Studies on the inhibitory substance of yeast growth (Part II ) Effect on the nitrogen uptake (항효모성 물질에 관한 연구 (제II보) 질소대사와의 관계)

  • The biochemical characteristics of Astradix -P, isolated from Astragalus membranaceus Bunge as yeaststatic substance, were reported on a previous paper. And on this report, some relation to the nitrogen metabolism of yeast was studied. Inorganic or organic source of nitrogen easily uptaking yeast did not show any antagonistic action to the inhibitory action of Astradix -P on the yeast growth. Especially an organic nitrogen source, arginine, histidine and lysine, classified to basic amino acid, was reacted as an antagonistic substance to the sample. But, ornithine, a basic amino acid, did not show any antagonistic action to the sample. In the mixed media containing neutral and acidic amino acids as a nitrogen source, yeast growth was inhibited strongly. If the basic amino acid was added to the same mixed media, the yeast growth was not inhibited by Astradix-P therefore, the antagonistic action of basic amino acid to the Astradix-p was readily observed. The yeast static action of Astradix-P was partially related to the isoelectric point of amino acid as a nitrogen source. Yeast cells which propagated under the media containing growth inhibitor, Astradix -p, did not bring any remarkable denaturation of cell structure by electro-microscopic observation.

Effects of phosphorus concentration and nitrogen sources on photoautotrophic microalgae Scenedesmus dimorphus applied wastewater treatment (인 농도 및 질소원이 광합성 미세조류인 Scenedesmus dimorphus를 이용한 하수고도처리에 미치는 영향)

  • A result of estimating the effects of initial phosphorus concentration and nitrogen sources on removal of nitrogen and phosphorus in wastewater treatment using Scenedesmus dimorphus shows that there was no difference in microalgae growth amount and nitrogen removal amount by phosphorus concentration(5 ~ 60 mg/L). On the other hand, as initial phosphorus concentration increased, phosphorus removal amount was increased by luxury uptake of microalgae. Scenedesmus dimorphus preferred to ammonia when ammonia(40 mg/L) and nitrate(40 mg/L) are simultaneously presented and nitrogen removal rate of ammonia was high at approximately 20 mg/L/day.

Changes of Soil Nitrogen Supply and Production of Upland Forage Crops by Cattle Manure during Conversion from Paddy to Upland Condition in Paddy Field (논의 밭전환 연차간 우분시용에 의한 질소공급 및 밭사료 작물의 생산력 변화)

  • The effect of cattle manure with the rates of 2 and 4 ton $l0a^{-1}$ for winter rye and summer corn cultivation, respectively, on the dry matter (DM) yield and nitrogen (N) uptake were investigated during successive three­year conversion period from paddy to upland condition in paddy field. The changes in soil properties and soil N sup­plying capacity during repetitive manure application were a1so examined. Growth and DM yield of upland forage crops, especially. winter rye were hindered highly by poor soil condition in the first year after conversion from paddy to upland condition, so apparent recovery of cattle manure N by crops was very low in the first conversion year. But, DM yield and N uptake of upland forage crops were increased linearly by accumulative input of cattle manure along with mineral N enrichment in soil, which also increased apparent recovery of cattle manure-No It seemed that those increases were mainly due to the improvement of soil properties such as soil mineral N, soil organic matter (soil carbon), potentially mineralizable N and bulk density by accumulative input of cattle manure rather than the increase of soil N supply according to accumulative conversion period from paddy to upland condition. It was derived that conversion period from paddy to upland condition over 2 years is needed to obtain proper DM yield in paddy field and accumulative inputs of cattle manure during the conversion period is more influential to the continuous increment of DM yield and N uptake of upland crop as well as of potential N supplying capacity of soil.

Growth and Quality Changes of Creeping Bentgrass by Application of Keratin Amino Acid Fertilizer (케라틴 아미노산 비료 시비에 따른 크리핑 벤트그래스의 생육과 품질 변화)

  • Amino acids in the plant were intermediate metabolites which produced by uptake and assimilation of nitrogen and these extracts which gained by bio-chemical digestion from protein of plant or animal were a source of functional fertilizer. This study was conducted to evaluate effects of keratin amino acid fertilizer (KAF) gained from animal hair or hoof on changes of turfgrass quality and growth by investigating turf color index, chlorophyll index, shoot number, clipping yield, and nutrient content in the turfgrass tissue. Treatments were designed as follows; non-fertilizer (NF), compound fertilizer (CF), keratin amino acid fertilizer treatments [CF + KAF $0.26ml\;m^{-2}$ (CKF), CF + KAF $0.52ml\;m^{-2}$ (2CKF)], and only keratin amino acid fertilizer treatment (KF). Shoot number, clipping yield, and nitrogen uptake of KF were higher than those of CF. As compared with CF, soil chemical properties, turf color index, chlorophyll index and clipping yield of keratin amino acid fertilizer were not significant, but shoot number and uptake of N and K were increased significantly. These results show that the application of keratin amino acid fertilizer increased shoot number and growth by increased uptake of nitrogen and potassium.

