NOG1 is a nucleolar GTPase that is critical for 60S ribosome biogenesis. Recently, NOG1 was identified as one of the downstream regulators of target of rapamycin (TOR) in yeast. It is reported that TOR is involved in regulating lifespan and fat storage in Caenorhabditis elegans. Here, we show that the nog1 ortholog (T07A9.9: nog-1) in C. elegans regulates growth, development, lifespan, and fat metabolism. A green fluorescence protein (GFP) promoter assay revealed ubiquitous expression of C. elegans nog-1 from the early embryonic to the adult stage. Furthermore, the GFP-tagged NOG-1 protein is localized to the nucleus, whereas the aberrant NOG-1 protein is concentrated in the nucleolus. Functional studies of NOG-1 in C. elegans further revealed that nog-1 knockdown resulted in smaller broodsize, slower growth, increased life span, and more fat storage. Moreover, nog-1 over-expression resulted in decreased life span. Taken together, our data suggest that nog-1 in C. elegans may be an important player in regulating life span and fat storage via the insulin/IGF pathway.
This experimental study was carried out to investigate on the safety of Nog-yong aqua-acupuncture solution in primary culture of adult rat hepatocytes. Primary culture of adult rat hepatocytes has been considered as a ideal model for toxicological studies because cultured hepatocytes maintained many liver-specific functions. In this research, we investigated the effects of Nog-yong aqua-aqupunture(1-10$\mu\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release assays. Hepatic glutathione level. glutathione-S-transferase activity, and albumin synthesis were not affected by treatment with Nog-yong aqua-aqupuntur alone. Nog-yong aqua-aqupuncoure solution(0.5-$10{\mu}\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release not significantly affected normal functional charaterists.
녹문 임성주는 18세기 조선후기 철학자이다. 녹문은 호락논쟁이 리(理)와 기(氣)를 분리해 보는 데서 발생했다고 판단하고, 자신의 관점에 따라 새로운 리기론과 심성론을 정립한다. 녹문 철학의 종지는 이기동실(理氣同實) 심성일치(心性一致)이고, 녹문은 담일한 기 본체를 바탕으로 자신의 종지를 논증한다. 그리하여 리기는 이원이 아니라 기일원으로 환원되고, 심성은 심으로 환원된다. 심의 존재근거는 기이기 때문에, 결국 녹문의 본체론은 기일원론(氣一原論)이 되는 것이다. 녹문은 정명도의 일원적 방법론을 원용하여, 자신의 거대한 일원론 철학을 제출한다. 본체의 보편성과 현상의 다양성을 '일원분수(一原分殊)'라는 논리구조로 설명한다. 녹문 철학의 특징은 일원과 분수가 모두 본연으로 동일하다는 데 있다. 즉, 체용일원 내지 본말일치를 의미한다. 심과 성도 본연으로 일치하고, 기와 심과 성도 본연으로 일치하고, 몸과 마음도 본연으로 일치한다는 내용이다. 성리학의 용어로 하면, 기질이 본연과 다르지 않다는 뜻이다. 그리하여 나는 녹문의 철학을 '성인(聖人)의 관점에서 건립한 철학' 이라고 정의 내린다.
$1.55{\cal}um$ 파장에서 동작하는 자기 광학적 가이딩 층(magnetooptic guiding layer)을 갖는 도파로형 아이솔레이터를 제작하기 위하여 신 개념의 간섭계형 아아솔레이터가 제안되고 디자인되었다. 이 소자는 다중모드 간섭 결합기(multimode interference coupler)로 구성되어 있으며 간섭계의 두 도파로가 서로 다른 적층 구조를 가지고 있다. 간섭계의 적층 구조는 각각 $HfO_2/CeY_2Fe_5O_{12}/NOG$와 $SiO_2/CeY_2Fe_5O_{12}/NOG$이다. 이러한 비대칭 적층 구조(asymmetric layer structure)로 인해 단일 방향의 자기장하에서 간섭계의 두 도파로가 서로 다른 비가역적 위상변위(nonreciprocal phase shift)를 갖게 함으로서 아이솔레이터로서의 동작을 가능하게 한다. 최적화된 아이솔레이터의 구조는 3D 광전송 방법(Beam Propagation Method)을 이용하여 결정되었다.
