Fresh sea squirt meat requires a modified processing and preservation process because it has a short shelf life due to its high moisture content and strong proteolytic enzyme activity. In this study, bottled sea squirt meat prepared in vegetable oil (BSMO) to enhance the consumer acceptability was exposed to ${\gamma}$-ray (Co60, 10 kGy/h) irradiation to extend the shelf life without the use of a heating process. Response surface methodology was used to determine the optimal mixing ratio of BSMO containing 5% dehydrated fresh meat. Texture analysis and nutritional evaluation were also performed on a control and BSMO samples. The volatile basic nitrogen (VBN) content and total cell count were measured to determine the shelf life of irradiated BSMO products during chilled storage at $4^{\circ}C$ for 60 days. According to a panel of 10 trained tasters (aged 20-29 years), the optimal mixing formulation was 80 g meat in 60 mL of mixed vegetable oil (30 mL of olive oil and 30 mL of sesame oil). The highest rated formulation, according to a panel of nine trained tasters (aged ${\geq}30$ years), was 80 g meat in 60 mL of mixed vegetable oil (42 mL of olive oil and 18 mL of sesame oil). Moisture, ash, and protein contents in BSMO did not change significantly (P < 0.05) compared with the control. A higher lipid content ($0.84{\pm}0.23$ to $2.13{\pm}0.61$; P < 0.05) was observed due to the presence of vegetable oil on the surface of BSMO. The vegetable oil raised the hardness, springiness, cohesiveness, gumminess, chewiness, and resilience of BSMO. BSMO products remained edible after 50 days of storage at $4^{\circ}C$ based on the VBN content (BSMO 1: $27.92{\pm}0.96$ mg/100 g, BSMO 2: $24.84{\pm}1.95$ mg/100 g) and total cell count (BSMO 1: $4.60{\pm}0.80$ log CFU/mL, BSMO 2: $3.65{\pm}0.20$ log CFU/mL) when compared with standard levels of VBN (25.00 mg/100 g) and total cell count (5 log CFU/mL), respectively. The results showed that irradiated BSMO products could help to expand the processed seafood market and increase the popularity of seafood among the younger generations.

The protein profiles of domestic potato cultivars were evaluated for total protein determination, amino acid composition, SDS-PAGE analysis and scanning densitometry. There were statistically significant differences in the levels of amino acids among potato cultivars. Total nitrogen amount was also significantly different among cultivars, ranging from 1.27 to 1.64%. SDS-PAGE analysis showed that there were significant differences in the content of major potato proteins such as papatin (40 kDa), trypsin inhibitor (20 kDa) and protease inhibitor (15 kDa) among cultivars (p<0.05). The amount of papatin among cultivars with a range of 22.16 to 25.81 mg/g d.w. was higher in Jopung, Shepody and Superior, whereas the amount of protease inhibitors including 15 kDa and 20 kDa was the highest in Jopung (37.0%). The Shepody contains the highest amount of papatin (25.8%) and the lowest of trypsin inhibitor (5.22%). Thus, it is suggested that Shepody is the most desirable cultivar for better nutrition based on the protein profile.

Two experiments, utilizing 204 broiler chicks of the Maniker strain, were conducted to study the effects of dried whey and chitin on the growth of chicks. In the first experiment, diets containing 5%, 10%, or 15% dried whey, or a control diet with no whey, were fed to chicks from 1 day to 4 weeks of age. There were no significant differences among the dietary groups, with respect to the rate of growth or protein or feed efficiency. In the second experiment, chicks were fed with diets containing no whey, 2% chitin, 20% dried whey, or 20% dried whey plus 2% chitin, from 1 day to 4 weeks of age. Adverse effects (diarrhea, crooked toes and enlarged cecum) were observed in the group fed 20% dried whey. The protein efficiency ratio (PER) and feed efficiency ratio (FER) tended to improve in the dietary groups with dried whey, as compared to the control group. No significant differences were observed in the total carcass nitrogen and lipid levels of dietary groups. But the chitin-supplemented diets tended to improve on the growth rate compared to the groups without chitin. The results of the two experiments suggest that the growing broiler chick can tolerate up to 15% dried whey in the diet, without any harmful effect on growth. Also, chitin may improve the poorer growth rate of chicks fed dried whey, but not all of the adverse effects of whey.
In order to make acquire a potential use for Opuntia humifusa as a natural functional food material, this study was performed to determine the quality properties of Jeung-pyun made with added Opuntia humifusa, or prickly pear powder. According to an analysis of its major components, we found that the prickly pear powder consisteds of nitrogen-free extracts (71.85%) and crude fiber (11.20%). Greater additions of prickly pear powder had resulted in significantly lower pH in the of Jeung-pyun. According to measurements on the degree of Jeung-pyun gelatinization, by means of ${\beta}$-amylase, greater additions of prickly pear powder led to the higher levels of isolated maltose, indicating that the gelatinization degree of the Jeung-pyun became higher. Also, samples with higher concentrations of prickly pear powder had a tendency toward lower water content, which allowed us to expect a longer storage duration for the Jeung-pyun. In the textural property tests the Jeung-pyun that had less hardness and greater adhesiveness (p<0.05) than the control group as the content of prickly pear powder became higher. Also, the Jeung-pyunhad lower gumminess and chewiness than the control group as the content of prickly pear powder became higher. Therefore, it is possible to prepare relatively soft Jeung-pyun using prickly pear powder. For the color differences of the Jeung-pyun samples, lower L- values, and higher a- and b -values (p<0.05) presented as the addition level of prickly pear powder became higher. According to SEM observations of the Jeung-pyun, the added prickly pear powder addition groups generally showed a smaller and more inconsistent pore size, but higher porosity, than the control group. According to sensory analyses of the Jeung-pyun, the P2 group scored highest for color item, and the P4 group generated the fermented scent. Higher additions content of prickly pear powder led to the lower score, but higher scores for adhesiveness. Finally, the P2 group achieved the highest score for overall taste.

