불임여성에서 NAT2, GSTM1, CYP1A1 유전자 다형성과 자궁내막증의 상관관계에 관한 연구 (Association between Endometriosis and Polymorphisms of N-acetyl Transferase 2 (NAT2), Glutathione S-transferase M1 (GSTM1) and Cytochrome P450 (CYP) 1A1 Genes in Korean Infertile Patients)

  • Objective: To investigate the association between endometriosis and polymorphisms of N-acetyl transferase 2 (NAT2), glutathione S-transferase M1 (GSTM1), and cytochrome P450 (CYP) 1A1 genes in Korean infertile patients. Materials and Methods: A total of 303 infertile patients who had undertaken diagnostic laparoscopy during January, 2001 through December, 2003 at Samsung Cheil Hospital enrolled in this study. The patients were grouped according to laparoscopic findings: minimal to mild endometriosis (group I: n=147), moderate to severe endometriosis (group II: n=57), normal pelvic cavity (n=99). Peripheral blood was obtained and genomic DNA was extracted. The genotypes of each genes were analyzed using polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP). For NAT2, RFLP was used to detect the wild type (wt) and mutant (mt) alleles, enabling classification into slow (mt/mt) or fast (wt/wt or wt/mt) acetylation genotypes. For GSTM1, PCR was used to distinguish active (+/- or +/+) from null (-/-) genotypes. For CYP1A1, MspI digestion was used to detect the wild type (A1A1), heterozygote (A1A2) or mutant (A2A2) genotypes. Result: The genotype frequencies of NAT2 slow acetylator was 12.8%, 10.9%, 12.8% in group I, group II and control, respectively. The genotype frequencies of GSTM1 null mutation was 55.3%, 41.8%, 53.2% in group I, group II and control, respectively. The genotype frequencies of CYP1A1 MspI polymorphism was 16.3%, 9.1%, 18.1% in group I, group II and control, respectively. No significant difference was observed between endometriosis and normal controls in the genotype frequencies of the NAT2, GSTM1, CYP1A1 MspI polymorphism. Conclusion: The NAT2, GSTM1, CYP1A1 gene polymorphism may not be associated with the susceptibility of endometriosis in Korean women.

Associations of CYP1A1, GSTM1 and GSTT1 Polymorphisms with Lung Cancer Susceptibility in a Northern Indian Population

  • Background: Susceptibility to lung cancer has been shown to be modulated by inheritance of polymorphic genes encoding cytochrome P450 1A1 (CYP1A1) and glutathione S transferases (GSTM1 and GSTT1), which are involved in the bioactivation and detoxification of environmental toxins. This might be a factor in the variation in lung cancer incidence with ethnicity. Materials and Methods: We conducted a case-control study of 218 northern Indian lung cancer patients along with 238 healthy controls, to assess any association between CYP1A1, GSTM1 and GSTT1 polymorphisms, either separately or in combination, with the likelihood of development of Lung cancer in our population. Results: We observed a significant difference in the GSTT1 null deletion frequency in this population when compared with other populations (OR=1.87, 95%CI: 1.25-2.80-0.73, P=0.002). However, GSTM1 null genotype was found associated with lung cancer in the non-smoking subgroup. (P=0.170). Conclusions: Our study showed the GSTT1 null polymorphism to be associated with smoking-induced lung cancer and the GSTM1 null polymorphism to have a link with non-smoking related lung cancer.

Genetic Polymorphism of Glutathione S-transferases M1 and T1, Tobacco Habits and Risk of Stomach Cancer in Mizoram, India

  • Aim: The incidence of stomach cancer in Mizoram is highest in India. We have conducted a population based matched case-control study to identify environmental and genetic risk factors in this geographical area. Methods: A total of 102 histologically confirmed stomach cancer cases and 204 matched healthy population controls were recruited. GSTM1 and GSTT1 genotypes were determined by PCR and H. pylori infections were determined by ELISA. Results: Tobacco-smoking was found to be an important risk factor for high incidence of stomach cancer in Mizoram. Meiziol (local cigarette) smoking was a more important risk factor than other tobacco related habits. Cigarette, tuibur (tobacco smoke infused water) and betel nut consumption synergistically increased the risk of stomach cancer. Polymorphisms of GSTM1 and GSTT1 genes were not found to be directly associated with stomach cancer in Mizoram. However, they appeared to be effect modifiers. Persons habituated with tobacco smoking and/or tuibur habit had increased risk of stomach cancer if they carried the GSTM1 null genotype and GSTT1 non-null genotype. Conclusion: Tobacco smoking, especially meiziol is the important risk factor for stomach cancer in Mizoram. GSTM1 and GSTT1 genes modify the effect of tobacco habits. This study is a first step in understanding the epidemiology of stomach cancer in Mizoram, India.

