Oct4-Transfection한 중간엽줄기세포 유래 핵이식 배반포의 Oct4 발현 분포 및 세포 자멸사의 변화에 관한 연구 (Study on Distribution of Oct4 Expression and Change of Apoptosis in Nuclear Transfer Blastocyst using Oct4-Transfected Mesenchymal Stem Cells)

  • There are various factors i.e. donor cell type, culture system as well as technical procedures which influence the pre-implantation embryonic development; however, may attempts have been made and still it is under investigation to improve the cloning efficiency using somatic cell nuclear transfer technique. It is has been investigated that stem cells like mesenchymal stem cell are able to more efficiently reprogram and reactivate the expression of early embryonic genes to promote nuclear transfer efficiency. In addition, Oct4 expression plays a pivotal role in early embryo development. In the present study, we investigated distribution of Oct4 expression and changes of apoptosis and total cell number in nuclear transfer blastocyst after using Oct4 transfected bone marrow stem cell as donor cells. Although Oct4-RFP expression was observed across blastocyst, more concentrated intensity was shown at hatched region in blastocyst on day 7. Reduction of apoptotic bodies was revealed in Oct4 transfected blastocyst by TUNEL staining, however, there was no significant difference in total cell number between Oct4 transfected and non-transfected nuclear transfer embryos. In conclusion, Oct4 transfected donor cells exhibited higher expression in hatching sight in day 7 blastocyst and were able to prevent apoptosis compared to non-transfected donor cells.

더덕의 체세포배로부터 식물체 재생과 사포닌 함량 변화 (Plant Regeneration and Saponin Contents in Codonopsis lanceolata L.)

  • 더덕 (Codonopsis lanceolata)의 자엽기의 체세포배 는 생장조절제 를 함유하지 않은 MS고체배지에 배양하였을 때 완전한 식물체로 분화되었다. 체세포배로부터 줄기 재분화에는 2% sucrose가, 뿌리의 분화에는 8% sucrose가 가장 좋았다. 줄기의 생장은 2% sucrose가, 뿌리의 생장은 4% sucrose 농도에서 가장 좋았다. 자연산 더덕뿌리, 재분화 개체의 뿌리, 배발생 캘러스, 비배발생 캘러스에서의 saponin계 물질의 함량을 HPLC로 조사한 결과 각 조직간의 물질 함량은 커다란 차이를 보였으며, 고농도의 sucrose 함유 배지에서 재분화된 뿌리의 saponin계 물질의 함량은 자연산뿌리와 비슷하였다. 세포분화가 진행됨에 따라 saponin계 물질의 함량은 급격히 감소하였다. 이러한 결과는 식물의 유용물질 생산에 효율적으로 활용될 수 있을 것이다.

  • 황우석 사태 이후 한국의 국가는 줄기세포 연구를 장려하고 시험관 아기 산업을 장려하겠다는 입장과 "글로벌 스탠다드"에 부합하는 윤리적 규제를 도입하겠다는, 많은 경우 서로 모순될 수밖에 없는 입장을 표명하여 왔다. 줄기세포 연구에 대한 윤리적 규제가 점점 강화되면서 인간배아세포 연구가 위축되면서, 연구 공동체와 바이오산업, 임상의사와 환자, 그리고 국가 자체를 위기로부터 구원해줄 대안으로 떠오른 것은 체세포 줄기세포였다. 그러나 한국 생명공학기술에 대한 연구들은 주로 배아줄기세포에 초점을 맞추고 있으며, 조혈줄기세포나 지방유래줄기세포와 같은 체세포 줄기세포에 대한 연구에는 상대적으로 관심이 적은 것으로 보인다. 배아줄기세포가 흔히 실험적이고 윤리적으로 논란거리로 여겨지는 반면에, 조혈모 혹은 간엽줄기세포와 체세포 줄기세포는 별다른 공적인 논의 없이 대중들의 일상 속으로 들어와 있다. 한국의 많은 일반인들은 조혈모 줄기세포 치료를 통해 백혈병으로부터 생명을 구한 환자들의 사례에 이미 익숙한가 하면, 다른 한편에서 지방유래줄기세포 치료를 선전하는 의사들의 수가 늘고 있고, 지방유래줄기세포의 개념을 활용하여 만든 화장품이 소비자들의 주목을 받고 있기도 한 현실이 이미 진행되고 있다. 이러한 맥락에서, 본 논문은 배아줄기세포나 국가 정책이나 연구 규제에만 집중되어 시장을 놓치고 있는 윤리적 논의는 한국에서 줄기세포 기술의 정치의 전모를 다루기에 한계가 크다는 사실을 주장하고자 한다.

