Development of transgenic cucumber expressing TPSP gene and morphological alterations

  • To develop transgenic cucumber tolerant to abiotic stress, a cotyledonary-node explants were co-cultivated with Agrobacterium tumefaciens (EHA101) carrying TPSP gene (pHC30-TPSP). After transfer to fresh medium every two week for eight weeks, putative transgenic plants were selected when shoots grown a length greater than 3 cm from the cotyledonary-node explants on selection medium supplemented with $5\;mgl^{-1}$ phospinotricin as selectable agent. The confirmation of transgenic cucumber was based on the Northern blot analysis. Thirty four shoots (5.2%) with resistance to phospinotricin were obtained from 660 explants inoculated. Of them, transformants were only confirmed from 11 plants (1.7%). Transgenic cucumber expressing TPSP gene was more synthesized at 3.8 times amounts of trehalose (0.014 mg g fresh $wt^{-1}$) than non-transformants (0.0037 mg g fresh $wt^{-1}$). However, all of transgenic plants showed abnormal morphology, including stunted growth (< height 15 cm), shrunken leaves, and sterility as compared with non-transgenic plants (> height 150 cm) under the same growth environment. These results lead us to speculate that the overproduction of trehalose was toxic for cucumber, even though that had known for rice as non-toxic.

Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Diabetic Rats ($\beta$-Carotene 첨가식이가 당뇨쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향)

  • This study investigated the effect of dietary $\beta$-carotene supplementation on lipid peroxidation and anti oxidative enzyme activity as indices of oxidative stress in diabetic rats. Fifty Sprague-Dawley male rats aging 7 weeks were used as experimental animals, which were divided into the non-diabetic control group and the diabetic group. The diabetic group received an intraperitoneal injection with streptozotocin to induce diabetes. Then the diabetic rats were divided into four dietary groups which contained different amounts of $\beta$-carotene; 0%, 0.002%, 0.02%, or 0.2% of the diet. The diabetic rats were fed the experimental diets and the non-diabetic rats were fed the basal diet without $\beta$-carotene supplementation for 2 weeks and then sacrificed. The diabetic group had a significantly higher blood glucose level than the non-diabetic group. However, blood glucose level were not significantly changed by the level of dietary $\beta$-carotene supplementation. Compared to the non-diabetic control group, the diabetic control group indicated a significant increase of plasma thiobarbituric acid reactive substance (TBARS). Liver TBARS level also tended to be higher in diabetic control group, although it was not significant. The $\beta$-carotene supplementation did not reduce plasma TBARS level. However, Liver TBARS level was significantly decreased when 0.02% or more $\beta$-carotene was supplemented in the diet. The liver lipofuscin level in the diabetic control group was higher than in the non-diabetic control group, but the effect of $\beta$-carotene supplementation did not show any differences. Superoxide dismutase activity was significantly lower in the diabetic group, but it was increased in groups receiving 0.02% or more $\beta$-carotene. Compared to the non-diabetic control group, lower activities of catalase and glutathione peroxidase were observed in the diabetic control group, although it was not significant. Catalase and glutathione peroxidase activities tended to increase as the levels of $\beta$-carotene supplementation increased, although it was not statistically significant. Therefore, it seems that dietary $\beta$-carotene supplementation might reduce diabetic complications by partly decreasing the lipid peroxidation and increasing the activity of antioxidative enzyme in diabetes.

Effects of n-3 Polyunsaturated Fatty Acids-enriched Diet Supplemented with Different Levels of α-Tocopherol on Lipid Metabolism in Laying Tsaiya Ducks

  • The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.

Hemato-biochemical and Cortisol Profile of Holstein Growing-calves Supplemented with Vitamin C during Summer Season

  • Effect of vitamin C (VC) on biochemical, hematological and cortisol profile of growing Holstein calves during summer was investigated. Eighteen calves between 14 and 16 weeks of age were divided equally into two groups. One group received a diet supplemented with VC (20 g/d) for 60 days, while other non-supplemented diet fed group served as a control (CON). The temperature humidity index (THI) was recorded and computed during the experiment. From days 0 to 60, the THI exceeded 70. Blood samples were collected from the jugular vein of each calf at days 0, 15, 30, 45 and 60. Serum albumin and total protein decreased (p<0.05) in CON and VC calves with age. Serum glutamic-oxaloacetic transaminase concentrations were not affected by treatments. Serum creatinine, albumin and glutamic-pyruvic transaminase concentrations were higher in calves in the VC group than the CON group. While red blood cells, hemoglobin and hematocrit were lower (p<0.05) in VC calves, mean corpuscular volume, mean corpuscular hemoglobin, red blood cell distribution width and mean platelet volume were higher (p<0.05) in these VC supplemented calves. Leukocyte parameters including white blood cells and full term for lymphocytes were not affected by the treatments. Also, serum cortisol was not affected by treatments. At day 15, 30 and 45, the total VC in plasma was higher (p<0.05) in calves fed with VC. In conclusion, serum cortisols were not affected by plasma VC concentration, while some blood parameters were positively influenced in calves fed with VC.