Glutathione S-transferase T1, M1 and P1 Genetic Polymorphisms and Susceptibility to Colorectal Cancer in Turkey

  • Colorectal cancer (CRC) is reproted to be the third most common cancer worldwide and the fourth most common cause of cancer related deaths. CRC is considered to be a multifactorial disease whose risk varies due to the complex interaction between individual genetic basis and disposure to multiple endogenous factors. Glutathione S-transferases are pro-carcinogenic in CRC and are required for the conjugation between chemotherapeutics and broad spectrum xenobiotics. One hundred and eleven patients with CRC and 128 control subjects without any cancer history were enrolled in this study. Multiplex PCR was applied to determine polymorphisms for the GSTT1 and M1 genes, and PCR-RFLP was applied for the GSTP1 (Ile105Val) gene polymorphism. Values p<0.05 were defined as statistically significant. We detected a significant high correlation between predisposition for CRC and presence of the Ile/Ile genotype of the GSTP1 (IIe105Val) gene polymorphism, but we did not find a significant relationship between predisposition for CRC and GSTT1 and M1 deletion polymorphisms. In addition, we did not determine a relationship between GSTT1, M1 and P1 gene polymorphisms and any clinicopathological features of CRC. GSTT1 null/GSTM1 positive and GSTT1 null/GSTM1 positive/GSTP1 Ile/Ile genotypes were significantly higher in the patient group. Our results revealed that there is no relationship among CRC, its clinicopathologic features, and GSTT1 M1 gene polymorphisms. However, there was a significant correlation between CRC and the GSTP1 Ile/Ile genotype. Further studies with larger patient groups are required to delineate the relationships between GST gene polymorphisms and the clinicopathologic features of CRC in Turkey.

Glutathione-S-Transferase Polymorphisms (GSTM1, GSTT1 and GSTP1) and Acute Leukemia Risk in Asians: a Meta-analysis

  • The association between glutathione-S-transferase polymorphisms (GSTM1, GSTT1 and GSTP1) and risk of acute leukemia in Asians remains controversial. This study was therefore designed to evaluate the precise association in 23 studies identified by a search of PubMed and several other databases, up to December 2013. Using random or fixed effects models odds ratios (ORs) with corresponding 95% confidence intervals (CIs) were calculated. Heterogeneity across studies was assessed, and funnel plots were constructed to test for publication bias. The meta-analysis showed positive associations between GST polymorphisms (GSTM1 and GSTT1 but not GSTP1) and acute leukemia risk [(OR=1.47, 95% CI 1.18-1.83); (OR=1.32, 95% CI 1.07-1.62); (OR=1.01, 95% CI 0.84-1.23), respectively] and heterogeneity between the studies. The results suggested that the GSTM1 null genotype and GSTT1null genotype, but not the GSTP1 polymorphism, might be a potential risk factors for acute leukemia. Further well-designed studies are needed to confirm our findings.

갈색종피와 녹색자엽 및 Tetra Null 유전자형을 가진 콩 계통 선발 (Selection of a Soybean Line with Brown Seed Coat, Green Cotyledon, and Tetra-Null Genotype)

  • 갈색종피와 녹색자엽을 가진 품종 및 유전자원의 성숙 종실에는 눈 건강에 유익한 루테인 성분과 항산화 효과를 가진 플라보노이드 성분이 많이 함유되어져 있다. 그러나, 렉틴, 7S α′ subunit, 리폭시게나제 및 쿠니츠 트립신 억제제(KTI) 단백질 같은 항영양 성분이 존재한다. 이러한 항영양 성분을 불활성화시키기 위하여 콩 식품 제조 및 가공시 고온 및 첨가제 처리가 필요하지만 여러 가지 단점이 수반된다. 따라서, 갈색종피와 녹색자엽이면서 성숙 종실에서 렉틴, 7S α′ subunit, 리폭시게나제 및 KTI의 4가지 단백질이 모두 부재한 tetra null 유전자형(lecgy1lox1lox2lox3ti)을 가진 계통을 선발하기 위하여 본 연구가 진행되었다. 4개의 품종과 1개의 유전자원을 이용하여 육종집단 창성을 위한 두 모본이 선발되었다. 전체 58개의 F2 식물체로부터 DNA 마커를 이용하여 lele 유전자형을 가진 개체가 선발된 후 갈색종피와 녹색 자엽이면서 7S α′ subunit 단백질이 부재한 F3 종자가 선발되었다. 선발된 F3 종자는 F3 식물체를 거쳐 3개의 계통으로 육성되었다. 3개의 선발 계통(S1, S2, S3)에 대하여 F6 종자에서 렉틴, 7S α′ subunit, 리폭시게나제 및 KTI의 4가지 단백질에 대한 유전적 부재가 검정되었다. 3개의 선발 계통은 갈색종피, 녹색자엽 및 흰색배꼽을 가지고 있으며 백립중은 26.4-30.9 g으로 대조품종인 '청자3호'의 36.0 g보다 작았다. S2 선발 계통은 백립중이 30.9 g으로 대립이며 콩에서 항영양성분으로 알려진 lectin, 7S α′ subunit, lipoxygenase 및 KTI의 4가지 단백질 모두 부재한 유색콩 품종육성을 위한 중간모본으로 이용될 수 있을 것으로 사료되었다.

