Availability of nutrients is known to influence marine primary production; and it is of general interest to see how nutrient limitation mediates phytoplankton responses to solar ultraviolet radiation (UVR, 280-400 nm). The red tide diatom Skeletonema costatum was cultured under nitrate (N)-limited and N-replete conditions and exposed to different solar irradiation treatments with or without UV-A (315-400 nm) and UV-B (280-315 nm) radiation. Its photochemical quantum yield decreased by 13.6% in N-limited cells as compared to that in N-replete ones under photosynthetically active radiation (PAR)-alone treatment, and the presence of UV-A or UV-B decreased the yield further by 2.8 and 3.1%, respectively. The non-photochemical quenching (NPQ), when the cells were exposed to stressful light condition, was higher in N-limited than in N-replete grown cells by 180% under PAR alone, by 204% under PAR + UV-A and by 76% under PAR + UV-A + UV-B treatments. Our results indicate that the N limitation exacerbates the UVR effects on the S. costatum photosynthetic performance and stimulate its NPQ.
Despite the therapeutic effect of mesenchymal stem cells (MSCs) in ischemic diseases, pathophysiological conditions, including hypoxia, limited nutrient availability, and oxidative stress restrict their potential. To address this issue, we investigated the effect of melatonin on the bioactivities of MSCs. Treatment of MSCs with melatonin increased the expression of peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α). Melatonin treatment enhanced mitochondrial oxidative phosphorylation in MSCs in a PGC-1α-dependent manner. Melatonin-mediated PGC-1α expression enhanced the proliferative potential of MSCs through regulation of cell cycle-associated protein activity. In addition, melatonin promoted the angiogenic ability of MSCs, including migration and invasion abilities and secretion of angiogenic cytokines by increasing PGC-1α expression. In a murine hindlimb ischemia model, the survival of transplanted melatonin-treated MSCs was significantly increased in the ischemic tissues, resulting in improvement of functional recovery, such as blood perfusion, limb salvage, neovascularization, and protection against necrosis and fibrosis. These findings indicate that the therapeutic effect of melatonin-treated MSCs in ischemic diseases is mediated via regulation of PGC-1α level. This study suggests that melatonin-induced PGC-1α might serve as a novel target for MSC-based therapy of ischemic diseases, and melatonin-treated MSCs could be used as an effective cell-based therapeutic option for patients with ischemic diseases.
It has been suggested that dietary fat increases the risk of breast cancer by elevating serum estrogen concentrations. However, studies on a relationship between fat intake and breast cancer risk have shown contradictory results, possibly because the levels of fat intake in study populations were too high to observe differences. Also, the effect of other dietary factors may present. The present study was performed to investigate the relationship between diet and estrogen metabolism in premenopausal women whose usual fat intake is relatively low compared to their western counterparts. Twenty lacto-ovo vegetarians (LOV) and twenty omnivores participated in the study. Three day food records including a Saturday or a Sunday were used to estimate nutrient intake. Serum lipids, estradiol, sex-hormone binding globulin, and urinary estradiol were measured. Study results showed $24.8\%$ and $20.9\%$ of energy intake were provided from fat in omnivorous and LOV subjects, respectively. Serum and urinary levels of estradiol were two times higher in omnivores. Fat intake was not related to either serum estradiol nor urinary estradiol when the Spearman correlation coefficient analysis was performed. Carbohydrate, total dietary fiber and soluble dietary fiber intakes were negatively related to serum estradiol concentration. Legumes, vegetables and fruit consumption showed significantly negative relationships with both serum and urinary estradiol concentrations. These results indicate lower estrogen availability may be associated with plant foodbased diets in premenopausal women whose usual diets contain less than $25\%$ of energy as fat.

This study was carried out to investigate the effects of Yerba Mate (YM) supplementation on nutrients' degradation, in vitro dry matter disappearance, gas production and rumen ammonia concentration. Three rumen-fistulated Holstein Friesian cows were used for the in situ incubations and provided rumen liquor for in vitro incubations. The inclusion of YM in a control diet (pasture+pellets) affected some in sacco degradation parameters. YM supplementation decreased the effective degradability and degradation rate of pasture crude protein (CP), and it seems to slow down the degradation of pasture neutral detergent fiber. A significant increase of degradation of pasture acid detergent fiber (ADF) was detected after YM inclusion in the control diet. YM supplementation reduced in vitro gas production of pasture and ammonia concentration of pellets. The addition of YM in ruminant diet could decrease ammonia production and increase protein availability for productive purposes. The moderate presence of tannins in YM could have affected the degradation kinetics of pasture CP and ADF and the ammonia production of pellets.
