Glycoproteins PAS-6(50 kDa) and -7(47 kDa) from the bovine milk fat globule membrane share a common protein core but differ in their carbohydrate moiety. We have analyzed and proposed the structures of the N-linked sugar chains of PAS-7 by Oxford Glyco System(OGS) RAAM2000. The N-linked sugar chains were liberated from PAS-7 by hydrazinolysis and, after modifying the reducing ends with 2-aminobenzamide(2-AB), were separated into one neutral(7N, 55%) and two acidic(7M, mono-, 43%; 7D, di-, 2%) sugar chain groups. 7N was finally separated into 5 chains(a, b, c, d, and e), respectively. The structure of this 2AB-neutral sugar chain was determined by sugar analysis, exoglycosidase digestion with OGS glycosidase Kit and OGS RAAM2000 system. The results show that fraction e was the same of reported 7N1A, the biantennary complex type with a fucose on reducing end and two N-acetyllactosamine branch on non-reducing end. Therefore, it was proved that OGS RAAM2000 method is in conformity with conventional analysis of sugar chain structure from bovine PAS-7.
Radiotheraphy may be considered as one of the most effective treatments for digestive tumors. This procedure has major side effects, especially in fast growing tissues like intestinal mucosa. For this reason, the morphological changes in mouse jejunal villi after whole body ${\gamma}-irradiation$ were analysed. Four week old ICR male mice were irradiatied with 6.5Gy or 12Gy of whole body ${\gamma}-irradiation$ and were sacrificed 3 days later. Jejunum among intestine were taken for morphology. Samples were carried out dehydration process and sliced sample covered with paraffin was hold on the slide and then light microscopy was observed by Hematoxylin & Eosin staining. villi at both irradiated doses were showed that the length of villi were shortened and thickened and that lumen were expanded in comparison with non-irradiated group. Since willi have an important role in digestion and very sensitive to radiation from this test, it has a role of test parameter for finding radioprotectors as well as evaluating the biological effect by radiaton.
Twelve dietary combinations were prepared using 70 parts of fermented wheat straw (FWS) as the sole roughage supplemented with 30 parts of either the low protein concentrate mixture (Conc.-I), high protein concentrate mixture (conc.-II), maize grains (M), solvent extracted mustard cake (DMC), deoiled rice bran (DRB), uromol bran mixture (UBM), deep stacked poultry litter (DSPL), dried poultry droppings (DPD), M-DMC mixture (50:50), M-UBM mixture (50:50), M-DPD mixture (50:50) or M-UBM-DPD mixture (50:25:25) and evaluated by in-sacco technique. The above dietary combinations were also evaluated by changing the roughage to concentrate ratio to 60:40. The digestion kinetics for DM and CP revealed that FWS:DPD had the highest, whereas, the FWS:M-DMC had the lowest rapidly soluble fraction. The potentially degradable fraction was found to be maximum in FWS:M and minimum in FWS:DPD dietary combinations. The higher degradation rate of FWS:DRB and FWS:UBM combinations was responsible for their significantly (p<0.05) higher effective degradability as compared to other combinations. The highest undegradable fraction noted in FWS:M-UBM-DPD followed by FWS:DMC was responsible for high rumen fill values. The FWS:DRB, FWS:UBM and FWS:DPD combinations had higher potential for DM intake. The dietary combination with higher concentrate level (60:40) was responsible for higher potentially degradable fraction, which was degraded at a faster rate resulting in significantly higher effective degradability as compared to the corresponding dietary combination with low concentrate level (70:30). The low undegradable fraction in the high concentrate diet was responsible for low rumen fill values, which predicted of high potential for DM intake. Out of 24 dietary combinations, FWS with either of UBM, DRB, DMC, Maize, M-DMC or DPD in 70:30 ratio supplemented with minerals and vitamin A in comparison to conventional feeding practice (roughage and concentrate mixture) could be exploited as complete feed for different categories of ruminants.