The Importance of Dissolved Organic Nutrients on the Interspecific Competition between the Harmful Dinoflagellate Cochlodinium polykrikoides and the Diatom Skeletonema sp. (유해 와편모조류 Cochlodinium polykrikoides와 규조류 Skeletonema sp.의 종간경쟁에서 용존 유기 영양염의 중요성)

  • We investigated the interspecific competition between the harmful dinoflagellate Cochlodinium polykrikoides and diatom Skeletonema sp. based on the utilization and uptake of dissolved organic nutrients. C. polykrikoides and S. costatum were able to grow using dissolved organic nitrogen (DON) and dissolved organic phosphorus (DOP) as well as dissolved inorganic nitrogen (DIN) and dissolved inorganic phosphorus (DIP). This result indicates that the utilization of dissolved organic nutrients may play a role in surviving strategy in the DIN or DIP-limited environments. The half-saturation constants (Ks) of urea and glycerophosphate (glycero-P) calculated from uptake kinetics experiment of C. polykrikoides was lower than those of Skeletonema sp. This result indicates that Skeletonema sp. have higher affinity for dissolved organic nutrients, such as urea and glycero-P, than C. polykrikoides. Although Skeletonema sp. have higher affinity of dissolved organic nutrients, C. polykrikoides could effectively uptake for urea and glycero-P at sub-saturating nutrient concentrations (${\alpha}$ (${\rho}_{max}/Ks$) of C. polykrikoides was higher than Skeletonema sp.. Therefore, C. polykrikoides by utilization and effectively uptake of dissolved organic nutrients under monoculture may have an advantageous position in the interspecific competition with Skeletonema sp. in the low nutrient environments.

Improvement of Nitrogen Efficiency by N Application at Early Tillering Stage in Direct-Seeded Rice

  • This study was conducted to establish the elaborate nitrogen fertilization method to enhance N use efficiency in direct-seeded rice on flooded paddy. The nitrogen uptake by rice plants was insignificant until 25 days after seeding, and increased gradually thereafter. During this early growth stage, rice plants absorbed only the $4\%$ of basal applied N, while the $45\%$ of N fertilizer remained in the paddy soil. The absorption of basal N by rice plants was almost completed at 46 days after application. Nitrogen top-dressed at 5-leaf stage was well matched to crop nutrient demand, so it could be absorbed so actively in 8days after application. As a result, we could cut down the amount of N fertilizer to $36\%$ of the basal N level without significant difference in yield. Plant recoveries of fertilizer $^{15}N$ applied with different application timings were $7.8\%$ for basal, $9.4\%$ for 5-leaf stage, $17.1\%$ for tillering stage, and $23.4\%$ for panicle initiation stage, respectively. When urea was applied with nitrogen fertilization practice based on basal incorporation (BN), plant recovery of $^{15}N$ at harvest was $31.0\%$, which was originated from $13.7\%$ for grain, and $21.3\%$ of the fertilizer $^{15}N$ remained in the soil, and the rest could be uncounted. Plant recovery of fertilizer $^{15}N$ applied with nitrogen fertilization practice based on topdressing at 5-leaf stage (TN), where N rate was reduced by $18\%$ compared with BN, was $35.1\%$ (grain $15.6\%$), and $19.9\%$ of the fertilizer $^{15}N$ remained in the soil, and the rest could be uncounted. TN showed a higher $^{15}N$ recovery than BN because it was to apply N fertilizer at a time to well meet the demand of rice plant direct-seeded on flooded paddy. We concluded that TN would be the nitrogen fertilization method to enhance N use efficiency in direct-seeded rice on flooded paddy.

Mineral N, Macro Elements Uptake and Physiological Parameters in Tomato Plants Affected by Different Nitrate Levels

  • The aim of this study was to know whether leaf nitrate can be a substitute of total leaf N to justify plant N status and how nitrate influences macro elements uptake and physiological parameters of tomato plants under different nitrogen levels. Leaf nitrate content decreased in low N, while showed similar value with the control in high N, ranging from 55 to $70mg\;g^{-1}$. Differences in nitrate supply led to nitrate-dependent increases in macro elements, particularly cations, while gradual decrease in P. Physiological parameters, photosynthesis rates and antioxidants, greatly responded in N deficient conditions rather than high N, which didn't show any significant differences compared the control. Considering nitrogen forms and physiological parameters, total-N in tomato plants represented positive relation with growth (shoot dry weight), nitrate and $CO_2$ assimilation, whereas negative relation with lipid peroxidation.