Optimization of Post-Activation Systems to Improve the Embryonic Development in Porcine Parthenogenesis and Somatic Cell Nuclear Transfer

  • This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB ($7.5{\mu}g/mL$), CB ($7.5{\mu}g/mL$) + CHX ($10{\mu}g/mL$), CB ($7.5{\mu}g/mL$) +DC ($0.4{\mu}g/mL$), and CB ($7.5{\mu}g/mL$) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

돼지 태아 섬유아 세포의 효과적인 배양 (Efficient Culture of Porcine Fetal Fibroblasts)

  • 체세포의 배양 방법은 체세포 핵이식에 의한 형질 전환 돼지 생산에 있어서 중요한 요인 중 하나이다. 본 연구에서는 돼지 태아 섬유 아세포의 효율적인 배양 방법을 수려하였다. 돼지 태아 섬유 아세포는 임신 33일째 태아로부터 제조하였으며, 돼지 태아 섬유아세포의 증식을 혈청과 배지 종류별로 분석하였다. 그 결과, 15% ES screened FBS가 포함된 DMEM 배지에서의 배양은 15% FBS보다 세포수의 증가가 훨씬 더 빠르게 나타났다. 또한, 태아 섬유아 세포는 DMEM/F-12와 다르게 ES midified DMEM과 DMEM 배지에서 $7{\sim}8$번째 계대까지 증식이 유지되었다. 이러한 배양 조건에서 PGK-neo 벡터(pKJ2)를 돼지 태아 섬유아 세포에 도입한 다음 12일간 $300\;{\mu}g/ml$ G418이 포함된 배지에서 선별하여 colony를 얻을 수 있었다. 이러한 결과는 본 연구에 이용된 배양 시스템이 transgenic vector를 도입시킨 돼지 체세포의 screening에 이용될 수 있음을 보여주고 있다.

모돈 한약재 급여에 따른 자돈 사산 및 폐사 감소효과 연구 (A study of reduction effects of stillbirth and mortality rates in pigs by medicinal herbs supplementation in sow)

  • This study was based on a survey of 100 sows which were raising in Jeonnam province for 11 months, from February to December 2008 in order to evaluate the medicinal herbs supplementation in sows on the effects of mortality in suckling pigs, and milk quality of sows. The medicinal herbs were supplemented in the feed of sows from 1 weeks before the expected date of delivery to 10 days after the delivery. As comparing stillbirth rate, test group was reduced 9.05%, and control group, 17.37%. It was certain that the medicinal herbs was very effective for reducing stillbirth rate. In comparison of mortality rate of suckling pig, test group was decreased by 1.52% as compared with control group. In analysis of milk components of sows, milk fat was 6.04% in test group and 5.22% in control group. Milk protein contents of test group and control group were 18.06% and 18.36%, respectively, showing no significant differences between two groups. Milk sugar contents showed also no difference between two groups. By milk somatic cell counts, test group ($582{\times}10^3$cells/ml) was found decreased number of milk somatic cell as compared with control group ($688{\times}10^3$cells/ml). These results imply that the medicinal herbs supplementation in sow might be effective for preventing stillbirth and mortality in piglets and mastitis in sow.