A series of in vitro and in vivo experiments were conducted to investigate the effects of the mixture of Tween 80 and cellulolytic enzymes (xylanase and cellulase) on total tract nutrient digestibility and rumen cellulolytic bacterial adhesion rates in Holstein steers. Ground timothy hay sprayed with various levels of Tween 80 and cellulolytic enzymes was used as substrates in an in vitro experiment to find out the best combinations for DM degradation. The application level of 2.5% (v/w) Tween 80 and the combination of 5 U xylanase and 2.5 U cellulase per gram of ground timothy hay (DM basis) resulted in the highest in vitro dry matter degradation rate (p<0.05). Feeding the same timothy hay to Holstein steers also improved in vivo nutrient (DM, CP, CF, NDF and ADF) digesibilities compared to non-treated hay (p<0.05). Moreover, Tween 80 and enzyme combination treatment increased total ruminal VFA and concentrations of propionic acid and isovaleric acid with decreased acetate to propionate ratio (p<0.001). However, adhesion rates of Fibrobacter succinogenes and Ruminococcus flavefaciens determined by Real Time PCR were not influenced by the treatment while that of Ruminococcus albus was decreased (p<0.05). The present results indicate that a mixture of Tween 80 and cellulolytic enzymes can improve rumen environment and feed digestibility with variable influence on cellulolytic bacterial adhesion on feed.
Background: Matrix metalloproteinases (MMPs) are linked with several complications such as metastasis of cancer progression, oxidative stress, and hepatic fibrosis. Brown seaweeds are being extensively studied for their bioactive molecule content against cancer progression. In this context, Sargassum horneri was reported to possess various bioactivities including antiviral, antimicrobial, and anti-inflammatory partly due to its phenolic compound content. Methods: In this study, potential of S. horneri was evaluated through anti-MMP effect in HT1080 fibrosarcoma cells. S. horneri crude extract was fractionated with organic solvents, namely, water ($H_2O$), n-buthanol (n-BuOH), 85% aqueous methanol (85% aq. MeOH), and n-hexane. The non-toxicity of fraction samples (Sargassum horneri solvent-partitioned extracts (SHEs)) was confirmed by cell-viability assay. SHEs were tested for their ability to inhibit MMP enzymatic activity through gelatin digestion evaluation and cell migration assay. Expressions of MMP-2 and MMP-9 and tissue inhibitors of MMP (TIMPs) were evaluated by reverse transcription and Western blotting. Results: All fractions inhibited the enzymatic activities of MMP-2 and MMP-9 according to gelatin zymography. Except $H_2O$ fraction, fractions hindered the cell migration significantly. All tested fractions suppressed both mRNA and protein levels of MMP-2, MMP-9, TIMP-1, and TIMP-2. Conclusion: Overall, current results suggested that S. horneri has potential to be a good source for anti-MMP agents, and further investigations are underway for better understanding of the action mechanism and isolation and elucidation of the bioactive molecules.
The present study investigated the effects of thermal pre-treatment on the enhancement of anaerobic biodegradability of waste activated sludge at varied TS concentration levels. The activated sludges were thermally oxidized for 30 minutes at $80{\sim}200^{\circ}C$ with varied TS concentrations (2%, 4% and 6%). and then, sludge characteristics, solubilization efficiency and methane production yield of thermally pre-treated sludges were analyzed. The higher the temperature in the thermal pre-treatment, the higher the concentration levels of dissolved matters such as $SCOD_{Cr}$, $NH_4{^+}$ and VFAs, which indicates that the thermal pre-treatment facilitates the hydrolysis and acid fermentation. Furthermore, the solubilization efficiency was increased in proportion to the temperature rise at all TS concentrations and was reached at 68.9%, 55.6% and 53.1%, respectively, at $200^{\circ}C$. In the BMP test of the pre-treated sludges, higher methane production yields were observed as 0.313. 0.314 and $0.299m^3\;CH_4/kg\;VS_{add}$ at the condition of TS 2% ($160^{\circ}C$), 4% ($160^{\circ}C$) and 6% ($180^{\circ}C$), respectively, and degradation rate was increased by 84%, 79% and 65% compared with non-pretreated waste activated sludge. These findings suggest the effectiveness of thermal pre-treatment of waste activated sludge for anaerobic biodegradable process.
This study was conducted to investigate the effects on chemical properties of soil and yield of chinese cabbage when the piggery liquid slurry (ADS) digested anaerobically in the biogas plant was sprinkled to the soils. Treatments were composed of non-nitrogen plot (PK), chemical fertilizer plot (NPK) and ADS plot, and were arranged by randomized block design with 3 replications. The ADS was an alkali (pH 8.6) fertilizer containing 0.55% of total nitrogen, optimal levels of phosphorus, potassium and other components. When based (100%) on the yield of chinese cabbage in the NPK, those of the ADS and the PK were 114% and 43%, respectively. The nitrogen absorption of chinese cabbage at 50-d after transplant was significantly greater (p<0.05) in the ADS than other treatment. Heavy metal contents of soil were not difference between before and after experiment. With supplementing phosphorus and potassium sources in it, the ADS should be an excellent fertilizer in cabbage culture.

In order to evlauate feasibility of the gene tagging by the maize transposable element Ac in heterologous plant systems, we have investigated physical distances and directions of transposition of the element in Arabidopsis thaliana and tobacco cultured cell line BY-2. We prepared a T-DNA construct that carried a non-autonomous derivative of Ac with a site for cleavage by endonuclease I-Scel (designated dAc-I-RS element). Another cleavage site was also introduced into the T-DNA region outside dAc-I-RS. A number of transgenic Arabidopsis plants were generated, each of which had a single copy of the T-DNA at a different chromosomal location. To examine the pattern of transposition, three out of these transgenic plants were crossed with the Arabidopsis plant that carried the gene for Ac transposase and progeny in which dAc-I-RS had been transposed were isolated. After digestion of the genomic DNA of these progeny with I-SceI, sizes of segment of DNA were determined byd pulse-field gel electrophoresis. We also performed linkage analysis for the transposed elements and sites of mutations near the elements. Our results with three transgenic lines showed that 50% of all transposition events had occurred within 1,700 kilo-base pairs (kb) on the same chromosome, with 35% within 200 kb, and that the elements transposed in both directions on the chromosome with roughly equal probability. The data thus indicate that the Ac-Ds system is most useful for tagging of genes that are present within 200 kb of the chromosomal site of Ac in Arabidopsis. In addition, determination of the precise localization of the transposed dAc-I-RS element should definitely assist in map-based cloning of genes around insertion sites. In the present paper, we report typical examples of such gene isolation studies.

This study investigated the effects of polyurethane foam on continuous hydrogen production from mixed wastes. Molasses was co-fermented with non-pretreated sewage sludge in a sequencing batch reactor. The results indicated that the addition of polyurethane foams as a microbial carrier in the reactor mitigated biomass loss at HRT 12 h, while most of the biomass was washed out during the operation period with no carrier. There was a stable hydrogen production rate of $0.4L-H_2/l/d$ in the carrier-sequencing batch reactor. Suspended biomass in the carrier-reactor indicated it possessed the highest specific hydrogen production rate ($241{\pm}4ml-H_2/g\;VSS/d$) when compared to that of biomass on the surface ($133{\pm}10ml-H_2/g\;VSS/d$) or inner carrier ($95{\pm}14ml-H_2/g\;VSS/d$).
