Effect of culture conditions on the fermentation of wheat flour solution by mixed lactic acid bacteria of Lactobacillus brevis, L. fermentum and L. plantarum was investigated. The optimum temperature for the fermentation of wheat flour solution was $35^{\circ}C$ because pH decreased the lowest value and TTA (total titrable acidity) increased the highest value at this temperature. In aerobic condition, fermentor was purged with air at 1.0 vvm and was purged with nitrogen gas at 1.0 vvm in anaerobic condition. The decrease of pH and the increase of TTA in aerobic condition were higher than those in anaerobic condition. In aerobic condition, the optimum condition of oxygen supply was found to be oxygen transfer rate coefficient of $60\;hr^{-1}$ which corresponded to agitation speed of 250 rpm in a 5 L fermentor. Repeated fed-batch cultures were performed using pH-stat in order to increase the productivity of fermented wheat flour. With increasing the repeated fraction of culture volume, mean cycle time increased but maximum operation time decreased. However, the volume of produced broth per culture volume per time and total volume of produced broth per culture volume were maximum at the repeated fraction of culture volume of 20%. In a repeated fed-batch fermentation of wheat flour solution using mixed lactic acid bacteria, the culture condition was optimum at temerature of $35^{\circ}C$, aeration rate of 1.0 vvm, oxygen transfer rate coefficient of $60\;hr^{-1}$, and repeated fraction of culture volume of 20%.

Pluripotency of human embryonic stem cell (hESC) is one of the most valuable ability of hESCs for applying cell therapy field, but also showing side effect, for example teratoma formation. When transplant multipotent stem cell, such as mesnchymal stem cell (MSC) which retains similar differentiation ability, they do not form teratoma in vivo, but there exist limitation of cellular source supply. Accordingly, differentiation of hESC into MSC will be promising cellular source with strong points of both hESC and MSC line. In this study, we described the derivation of MSC like cell population from feeder free cultured hESC (hESC-MSC) using direct differentiation system. Cells population, hESC-MSC and bone marrow derived MSC (BM-MSC) retained similar characteristics in vitro, such as morphology, MSC specific marker expression and differentiation capacity. At the point of differentiation of both cell populations, differentiation rate was slower in hESC-MSC than BM-MSC. As these reason, to verify differentially expressed molecular condition of both cell population which bring out different differentiation rate, we compare the molecular condition of hESC-MSC and BM-MSC using 2-D proteomic analysis tool. In the proteomic analysis, we identified 49 differentially expressed proteins in hESC-MSC and BM-MSC, and they involved in different biological process such as positive regulation of molecular function, biological process, cellular metabolic process, nitrogen compound metabolic process, macromolecule metabolic process, metabolic process, molecular function, and positive regulation of molecular function and regulation of ubiquitin protein ligase activity during mitotic cell cycle, cellular response to stress, and RNA localization. As the related function of differentially expressed proteins, we sought to these proteins were key regulators which contribute to their differentiation rate, developmental process and cell proliferation. Our results suggest that the expressions of these proteins between the hESC-MSC and BM-MSC, could give to us further evidence for hESC differentiation into the mesenchymal stem cell is associated with a differentiation factor. As the initial step to understand fundamental difference of hESC-MSC and BM-MSC, we sought to investigate different protein expression profile. And the grafting of hESC differentiation into MSC and their comparative proteomic analysis will be positively contribute to cell therapy without cellular source limitation, also with exact background of their molecular condition.
On the development of hirame(Paratichtys olivaceus) culture, outbreak of scuticociliata infection was reported to cause severe damage in Japan. To establish effective measures for isolation and cultivation of this ciliate, we tried to culture this pathogenic ciliate using medium for bacteria and fish cell lines in vitro. Scuticociliata from the brain tissues of infected fish was aseptically inoculated to CHSE-214 cells cultured in MEM-10 without antibiotic. Scuticociliata grew well and the number of ciliate reached $10^6\;cells/ml$ at temperatures of $15^{\circ}C$ to $20^{\circ}C$ for 10d. The number of ciliate cultured in the cell lines is 10 times higher than the numbers cultured in the liquid medium alone. This ciliata could be cloned by dilution method. Scuticociliata isolated could grow well on 42 different cell lines that were established from marine fish, warm freshwater fish, and salmonids. This ciliate could be preserved in liquid nitrogen for more than 6 months. Subsequently, we observed the optimal temperature and salinity for growth, and tested the sensitivities of this organism to formaldehyde, flagyl(Metronidazole), Ekuteshin(Combination compound of sulfamonometoxin and ormethoprim), and ozonixation. Optimal temperature for growth was $25^{\circ}C$ and salinity was 1.0 to 1.5%. Washed scuticociliata was killed by formaldehyde at the concentration of 50ppm for 10min, but was not completely killed even at a high concentration of 400ppm for 20min in MEM-5. Flagyl and Ekuteshin can inhibit the growth of scuticociliata at the concentration of 1,000 and $100{\mu}g/ml$ in MEM-10, respectively. More than 99% of this scuticociliata could be killed by ozonization at a dose equivalent to $1.0mg/\ell$ oxidant for 30sec in sea water. Isolated scuticociliata showed the pathogenicity to the cultured hirame by artificial infection(I. P. injection, $10^5\;cells$/fish). The number of scuticociliata in the water could be counted by most probable number(MPN) method using tissue culture, and the minimum detectable number was $1.8\;cells/\ell$. The number in the reservoir tank for water supply to the culture tank was 110 cells/l. After cleaning by elimination of the sediments from of the reservoir tank and disinfected with formaldehyde, number of scuticociliata decreased and was counted less than $1.8\;cells/\ell$ and infection rate of cultured hirame was decreased.

