EFFECTS OF DIETARY LEVELS OF CHROMIUM PICOLINATE ON GROWTH PERFORMANCE, CARCASS QUALITY AND SERUM TRAITS IN BROILER CHICKS

  • An experiment was conducted to evaluate the effects of dietary levels of chromium in the form of chromium picolinate on growth performance, nutrient utilizability, carcass composition, serum traits, and in vitro lipolysis and lipogenesis in adipose tissues of Arbor Acre broiler chicks. Experimental diets containing six different levels of chromium (0, 100, 200, 400, 600 and 800 ppb) were fed for 6 weeks. Individual treatment had six replicates of eight birds each and their average initial weight was 59.2 g. Dietary addition of chromium did not affect growth performance and nutrient utilizability. However, mortality appeared to be reduced with addition of chromium to the diet. It was obvious that chromium supplementation significantly decreased serum cholesterol and increased serum HDL cholesterol (p < 0.05), but serum insulin, glucose, triglyceride and non-esterified fatty acid concentrations were inconsistent among dietary supplementation levels of chromium. The in vitro lipolysis and lipogenesis in adipose tissues were significantly influenced by dietary addition of chromium (p < 0.05). Chicks fed diets containing 200 or 400 ppb chromium showed the highest protein content and the lowest fat content in their carcass.

Study on Changes in Racehorses' Metabolites and Exercise-related Hormones before and after a Race

  • Physiological changes in thoroughbred racehorses during the race were investigated by measuring concentrations of metabolites and exercise-related hormones before and after a race. The conversion point from anaerobic to aerobic exercise during the race was estimated subsequently. Blood samples were taken from the jugular vein of 53 thoroughbreds at different times -three h before and 45 min after- for measuring the concentrations of glucose, non-esterified fatty acids (NEFA), lactate, uric acid, ammonia, insulin, adrenocorticotrophin (ACTH) and cortisol according to the race distance. In accordance with the race distance, each metabolite increased in concentration compared with the level before the race. The level of glucose, in particular, increased from $56.18{\pm}3.20$ mg/dl before the race to $148.82{\pm}8.82$ mg/dl after the race for horses that raced 1,400 m, showing a significant increase of 165% (p<0.001). The concentration of NEFA rose from $76.77{\pm}5.59$ uEq/L to $335.85{\pm}35.39$ uEq/L, up 337% (p<0.01) after a 1,400 m race. Exercise-related hormones also showed similar changes. The level of insulin dropped the most in horses that raced 1,400 m, by 42%, from $0.97{\pm}0.18$ to $0.56{\pm}0.05\;{\mu}g/L$ (p<0.5); however, ACTH and cortisol jumped significantly at 1,800 m, from $20.17{\pm}2.12$ to $551.45{\pm}91.33$ pg/ml (p<0.5) and $1.13{\pm}0.16$ to $5.66{\pm}0.45\;{\mu}g/dl$ (p<0.01), respectively, representing the highest increase. Therefore, based on the changes in glucose, NEFA and insulin levels before and after the race, it was concluded that the race distance of 1,400 m represents the point where racehorses make a conversion from anaerobic to aerobic exercise.

Changes in Maternal Blood Glucose and Plasma Non-Esterified Fatty Acid during Pregnancy and around Parturition in Twin and Single Fetus Bearing Crossbred Goats

  • The effects of fetal number (single or twin) on blood glucose and plasma NEFA during pregnancy and around parturition were studied on ten Alpine ${\times}$ Beetal crossbred goats in their first to third lactation. The animals were divided in-groups 1(carrying single fetus, n=4) and 2(twin fetus, n=6). The samples were drawn on day1 after estrus and then at 14 days interval (fortnight) for 10 fortnights. Around parturition the samples were taken on days -20, -15, -10, -5, -4, -3, -2, -1 prior to kidding and on day 0 and +1, +2, +3, +4, +5, +10, +15, +20 days post kidding. In twin bearing goats the blood glucose concentration continued to increase from 1st until 4th fortnight and thereafter gradually decline from 5th upto 8th fortnight. In single bearing goats there was increase in levels from 2nd upto 4th fortnight and thereafter it declined from 5th uptill 9th fortnight. The difference in sampling interval was highly significant (p<0.01) in both the groups. However the values were higher in single than in twin bearing goats. The plasma NEFA concentration was low in both the groups' upto 4th fortnight and thereafter it is continuously increased upto 9th fortnight. During prepartum period the blood glucose was higher in single than in twin bearing goats. The values were minimum on the day of kidding in both the groups. During postpartum period the values were significantly (p<0.01) higher in twin than in single fetus bearing goats. The plasma NEFA was significantly (p<0.05) higher in twin than in single fetus bearing goats. The blood glucose and plasma NEFA concentration can be used as index of nutritional status during pregnancy and around parturition in goats.

