  • Wild type nodulin 26(nod 26) cDNA(S262) and phodphorylation aite mutant(S262D) were constructed by a yeast expression system using pYES2 plasmids(pTES2-D262 and pTES2-S262D) were sc-reened by restriction mapping with BamHI of KpnI. S262 nod 26 contained a sreine residue at position 262 and S262D nod 26 contained the substitution mutation of serine to aspartic acid residue at position 262 were verified by automated floursent DNA sequencing.

  • Megaplasmids of Azospirillum strains isolated in the Korean paddy field were identified. Five megaplasmids were identified from Azospirillum lipoferum AS192. Homology between nod ABC gene of Rhizobium meliloti and megaplasmids of Azospirillum lipoferum AS192 and Azospirillum brasilense AS112 was found. This observation might have reflected a common mechanism in the early process of soil bacterial association with plants.

  • 컴퓨터와 통신망 기술의 발전으로 많은 언론 기관에서 전자신문 서비스를 제공하고 있다. 그러나 현재 서비스되는 전자신문은 텍스트 위주의 정적인 정보를 주고 서비스 하며, 사용자가 필요한 정보를 찾아 다니는 pull 기술을 기반으로 서비스한다. 그리고 사용자 맞춤 기능을 제공하기 못하므로, 불필요한 정보를 사용자에게 전송함으로써 시스템 자원을 낭비하는 단점을 가지고 있다. 본 논문에서는 멀티미디어 데이터를 지원하는 NOD(News On Demand)시스템에서 텍스트 데이터 뿐아니라 오디오/비디오 데이터를 push 하므로써 서버의 load를 분배시키고 사용자에게 실시간성을 제공하는 NOD 분배 서버를 설계하고 프로토타입을 구현하였다. 특히 본 논문에서는 대용량 데이터인 오디오/비디오 데이터를 사용자 시스템으로 Push할 때, 사용자 시스템의 디스크 공간상태 등을 고려하여 실시간성을 유지할 수 있는 적정 Push 량을 시뮬레이션을 통해 측정하였다.

  • Blau syndrome (BS) is a rare autosomal dominant, inflammatory syndrome that is characterized by the clinical triad of granulomatous dermatitis, symmetric arthritis, and recurrent uveitis. Mutations in the nucleotide oligomerization domain 2 (NOD2 ) gene are responsible for causing BS. To date, up to 30 Blau-associated genetic mutations have been identified within this gene. We report a novel NOD2 genetic mutation that causes BS. A girl, aged 8 years, and her brother, aged 10 years, developed erythematous skin rashes and uveitis. The computed tomography angiogram of the younger sister showed features of midaortic dysplastic syndrome. The brother had more prominent joint involvement than the sister. Their father (38 years) was also affected by uveitis; however, only minimal skin involvement was observed in his case. The paternal aunt (39 years) and her daughter (13 years) were previously diagnosed with sarcoidosis. Mutational analysis revealed a novel c.1439 A>G mutation in the NOD2 gene in both siblings. The novel c.1439 A>G mutation in the NOD2 gene was found in a familial case of BS. Although BS is rare, it should always be considered in patients presenting with sarcoidosis-like features at a young age. Early diagnosis of BS and prompt multisystem workup including the eyes and joints can improve the patient's outcome.

  • Since the identification and characterization of toll-like receptors (TLR) in Drosophila, numerous scientific studies have examined the role of TLRs in host innate immunity. Recent studies have suggested a convergence of the nuclear factor kappa B (NF-${\kappa}B$) signaling and cytokine production regulated by the cytosolic elicitor known as NLRs (nucleotide-binding domain and leucine-rich repeat containing domain receptors) as a key modulator in inflammatory diseases. Among the NLRs, NOD1 and NOD2 have been intensively investigated for its role in inflammatory bowel disease (IBD). On the other hand, NLRs such as NLRP3, NLRP1, and NLRC4 (also known as IPAF) have been identified to form the inflammasome to activate downstream signaling molecules in response to pathogenic microbes. There is evidence to suggest that substantial crosstalk exists for the TLR and NLR signaling pathway in response to pathogen associated molecular pattern (PAMP). However, the substrate and the mechanistic role of NLRs are largely unknown in innate immune response. Understanding the signaling mechanisms by which NLRs recognize PAMP and other danger signals will shed light on elucidating the pathogenesis of various human inflammatory diseases such as IBD.