소 체내포 핵이식에 의한 핵-세포질 상호작용에 관한 연구 (Study on Nucleo-Cytoplasmic Interaction by Somatic Cell Nuclear Transfer in Bovine)

  • This study was conducted to investigate the effects of quiescent treatment of donor cells and activation treatment time of recipient cytoplasm on nuclear remodeling and in vitro development of somatic cell-cloned bovine embryos. Serum starved, confluent and nonquiescent cycling adult skin cells were teansferred into enucleated oocytes. Nuclear transfer oocytes were activated at 30 min, 1 and 2 hrs after electrofusion. Some nuclear transfer embryos(23% to 35%) extruded a polar body, which was not affected by quiescent treatment of donor cells and activiation time of recipient cytoplasm. About 68% of nuclear transfer embryos fused with a serum starved cells has a chromatin clump, but which was not different from embryos fused with confluent(51%) and nonquiescent(47%) cells. The proportion of embryos with a single chromatin clump was sightly increased when nuclear transfer embryos were activated within 30 min after fusion(69%) compared to those were activated at 1 and 2 hrs after fusion, but there was not significantly different. Development rates to the blastocyst stage were 8.6% and 15.9% when serum starved and confluent cells were transferred, which were higher than that of control group. Developmental rate to the blastocyst stage was higher in embryos were activated within 30 min after fusion (17.3%) compared to those of embryos were activated at 1 and 2 hrs after fusion (P<0.05). From the present result, it is suggested that quiescent treatment of donor cells and activation time of recipient cytoplasm can affect the in vitro development. Quiescent plasm activation within 30 min after fusion could increase the number of embryos with a normal chromation structure, which results in increased in vitro development.

돼지 공여세포의 조건이 핵이식 수정란의 체외발달에 미치는 영향 (Effects of Donor Somatic Cell Conditions on In Vitro Development of Nuclear Transplanted Porcine Embryos)

  • 본 연구는 복제 돼지의 생산성 향상과 형질전환에 의한 대체상기용 복제 돼지 생산에 기여하기위한 기초연구로 공여세포의 조건, 핵이식 수정란의 융합 및 활성화와 체외발달에 미치는 각종 요인들은 조사하였다. 공여세포는 생후 10개월 된 Landrace 종으로부터 귀 세포조직(5$\times$5mm)을 채취하여 0.05%의 trypsin과 EDTA가 첨가된 D-PBS로 세포를 분리하여 10% FBS가 첨가된 TCM-199 배양액으로 계대배양을 실시하여 사용하였다. 핵이식은 laser system 으로 투명대를 drilling하여 수핵난자의 극체와 핵을 제거한 후 공여세포를 주입하였으며. 핵이식란은 DC 1.9kv/cm, 30$\mu$sec 1회의 전기자극으로 융합과 1시간 후 AC 1.50kv/cm, 30$\mu$sec 1회의 조건으로 활성화를 실시하여 분할을 유도하였다. 분할된 핵이식 수정란은 10% FBS가 첨가된 NCSU-23 배양액으로 $CO_2$배양기에서 6~8일 동안 체외배양을 실시하여 배반포기로 발달한 수정란을 Hoechst 33342로 핵염색을 하여 할구수를 조사하였다. 공여세포의 기아배양을 3~4 및 5~6 일간 실시하여 핵이식 후 전기자극으로 융합을실시하였을 때 융합율은 각각 45.6 및 36.8%로써 기아배양 기간에 따른 차이는 없었다. 융합 및 활성화가 유기된 핵이식란의 분할율은 3~4일간 기아배양을 실시한 공여세포가 67.1%로써 가장 높았으며(P<0.05), 5~6일간 기아배양을 실시한 공여세포의 57.1%와는 차이가 없었다. 공여세포를 1~2, 5~6 및 13~14대 계대배양한 것을 사용한 핵이식란의 융합율은 각각 52.7, 53.0 및 51.7%로써 차이가 없었다. 융합이 이루진 핵이식란을 활성화를 유도했을 때 1~2, 5~6 및 13~14대 계대배양한 공여세포의 분할율도 각각 42.7, 46.8 및 45.5%로써 차이가 없었다. 25$\mu$m$\geq$ 크기의 공여세포를 사용하였을 때 핵이식란의 융합율은 65.3%로써 25~30$\mu$m 및 30$\mu$m $\leq$ 크기 공여세포의 융합율 42.5 및 45.5% 보다는 유의적(P<0.05)으로 높았다. 융합과 활성화가 유기된 핵이식란의 분할율은 25$\mu$m $\geq$, 25~3o$\mu$m 및 30$\mu$m $\leq$ 크기에서 각각 56.5, 68.8 및 58.5%로써 공여세포의 크기에 따른 분할율은 유의적인 차이가 없었다. 체외수정란과 체세포 핵이식 수정란의 발달에 있어서는 분할율이 각각 80.1%와 64.0%로써 핵이식 수정란이 체외수정란 보다 낮았으나, 배반포기로의 발달율에 있어서는 각각 12.4%와 10.5%로써 차이가 없었다.

