단계별 성장 개념의 기질 이용과 미생물 호흡모델 개발 (Development of the Substrate Utilization and Respiration Model by the Step Growth Concept)

  • 최근, 활성 슬러지 공법에 대한 수학적 모델링은 폐수처리장의 설계와 운영에 있어서 매우 중요한 인자로 인식되고 있다. 그럼에도 불구하고, BNR 공정에서 미생물의 성장 및 호흡과 관련한 내 외부 기질의 이용 경로에 관한 정보는 여전히 부족한 실정이다. 본 논문에서는 ASM No.3와 비교되는 새로운 개념의 활성슬러지 모델을 제시하고자 한다. 미생물의 단계별 성장이론에 근거한 이 모텔의 구조는 호기성 저장, 내부저장물질(ISCs; Intercellular Storage Compounds)과 외부기질 활용에 따른 미생물의 성장, 내생호흡과 내부저장물질을 이용한 호기성 호흡 등 5 단계로 구성되어 있다. 단계적 성장모델에 기초한 예측결과는 산소이용율(OUR)과 TCOD에 의한 실험결과에 있어 ASM No.3의 결과보다 더욱 일치함을 나타냈다.

패류 건제품의 저장 중 지질산화 (Lipid Oxidation in Dried Shellfish during the Storage)

  • 패류 건제품의 지질산화는 주로 단백질과의 상호반응에 의해 변색, 산패취 등을 일으키며 또한 산화로 인해 hydroperoxide나 불포화 aldehyde 등과 같은 독성물질을 생성한다고 알려져 있다. 특히 건제품의 저장 중에 발생하는 지질과 단백질의 반응에 의한 품질저하는 상품가치의 저하는 물론 영양가 손실 등을 야기한다. 본 실험에서는 이들 패류 건제품의 저장온도 ($4{\pm}2^{\circ}C$$25{\pm}2^{\circ}C$)에 따른 지질산화 경향을 지질산화생성물과 단백질 등의 amino 화합물과의 반응정도를 조사하였다. 산가의 변화는 홍합이나 바지락 모두 $25^{\circ}C$에서 저장한 것이 $4^{\circ}C$에서 저장한 것보다 높았으며 또한 저장온도에 관계없이 바지락이 홍합보다 다소 높았다. 그러나 생성된 유리지방산은 저장 60일까지 증가한 후 거의 변화가 없었다. 과산화물가는 $25^{\circ}C$ 저장구의 경우 홍합 및 바지락이 각각 저장 60일과 30일까지 증가한 후 감소하였으나, $4^{\circ}C$ 저장구에서는 120일간의 저장기간 중 과산화물이 분해되지 않고 지속적으로 축적되었다. TBA가와 카르보닐가는 홍합의 경우 $4^{\circ}C$ 저장구가 저장 30일경에만 $25^{\circ}C$ 저장구보다 낮은 값을 나타내었을 뿐 그 이후는$25^{\circ}C$ 저장구보다 높은 값을 나타내며 증가하는 경향이었다. 또한 바지락도 $4^{\circ}C$ 저장구가 $25^{\circ}C$ 저장구보다 다소 높은 값을 유지하였다. 아미노질소의 함량은 두 제품모두 저장 60일까지 증가한 후 감소하였으며, $25^{\circ}C$ 저장시료가 $4^{\circ}C$ 저장시료보다 감소폭이 컸다. 형광강도는 $25^{\circ}C$ 저장시료가 $4^{\circ}C$ 저장시료보다 그리고 바지락이 홍합보다 높았으며, 저장 60일 이후 감소하는 경향을 나타내었다.

Fitness of Dicarboximide-Resistant and Sensitive Monilinia fructicola Isolated from Peach in Korea

  • Dicarboximide-resistant isolates of Monilinia fructicola grew readily on media amended with dicarbosimide fungicides, and showed cross-resistance to pentachlo-ronitrobenzene (PCNB). The fitness of resistant isolates was inferior to that of sensitive isolates. Mycelial growth on fungicide-free medium was not significantly different between the dicarboximide-resistant and sensitive isolates. The originally high $\textrm{EC}_{50}$ values of the resistant isolate decreased after storage for 16 weeks at $4^{\circ}$. After inoculation with the mixture of spore suspensions of resistant and sensitive isolates, the re-isolation rate of the resistant spores was significantly reduced regardless of the mixing ratio. From the results, it could be concluded that the competitive ability of the resistant isolates is inferior to the sensitive ones.

  • Ultastructural studies of oocyte degeneration and follicle cells in female Spisula sachalinensis are described for clams collected from Jumunjin, Gangwondo, Korea. The follicle cells playan integral role in vitellogenesis and oocyte degeneration by assimilating products originating from the degenerated oocytes (thus allowed the transfer of yolk precursors needed for vitellogenesis). The functions of the follicle cells include phagocytosis and intracellular digestion of products originating from oocyte degeneration. During the period of oocyte degeneration, follicle cells of this species probably have lysosomal systems for the breakdown and reabsorption of various phagosomes(phagolysosomes) in the cytoplasm for nutrient storage; this process has been observed in other bivalves.

Isolation and Characterization of Edestin from Cheungsam Hempseed

  • Edestin, a major hempseed storage protein from Cheungsam, was isolated to apparent homogeneity by acid precipitation and gel filtration chromatography. The native molecular weight of purified edestin was approximately 300 kDa by Sephacryl S-300 gel filtration. Upon adding 2-mercaptoethanol, the isolated edestin of 56.7 kDa on the non-reduced sodium dodecyl sulfate polyacrylamide gel was converted into subunits, suggesting that the protein might be composed of subunits linked by disulfide bond. Cheungsam edestin was rich in essential amino acids and it has 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity. The results suggest that Cheungsam edestin might be utilized as a superior antioxidative nutrient.