Alterations in Lipid Metabolism between the Growing and the Laying Periods of White Leghorn Layers

  • This study compared the lipid metabolism of white Leghorn layers between the growing and the laying periods. The study was conducted on thirty layers in the growing period (14 weeks old) and in the initial laying period (36 weeks old; 14 weeks from the onset of laying). Results indicated that all plasma lipid traits were significantly different (p<0.05) between the two periods. The estrogen concentration in the laying period was about 3 fold that in the growing period. Triacylglycerol (TG) concentration in the laying period was about 12 fold that in the growing period. The phospholipids, cholesterol, glycerol and non esterified fatty acid (NEFA) in the laying period were also higher than those in the growing period (p<0.05). Lipogenic enzyme activities in the laying period were higher than in the growing period (p<0.05). High-density lipoprotein (HDL) was the largest lipoprotein portion in growing layers. In laying hens, the major lipoprotein portion was very low-density lipoprotein (VLDL). It was also shown that protein is the primary component of HDL and TG is the largest portion of VLDL in both periods. In laying hens, VLDL contained more TG than that in growing layers (p<0.05). The VLDL particle size in laying hens was larger than that in growing layers (p<0.05). This study indicates that the lipid metabolism traits of a layer in the laying period were different from those in the growing period. The lipogenesis related traits in the laying period were markedly greater than in the growing period.

Effects of Chromium Picolinate (CrP) on Growth Performance, Carcass Characteristics and Serum Traits in Growing-Finishing Pigs

  • An experiment was carried out to evaluate the effects of feeding graded levels of chromium in the form of chromium picolinate on growth performance, blood components, carcass grade, in vitro lipogenesis and lipolysis, and in vitro protein degradation and synthesis in growing-finishing pigs. There were no significant differences for daily weight gain, feed intake and feed conversion among treatments during growing phase, while in the finishing phase, feed intake was lower in groups fed diets with 200 ppb chromium than in other treatment (p < 0.1). Feed conversion was improved in the groups fed diets with chromium compared with control. Carcass weight was similar among treatments while carcass length was longer in groups fed diets with 200 ppb chromium (p < 0.05). Thinner carcass fat was found with groups fed diets with chromium compared to control. Three A grade of carcasses were from groups fed chromium compared to control. No significant differences were observed with blood glucose, insulin, total cholesterol, triglycerides and non-esterified fatty acid at 60kg body weight. While, at 100 kg body weight, blood triglyceride was lower in groups with 200 and 400 ppb chromium but higher in groups with 100 ppb chromium (p < 0.05). In vitro lipolysis and protein synthesis in adipose tissues were increased as dietary chromium was increased from 0 to 200 ppb (p < 0.1). As a result, 200 ppb chromium in a growing-finishing diet could improve feed efficiency and carcass traits; an increase to 400 ppb has no further effect.

