Aspergillus nidulans내에서 Aspergillus awamori의 Glucoamylase 유전자 발현 (Expression of Aspergillus awamori Glucoamylase Gene in Asperillus nidulans)

  • A awamori 의 glucoamylase 유전자와 A. nidolans 의 trpC maker 유전자를 가진 A. nidulans 의 expression vector 를 만들었다. 이재조합 plasmid 를 트립토판 영양요구주의인 A. nidulans B17 에 형질전환시켰다. Southern blotting 으로 vector DNA 가 A. nidulans 의 chromosomal DNA 에 integration 된 것을 확인하였다. Northern blotting 의 결과, glucoamylase 유전자는 induction 조건에서 mRNA 를 합성하였다. glucoamylase 의 역가가 형질전환체에서 증가하였으며, nondenaturing polyacrylamide gel 에서 glucoamylase 의 활성이 나타났다.

First report of the branchiobdellidan Holtodrilus truncatus (Annelida: Clitellata) found on the freshwater atyid shrimp Neocaridina sp. from Korea

  • The branchiobdellidan species Holtodrilus truncatus (Liang, 1963) is reported for the first time in Korea. The genus Holtodrilus Gelder and Brinkhurst, 1990 is monotypic within the family Xironodrilidae, and the genus and family of this species are also new to the Korean fauna. The branchiobdellidan specimens were collected from the exoskeletons of the freshwater atyid shrimp, Neocaridina sp., sampled from the 'Andeok Valley', Jeju-do Province, South Korea. Holtodrilus truncatus is characterized by its terete body form, 7/7 dental formula (with a large medial tooth and three pairs of smaller lateral teeth), and no trunk appendages. In addition, we determined a partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene as a DNA barcode marker for H. truncatus. The present study is the first record of the branchiobdellidan occurrence on a non-crayfish host (shrimp) in Korea. To date, seven species (four genera in three families) of branchiobdellidans have been reported in the Korean fauna.

An overview of the endocrine functions of osteocalcin

  • Osteocalcin is the most abundant non-collagenous protein produced in bone. It has traditionally been regarded as a marker of bone turnover and is thought to act in the bone matrix to regulate mineralization. However, emerging knowledge regarding osteocalcin has expanded to include functions in energy metabolism, fertilization, and regulation of cognition. Fully carboxylated osteocalcin binds to hydroxyapatite, thereby modulating bone turnover, whereas undercarboxylated osteocalcin in the circulation binds to osteocalcin-sensing receptors and acts as a hormone that affects multiple physiological aspects. In this review, we summarize the current knowledge regarding the hormonal actions of osteocalcin in various organs and potential cellular downstream signaling pathway that may be involved.

보행 중 인체 슬관절의 3차원 접촉 모델 개발 (Development of Three-Dimensional Contact Model of Human Knee Joint During Locomotion)

  • The human knee joint is the intermediate joint of the lower limb that is the largest and most complex joint in the body. Understanding of joint-articulating surface motion is essential for the joint wear, stability, mobility, degeneration, determination of proper diagnosis and so on. However, many studies analyzed the passive motion of the lower limb because of the skin marker artefact and some studies described medial and lateral condyle of a femur as a simple sphere due to the complexity of geometry. Thus, in this paper, we constructed a three-dimensional geometric model of the human knee from the geometry of its anatomical structures using non-uniform B-spline surface fitting as a study for the kinematic analysis of more realistic human knee model. In addition, we developed and verified 6-DOF contact model of the human knee joint using $C^2$ continuous surface of the inferior region of a femur, considering the relative motion of shank to thigh during locomotion.

Enhancement of UV-induced nucleotide excision repair activity upon forskolin treatment is cell growth-dependent

  • Forskolin (FSK), an adenylyl cyclase activator, has recently been shown to enhance nucleotide excision repair (NER) upon UV exposure. However, our study revealed that this effect was detected in human skin epithelial ARPE19 cells only in growing cells, but not in non-cycling cells. When the cells were grown at low density (70% confluence), FSK was capable of stimulating cAMP responsive element binding (CREB) phosphorylation, a marker for FSK-stimulated PKA activation, and resulted in a significant increase of NER activity compared to control treatment. However, cells grown under 100% confluent conditions showed neither FSK-induced CREB phosphorylation nor the resulting NER enhancement. These findings indicate that cellular growth is critical for FSK-induced NER enhancement and suggest that cellular growth conditions should be considered as a variable while evaluating a reagent's pharmacotherapeutic efficacy.

