On the use of time-dependent success criteria within risk-informed analyses. Application to LONF-ATWS sequences in PWR reactors

  • The classical Probabilistic Safety Analysis (PSA) does not include any time dependence explicitly. However, the success criteria (SC) could evolve during the cycle for some initiating events. In that sense, there is a type of sequence in which this time-dependency is quite important, the family of Anticipated Transient without Scram (ATWS) sequences in Pressurized Water Reactors. Therefore, a new risk-informed approach is proposed in this paper, which makes it possible to obtain the time-dependent SC evolution of the safety functions affected by the Moderator Temperature Coefficient (MTC) value. Then, the evolution of the ATWS conditional core damage probability (CCDP) could be obtained using a PSA model. To quantify the CCDP, the average values of the time-dependent failure probabilities must be computed. Finally, the comparison between the CCDP obtained through the application of the classical PSA approach and the new one makes it possible to quantify the impact of time-dependence on the SC of the headers that this new risk-informed ATWS approach can provide.

  • The lengthened average span of human life by virtue of recent developments in medicine has caused the Population of elders to increase. The development of modern industrial society has transformed family structure from the large family system to that of a nuclear family. Due to the shift in family structure, the problem of support for the aged has surfaced as a nursing problem as well as a social problem. With regard to this problem, this study aims to investigate the adolescent's understanding of elders and aging. By identifying their understanding and classifying their attitude patterns, this study will help the nursing assessment of the support of elders in the family. This study employed Q-methodology and the research was conducted from December 1998 to May 1999. One method of the research included deep interviews with elders, those who are in their 50's. 40's or 30's. and the adolescent. 183 Q-Populations taken from literary works such as poems or novels were also formed as another method. Finally. 36 Q-cards were made after consultation with Professors of the nursing department. The subjects of the P- sample were 30 high sohoolboys/girls - who were in first, second, and third years. The result showed that 3 factors provided an explanation for 59.14% of the whole variables: the first factor, 41.37%; the second factor. 11.49%, and the third factor. 6.28%. These three factors were analyzed and categorized as three types. Twenty subjects out of the 30 were included in Type 1: Respecting Elders. The statements which showed the most positive consent were as follows: 'The declining age is a perfect time to prosper completing a worthy life' ; 'Getting old. one needs financial stability' and 'Elders wish the best for their children' The statements showing the most negative response were as follows: 'It is better to die than to live as an older person' ; 'Elders are insignificant' ; and 'Getting old is the worst unhappiness that tortures human.' Four subjects were included in Type 2. Resenting Elders. The statements which showed the most positive consent were as follows: 'Aging is a process of dying that nobody can escape from'. 'Elders should be concerned about his health and try to maintain their health' ; and 'When you set older. you regret about the life in the Past.' The statements showing the most negative response were as follows: 'When You get older. You should stand aloof greed and worldly things' 'When You got older, You become generous and gentle' ; and 'When You set he gets old. You change to become a comfortable and warm person.' Six out of 30 subject were included in Type 3 Caring Elders. The statements which showed the most positive consent were as follows: 'Elders should be concerned about his health and try to maintain their health' ; 'Elders wish the best for their children' ; and 'Elders deserve to be treated with filial respects.' The statements showing the most negative response were as follows 'Elders are insignificant' ; 'Elders have freedom and plenty of free time.' and 'Elders are alienated form and drove out of the society.' The above-mentioned results show that most adolescents in Korea recognize aging as the time of fruition and development: it is a time of benefiting and giving back to society. Aging can also be seen as a time of generosity and magnanimity and the time of respect and favorable treatment from society. despite the change of modern society and the ostensible transformation of a family system. Their recognition seems deeply rooted in the traditional confucian values and the dual family system which is Peculiar especially to the Korea - one which maintains both the superficial form of nuclear family and the substantial mode of the enlarged family system. In sum, many Korean adolescents attribute the meaning of the elderly and aging to the type of the respect with the elderly and the type of the elderly's caretaking.