Pectinase-Processed Ginseng Radix (GINST) Ameliorates Hyperglycemia and Hyperlipidemia in High Fat Diet-Fed ICR Mice

  • To develop a ginseng product possessing an efficacy for diabetes, ginseng radix ethanol extract was treated with pectinase and obtained the GINST. In the present study, we evaluate the beneficial effect of GINST on high fat diet (HFD)-induced hyperglycemia and hyperlipidemia and action mechanism(s) in ICR mice. The mice were randomly divided into five groups: regular diet group (RD), high fat diet group (HFD), HFD plus GINST at 75 mg/kg (GINST75), 150 mg/kg (GINST150), and 300 mg/kg (GINST300). Oral glucose tolerance test reveals that GINST improves the glucose tolerance after glucose challenge. Fasting plasma glucose and insulin levels were decreased by 4.3% and 4.2% in GINST75, 10.9% and 20.0% in GINST150, and 19.6% and 20.9% in GINST300 compared to those in HFD control group. Insulin resistance indices were also markedly decreased by 8.2% in GINST75, 28.7% in GINST150, and 36.4% in GINST300, compared to the HFD control group. Plasma triglyceride, total cholesterol and non-esterified fatty acid levels in the GINST300 group were decreased by 13.5%, 22.7% and 24.1%, respectively, compared to those in HFD control group. Enlarged adipocytes of HFD control group were markedly decreased in GINST-treated groups, and shrunken islets of HFD control mice were brought back to near normal shape in GINST300 group. Furthermore, GINST enhanced phosphorylation of AMP-activated protein kinase (AMPK) and glucose transporter 4 (GLUT4). In summary, GINST prevents HFD-induced hyperglycemia and hyperlipidemia through reducing insulin resistance via activating AMPK-GLUT4 pathways, and could be a potential therapeutic agent for type 2 diabetes.

전사체 프로파일을 이용한 고려 홍삼의 항당뇨 기전 연구 (Anti-diabetic Mechannism Study of Korean Red Ginseng by Transcriptomics)

  • This study was designed to investigate the anti-diabetic effect and mechanism of Korean red ginseng extract through transcriptomics in C57BL/KsJ db/db mice. The db/db mice were randomly divided into six groups: diabetic control group (DC), red ginseng extract low dose group (RGL, 100 mg/kg), red ginseng extract high dose group (RGH, 200 mg/kg), metformin group (MET, 300 mg/kg), glipizide group (GPZ, 15 mg/kg) and pioglitazone group (PIO, 30 mg/kg), and treated with drugs once per day for 10 weeks. At the end of treatment, we measured blood glucose, insulin, hemoglobin A1c (HbA1c), triglyceride (TG), adiponectin, leptin, non-esterified fatty acid (NEFA). RGL-treated group lowered the blood glucose and HbA1c levels by 19.6% and 11.4% compared to those in diabetic control group. In addition, plasma adiponectin and leptin levels in RGL-treated groups were increased by 20% and 12%, respectively, compared to those in diabetic control. Morphological analyses of liver, pancreas and epidydimal adipose tissue were done by hematoxylin-eosin staining, and pancreatic islet insulin and glucagon levels were detected by double-immunofluorescence staining. RGL-treated group revealed higher insulin contents and lower glucagon contents compared to diabetic control. To elucidate an action mechanism of Korean red ginseng, DNA microarray analyses were performed in liver and fat tissues, and western blot and RT-PCR were conducted in liver for validation. According to hierarchical clustering and principal component analysis of gene expression Korean red ginseng treated groups were close to metformin treated group. In summary, Korean red ginseng lowered the blood glucose level through protecting destruction of islet cells and shifting glucose metabolism from hepatic glucose production to glucose utilization and improving insulin sensitivity through enhancing plasma adiponectin and leptin levels.

Streptozotocin 유발 당뇨병성 쥐에대한 $Ginsenoside-Rb_2$의 연속투여 효과 (Effect of Serial Administration of $Ginsenoside-Rb_2$ on Streptozotocin-diabetic Rats)