Objective : This research investigates the effect of the NogYongDaeBoTang,(NYDBT) on Alzheimer's disease. Method : The effects of the NYDBT extract on (1) $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ mRNA of PC-12 cells treated with LPS; (2) acetylcholinesterase(AChE), amyloid precursor proteins(APP), and glial fibrillary acidic protein(GFAP) mRNA the AChE activity and the APP production of PC-12 cell treated with CT-105; (3) the behavior; (4) expression of $IL-1{\beta}$, $TNF-{\alpha}$, MDA, $IL-1{\beta}$ mRNA, and $TNF-{\alpha}$ mRNA; (5) the infarction area of the hippocampus, and brain tissue injury in Alzheimer‘s diseased mice induced with ${\beta}A$ were investigated. Results : 1. The NYDBT extract suppressed the expression of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNA in BV2 microglia cell line treated with LPS. 2. The NYDBT extract suppressed the expression of $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ protein production in BV2 microglia cell line treated with LPS. 3. For the NYDBT extract group a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by $A{\beta}$ in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 4. The NYDBT extract suppressed the over-expression of $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, and CD68/CD11b, in the mice with Alzheimer's disease induced by $A{\beta}$. 5. The NYDBT extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by $A{\beta}$. 6. The NYDBT extract reduced the Tau protein, GFAP protein, and presenilin1/2 protein (immunohistochemistry) of hippocampus in the mice with Alzheimer's disease induced by $A{\beta}$. Conclusions : These results suggest that the NYDBT extract may be effective for the prevention and treatment of Alzheimer's disease.
고령기의 생활을 위협하는 비만 여성고령자를 대상으로 sarcopenia 동반에 따른 16주간의 운동중재가 신체조성 및 심폐체력에 미치는 변화를 알아보기 위한 실험이다. 비만 여성고령자를 sarcopenia 동반유무에 따라 sarcopenic obestiy group(SOG)과 non-sarcopenic obesity group(NOG)으로 나누어 16주간, 주 3회, 50-60분간 복합운동 실시 후 신체조성 및 심폐체력에 미치는 영향을 분석하여 다음과 같은 결론을 얻었다. 신체조성의 경우 시기와 그룹간 상호작용효과는 fat free mass(FFM)에서만 나타났으며, 운동 전 후의 변화에서는 SOG의 경우 % body fat은 감소하였고, FFM과 ASM에서는 차이가 없었다. NOG의 경우 %body fat은 감소하였으며, FFM과 ASM에서는 증가하였다. 심폐체력에서는 두 그룹 모두 $\dot{V}O2peak$가 증가하였다. 따라서, sarcopenic obesity 여성고령자가 non-sarcopenic obesity 여성고령자에 비해 근육량(FFM, ASM)에서 운동효과가 지연되는 것을 발견할 수 있었다.

The Asian honeybee, Apis cerana, is a native honeybee species in Korea which is important in agriculture for pollination and honey production. For better understanding of the physiology of A. cerana, high-throughput Illumina transcriptome sequencing was performed to analyze the gene expression profiles of queen, worker, and larva. A total of 219,799,682 clean reads corresponding to 22.2 Gb of nucleotide sequences was obtained from the whole body total RNA samples. The Apis mellifera reference mRNA sequence database was used to measure the gene expression level with Bowtie2 and eXpress software, and the Illumina short reads were then mapped to 11,459 out of 11,736 A. mellifera reference genes. Total of 9,221 genes with FPKM value greater than 5 of each sample group were subjected to eggNOG with BLASTX for gene ontology analysis. The differential gene expression between queen and worker, and worker and larva were analyzed to screen the overexpressed genes in each sample group. In the queen and worker sample group, total of 1,766 genes were differentially expressed with 887 and 879 genes overexpressed over two folds in queen and worker, respectively. In the worker and larva sample group, total of 1,410 genes were differentially expressed with 1,009 and 401 genes overexpressed over two folds in worker and larva, respectively.