In order to study changes in the marine ecosystem of the East China Sea derived by the global warming and construction of the Three Gorges Dam in the middle of the Changjiang, temperature, salinity, nutrients, and chlorophyll-a were studied intensively in the northern part of the East China Sea during the summer of 2003 and spring of 2004. According to the previous studies, the upwelling of the Kuroshio Current and the Changjiang resulted in a major inputs of nutrients in the East China Sea, but these two inputs may not contribute gently to a build up of nutrients in the northern East China Sea. In spring, relatively high concentrations of nitrates and phosphates were observed in the western part of the study area, which resulted from the supply of high concentrations of nutrients showing up in the surface waters as a result of vertical mixing from the ocean bottom. The concentrations of nitrates and phosphates observed in summer were lower than those in spring, since the surface waters were well stratified by the larger discharge of fresh water from the Changjiang in summer. The surface nitrate/phosphate ratios ranged from 1.3 to 16 in spring and from 1.1 to 15 in summer and were lower than the Redfield ratio of 16, indicating that the growth of phytoplankton is limited by nitrogen. This results are contrary to the previous results, in which the growth of phytoplankton was limited by phosphate in the East China Sea. The reason for this contrary result is that most nutrients in the surface waters are supplied by vertical mixing from the bottom waters with low nitrate/phosphate ratios, not directly influenced by the Changjiang with high nitrate/phosphate ratios. The depth-integrated chlorophyll observed in summer was similar to the previous results, but those measured in spring were almost twice as high as those found in previous results. The depth-integrated chlorophyll in spring was higher than that of summer, which results from high concentrations of nitrates and phosphates in the surface waters in spring due to active vertical mixing.
The growth response, lipid deposition, fat free body mass and energy expenditure of weanling rats fed the equal amount of isocaloric diets containing 8%, 13% and 18% casein were investigated. After a period of 30day feeding, the rats fed low level of protein diet were 43.01g lighter than 18% protein group (weight gains of ${85.57}{\pm}{7.50g}$ vs. ${128.58}{\pm}{11.64g}$, p<0.01). Despite of the smaller body size, there were no significant differences in lipid deposition in grams per carcass. Whereas, nitrogen accumulation was significantly greater in 13% and 18% protein fed groups compared to 8%. The estimated energy expenditure were 4,576.61 kJ, 5,440.80kJ and 5,607.67kJ for 8%, 13% and 18% protein groups respectively. The part of excess energy consumed by the low protein group may have been dissipated. The malic enzyme activity in the liver of rats was found to be unaltered by different dietary treatments. From these observations, it was conluded that the retarded growth response in lower protein level may have been originated from the shortage ge of protein supply rather than that of the energy. The protein restriction appeared to be resulted in the lower fat free compartment without affecting the ability of rats to synthesize body lipid in a similar rate to the higher protein group when energy intakes were equalized.

The effects of compost inoculation on the degradation of cellulosic fraction in composting of food waste and paper mixture were investigated by bench-scale composting. With the increase of seed inoculation, the time to reach the peaks of temperature, $CO_2$ evolution rate, and ammonia evolution rate was reduced, indicating that seed compost had beneficial effects on the enhanced degradation of organic materials at the early stage of composting. However, the final conversion of organic matters and the loss of ammonia were not affected by the amount of seed compost inoculated. The increasing of seed inoculum also resulted in the higher level of cellulase activity at initial stages and rapid rise to the maximums, suggesting that initial supply of sufficient cellulolytic microorganisms might facilitate the evolution of cellulase activity. The cellulose was degraded substantially during the increasing phase of cellulase activity, while they showed similar values at the end of 20 days composting. As a result, the seed inoculation seemed to be effective to the enhanced evolution of cellulase activity and cellulose degradation at initial stage of composting. But it did not contribute to increase the final degradation of cellulose after the entire composting reaction of 20 days.
Almost all of the natural gas consumed in South Korea is compressed into very high pressure for the transportation through the underground pipelines, then reduced in pressure regulation stations before delivery to the consumer. For pressure reduction, expansion valves have been used due to the simple and effective installation, but recover none of the energy in the gas during compression. Hence, turbo-expanders are proposed instead of the valves to accomplish the same pressure letdown function and recover some of the compression energy in the form of shaft work converting into electric powers. Here we have theoretically calculated the electric powers at the pressure reduction from 68.7 bar to 23 bar (which are the average values taken at the inlet and outlet points of the expansion valve in medium-pressure regulation stations) according to the inlet conditions of temperature and flow rate. The natural gas is considered as two cases of a pure methane and the mixture of hydrocarbons with a very small amount of nitrogen, and the Peng-Robinson equation of state is employed for the calculation of required thermodynamic properties. The electric energy is recovered as much as 1596 MW(methane) and 1567 MW(mixture) based on the total supply of natural gas in 2013.