Effects of Selenium, Copper and Magnesium on Antioxidant Enzymes and Lipid Peroxidation in Bovine Fluorosis

  • The antioxidant enzymes, lipid peroxidation and free radicals assessment were made of the effects of selenium, copper and magnesium on bovine endemic fluorosis under high fluoride, low selenium and low copper productive conditions. Thirty-two beef cattle were selected from high fluoride area, and randomly divided into four groups with eight cattle each as follows: (1) high fluoride control group (HFC); (2) supplemented group with 0.25 mg/kg selenium (HFSe); (3) supplemented group with 15 mg/kg copper (HFCu) and (4) supplemented group with 0.25 mg/kg selenium+15 mg/kg copper+1 mg/kg magnesium (HFSeCuMg) per day for 83 days. Moreover, eight beef cattle were selected from non-high fluoride area as normal control group. Blood samples were collected from cattle on 0 d, 30 d and 83 d respectively, to analyze the enzyme activities and concentration of GSH-px, CAT, SOD, MDA and free radicals. The results showed that the contents of free radicals and MDA in HFC group were significantly higher, and the whole blood GSH-px, CAT, erythrocyte SOD activities were lower than the normal control group. Free radicals, metabolic imbalance and antioxidant disorder therefore, play an important role in fluorosis. However, GSH-px, CAT and SOD activities in HFSe group and HFSeCuMg group at 30 d and 83 d were markedly higher than the same groups at the 0 d and the HFC group at the same time. Likewise, there was a corresponding reduction in the contents of free radicals and MDA. These findings indicated that supplementation with selenium, copper and magnesium elevated high fluoride bovine antioxidant enzymes, and decreased MDA and free radicals contents. But, the activities of supplementation selenium group did not increase until day 83. These results demonstrated that fluorosis was associated with lower serum Se and Cu levels than in the control, and it was therefore concluded that fluorosis is associated with decreased serum levels of these minerals. Long-term high fluoride intake under productive condition enhances oxidative stress in the blood, thereby disturbing the antioxidant defense of cattle. Increased oxidative stress could be one of the mediating factors in the pathogenesis of toxic manifestations of fluoride. It is benefical for high fluoride cattle supplemented with proper selenium, copper and magnesium to increase fluoride excretion and obtain the protective impact of the activity of oxidative enzymes, and to decrease lipid peroxidation and free radicals contents.

  • The variations of both thiamine and riboflavin value in the organs, viz. liver, small intestine, spleen and kidney of the rats were measured for observing some metabolic changes in the animals during fasting and feeding different quality of diets without V-E supplement. The animal used for the experiment was adult female ablino rat from a pure strain, weighing 225-280g. The animals were divided into 6 groups; the control group, the high carbohydrate diet group, the high carbohydrate diet with V-E group, the high protein diet group, the high protein diet with V-E group, and fasting group. The result obtained are summarized as follows; 1. The thiamine contents in the liver were once increased during early stage of starvation compared with the control group, thereafter they were decreased on the 8 days fasting while the contents in the small intestine and spleen were decreased during 1 to 8 days fasting. 2. The riboflavin contents in the liver and kidney were increased during starvation and the content in the small intestine was no marked change compared with control group. 3. The thiamine contents in the liver and small intestine during feeding the high carbohydrate with V-E supplement diet group were lower than that of the diet without V-E group and the content in the spleen was increased by feeding V-E enriched high carbohydrate diet. 4. The thiamine contents in the liver, small intestine and spleen during feeding the V-E supplemented diets were lower than that of the non-supplemented one's. 5. The riboflavin contents in the liver, small intestine, and kidney were increased during feeding the high carbohydrate diet compared to the control group, and they were decreased during feeding the V-E enriched high carbohydrate diet. 6. The riboflavin contents in each organ were increased during feeding the high protein diet compared to the control group, and they were much increased during 20 to 30 days of feeding the V-E supplemented high protein diet. 7. Therefore, as the above results showed, the variation of thiamine value in the each organs were not markedly changed during feeding different quality of the diets. The thiamine and riboflavin contents in the each organ in the V-E enriched high carbohydrate diet group were lower than without V-E supplemented one's The riboflavin contents in each organ were increased during feeding the high protein diet compared with the control group and the centents were increased during 20 to 30 days of the feeding V-E enriched high protein diet.