최근 경제위기의 여파로 국내 산업에서 소상공인이 차지하고 있는 비중은 전체 사업체 중 86%를 차지할 정도로 소상공인 창업은 매년 증가하고 있으나, 1년 내 40% 정도 폐업하는 등 사업의 성공률은 낮아 소상공인의 사업 성공률을 높이기 위한 다양한 방안을 살펴 볼 필요가 있다. 소상공인의 창업 성공에 대한 대부분의 선행연구들은 소상공인의 성공 및 성장에 영향을 주는 요인이 무엇인지를 확인하는 것에 집중하고 있다. 이러한 기존 연구는 대부분 정량적 분석이며, 실제 사례연구 등의 정성적 분석은 학술논문에서는 찾아보기 힘들다. 본 연구에서는 소상공인의 창업 성공에 대한 광범위한 사례를 분석한 후, 유형 별로 대응방안을 도출하고 이를 토대로, 소상공인이 창업하는 준비 과정에서 적절하게 대응할 수 있는 실용적 교훈을 제시하였다.
Leptospirosis is a zoonotic disease caused by spirochetes from the genus Leptospira. In Thailand, Leptospira interrogans is a major cause of leptospirosis. Leptospirosis patients present with a wide range of clinical manifestations from asymptomatic, mild infections to severe illness involving organ failure. For better understanding the difference between Leptospira isolates causing mild and severe leptospirosis, illumina sequencing was used to sequence genomic DNA in both serotypes. DNA of Leptospira isolated from two patients, one with mild and another with severe symptoms, were included in this study. The paired-end reads were removed adapters and trimmed with Q30 score using Trimmomatic. Trimmed reads were constructed to contigs and scaffolds using SPAdes. Cross-contamination of scaffolds was evaluated by ContEst16s. Prokka tool for bacterial annotation was used to annotate sequences from both Leptospira isolates. Predicted amino acid sequences from Prokka were searched in EggNOG and David gene ontology database to characterize gene ontology. In addition, Leptospira from mild and severe patients, that passed the criteria e-value < 10e-5 from blastP against virulence factor database, were used to analyze with Venn diagram. From this study, we found 13 and 12 genes that were unique in the isolates from mild and severe patients, respectively. The 12 genes in the severe isolate might be virulence factor genes that affect disease severity. However, these genes should be validated in further study.
The identification of oleaginous yeast species capable of simultaneously utilizing xylose and glucose as substrates to generate value-added biological products is an area of key economic interest. We have previously demonstrated that the Cutaneotrichosporon dermatis NICC30027 yeast strain is capable of simultaneously assimilating both xylose and glucose, resulting in considerable lipid accumulation. However, as no high-quality genome sequencing data or associated annotations for this strain are available at present, it remains challenging to study the metabolic mechanisms underlying this phenotype. Herein, we report a 39,305,439 bp draft genome assembly for C. dermatis NICC30027 comprised of 37 scaffolds, with 60.15% GC content. Within this genome, we identified 524 tRNAs, 142 sRNAs, 53 miRNAs, 28 snRNAs, and eight rRNA clusters. Moreover, repeat sequences totaling 1,032,129 bp in length were identified (2.63% of the genome), as were 14,238 unigenes that were 1,789.35 bp in length on average (64.82% of the genome). The NCBI non-redundant protein sequences (NR) database was employed to successfully annotate 11,795 of these unigenes, while 3,621 and 11,902 were annotated with the Swiss-Prot and TrEMBL databases, respectively. Unigenes were additionally subjected to pathway enrichment analyses using the Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), Cluster of Orthologous Groups of proteins (COG), Clusters of orthologous groups for eukaryotic complete genomes (KOG), and Non-supervised Orthologous Groups (eggNOG) databases. Together, these results provide a foundation for future studies aimed at clarifying the mechanistic basis for the ability of C. dermatis NICC30027 to simultaneously utilize glucose and xylose to synthesize lipids.