The pre-concentration and determination of ultratrace arsenic in water samples was studied by the precipitate flotation technique. The arsenic in 1.0l of water sample, in which all suspended materials were filtered out, was coprecipitated together with La(OH)$_3$ precipitates at pH 8.5${\pm}$0.1. After the precipitate was made to be hydrophobic by adding mixed surfactant of 1 : 8 mole ratio of sodium oleate and sodium dodecyl sulfate, it was floated with the aid of tiny bubbles of nitrogen gas in a flotation cell. The floated precipitate was quantitatively collected on a micropore glass filter by the suction, dissolved with small volume of 1.0M sulfuric acid, and accurately diluted to 25.00ml with a de-ionized water. Total arsenic was spectrophotometrically determinated by forming silver diethyldithiocarbamate complex of arsine generated from arsenic in the concentrated solution. The calibration curve was linear up to 20ng/ml in the original solution. Analytical results showed that contents of arsenic in a campus wastewater and a river water were 8.2ng/ml and l.0ng/ml, respectively, and their recoveries were 93${\%}$ and 90${\%}$ in water samples which a given amount of arsenic was added into. From above result, it could be concluded that this method was applicable to the determination of arsenic in various kinds of water at low ng/ml levels.

As a new practical method for preserving freshness of fish, partially frozen storage has been reported to be useful in terms of K-value, TBA value, sensory evaluation etc. In order to develop a storage procedure to be used in place of cooled or frozen storage for the preservation of precooked fish food, partial freezing for up to two to three weeks was examined using baked mackerel. The criteria for evaluation were made according to the changes in volatile basic nitrogen (VBN), thiobarbituric acid (TBA) value, viable bacterial number, pH, color difference and sensory evaluation in fish muscle. The changes in TBA value of baked muscle during storage at $-3^{\circ}C$ differed slightly from those obserbed during cooled ($5^{\circ}C$) and frozen storage ($-20^{\circ}C$). Partial frozen storage ($-3^{\circ}C$) was effective in prolong an induction period of lipid oxidation during early storage. VBN of baked muscle tends to increase slowly while pH value was decrease during storage and there was no observed significant differences among three different storage condition. Viable bacterial number of the baked mackerel muscle stored at $-3^{\circ}C$ showed significantly less than that stored at $5^{\circ}C$, and similar to that stored at $-20^{\circ}C$ (the levels of $10^2/g$). Judging from the results of sensory evaluation and experimental data, partial frozen storage at $-3^{\circ}C$ seems to be effective as means of short-period preserving baked mackerel.
The purpose of this study aimed to study effects of fire on vegetation and soil properties after the first growing season in Mt. Chiak. 1. With the basis of importance value of species in each stand, status of species was assessed for three categories; Increaser species, Decreaser species, and Neutral species. 2. Biomass was 2.2 times higher on burned area than unburned. This indicates that biomass was remarkably increased after fire. 3. To evaluate similarity, coefficients of similarity among communities were obtained, and correlation coefficients were also estimated. These indices showed that burned and unburned community were markedly different.. $B_1$-stand and $B_1$-stand appeared most similar to each other among stands. 4. Species diversity was greater in burned than unburned stands. 5. Soil pH value and organic matter content in burned area were significantly higher than those in unburned area. However, soil water content was lower in burned area. There was no effect of burning on soil pH value and water content at 15~20 cm depth of soil. 6. All chemical compositions except sodium were much higher in soil surface. The decreases in sodium levels at surface were probably resulted from the rapid leaching due to the increased solubility and decreased capacity for adsorption of sodium in comparison with potassium or calcium. Among chemical compositions of soil amount of nitrogen showed least difference between the burneb and unburned surfaces soil.