Identification and Functional Characterization of P159L Mutation in HNF1B in a Family with Maturity-Onset Diabetes of the Young 5 (MODY5)

  • Mutation in HNF1B, the hepatocyte nuclear factor-$1{\beta}$ (HNF-$1{\beta}$) gene, results in maturity-onset diabetes of the young (MODY) 5, which is characterized by gradual impairment of insulin secretion. However, the functional role of HNF-$1{\beta}$ in insulin secretion and glucose metabolism is not fully understood. We identified a family with early-onset diabetes that fulfilled the criteria of MODY. Sanger sequencing revealed that a heterozygous P159L (CCT to CTT in codon 159 in the DNA-binding domain) mutation in HNF1B was segregated according to the affected status. To investigate the functional consequences of this HNF1B mutation, we generated a P159L HNF1B construct. The wild-type and mutant HNF1B constructs were transfected into COS-7 cells in the presence of the promoter sequence of human glucose transporter type 2 (GLUT2). The luciferase reporter assay revealed that P159L HNF1B had decreased transcriptional activity compared to wild-type (p < 0.05). Electrophoretic mobility shift assay showed reduced DNA binding activity of P159L HNF1B. In the MIN6 pancreatic ${\beta}$-cell line, overexpression of the P159L mutant was significantly associated with decreased mRNA levels of GLUT2 compared to wild-type (p < 0.05). However, INS expression was not different between the wild-type and mutant HNF1B constructs. These findings suggests that the impaired insulin secretion in this family with the P159L HNF1B mutation may be related to altered GLUT2 expression in ${\beta}$-cells rather than decreased insulin gene expression. In conclusion, we have identified a Korean family with an HNF1B mutation and characterized its effect on the pathogenesis of diabetes.

Deficiency of Anoctamin 5/TMEM16E causes nuclear positioning defect and impairs Ca2+ signaling of differentiated C2C12 myotubes

  • Anoctamin 5 (ANO5)/TMEM16E belongs to a member of the ANO/TMEM16 family member of anion channels. However, it is a matter of debate whether ANO5 functions as a genuine plasma membrane chloride channel. It has been recognized that mutations in the ANO5 gene cause many skeletal muscle diseases such as limb girdle muscular dystrophy type 2L (LGMD2L) and Miyoshi muscular dystrophy type 3 (MMD3) in human. However, the molecular mechanisms of the skeletal myopathies caused by ANO5 defects are poorly understood. To understand the role of ANO5 in skeletal muscle development and function, we silenced the ANO5 gene in C2C12 myoblasts and evaluated whether it impairs myogenesis and myotube function. ANO5 knockdown (ANO5-KD) by shRNA resulted in clustered or aggregated nuclei at the body of myotubes without affecting differentiation or myotube formation. Nuclear positioning defect of ANO5-KD myotubes was accompanied with reduced expression of Kif5b protein, a kinesin-related motor protein that controls nuclear transport during myogenesis. ANO5-KD impaired depolarization-induced $[Ca2^{+}]_i$ transient and reduced sarcoplasmic reticulum (SR) $Ca^{2+}$ storage. ANO5-KD resulted in reduced protein expression of the dihydropyridine receptor (DHPR) and SR $Ca^{2+}-ATPase$ subtype 1. In addition, ANO5-KD compromised co-localization between DHPR and ryanodine receptor subtype 1. It is concluded that ANO5-KD causes nuclear positioning defect by reduction of Kif5b expression, and compromises $Ca^{2+}$ signaling by downregulating the expression of DHPR and SERCA proteins.

A study on the classification of consumer types based on food safety attitudes: focusing on the Fukushima nuclear accident

  • The purposes of this study were to classify consumer types and to analyze purchase behavior depending on food safety attitudes related to the Fukushima nuclear accident. Based on the consumer survey data, factor analysis and cluster analysis were done to classify consumer types. Applying factor analysis, six factors with a total variance of 73.4% were extracted from 22 basic variables. The 6 factors were termed 'anxiety from ingesting agricultural and fishery products after the nuclear accident', 'importance of food safety when purchasing food', 'reduced rate of purchases of agricultural and fishery products after the accident', 'importance of food prices', 'socio-demographic characteristics' and 'family composition'. Using cluster analysis, consumers were classified into 5 types using the factor scores of 6 factors. The main results are as follows. For every consumer type, the anxiety about imported agricultural and fishery products was higher than that for domestic products after the nuclear accident. The food safety attitudes of consumers were mainly formed by the socio-demographic characteristics. Consumer types with high-education and high-income responded flexibly to the food safety concerning the accident; especially, the consumer type with a high-income and kids under 19 responded very reasonably to the accident. However, consumer types with a low-income and aged over 50 or less than 30 years were the least responsive to the accident, mainly due to financial restrictions. Thus, we suggest some food safety strategies and policy implications based on consumer type for food safety after the accident.