  • The ultrastructure of germ cells and follicle cells during oogenesis, oocyte degeneration, reproductive cycle, and first sexual maturity in female Cyclina sinensis were investigated for clams collected from Simpo, Jeollabuk-do, Korea, by cytological and histological observations. Vitellogenesis occured by way of endogeous autosynthesis and exogenous heterosynthesis: vitellogensis occurred through a process of autosynthesis, which involves a combined activities of the Golgi complex, mitochondria, and rough endoplasmic reticulum. The process of heterosynthesis involved endocytotic incorporation of extraovarian precursors into the basal region of the early vitellogenic oocytes prior to the formation of vitelline envelope. The follicle cells appear to play an integral role in vitellogenesis and oocyte degeneration, functioning in phagocytosis and digestion of products originating from the degenerated oocytes: these functions can permit the transfer of yolk precursors needed for vitellogenesis. Follicle cells might have a lysosomal system for breakdown and might also resorb phagosomes in the cytoplasm for nutrient storage during oocyte degeneration.

Biological Characteristics of Benzimidazole-Resistant and-Senstive Isolates of Monilinia fructicola from Peach Fruits in Korea

  • Fungicide-resistant isolates of Monilinia fructicola grew readily on media amended with 0.1, 1.0, 10, 100 and $1,000\mu\textrm{g}$ a.i./ml of carendazim, benomyl, or thiophanate-methyl. However, sensitive isolates did not grow on media amended even with $0.1\mu\textrm{g}$ a.i./ml of carbendazim, $1.0\mu\textrm{g}$ a.i./ml of benomyl or thiophanate-methyl. The fitness compositions including mycelial growth on fungicide-free medium, sporulation on fungicide-free medium and pear, and virulence on pear were not different between resistant and sensitive isolates. The resistant isolates persisted carbendazim resistance during multiple subdulturing and long term storage. The competitive ability of resistant isolates obtained from peach orchards in Korea was similar to those of sensitive isolates.

Ustilago maydis의 소생자 형성에 미치는 영양원, 온도, 광의 효과 및 자기보존 후의 생존력 (Effects of nutrient Source, Temperature, pH, and Light on Sporidial Production of Ustilago maydis and It Viability for Long-term Storage)

  • 옥수수 깜부기병균 Ustilago maydis 동포자를 풍건한 후 모래나 silica gel을 보존매체로 해서 4$^{\circ}C$나 그 이하 온도에서 24개월 두었을 때 동포자 생존율은 40~45%였다. 동포자는 28$^{\circ}C$ PDA상에서 쉽게 발아하여 많은 소생자를 형성하였다. U. maydis 동포자로부터 소생자 형성의 최적배지는 corn meal broth였고, carrot broth와 potato-dextrose broth의 순으로 좋았다. 소생자 형성에 가장 적합한 조건은 온도 28~3$0^{\circ}C$, pH5~7이었다. 소생자 형성은 암조건에서 보다 좋았다.

  • Recent public concern about air pollution caused by swine production facilities has forced to develop the methods to reduce and control the swine odors. Swine odors were affected the life of pig farm neighborhoods, swine productivity, pig health, diseases, and human right, safety, sanity as negatively. The first approaches of control of swine odors are the change or improve of the classical management systems, which are manure treatment method, manure storage facility, phase feeding, sex-divided feeding, feeder type, liquid-slurry feeding, environment control of swine building and dust control of indoor swine facility. The methods to control odor emission from manure have to include the diet modification as nutritional basis. In recent, research emphasis has focused on manipulating the swine diet to increase the nutrient utilization of the diet to reduce excretion products and reduction of odors. There are lots of feed additives and pit additives introduced as practical basis for reducing odor emissions. The ozone treatment method is candidate as the good system for reducing swine odor. But this system is still too expensive to practice in present.

조사료의 곰팡이 발생과 곰팡이독소 오염 (Mold Growth and Mycotoxin Contamination of Forages)

  • In order to ensure good animal health and performance, it is essential to produce forages with high feeding value and good hygienic quality. However, huge amounts of forages consumed by ruminants are contaminated with mold prior to harvest or during storage as hay, straw or silage. These mold can grow in forages only when nutrients are available, correct temperature exist, oxygen is present, and unbound water is available. Fungal 'species can be divided into two groups: field fungi and storage fungi. Field fungi invade the forages while the crop is still in the field, require high moisture conditions, and are such as species of Fusarium, Alternaria, Clodosporium, Diplodia, Gibberrella and Helminthosporium. Storage fungi invade forages during storage and need less moisture than field fungi. These such as species of Aspergillus and Penicillium usually do not occur any problem before harvest. Mold growth can spoil the nutritional aspects of the forages and also results in secondary metabolites that are highly toxic to animal, humans and plants. Moldy feeds are less palatable and may reduce dry matter intake. This, in turn, leads to a reduction of nutrition intake, reducing weight gains or milk production. Performance losses of 5 to 10 percent are typical with moldy feeds. Mycotoxins are toxic substances produced by fungi (molds) growing on crops in the field or storages. While greater than 400 mycotoxins have been chemically identified, the biological or veterinary medical impact of only several mycotoxins is known. Mycotoxins have attracted considerable attention as potential causes for poor performance and health disorders in domestic livestock. They can be carcinogenic, hepatotoxic, hematotoxic, immunosuppressive, estrogenic, or mutagenic. So, feeding moldy forages has adverse effects on animal health and milk consumers. Also, this author reported that rice straw hay was contaminated mycotoxigenic fungi such as Penicillium roqueforti and Fusarium culmorum in Korea. Therefore, it is an urgent need to develop an improved post harvest storage method to reduce nutrient loss and mycotoxin contamination of forages, which will have a positive impact on human health.