Organic matter in rice-paddy soils exercises two antagonistic effects on the rice plant under water-logged conditions in growing season in the course of its decomposition: It liberates mineral nutrients and promotes soil fertility. On the other hand, however, it demands oxygen for its decay and therefore competes with rice roots for this element, when applied in large quantity of fresh status. For the practical end of rice culture, it is most desirable that these two effects should not contend with each other. To determine the proper content of organic materials to be applied, the influences of varied amounts of a homogeneous mixture of dried green manure, ranging from 0 to 20g/pot (1/20,000 tanbo), upon hte growth of rice was investigated in a sand culture. Labeled phosphorus fertilizer was also used in the form of KH232PO4 to evaluate the availability of this nutrient in the soil. Under the present experimental conditions, green mature seems to have influenced little on the growth of rice, except on number of grains produced and grains/straw ratio. Moreover, no sympton of growth inhibition is obsrvable even by the largest amount of its application. The available phosphorus, as estimated by A-value, appears to have increased, as the amount of organic materials applied increases. In view of the fact that pure sand instead of a paddy soil is used in this culture, the present results would not be directly applicable to practical rice farming. Besides, the estimated A-value is in need of further study, since it varies according to method of application, as suggested by Nishigki et. al. (1958).
Nutrient availability is a critical component of agroecosystems, and is relevant to both above- and below- ground interactions. The principal objective of this study was to determine how the cessation of fertilization affects the communities of weeds and soil organisms in a corn/wheat field. Changes in dominant weed species, substrate-induced respiration, and the population density of nematodes and microarthropods were evaluated. Microbial substrate-induced respiration (SIR) and the population density of microarthropods decreased following the cessation of fertilization and were partly correlated with the aboveground weed biomass. The cessation of organic fertilizer application but continuing application of inorganic fertilizer reduced the population density of nematodes. In response to the cessation of fertilization, weed communities were dominated by species with little dependency on fertilization. Amaranthus retroflexus was identified as the most dominant species in the corn field; however, it was replaced by Digitaria ciliaris after the cessation of fertilization. In the wheat field, the cessation of fertilization led to a rapid reduction in the biomass of most weeds, except for Vicia angustifolia, supposedly as the result of symbiotic nitrogen fixation. Additionally, the fact that weed biomass was partially correlated with SIR or the population density of microarthropods may reflect a mutual feedback between soil organisms and weeds. The results indicate that the cessation of fertilization alters communities of weeds and soil organisms through changes in weed biomass and interactions with symbiotic microorganisms.
Iron availability is limited in the environment and most bacteria have developed a system to acquire iron from host hemoproteins. Heme oxygenase plays an important role by degrading heme group and releasing the essential nutrient iron. The structure of Bacillus subtilis HmoB was determined to 2.0 ${\AA}$ resolution. B. subtilis HmoB contains a typical antibiotic biosynthesis monooxygenase (ABM) domain that spans from 71 to 146 residues and belongs to the IsdG family heme oxygenases. Comparison of HmoB and IsdG family proteins showed that the C-terminal region of HmoB has similar sequence and structure to IsdG family proteins and contains conserved critical residues for heme degradation. However, HmoB is distinct from other IsdG family proteins in that HmoB is about 60 amino acids longer in the N-terminus and does not form a dimer whereas previously studied IsdG family heme oxygenases form functional homodimers. Interestingly, the structure of monomeric HmoB resembles the dimeric structure of IsdG family proteins. Hence, B. subtilis HmoB is a heme oxygenase with a novel structural feature.
Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.
Healthy aging has become a major goal of public health. Many studies have provided evidence and theories to explain molecular mechanisms of the aging process. Recent studies suggest that epigenetic mechanisms are responsible for life span and the progression of aging. Epigenetics is a fascinating field of molecular biology, which studies heritable modifications of DNA and histones that regulate gene expression without altering the DNA sequence. DNA methylation is a major epigenetic mark that shows progressive changes during aging. Recent studies have investigated aging-related DNA methylation as a biomarker that predicts cellular age. Interestingly, growing evidence proposes that nutrients play a crucial role in the regulation of epigenetic modifiers. Because various nutrients and their metabolites function as substrates or cofactors for epigenetic modifiers, nutrition can modulate or reverse epigenetic marks in the genome as well as expression patterns. Here, we will review the results on aging-associated epigenetic modifications and the possible mechanisms by which nutrition, including nutrient availability and bioactive compounds, regulate epigenetic changes and affect aging physiology.
Gastrointestinal tract of ruminants as well as monogastric animals are colonised by a variety of microorganisms including bacteria, fungi and protozoa. Gastrointestinal ecosystem, especially the rumen is emerging as an important source for enrichment and natural selection of microbes adapted to specific conditions. It represents a virtually untapped source of novel products (e.g. enzymes, antibiotics, bacteriocins, detoxificants and aromatic compounds) for industrial and therapeutic applications. Several gastrointestinal bacteria and fungi implicated in detoxification of anti-nutritional factors (ANFs) can be modified and manipulated into promising system for detoxifying feed stuffs and enhancing fibre fermentation both naturally by adaptation or through genetic engineering techniques. Intestinal lactobacilli, bifidobacteria and butyrivibrios are being thoroughly investigated and widely recommended as probiotics. Restriction endonucleases and native plasmids, as stable vectors and efficient DNA delivery systems of ruminal and intestinal bacteria, are increasingly recognised as promising tools for genetic manipulation and development of industrially useful recombinant microbes. Enzymes can improve the nutrient availability from feed stuffs, lower feed costs and reduce release of wastes into the environment. Characterization of genes encoding a variety of commercially important enzymes such as cellulases, xylanases, $\beta$-glucanases, pectinases, amylases and phytases will foster the development of more efficacious and viable enzyme supplements and enzyme expression systems for enhancing livestock production.