L-ascorbic acid induces apoptosis in human laryngeal epidermoid Hep-2 cells by modulating the nuclear factor kappa-light-chain-enhancer of activated B cells/mitogen-activated protein kinase/Akt signaling pathway

  • L-ascorbic acid (L-AA; vitamin C) induces apoptosis in cancer cells. This study aimed to elucidate the molecular mechanisms of L-AA-induced apoptosis in human laryngeal epidermoid carcinoma Hep-2 cells. L-AA suppressed the viability of Hep-2 cells and induced apoptosis, as shown by the cleavage and condensation of nuclear chromatin and increased number of Annexin V-positive cells. L-AA decreased Bcl-2 protein expression but upregulated Bax protein levels. In addition, cytochrome c release from the mitochondria into the cytosol and activation of caspase-9, -8, and -3 were enhanced by L-AA treatment. Furthermore, apoptosis-inducing factor (AIF) and endonuclease G (EndoG) were translocated into the nucleus during apoptosis of L-AA-treated Hep-2 cells. L-AA effectively inhibited the constitutive nuclear factor-κB (NF-κB) activation and attenuated the nuclear expression of the p65 subunit of NF-κB. Interestingly, L-AA treatment of Hep-2 cells markedly activated Akt and mitogen-activated protein kinase (MAPK; extracellular signal-regulated kinase 1/2, p38, and c-Jun N-terminal kinase [JNK]) and and LY294002 (Akt inhibitor), SB203580 (p38 inhibitor) or SP600125 (a JNK inhibitor) decreased the levels of Annexin V-positive cells. These results suggested that L-AA induces the apoptosis of Hep-2 cells via the nuclear translocation of AIF and EndoG by modulating the Bcl-2 family and MAPK/Akt signaling pathways.

경남 일부지역 노인복지시설 입소자의 구강건강 실태조사 (Study on actual condition of oral hygiene among admitted aged person at the some of Gyeongnam welfare facilities)

  • In Korea, respect for the aged and filial devotion is treated as basic ethics for human life, and family takes care of the aged person mainly. Nowadays, family support on the aged person is prioritized than the others. However, number of aged person is growing, and family, which is used to protect the aged, becomes nuclear through industrialization. In addition, social advancement of female induces weakened supporting function of family, and all these issues generate the problem of protection for the aged as significant social problems. Author conducted oral inspection and questionnaire for the aged in some welfare facilities in Gyeongnam location from December 2005 to February 2006 to improve quality of life and oral hygiene of the aged. Through gathered data, the actual condition of oral hygiene management on the aged person in welfare facility was evaluated as fundamental data for project development on oral hygiene of aged person. Through the analysis of inspected data, the following conclusions are derived. 1. Management status of oral hygiene is mostly not good, and toothbrushing per day is 'one time' for the most cases. 2. In free-of-charge facility, monthly allowance is 'under 50 thousand won' for the most cases. 3. Time for visiting oral treatment facility is on 'when toothache is occurred' for the most cases. For treatment content, 'prosthetic dentistry' takes 36.8% in charged facility, and 'tooth extraction' is 27.0% for free-of-charge facility. 4. Average DMFT index of the aged in charged facility is 16.81, and free-of-charge facility is 21.71. 5. Average number of functional teeth in charged facility is 15.22, and free-of-charge facility shows 7.29. 6. Average number of remained teeth in charged facility is 15.71, and the umbe in free-of-charge facility is 9.04. 7. Average number of extractable teeth in charged facility is 0.48, and for free-of-charge facility, the number goes up to 1.70.

김치에 관한 원주지역 주부들의 식생활 실태조사 (A Survey on the Dietary Life toy Kimchi of Housewives in Wonju Area)

