Effects of raw soy flour(RSY) and black(RSB) feeding on protein concentration of liver and serum, and GOT, GPT and alkaline phosphatase activities of serum in diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley), mean weight of (338.4$\pm$19.2g) were assigned to six dietary groups and fed with the assigned diet for 28 days. For each experimental, some rats were injected with streptozotocin intraperitoneally(L.P.) to induce diabets, and other rats were injected with buffer L.P. as a control group. The liver, kidney and spleen weights relative to bo요 weigth were higher in raw yellow soy flour diet diabetes(D-RSY) and black soy flour diet diabetes(D-RSB) groups than control, but the body weights were lower than control. The protein and albumin concentrations of liver and serum were lower in D-RSY and D-RSB groups than control. The albumin concentration of serum in D-RSB group was lower than control. The GOT activities of serum in RSY and RSB groups were increased compared with control, but the GPT activities were lower in diabetic control, D-RSY and D-RSB groups than control. The alkaline phosphatase activities of serum in RSY and RSB groups were higher than control, but those in D-RSY and D-RSB groups were lower than diabetic control.
Much research evidence supports the hypothesis that chronic, low-grade inflammation related to innate immunity may play an important role in the pathophysiology of type 2 diabetes mellitus (T2DM). Runt-related transcription factor 2 (RUNX2; MIM# 600211) acts as a scaffold that controls the integration, organization, and assembly of nucleic acids. To examine whether the novel promoter variant in RUNX2 is associated with the risk of T2DM and related phenotypes, RUNX2-742G > T was genotyped in 378 T2DM patients and 382 normal controls recruited in the Korean T2DM Study. Statistical analysis revealed that RUNX2-742G > T was associated with serum triglyceride level (TG) in nondiabetic controls, although it was not associated with the risk of T2DM. Individuals who carry T/T, T/G, and G/G genotypes had the highest ($2.061{\pm}0.20$), intermediate ($2.01{\pm}0.19$), and the lowest ($1.97{\pm}0.18$) levels of log [TG (mmol/l)] (P = 0.007), respectively. Our data on this important variant of RUNX2 suggest that lipid metabolism might be affected by genetic polymorphisms in the promoter region.
In this study. the importance of body fat distribution as an indicator of metabolic aberrations in diabetics was'evaluated. Skinfold thicknesses at eight sites and circumferences at five sites and total body fat content were measured on 105 diabetics. 1) The waist/thigh girth ratio(WTR) was positively correlated(r=0.38-0.54) to plasma glucose levels during oral glucose loading in diabetic men alone. 2) There was no significant difference in serum total cholesterol of diabetics and nondiabetics. However, HDL-cholesterol was significantly lower(p : 0.000. in male and female) in diabetics(male : 36.31$\pm$ 16.98mg/dl, female: 37.89$\pm$14.67mg/dl) than nodiabetics(male 61.75$\pm$14.08mg/dl, female : 62.29$\pm$12.65mg/dl) and serum triglyceride was significantly higher(p=0.0212) in diabetic women(171.90$\pm$ 76.61mg/dl) than nondiabetic women(111.10$\pm$42.84mg/dl) . 3) In both sexes. anthropometric measurements that significantly correlated to serum triglyceride concentration were percentage of ideal body weight. body mass index(BMI) and waist circumference. Positive and significant correlations were found between serum cholestrol, and BMI(R: 0.31, 0.34) and waist circumference(r=0.29) in diabetic men. Moreover, skinfolds of trunk area(r=0.29~0.32) especially abdomen, were closely associated with serum lipids than other fat deposits. Increasing percentage of total body fat content was accompanied by progressively increasing serum triglyceride concentration(r=0.41) in dieabetic men. This study shows that knowledge of body fat localization may help identify risks of carbohydrate intolerance and hyperlipidemia in diabetics.