3차원 미니밴 형상 주위의 비압축성 점성 유동 해석 (Incompressible Viscous Flow Analysis Around a Three Dimensional Minivan-Like Body)

  • The flow field around a three dimensional minivan-like body has been simulated. This study solves 3-D unsteady incompressible Navier-Stokes equations on a non-orthogonal curvilinear coordinate system using second-order accurate schemes for the time derivatives, and third/second-order scheme for the spatial derivatives. The Marker-and-Cell concept is applied to efficiently solve continuity equation. The fourth -order artificial damping is added to the continuity equation for numerical stability. A H-H type multi-block grid system is generated around a three dimensional minivan-like body. Turbulent flows have been modeled by the Baldwin-Lomax turbulent model. The simulation shows three dimensional vortex-pair just behind body. And the flow separation is also observed the rear of the body. It has concluded that the results of present study properly agree with physical flow phenomena.

  • This paper describes analysis of complex flow around Car-like body using Chimera grid technique. As a computational algorithm, Pullboat and Chaussee's Diagonal algorithm is selected to reduce computational time. Introducing hole points flag to this Diagonal algorithm, an algorithm for Chimera grid is generated easily.   • Nowadays, terms such as 'Smart Textile', 'Intelligent Textile' and 'Wearable Computing' are commonly used in everyday contexts. And radio-frequency identification (RFID) is the use of a wireless non-contact system that uses radio-frequency electromagnetic fields to transfer data from a tag attached to an object, for the purposes of automatic identification and tracking. These products are required technologies which are textile treatments, printing, ink, etc. Durability of textile substrates is an essential marker for conductive ink printing process. Especially, heat stability is important, since conductive ink should be processable (annealing, curing) at temperatures below $150^{\circ}C$. This study was application of RFID on textiles. The textile pre-treatment processes should be carried out to use RFID antenna on textiles.

Improved Transformation of the Filamentous Fungus Aspergillus niger Using Agrobacterium tumefaciens

  • Since it is known that Agrobacterium tumefaciens, which has long been used to transform plants, can transfer the T-DNA to yeast Saccharomyces cerevisiae during tumourigenesis, a variety of fungi were subjected to transformation to improve their transformation frequency. In this study, I report the A. tumefaciens-mediated transformation of filamentous fungus Aspergillus niger. Transfer of the binary vector pBIN9-Hg, containing the bacterial hygromycin B phosphotransferase gene under the control of the Aspergillus nidulans trpC promoter and terminator as a selectable marker, led to the selection of $50{\sim}100$ hygromycin B-resistant transformants per $1{\times}10^7$ conidia of A. niger. This efficiency is improved $10{\sim}20$ fold more than reported elsewhere. In order to avoid the difficulties in selection transformant from the over-growing non-transformant, I used top agar containing 900 ${\mu}g/ml$ of hygromycin. Genomic PCR and Southern analysis showed that all transformants contained single T-DNA insert per fungal genome. This technique offers an easier and more efficient method than that of using protoplast.

A Database of Gene Expression Profiles of Korean Cancer Genome

  • Because there are clear molecular differences entailing different treatment effectiveness between Korean and non-Korean cancer patients, identifying distinct molecular characteristics of Korean cancers is profoundly important. Here, we report a web-based data repository, namely Korean Cancer Genome Database (KCGD), for searching gene signatures associated with Korean cancer patients. Currently, a total of 1,403 cancer genomics data were collected, processed and stored in our repository, an ever-growing database. We incorporated most widely used statistical survival analysis methods including the Cox proportional hazard model, log-rank test and Kaplan-Meier plot to provide instant significance estimation for searched molecules. As an initial repository with the aim of Korean-specific marker detection, KCGD would be a promising web application for users without bioinformatics expertise to identify significant factors associated with cancer in Korean.