  • The purpose of this study was to investigate the dietary life about Kimchi of the housewives. Self-administrated questionnaires were completed by 322 housewives in Wonju area. The results were as follows: In the point of housewives' domestic duties, over 50% of housewives took care of their domestic duties alone, and the rate who learned how to manage domestic duties from their mothers before marriage decreased as the age got younger. 77.6% of housewives responded that both man and woman could manage a dietary life at home in the future, and the rate of such response was higher as their educational careers were higher and they were younger. In the point of housewives' basic situation about Kimchi, the rate of housewives who knew how to prepare kimchi was high as they were older, and the holding rate of a refrigerator for Kimchi was high as much as 72.4%. In the point of preparation for Kimchi, the rate who made Kimchi by themselves was high, but the rate of preparing Kimchi by their parents for them was higher in the twenties. Frequency of making Kimchi, an amount or a kind of Kimchi were notably different according to housewives' age, a type of their family, the number of their family, and their having a job or not. Housewives' opinion of a market kimchi was that it was expensive, and not sanitary, and rate of purchasing Kimchi was also low. In the point of the prepartion method of Kimchi for the winter, the rate who gained Kimchi from their parents was higher as they lived in an apartment, and had a nuclear family, and had the lesser family members. In an amount of Kimchi prepared for the winter, 11∼22 heads were 23.9% and 21∼30 heads were 21.9%. In the point of the method preparing Kimchi in the future, 53.8% of housewives responded that they would make by themselves, and 28.0% answered they would prepare or purchase. 77.4% of housewives answered that they would teach how to make Kimchi to their children. In conclusion, in housewives of Wonju area, wanted to make Kimchi by themselves and wanted their children to learn how to make Kimchi.

초국가적 입양과 탈경계적 정체성 -제인 정 트렌카의 『피의 언어』 (Transnational Adoption and Beyond-Borders Identity: Jane Jeong Trenka's The Language of Blood)

  • This paper elucidates the characteristics of transnational adoption, estimates the possibility of beyond-borders identity of transnational adoptees, and tries to analyze Jane Jeong Trenka's The Language of Blood in its context. Though it has been regarded as one of the most humanitarian ways of helping orphans and poor children of the world, transnational adoption, a one-way flow of children from poor Asian countries to rich white countries, has been operated under the market logic between countries. Transnational adoptees, who had been abandoned and forced to be taken away from their birth mother, and later, to fulfill the desire of white parents for a perfect family, perform an ideological labor, serving to make the heterogeneous nuclear family complete. Korean transnational adoptees, forced to transcend the borders of nation, culture, and ethnicity, experience racial conflict and alienation in white adoptive family and society. Their diaspora experience of violent dislocation creates frustration and confusion in establishing their identity as a whole being. When they return to Korea to find their birth mother and their true identity, Korean adoptees, however, are faced with other obstructing issues, such as language problem, culture conflict, and maternal nationalism. Finally, Korean transnational adoptees reject Korean nationalism discourse based on blood, and try to redefine themselves as beyond-borders subjectivities with new and fluid identities. Jane Jeong Trenka's The Language of Blood, an autobiographical novel based on her experiences as a transnational adoptee, represents a Korean adopted girl's personal, cultural, and racial conflict within her white adoptive family, and questions the image of benevolent white mother and the myth of multiculturalism. The novel further represents Jane's return to Korea to find out her true identity, and shows Jane's disappointment and alienation in her birth country due to her ignorance of language and culture. Returning to USA again, and trying to be reconciled with her American mother, Jane shows the promise of accepting her new identity capable of transcending the borders, and thus, the possibility of enlarging the category of belonging.

Assessment of Biomarkers in Acetaminophen-Induced Hepatic Toxicity by siRNA

  • We investigated global gene expression from both mouse liver and mouse hepatic cell lines treated with acetaminophen (APAP) in order to compare in vivo and in vitro profiles and to assess the feasibility of the two systems. During our analyses of gene expression profiles, we picked up several down-regulated genes, such as the cytochrome P450 family 51 (Cyp51), sulfotransferase family cytosolic 1C member 2 (Sult1c2), 3-hydroxy-3-methylglutaryl-Coenzyme A synthase 1 (Hmgcs1), and several genes that were up-regulated by APAP, such as growth arrest and DNA-damage-inducible 45 alpha (Gadd45a), transformation related protein 53 inducible nuclear protein 1 (Trp53inp1) and zinc finger protein 688 (Zfp688). For validation of gene function, synthesized short interfering RNAs (siRNAs) for these genes were transfected in a mouse hepatic cell line, BNL CL.2, for investigation of cell viability and mRNA expression level. We found that siRNA transfection of these genes induced down-regulation of respective mRNA expression and decreased cell viability. siRNA transfection for Cyp51 and others induced morphological alterations, such as membrane thickening and nuclear condensation. Taken together, siRNA transfection of these six genes decreased cell viability and induced alteration in cellular morphology, along with effective inhibition of respective mRNA, suggesting that these genes could be associated with APAP-induced toxicity. Furthermore, these genes may be used in the investigation of hepatotoxicity, for better understanding of its mechanism.