Comparative Analysis on Concentration and Uptake Amount of Major Mineral Nutrients in Plant Tissues and Years Old of Panax ginseng C. A. Meyer (인삼의 연생과 식물체 부위별 무기영양성분 함량과 흡수량 비교)

  • The management and the use of major mineral nutriments such as nitrogen, phosphorous, and potassium, etc have been practiced and improved in various cultivating methods of Panax ginseng C. A. Meyer. The purposes of this study were to examine the content of major mineral nutrients on different ginseng aging from 1 to 6 years old, to analyze their uptake and utilization in tissues of ginseng, and to find out their proper managing techniques throughout the cultivation of ginseng. In case of the leaves, the N content was not clearly different from 1 to 6 years old, while the content of P and K was generally decreased throughout the cultivating years. In case of the roots, the content of N and K was gradually decreased from 1 to 6 years old, while the P content was increased until 3 years old, decreased at 4 years old, increased again at 5 years old, and decreased again at 6 years old. The uptake amount of N was increased in root of ginseng from 1 to 6 years old, 0.02 to 2.79kg/10a based on dry weight, respectively. Other minerals of P, K, Ca, and Mg were increased for the cultivating year. Comparing the uptake amounts of N, P, K with different cultivating year, they were the highest uptake amount at 4 years old and then were decreased. The management techniques of major mineral in cultivation of ginseng would be studied and evaluated more in order to have better ginseng production.

Responses of Pea Varieties to Rhizobium Inoculation: Nitrogenase Activity, Dry Matter Production and Nitrogen Uptake

  • The responses of five varieties and three cultivars of pea (Pisum sativum) to Rhizobium inoculation on nodulation, growth, nitrogenase activity, dry matter production and N uptake were investigated. The pea varieties were IPSA Motorshuti-l, IPSA Motorshuti-2, IPSA Motorshuti-3, BARI Motorshuti-l, BARI Motorshuti-2 and the cultivars were 063, Local small and Local white. Fifty percent seeds of each pea variety/cultivar were inoculated with a mixture of Rhizobium inoculants at rate of 15g/kg seed and the remaining fifty percent seeds were kept uninoculated. The plants inoculated with Rhizobium inoculant significantly increased nodulation, growth, nitrogenase activity, dry matter production and N uptake. Among the varieties/cultivars, BARI Motorshuti-l performed best in almost all parameters including nitrogenase activity of root nodule bacteria of the crop. There were positive correlations among the number and dry weight of nodules (r=$0.987^{**}$, $0.909^{**}$), nitrogenase activity of root nodule bacteria (r=$0.944^{**}$, $0.882^{**}$), dry weight of shoot (r=$0.787^{**}$, $0.952^{**}$), N content (r=$0.594^{**}$, $0.605^{**}$) and N uptake (r=$0.784^{**}$, $0.922^{**}$) by shoot both at flowering and pod filling stages of the crop, respectively. It was concluded that BARI Motorshuti-l in symbiotic association with Rhizobium inoculant performed best in recording nitrogenase activity, dry matter production and N uptake by pea.

Changes of Nitrogen Uptake, Growth and Activities of Nitrogen Metabolizing Enzymes by Different Source of Nitrogen in Tobacco (담배에서 질소 형태에 따른 흡수 양상 및 생육과 질소대사 효소의 활성 변화)

  • Tobacco plant was grown for 40 days hydroponically in nutrient solutions composed of different forms of nitrogen, like NO$_3$$^{[-10]}$ -N, NH$_4$$^{+}$-N, and a mixed formulation of NO$_3$$^{[-10]}$ -N and NH$_4$$^{+}$-N. Uptake response, nitrate reductase, and glutamine synthetase activity at growth stage were investigated to understand the basic knowledge of nitrogen metabolism. The better growth of shoot and root was observed in the mixed nutrient solution than NO$_3$$^{[-10]}$ -N or NH$_4$$^{+}$-N, alone. The plant growth in NH$_4$$^{+}$-N nutrient solution was poor due to ammonium toxicity. The pH of nutrient solution containing NO$_3$$^{[-10]}$ -N increased up to 40 days after transplanting. But the pH of solution containing NO$_3$$^{[-10]}$ -N decreased drastically to 3.42 at 20 days after transplant. The pH in the mixed formulation dropped to pH 3.64 at 30 days after transplant and showed re-increase. It is assumed that nitrogen of NH$_4$$^{+}$-N form was taken up preferentially at early stage and NO$_3$$^{[-10]}$ -N form was taken up preferentially at middle stage in the treatment with the mixed solution. The result indicates that the relative proportion of nitrogen forms affected the uptake patterns at each growth stages. The contents of chlorophyll and soluble protein were high with the mixed solution. Total nitrogen content was the highest in NH$_4$$^{+}$-N solution and the content also increased by the application of the mixed type of nitrogen. The amount of nitrate in leaves was high in NO$_3$$^{[-10]}$ -N treatment and the amount of ammonium was high in NH$_4$$^{+}$-N treatment. The activity of nitrate reductase or glutamine synthetase was highest in the leaves grown in mixed nutrient solution than in those with any other single of nitrogen form.