  • Streptozotocin으로 유발시킨 당뇨병성 쥐(혈당수준$430{\pm}30ml/dl$)에게 ginsenoside-$Rb_{2}$를 연속적으로 복강투여 하였다. Ginsenoside $Rb_{2}$ 투여군은 현저한 혈당감소를 보였으며 이군들의 혈당수준은 6-9일 투여로 약 160mg/dl로 떨어졌다. 한편 대조군보다 먹이섭취량이 적지만 체중은 ginsenoside-$Rb_{2}$를 투여한 당뇨쥐에서 상당히 증가하였다. Ginsenoside-$Rb_{2}$투여군은 과식, 다뇨, 당뇨 등과 같은 당뇨병 증상이 개선 되었다. 혈당을 떨어뜨리는 작용기전을 연구하기위한 일환으로 간 및 지방조직 그리고 혈청 등에 있는 탄수화물 대사물질과 지질성분 등에 미치는 ginsenoside-$Rb_{2}$의 효과를 조사하였다. 당뇨쥐에서는 ginsenoside-$Rb_{2}$가 먼저 정상쥐에서와 같이 간에서 혈당의 이용을 촉진 시키고 그결과 지방조직에 triglyceride 들이 증가되는 것으로 추측된다.

Risk Factors for Displacement of the Abomasum in Dairy Cows and its Relationship with Postpartum Disorders, Milk Yield, and Reproductive Performance

  • We determined the risk factors for displacement of the abomasum (DA), and the relationships between DA and postpartum disorders, milk yield, and reproductive performance in dairy cows. Initially, we identified the risk factors for DA using data regarding cow health and calving season from 2,208 lactations. Then, we compared the incidence of postpartum disorders, culling, death, and reproductive performance between cows with DA and their control herdmates (each n = 57). In addition, serum metabolites concentrations and milk yield were compared between cows with DA and controls (each n = 33). Ketosis (odds ratio [OR] = 9.27, p < 0.0001) and twin calves (p = 0.06) increased the risk of DA. Cows with a parity of three had a higher risk (OR = 5.23, p < 0.01) of DA than primiparous cows. Serum total cholesterol concentration was lower but non-esterified fatty acid, ${\beta}-hydroxybutyrate$, and alanine aminotransferase concentrations were higher after calving in cows with DA than in controls (p < 0.05). The removal rate from the herd by 2 months after calving was higher (p < 0.05) but milk yield 1 and 2 months after calving (p < 0.01) and the rate of first insemination by 150 days postpartum were lower (hazard ratio = 0.49, p < 0.05) in cows with DA than controls. In conclusion, higher parity, twin calves, and ketosis are risk factors for DA in dairy cows, which is associated with a higher removal rate from the herd, lower milk yield, a longer calving to first insemination interval, and unfavorable levels of metabolites related to energy and liver function.

Nutritional condition in the dry period is related to the incidence of postpartum subclinical endometritis in dairy cattle

  • Objective: Endometritis is a major disease, that causes infertility in cattle, and is usually categorized as clinical or subclinical endometritis (SCE). The nutritional condition during the dry period is important for recovery after the last stage of the lactation period, and for postpartum production and reproduction. This study aimed to clarify the relationship between nutritional and metabolic characteristics in the dry period, and the risk of postpartum SCE. Methods: Multiparous Holstein dairy cows (n = 25, raised in a tied stall) were used. Endometrial cytological analysis was performed around 30 days post-partum, with 5% to 14% polymorphonuclear (PMN) as a cut-off point to define SCE. Serum levels of glucose, non-esterified fatty acids, β-hydroxybutyric acid (BHBA), blood urea nitrogen, total cholesterol, aspartate aminotransferase, γ-glutamyl transpeptidase, calcium, phosphorus, and magnesium were measured in the cows at the dry period to evaluate energy status, protein metabolism, and mineral metabolism. Results: The incidence of SCE in the cows was 60.0% (n = 15/25) and the mean PMN% in postpartum cows diagnosed as SCE was 8.05%±2.6%. Overall, 17 and 8 samples were collected from the cows in the far-off and close-up periods, respectively. The serum concentration of BHBA in the far-off period and serum glucose concentration in the closeup period were correlated with postpartum PMN% (r = 0.62, p<0.01; r = -0.74, p<0.05, respectively). Serum levels of calcium and magnesium in the dry period were associated with the incidence of postpartum SCE (healthy vs SCE cows, p<0.05). Conclusion: Blood levels of glucose, BHBA, calcium, and magnesium in dry periods could be useful parameters for predicting the risk of postpartum SCE. The present study also suggests that management in the close-up period is essential for promoting recovery from calving fatigue.