Cellular and Molecular Roles of $\beta$ Cell Autoantigens, Macrophages and T Cells in the Pathogenesis of Automimmune Diabetes

  • Type I diabetes, also known as insulin-dependent diabetes mellitus (IDDM) results from the destruction of insulin-producing pancreatic $\beta$ cells by a progressive $\beta$ cell-specific autoimmune process. The pathogenesis of autoimmune IDDM has been extensively studied for the past two decades using animal models such as the non-obese diabetic (NOD) mouse and the Bio-Breeding (BB) rat. However, the initial events that trigger the immune responses leading to the selective destruction of the $\beta$ cells are poorly understood. It is thought that $\beta$ cell auto-antigens are involved in the triggering of $\beta$ cell-specific autoimmunity. Among a dozen putative $\beta$ cell autoantigens, glutamic acid decarboxylase (GAD) has bee proposed as perhaps the strongest candidate in both humans and the NOD mouse. In the NOD mouse, GAD, as compared with other $\beta$ cell autoantigens, provokes the earliest T cell proliferative response. The suppression of GAD expression in the $\beta$ cells results in the prevention of autoimmune diabetes in NOD mice. In addition, the major populations of cells infiltrating the iselts during the early stage of insulitis in BB rats and NOD mice are macrophages and dendritic cells. The inactivation of macrophages in NOD mice results in the prevention of T cell mediated autoimmune diabetes. Macrophages are primary contributors to the creation of the immune environment conducive to the development and activation of $\beta$cell-specific Th1-type CD4+ T cells and CD8+ cytotoxic T cells that cause autoimmune diabetes in NOD mice. CD4+ and CD8+ T cells are both believed to be important for the destruction of $\beta$ cells. These cells, as final effectors, can kill the insulin-producing $\beta$ cells by the induction of apoptosis. In addition, CD8+ cytotoxic T cells release granzyme and cytolysin (perforin), which are also toxic to $\beta$ cells. In this way, macrophages, CD4+ T cells and CD8+ T cells act synergistically to kill the $\beta$ cells in conjunction with $\beta$ cell autoantigens and MHC class I and II antigens, resulting in the onset of autoimmune type I diabetes.

A Push-Caching and a Transmission Scheme of Continuous Media for NOD Service on the Internet (인테넷상에서 NOD 서비스를 위한 연속미디어 전송 및 푸쉬-캐싱 기법)

  • In multimedia new service on the internet, there are problems such as server overload, network congestion and initial latency. To overcome these problems, we propose a proxy push-caching scheme that stores a portion of continuous media stream or entire stream, and a transmission scheme of NOD continuous media, RTP-RR and RTP-nR to exploit push-caching scheme. With the proposed push-caching scheme, NOD server pushes fixed portion of stream to a proxy when new data is generated, and the cached size of each stream changes dynamically according to the caching utility value of each stream. As a result, the initial latency of client side could be reduced and the amount of data transmitted fro ma proxy server to client could be increased. Moreover, we estimate a caching utility value of each stream using correlation between disk space occupied by the stream and the amount of data stream requested by client. And we applied the caching utility value ot replacement policies. The performance of the proxy push-caching and continuous media transmission schemes proposed were compared with other schemes using simulations. In the simulation, these schemes show better results than other schemes in terms of BHR (Byte Hit Rate), initial latency, the number of replacement and packet loss rate.

  • Bone is a dynamic tissue that is regulated by the balance between bone-resorbing osteoclasts and bone-forming osteoblasts. Curcumin isolated from Kang-hwang (Turmeric) is widely used as a foodstuff, cosmetic, and medicine. However, the effect of curcumin isolated from Kang-hwang in osteoclast differentiation remains unknown. In this study, we sought to examine the role of curcumin in osteoclast differentiation. Here we show that curcumin greatly inhibited RANKL-mediated osteoclast differentiation in osteoclast precursors without cytotoxicity. RANKL induced the phosphorylation of p38 and JNK mitogen-activated protein kinase (MAPK) and mediated $I-{\kappa}B$ degradation in bone marrow macrophages (BMMs). However, RANKL-mediated p38 MAPK phosphorylation was inhibited by the addition of curcumin. Curcumin inhibited the mRNA expression of TRAP, c-Fos, and NFATc1 in BMMs treated with RANKL. Furthermore, the protein expression of c-Fos and NFATc1 induced by RANKL was suppressed by curcumin treatment. Taken together, our results suggest that curcumin may have a potential therapeutic role in bone-related diseases such as osteoporosis by inhibiting osteoclast differentiation.

A Speaker Change Detection Experiment that Uses a Statistical Method (통계적 기법을 이용한 화자변화 검출 실험)

  • In this paper, we experimented with speaker change detection that uses a statistical method for NOD (News On Demand) service. A specified speaker's change can find out content of each data in speech if analysed because it means change of data contents in news data. Speaker change detection acts as preprocessor that divide input speech by speaker. This is an important preprocessor phase for speaker tracking. We detected speaker change using GLR(generalized likelihood ratio) distance base division and BIC (Bayesian information criterion) base division among matrix method. An experiment verified speaker change point using BIC base division after divide by speaker unit using GLR distance base method first. In the experimental result, FAR (False Alarm Rate) was 63.29 in high noise environment and FAR was 54.28 in low noise environment in MDR (Missed Detection Rate) 15% neighborhood.