Objectives : This study was undertaken to verify the effects of Wolbigachul-tang1(WBCEx1) on obesity using high fat diet-induced male mice and to investigate the molecular mechanisms involved. Methods : 8-week old C57BL/6 mice were divided into 5 groups; lean control, obese control, WBCEx1, 2, 3. After mice were treated with WBCEx1(water extract), 2(30% ethanol extract), 3(water extract; Ephedra sinica Stapf., Gypsum fibrosum) for 12 weeks, body weight gain, feeding efficiency ratio, plasma lipid and glucose metabolism, the messenger RNA(mRNA) expression of peroxisome proliferator activated receptor(PPAR)$\alpha$ target genes were measured. In addition, $PPAR{\alpha}$ target gene expression was examined in liver, white adipose tissue and skeletal muscle. Results : 1. WBCEx1-treated mice had significantly lower body weight gain and feeding efficiency ratio. 2. Consistent with the effects on body weight gain, WBCEx1 decreased the weights of epididymal and retroperitoneal white adipose tissue, inguinal subcutaneous adipose tissue, and brown adipose tissue. 3. WBCEx1 significantly decreased plasma triglyceride and total cholesterol levels. 4. The size of adipocytes were significantly decreased by WBCEx1, whereas the adipocyte number per unit area was increased. Hepatic lipid accumulation was decreased by WBCEx1. 5. WBCEx1 did not affect the mRNA expression of $PPAR{\alpha}$ target genes in liver, adipose tissue, and skeletal muscle. 6. Plasma asparate aminotransferase(AST), alanine aminotransferase(ALT), blood urea nitrogen(BUN) and creatine concentrations were in the physiological range. Liver and kidney weights were significantly lower following WBCEx treatment compared with obese controls, indicating that WBCEx does not show any toxic effects on liver and kidney. Conclusions : These results suggest that WBCEx1-induced body weight reduction is associated with appetite control and mediated by a mechanism other than the activation of $PPAR{\alpha}$.

Physical and chemical analyses of water discharged from 4 crowded farms (Sungsan, Pyosun, Wimi and Daejung) in Jeju island were performed from July, 2006 to Dec, 2006, and the result of the analyses showed that hydrogen ion concentrations (pH) for water discharged from Sungsan farm was 7.74, Pyosun was 7.68, Wimi was 7.68 and Daejung was 7.7. Salinity levels for Sungsan, Pyosun and Wimi had an average of 31$\sim$33 $^\circ$/$_\circ$$_\circ$ indicating characteristics of far distance areas, whereas that of Daejung was 28.81 %, which was far lower compared to regular sea water salinity. As the result of measuring dissolved oxygen (DO) for each area, each area showed first graded DO for each discharged water based on water quality level for each sea district. The result of measuring the temperature for discharged water showed that water temperatures for summer were 23$\sim$25$^\circ$C, and those for winter were 16$\sim$ 18$^\circ$C. Nitrogen concentrations for discharged water exceeded each sea area's water quality level in all farms. In the case of phosphate, its average value was 0.48 mg/l for Sungsan, 0.55 mg/I for Pyosun, 0.66 mg/I for Wimi, and 0.44 mg/l for Daejung, and chemical oxygen demand (COD) was shown to be 1.5 mg/l 1.8 mg/I, 1.6 mg/I and 2.3 mg/I for Sungsan, Pyosun, Wimi and Daejung respectively. For suspended solids (SS), the average concentration was 19.3 mg/I, 21.2 mg/I, 21.3 mg/I and 18.5 mg/I for Sungsan, Pyosun, Wimi and Daejung respectively. The results of physical and chemical analyses for discharged water in farms based on time showed that almost all items were shown to increase in the forenoon and decrease, overall, in the afternoon.

Biochar amendment to agricultural soil is regarded as a promising option to mitigate climate change and enhance soil quality. It could sequester more carbon within the soil system and increase plant yield by changing soil physicochemical characteristics. However, sustainable use of biochar requires comprehensive environmental assessment. In this sense, it is important to measure additional greenhouse gas emission from soils after biochar addition. We investigated emissions of $CO_2$, $N_2O$, and $CH_4$ from incubated soils collected from rice paddy and cultivated grassland after amendment of 3% biochar (wt.) produced from rice chaff. During incubation, soils were exposed to three wet-dry cycles ranging from 5~85% soil gravimetric water content (WC) to investigate the changes in effect of biochar when influenced by different water levels. The $CO_2$ emission was reduced in biochar treatment compared to the control at WC of 30~70% both in rice paddy and grassland soils. This indicates that biochar could function as a stabilizer for soil organic carbon and it can be effective in carbon sequestration. The $N_2O$ emission was also reduced from the grassland soil treated with biochar when WC was greater than 30% because the biochar treated soils had lower denitrification due to better aeration. In the rice paddy soil, biochar addition resulted in decrease in $N_2O$ emission when WC was greater than 70%, while an increase was noted when WC was between 30~70%. This increase might be related to the fact that available nutrients on biochar surface stimulated existing nitrifying bacterial community, resulting in higher $N_2O$ emission. Overall results imply that biochar amendment to agricultural soil can stabilize soil carbon from fast decomposition although attention should be paid to additional $N_2O$ emission when biochar addition is combined with the application of nitrogen fertilizer.