Effect on Cryopreservation Stability of Kimchi Duruchigi Supplemented with Rubus coreanus Miquel Extract (복분자 추출물을 포함하는 김치 두루치기의 저온 저장 특성)

  • The goal of this study was done for evaluating stability according to low-temperature storage of kimchi duruchigi supplemented with a hot water extract of Rubus coreanus Miquel (RCM). Total polyphenol and flavonoid contents in the RCM extract prepared from the hot water were detected by 293.34 μg CA/mg and 90.57 μg quecetin/mg, respectively. DPPH and superoxide radical scavenging activities of the extract were showed by relatively high values of 70.63 and 57.87%, respectively. Kimchi duruchigi was designed by control (non-treated), T1 (3% RCM extract), T2 (6% RCM extract), and T3 (0.1% ascorbic acid, a positive control). When compared with control and T3 groups, pHs of T1 and T2 groups supplemented with the RCM extract were gently changed depending on the storage time, and water holding capacities of T1 and T2 groups were improved in comparison with control group. Although meat color showed a tendency to most of increase according to the elapsed time, T1 and T2 groups showed less changes than that of control group. Lipid peroxidation appeared in a little bit changes regardless of the processing and storage days, but protein spoilages in T1 and T2 groups were found by lower changes when compared with the control group. As the results of sensory evaluation, T1 and T2 groups during storage had the better taste, flavor and acceptability than those of control and T3 groups. Therefore, we suggest that kimchi duruchigi supplemented with the RCM extract is a possible of improving the storage stability and product preference.

Influence of Dietary Zinc, Copper and Cadmium Levels on Rat Liver Aryl Sulfotransferase IV Activity

  • Aryl sulfotransferase (AST) IV is a liver enzyme involved in detoxication and has been shown to be susceptible to down regulation by a number of hepatotoxic xenobiotics. Studies presented here to investigate the ability of biological and non-biological divalent metal cations on AST IV activity showed that AST IV was strongly inhibited following in vitro or in vivo exposure to. Zn ( II ), Co ( II ) or Cd ( II ). It was found that $0.025\sim$2.5 uM of these metal ions were sufficient to cause $50\%$ of inhibition in vitro in purified AST IV and $0.25\sim$25 uM of these metal ions in liver cytosolic fractions. For the in vivo study, 1,000 mg Cu ( II )/kg, 2,000 mg Zn ( II )/kg or 250 mg Cd( II )/kg was added to individual diets and administered to three (3) group; of mts over a 7 week period The Co ( II )-supplemented diet produced no apparent change in rat growth rate and resulted in 30-fold increase in liver cytotolic Cu ( II ) levels, suggesting that elevated levels of Cu ( II ) ion in the liver were responsible for the loss of AST IV activity. In contrast, the Zn ( II )-supplemented diet caused a decrease in rat growth rates and resulted in zero increase in liver Zn ( II ) levels, which suggested an indirect inhibition mechanism was caused by Zn ( II ) in the liver. Rats were fed the Cd-supplemented diet also displayed a decrease in growth rate with little or no change in liver Cu ( II ) or Zn ( II ) levels. When the liver cytosols of mts from the metal ion diets were immunochemically analyzed for the AST IV and albumin contents, no significant changes were observed in albumin levels. However, AST IV contents in the cytosols of mts fed the Zn ( II )-supplemented diets showed a slight decrease in amount These results showed that AST IV activity in vitro and in vivo can be inhibited by Co ( II ), Zn ( II ), and Cd ( II ) by apparently different mechanisms. The immediate response to a Zn injection showed a decrease in AST IV activity but not in the AST IV content in liver cytosol. These mechanisms appeared to involve direct actions of the metal ion on AST IV activity and indirect actions affecting AST IV amount.

Studies on the detrimental factors affecting in vitro maturation and fertilization of bovine follicular oocytes (소 난포란(卵胞卵)의 체외성숙(體外成熟) 및 체외수정(體外受精)에 영향(影響)을 미치는 요인(要因)에 관한 연구(硏究))