Fertigation with drip irrigation provides effective and cost-efficient way to supply both nutrient and water to crop. However, inappropriate management of fertigation systems may cause inefficient nutrient and water use, thereby diminishing expected yield benefits as well as contributing to deterioration of soil properties. In this study, greenhouse experiments were conducted to investigate the optimal concentration of N, P and K fertigation solution for maximum production of oriental melon (Cucumis melo L.) using a response surface methodology, to evaluate an efficiency of nutrients uptake and an effect on soil chemical properties. Canonical analysis of response surface and contour plot interpretation revealed that $108.3mg\;L^{-1}$ of nitrogen (N), $54.8mg\;L^{-1}$ of phosphorous (P) and $158.3mg\;L^{-1}$ of potassium (K) resulted in maximim yield of oriental melon ($2,966kg\;10a^{-1}$). Compared to conventional practice, fertigation increased fruit yield up to 23.0% (p<0.001), uptake of N and K by plant also up to 33.3% (p<0.001) and 15.7% (p<0.01), respectively. These results suggest that fertigation has the advantage of the increase in yield and fertilizer use efficiency.

The use of heterotrophic denitrification as an alternative method for supplying alkalinity during sulfur-utilizing autotrophic denitrification was evaluated by examining the effects of external carbon source (both type and concentration) and HRT on denitrification efficiency. Concentrations of $NO_3{^-}-N$ and $COD_{Cr}$ of nitrified landfill leachate used for experiment were 700-900mg/L and 900-2500mg/L. respectively, All experiment was conducted with sulfur packed bed reactors (SPBRs) which were operated at $35^{\circ}C$. The fraction of $NO_3{^-}-N$ removed by heterotrophic denitrification ($HDNR_{fraction}$) to balance the alkalinity consumption by autotrophic denitrification varied with the type of external carbon source. When methanol and sodium acetate was added at theoretical HDNRfraction value. 100% denitrification was achieved without alkalinity addition. However, glucose and molasses require $HDNR_{fraction}$ value greater than theoretical value for complete denitrification. The EBCT and volumetric loading rate at which 100% denitrification efficiency could be achieved were 6.76 h and $2.84kg-NO_3{^-}-N/m^3{\cdot}d$, respectively, based on the fact that 100% denitrification occurred within the bottom 11.5 cm layer of the SPBR. The maximum nitrogen removal rate occurred with 89% removal efficiency at loading rate of $5.05kg-NO_3{^-}-N/m^3{\cdot}d$. However, at short EBCT, clogging of SPBR was observed with excess growth of heterotrophic denitrifiers. This problem may be eliminated by back washing or by separating of heterotrophic denitrification from sulfur-utilizing denitrification.
The study was performed to investigate the effect of incorporation of green manures (GM) into a sandy loam soil for organic vegetable production in the condition of plastic film house, relating to growth and yield of tomato crop. Three species of GM as perennial ryegrass, sudangrass and soybean are cultivated during the rest time of summer season and incorporated into soil just after the harvest. Thereafter tomato crop was transplanted as the following crop to soil incorporated GM. Among GM, soybean was proper as GM crop for organic farming, due to the effect of yield increase by continuous supply of nitrogen on following the tomato crop. Yield of tomato crop after soybean incorporation into soil was $4.2Mg\;ha^{-1}$ similar to $4.4Mg\;ha^{-1}$ of N-P-K standard fertilization (conventional) treatment. But perennial ryegrass and sudangrass were improper, because the biomass yield of perennial ryegrass was very low due to growth retardation by high temperature during summer season and soil incorporation of sudangrass as GM results in yield decrease of following the tomato crop caused by high C/N ratio of sudangrass itself. In conclusioa soybean incorporation into soil had advantage of producing conventional level on following the tomato yield and therefore it could recommend as GM for organic vegetable production.