Few research had investigated the secretion of phospholipase and aspartyl proteinase from Candida spp. causing infection in females with type 2 diabetes mellitus. This research aimed to investigate the prevalence of vulvovaginal candidiasis (VVC) in diabetic versus non-diabetic women and compare the ability of identified Candida isolates to secrete phospholipases and aspartyl proteinases with characterization of their genetic profile. The study included 80 females with type 2 diabetes mellitus and 100 non-diabetic females within the child-bearing period. Candida strains were isolated and identified by conventional microbiological methods and by API Candida. The isolates were screened for their extracellular phospholipase and proteinase activities by culturing them on egg yolk and bovine serum albumin media, respectively. Detection of aspartyl proteinase genes (SAP1 to SAP8) and phospholipase genes (PLB1, PLB2) were performed by multiplex polymerase chain reaction. Our results indicated that vaginal candidiasis was significantly higher among the diabetic group versus nondiabetic group (50% versus 20%, respectively) (p = 0.004). C. albicans was the most prevalent species followed by C. glabrata in both groups. No significant association between diabetes mellitus and phospholipase activities was detected (p = 0.262), whereas high significant proteinase activities exhibited by Candida isolated from diabetic females were found (82.5%) (p = 0.000). Non-significant associations between any of the tested proteinase or phospholipase genes and diabetes mellitus were detected (p > 0.05). In conclusion, it is noticed that the incidence of C. glabrata causing VVC is increased. The higher prevalence of vaginal candidiasis among diabetics could be related to the increased aspartyl proteinase production in this group of patients.
Purpose: This study was performed to identify changes in blood glucose at preoperative fasting time in surgical patients over 60 yr. Methods: Data collection was performed from July, 2008 through July, 2009. Participants consisted of 80 nondiabetic surgical patients. Blood glucose was checked from 3 to 5 times. The 5 times were 2-hr fasting on the pre-operative day (T1, n=80), 8 hr (T2, n=80), 10 hr (T3, n=17), 12 hr (T4, n=34) and 14 hr fasting on the day of the operation (T5, n=29). Results: Of the patients, 27.5% had a blood glucose level of less than 79 mg/dL at T2; 17.6% at T3; 32.4% at T4; and 17.2% at T5. Mean blood glucose levels were 93.8 mg/dL at T1; 88.4 mg/dL at T2; 91.7 mg/dL at T3; 87.4 mg/dL at T4: and 94.1 mg/dL at T5. Blood glucose was the lowest at T2 (p<.001). Conclusion: As 17.6-32.4% of the patients showed the blood glucose level of less than 79 mg/dL at 8-14 hr pre-operative fasting, the authors recommend that surgical patients p>60 yr-of-age be observed for hypoglycemia during pre-operative fasting of more than 10 hr and that surgical patients >60 yr-of-age with risks for hypoglycemia be scheduled for operation within 10 hr preoperative fasting.
The purpose of this study was to determine the dose effects of taurine supplementation and effects of taurine depletion in diabetic rats. Sprague-Dawley male rats were fed the purified diets and supplemented with 1, 2 or 3% taurine in drinking water for 7 weeks(E1, E2 and E3, respectively). To induce taurine depletion, rats were treated with 5% $\beta$-alanine in drinkeng water (EA). Diabetes was induced by streptozotocin injection(50mg/kg B.W) after 3 weeks. Taurine-depleted diabetic rats(EA)had significantly lower body weight compared to those of nondiabetic(CO), nontaurine-supplemented diabetic(E0) and taurine-supplemented diabetic rats(E1, E2 and E3). E0 had significantly higher food intake compared to that of CO and EA. E1, E2 and E3, however, had significantly lower food intake compared to that of E0, and E3 had significantly lower food intake compared to that of E1 and E2. E0 had significantly higher water intake compare to that of CO and EA. EI had significantly lower water intake compared to that of E0. Blood glucose concentration of E0 was significantly increased compared to that of CO and EA. E2 and E3 had significantly lower blood glucose concentration compared to E0 and E1. Also there was a dose effect in blood glucose concentration between E2 and E3. The total cholesterol and LDL-cholesterol concentrations of EA were significantly increased compared to those of other groups. Therefore, it may be suggested that taurine supplementation is necessary for diabetes in order to prevent diabetic complications such as cardiac vascular diseases.
Recent evidence has strongly suggested that the CAP/TC10 pathway is involved in the trafficking, docking, and fusion of vesicles containing the insulin-responsive glucose transporter Glut4 to the plasma membrane. However, little is known about how the genes employed in the CAP/TC10 pathway are associated with the development of type 2 diabetes mellitus. In this study, we sequenced 4 genes of the CAP/TC10 pathway [SORBS1, CBL, CRK, and RHOQ] in 24 individuals to identify genetic variations in these loci. A total of 48 sequence variants were identified, including 23 novel variations. To investigate the possible association with type 2 diabetes mellitus, 3 single nucleotide polymorphisms from SORBS1, 3 from CBL, and 4 from RHOQ were genotyped in 1122 Korean type 2 diabetic patients and 1138 nondiabetic controls. Using logistic regression analysis, 1 significant association between SNP rs1376405 in RHOQ and type 2 diabetes mellitus [OR = 8.714 (C.I. 1.714-44.29), p = 0.009] was found in the recessive model. Our data demonstrate a positive association of the RHOQ gene in the CAP/TC10 pathway with T2DM in the Korean population.