  • The genetic polymorphism of GSTM1, GSTT1, GSTP1, NATII and CYP1A1 genes among 33 asthma patients and 66 controls were investigated to find the association between the polymorphism and the risk of asthma. The frequency of the GSTT1 null genotype was slightly higher in asthma patients than that in the control, but this difference was not significant.(omitted)

  • 서 론 : 폐암의 80-90%는 흡연과 관계가 있으나 흡연자의 일부에서만 폐암이 발생하는 현상은 개체의 유전적 소인이 폐암발생을 결정하는 주요 요인임을 시사한다. 저자들은 한국인에서 발암물질 대사효소계의 유전자 다형성에 따른 폐암의 위험도를 조사하고자 연구를 시행하였으며 본 연구에서는 담배 내에 존재하는 benzo(a)pyrene 등의 polycylic aromatic hydrocarbon의 대사에 관여하는 GSTM1 과 CYP1A1 유전자 다형성에 따른 폐암의 상대위험도를 조사하였다. 방 법 : 1998년 1월부터 1998년 9월까지 경북대학교병원내과에서 병리학적으로 폐암으로 확진된 환자를 대상으로 하였으며 악성종양으로 진단받은 과거력이 있는 사람은 제외하였다. 대조군은 1998년 1월부터 1999년 8월까지 경북대학교병원 건강검진센터를 방문한 40세 이상의 검진자들을 대상으로 하였으며 호흡기질환이나 악성종양이 있는 경우는 제외하였다. 대상인의 나이, 성, 흡연력, 과거력 등은 면접이나 병력지를 통해 얻었으며, 시료는 전혈 5cc에서 DNA를 추출하고 PCR과 RFLP법을 통해 GSTM1과 CYP1A1의 유전자 다형성을 조사하였다. 결 과 : GSTM1(-) 형인 경우 소세포폐암의 대응비가 1.772로 높았으나 통계적 유의성은 없었다. CYP1A1 MspI 유전자형이 m2/m2 인 경우 m1/m1 형인 경우에 비해 소세포폐암의 대응비가 3.374(95% CI=1.092-10.421)로 유의하게 높았다. 결 론 : GSTM1과 CYP1A1 유전자형은 폐암의 위험도를 결정하는 인자로 생각되나, 보다 많은 예를 대상으로 한 연구가 필요할 것으로 생각된다.

CYP1A1, GSTM1, GSTT1 and TP53 Polymorphisms and Risk of Gallbladder Cancer in Bolivians

  • The Plurinational State of Bolivia (Bolivia) has a high incidence rate of gallbladder cancer (GBC). However, the genetic and environmental risk factors for GBC development are not well understood. We aimed to assess whether or not cytochrome P450 (CYP1A1), glutathione S-transferase mu 1 (GSTM1), theta 1 (GSTT1) and tumor suppressor protein p53 (TP53) genetic polymorphisms modulate GBC susceptibility in Bolivians. This case-control study covered 32 patients with GBC and 86 healthy subjects. GBC was diagnosed on the basis of histological analysis of tissues at the Instituto de Gastroenterologia Boliviano-Japones (IGBJ); the healthy subjects were members of the staff at the IGBJ. Distributions of the CYP1A1 rs1048943 and TP53 rs1042522 polymorphisms were assayed using PCR-restriction fragment length polymorphism assay. GSTM1 and GSTT1 deletion polymorphisms were detected by a multiplex PCR assay. The frequency of the GSTM1 null genotype was significantly higher in GBC patients than in the healthy subjects (odds ratio [OR], 2.35; 95% confidence interval [CI], 1.03-5.37; age-adjusted OR, 3.53; 95% CI, 1.29-9.66; age- and sex-adjusted OR, 3.40; 95% CI, 1.24-9.34). No significant differences were observed in the frequencies of CYP1A1, GSTT1, or TP53 polymorphisms between the two groups. The GSTM1 null genotype was associated with increased GBC risk in Bolivians. Additional studies with larger control and case populations are warranted to confirm the association between the GSTM1 deletion polymorphism and GBC risk suggested in the present study.
