration (LOEC). As a result, NOEC on algae growth inhibition test for octacholorostyrene and 2-, 3-chlorostyrene was determined as $0.50\;mg\;L^{-1}$, and NOEC for 4-chlorostyrene was determined as $0.13\;mg\;L^{-1}$. NOEC on daphnia, acute immobilisation test for octachlorostyrene and 2-, 3-chlorostyrene was determined as $5.00\;mg\;L^{-1}$ and $EC_{50}$ for 4-chlorostyrene was determined as $2.128\;mg\;L^{-1}$. NOEC on Oryzias Latipes, acute toxicity test for octachlorostyrene was determined as $80.0\;mg\;L^{-1}$ and NOEC for 2-, 3-chlorostyrene was determined as $60.0\;mg\;L^{-1}$. $LC_{50}$ for 4-chlorostyrene was determined as $39.0\;mg\;L^{-1}$ (48h) and $22.6\;mg\;L^{-1}$ (96h).

  • Octachlorostyrene(OCS) has been persisted in environment because it has not been decomposed easily. And, it has been known as highly toxic compounds to the environment and human as well as accumulated as high concentrations in a biota through a food chain. Therefore, OCS was monitored for water, soil and fish sampled from the areas where were able to be contaminated with OCS. The recoveries of octachlorostyrene were 93.1${\sim}$98.6% in water, 90.4${\sim}$94.8% in soil and 81.5${\sim}$90.2% in fish and detection limits were 0.0004 mg $L^{-1}$ for water, and 0.002 mg $kg^{-1}$ for soil and fish, respectively. OCS was not detected in water, sediment, soil and fish samples from Ulsan, Yeosu, Daejeon and Sihwa industrial complex and in soil sampled nearby incineration plants in all parts of the country. Accordingly, we estimated that there is no risk from exposure of OCS.

붕어 및 횡소개구리 체내의 Organochlorine pesticides의 분석

  • 유기염소계농약류 25종을 분석한 결과 HCHs(4종, endosulfan(2종), octachlorostyrene은 모든 시료에서 검출되지 않았으며 trans-nonachlor, heptachlor는 각각 한 지점의 붕어에서 검출되었고 나머지는 검출되지 않았다. Chlordane의 경우 두 이성질체가 한 지점의 붕어에서 검출되었고, DDT는 6개 이성질체 중 p,p’-DDT와 p,p’-DDD가 한 지점의 붕어에서 검출되었으며 o,p’-DDT는 한 지점의 황소개구리에서 검출되었다. 그 외의 이성질체는 모든 시료에서 검출되지 않았다.