Potential functional roles of follistatin on bovine somatic cell nuclear transfer embryos

  • To demonstrate that follistatin treatment enhances the efficiency of nuclear transfer (SCNT), cell allocation and preimplantational development were determined in bovine SCNT embryos in the present study. Treatment of activated SCNT embryos with 10 ng/ml follistatin significantly increased the proportion of blastocyst development compared to untreated SCNT embryos. In addition, an increase in trophectoderm (TE) cell numbers and relatively higher proportion of TE cells to total cells were observed, but the number of inner cell mass (ICM) cell and total cell numbers were not changed (P < 0.05). No significant effect of other doses of follistatin was observed for the above endpoints. However, treatment with 1 and 10 ng/ml follistatin reduced the proportion of nuclear transfer blastocysts with an ICM ratio of > 60% relative to untreated nuclear transfer blastocysts at Day 7. No significant effect of follistatin treatment on proportions of nuclear transfer blastocysts with ICM ratio of 20-40% or 40-60% was observed. Taken together, these results suggested that follistatin can be used to increase developmental competence of SCNT embryos in terms of cell allocation, particularly TE cells, during preimplantation stages, subsequently enhancing placentation and birth of live offspring.

Morphological Characteristics of Pig Blastocysts Produced by Somatic Cell Nuclear Transfer

  • Blastocyst formation, consisting of the inner cell mass (ICM) and trophectoderm (TE), is the first differentiation process during embryonic development in mammals. It has been hypothesized that the proportion of ICM to TE in the blastocyst may be crucial for subsequent developmental competence of early embryos, which it may be expressed as a sensitive indicator for evaluating in vitro systems. In this study ICM/total cell ratio of nuclear transfer (NT) embryos was compared with IVF-derived and in vivo embryos. Somatic cell nuclei obtained from a fetus at Day 40 of gestation were transferred into the enucleated oocyte and then cultured in NCSU 23 medium for 6 days as previously described (Koo et al., Biol. Reprod. 2000; 63:986-992). ICM and TE cells of blastocysts were determined by using a differential staining method (Han et al., Biol. Reprod. 1999; 60:1110-1113). Development rate (9.8$\pm$2.5%, 23/225) to the blastocyst stage of NT embryos was lower than IVF embryos (23.8$\pm$2.7%, 53/223). Thus, a difference was detected in the in vitro developmental rate to blastocyst stage between NT and IVF-derived embryos (P<0.05). In the next experiment, we investigated ICM and TE nuclei to assess the quality of blastocysts that produced by NT, IVF and in vivo, respectively. NT blastocysts (27.6$\pm$8.3) showed a smaller total cell number than IVF-derived (42.6$\pm$17.4) and in vivo embryos (283.9$\pm$103.5) (P<0.05). Ratios of ICM/total cells in NT, IVF and in vivo blastocysts were 15.1$\pm$ 18.6% (n=56), 12.3$\pm$9.2% (n=57) and 30.4$\pm$6.8% (n=40), respectively. Individual blastocysts for the ratio of ICM/total cells were assigned to 3 groups (I; <20%, II; 20 to 40% and III;>40%). As the results, most in vivo blastocysts (97.5%, 39/40) were distributed into group II while most NT (78.6%, 44/56) and IVF-derived blastocysts (82.5%, 47/57) were allocated to group I. Thus, our data show that NT or IVF-derived embryos have aberrant morphology during early development in vitro systems, suggesting that these anomalies may result in developmental failures of the NT embryos to term.