This study was designed to examine the effects of sea buckthorn (SBT) on the plasma blood glucose and cholesterol level in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200~220 g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of SBT powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-SBT 10% and N-SBT 20% treated groups, STZ-control, STZ-SBT 10% and STZ-SBT 20% treated groups. The rats' body weight, aminotransferase activities and hematocrit (Hct) values were measured along with plasma levels of blood glucose and cholesterol. Body weight losses were observed by diabetic groups While the nondiabetic rats gained weight. There were significant differences between the control group and the diabetic groups in the weight of kidney. Aspartate aminotransferase activity was lower in the non-diabetic group compared to diabetic experimental groups. The blood glucose were significantly decreased in the 10% SBT of diabetic group. The cholesterol level of STZ-SBT 10% and STZ- SBT 20% were significantly lower than for the STZ-control group. These results show that the supplementation of sea buckthorn leave powder may have favorable influence on reducing blood glucose and cholesterol level in STZ-induced diabetic rats.
Objectives : This study aimed to evaluate the anti-diabetic effect of Wen-Pi-Tang-Hab-Wu-Ling-San (WHW) extract in streptozotocin(STZ)-induced type-1 diabetic rats. Methods : Experimental diabetes were induced by intraperitoneal injection of streptozotocin (60 mg/kg). Two groups of STZ-induced diabetic rats were given the following treatments for 2 weeks by oral Administrations : (1) WHW 10 mg/kg, (2) WHW 100 mg/kg. In addition, vehicle-treated diabetic and nondiabetic controls were used in the experiment. The effects of WHW extract on STZ-induced diabetes were observed by measuring the changes of body weights and the levels of fasting blood glucose, insulin, urea nitrogen (BUN) and creatinine level in sera of rats, respectively. Results : In comparison control group, WHW-treated groups (100 mg/kg) were significantly decreased fasting blood glucose levels and increased serum insulin levels in STZ-induced diabetic rats. Moreover, WHW-treated groups (100 mg/kg) were reduced s-creatinie levels in STZ-induced diabetic rats. In addition, the changes related to diabetic nephropathy with body weight were significantly lower in WHW extract-dosing groups than in the diabetic control. Conclusions : The study thus showed that WHW extract enhanced the anti-diabetic effect in STZ-induced diabetic rats by improving the hypoglycemia. It also increased pancreatic insulin content in these rats.
This study was an attempt to compare the energy, nutrient intake, blood lipids, and body fat distribution of diabetics with those of normal control subjects. Daily energy and nutrient intake were measured by convenient method. The following anthropometric measurements were made on all participants : weight, height, and waist and hip circumference. We matched 31 pairs with diabetics and nondiabetic control subjects by sex, age, body weight and height, and made comparisons between two groups to observe the effect of diabetes in lipid profiles. Aslo the effect of exercise in diabetes was investigated. The results obtained were as follows : 1.The average of daily energy intake of diabetics was 1, 927 kcal and that of nondiabetics was 2, 159 kcal. Percentage of energy in diabetics is that carbohydrate : protein : fat = 69:13:18 2. When diabetics were divided into obese and nonobese group according to RBW, 33% of diabetics were obese and energy intake was not significantly different between the two groups, but VLDL - cholesterol concentration was significantly higher in the obese group. 3. Energy intake, BMI, RBW and total cholesterol level of upper obesity group were significantly higher than those of lower body obesity group in male diabetic subjects. 4. When matching 31 pairs of diabetics and nondiabetics according to sex, age, body weight and height triglyceride concentration and WHR of diabetics were measured to be significantly higher than those of nondiabetics. 5. Although energy intake was not significantly different between exercise and nonexercise groups, triglyceride, blood glucose, RBW, BMI were significantly lower in the exercise and nonexercise groups, triglyceride, blood glucose, RBW, BMI were significantly lower in the exercise group than the nonexercis group in diabetics. 6. BMI and RBW were not correlated with cholesterol or triglyceride, but WHR correlated significantly with both cholesterol and triglyceride. In conclusion, diabetics had higher triglyceride concentration and WHR than nondiabetics. WHR may be better than BMI or RBW for predicting alteration in lipid profile and exercise seems beneficial for controlling blood glucose and lipids in diabetics.