  • These studies were carried out to investigate the effects of the size of follicles, semen types, capacitation methods, and additions of hormones, estrous cow serum(ECS), fetal calf serum(FCS), bovine follicular fluid(BFF) and matured cumulus cell(MCC) to the medium on in vitro maturation and fertilization of bovine follicular oocytes. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluid from the visible follicles of diameter 3~5mm. The follicular oocytes were cultured in TCM-199 medium containing hormones, FCS, ECS, BFF and MCC for 24~48hrs. in an incubator with 5% $CO_2$ in air at $38.5^{\circ}C$ and then matured oocytes were again cultured for 18~20 hrs. with motile capacitated spermatozoas the TCF (Tyrode calcium free) solution containing $100{\mu}g/ml$ of heparin. The results obtained in these experiments were summarized as follows: 1. The oocytes classified as "A,B,C,D and Degenerative" depending morphological integrity and those 61.4%, 12.1%, 19.2%, 4.2% and 3.0% of the total oocytes recovered, respectively. The maturation and fertilization rate of the A, B, C class follicular oocytes, cultured in the TCM-199 medium supplemented with 10% FCS were 89.1%, 78.0%, 52.6% and 78.1%, 66.1, 33.3%, respectively. 2. The average number of the follicular oocytes recovered from follicles size, 1~2mm, 3~5mm and above 5mm in diameter were 67, 98 and 63, respectively. The maturation and fertilization rate of the follicular oocytes, cultured in the TCM-199 medium were 56.7%, 82.5%, 46.0% and 44.8%, 71.4%, 28.6%, respectively. 3. The fertilization and cleavage rate of the follicular oocytes, inseminated with spermatozoas of epididymal cauda, neat and frozen semen were 63.3%, 73.3%, 70.0% and 32.7%, 37.8% 38.3%, respectively. 4. The fertilization and cleavage rate of follicular oocytes, fertilized with capacitated spermatozoas by heparin, BFF and HIS methods were 70.0%, 53.8%, 34.2% and 38.3%, 23.1%. 17.1%, respectively. And the fertilization and cleavage rate were higher method of heparin. 5. The maturation and fertilization rate of follicular oocytes, cultured in the TCM-199 medium supplemented with 5%, 10%, 15%, 20% and FSH, HCG, 17, $\beta$-estradiol were 76.0~82.3% and 26.2~70.0%, and those values were higher the supplementation than non-supplementation. 6. The maturation and fertilization rate of the follicular oocytes cultured in TCM-199 medium supplemented with 5~20% ECS and FCS were 74.0~80.6%, 26.2~30.0% and 71.7~76.9%, 51.9~58.0%, and the values were higher the supplement of ECS than FCS. 7. The maturation rate (68.0~64.6%) and fertilization rate(59.6~60.4%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and 20~30% BFF were higher than those of follicular oocytes cultured TCM-199 medium supplemented with 10% FCS and 10% and 50% BFF. 8. The maturation rate(76.5%) and fertilization rate (61.7%) of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and $1{\times}10^6/ml$ MCC were higher than those of follicular oocytes cultured in TCM-199 medium supplemented with 10% FCS and $1{\times}10^{{4}{\sim}{5}}/ml$ and $1{\times}10^8/ml$ cumulus cells.

Mechanism of Heat-Libile E. coli Enterotoxin Production (대장균의 이열성장독소 생산기전)

  • Enterotoxigenk E. coli is one of the major causative agents of the infantile diarrhea and traveler's diarrhea. The heat-labile enterotoxin is thought to be a virulence factor in the pathogenesis of the diarrhea and to be a marker for identification of the enterotoxigenic E. coli from non pathogenic E. coli. Therefore knowledge about the heat-labile enterotoxin is essential not only for understanding the pathogenesis but also for the diagnosis of the diarrhea. However the in-vitro heat-labile enterotoxin production is reported to be greatly affected by the cultural condition. In this regards, this study was designed to know the optimal conditions for the production of the heat-labile enterotoxin by assaying the permeability factor in the 18 hours culture supernatant of E. coli 08K25(B2) H9 and of E. coli 015 H11. Results obtained were summerized as follows: 1. Amounts of heat-labile enterotoxin produced were greater at initial pH 8.5 than at 7.0 of CYES-2 broth culture. However, the bacterial growth itself was more abundant at 7.0 than at 8.5. 2. Heat-labile enterotoxin per unit volume of culture supernatant was greater at shaking culture than at standing culture condition, but ratio of the enterotoxin produced over the unit mass of E. coli calculated was greater at standing culture than shaking culture condition, indicating that the greater yields of the toxin produced at shaking culture was due to increase in E. coli cell mass compared to the standing culture condition: 3. The enterotoxin produced in the lincomycin(128 microgram/ml) supplemented media was 5 or 11 times greater on the basis of enterotoxin per unit mass of E. coli, compared to the lincomycin-non-supplemented media, indicating that lincomycin itself increases the enterotoxin production. 4. Treatment of 18 hours culture of E. coli with polymyxin B(0.2 mg/ml) for 1 hour increased the yields of enterotoxin amounting to 2 or 5 times of the non-treated control cultures.
