Studies on the amino acid metabolism of young rice root (Part 3) - Effects of nitrogen, phosphorus, potassium and respiratory inhibitor on the enzyme activities of rice root - (수도근(水稻根)의 Amino산(酸) 대사(代謝)에 관한 연구 -제(第) 3 보(報) 수도근(水稻根)의 몇가지 효소(酵素)의 활성(活性)에 미치는 삼요소(三要素)및 호흡저해제(呼吸沮害劑)의 영향(影響))

  • Some effects of nitrogen, phosphorus, potassium and respiratory inhibitor on growth of rice plant and activity of GOT, GPT and peroxidase for the rice root were investigated. Obtained results were summarized as follows: 1. Growth of rice root and plant applied with $NO_3$-N in culture solution was generally increased in the length and weight compared with that of $NH_4$-N plot. On the other hand, the GOT, GPT and peroxidase activity was more increased in the $NH_4$-N plot than in the $NO_3$-N plot. 2. Oxidative power of ${\alpha}$-naphthylamine in rice root was stronger in the $NO_3$-N plot than in the $NH_4$-N plot. 3. When rice plant was cultured in the medium which did not supplied nitrogen, phosphorus or potassium, respectively, GOT activity was more decreased than GPT activity, while peroxidase activity was increased mostly in the potassium-free plot. 4. When rice plant was cultivated in the culture solution added respiratory inhibitor, NaF, plant height was shortened in the order of nitrogen-free > $NH_4$-H > urea-N > $NO_3$-N plot, and GOT and GPT activity was also decreased in the order of nitrogen-free > $NH_4$-N > urea-N > $NO_3$-N plot.

Long-term Variation of Water Quality in Lake Andong (안동호 수질의 장기적인 변화)

  • Water quality parameters were surveyed in Lake Andong. Turbidity, temperature, secchi disc transparency (SD), phosphorus, nitrogen and chlorophyll a concentration were measured at dam site from July 1993 to December 1998. Minimum transparency in summer was only about 2 meters in 1993 and 1994, but it decreased to about 1 meter in 1997 and 1988. Total phosphorus concentration of the epilimnion increased slightly from $11{\sim}30\;mgP/m^3$ in 1993 to $18{\sim}42\;mgP/m^3$ in 1998. Total nitrogen concentration of the epilimnion decreased slightly from $1.81{\sim}2.96\;mgN/L$ in 1993 to $1.48{\sim}2.57\;mgN/L^3$ in 1998. TN/TP weight ratio decreased from $82{\sim}281$ in 1993 to $21{\sim}143$ in 1998 due to the increase of phosphorus concentration and the decrease of nitrogen concentration. Dissolved inorganic phosphorus concentration and the decrease of nitrogen concentration. Dissolved inorganic phosphorus and nitrate nitrogen concen tration of the epilimnion were in the range of $0.9{\sim}5.3\;mgP/m^3$ and $1.36{\sim}1.68\;mgN/L$, respectively. Chlorophyll a concentration in summer was in the range of $11.0{\sim}19.1\;mg/m^3$ in 1994, 1996 and 1997, but it decreased to $2.3{\sim}6.5\;mg/m^3$ in 1998. Trophic state of Lake Andong can be classified as mesotrophic to eutrophic from TP, TN and chlorophyll a concentration.

Carbon and nitrogen status in litterfall of a red pine stand with varying degrees of damage from pine wilt disease

  • We evaluated the carbon (C) and nitrogen (N) status of litterfall in a natural red pine (Pinus densiflora) stand damaged by pine wilt disease in Jinju City, which was one of the areas severely affected by the disease in Korea. A significant correlation (P < 0.05) was found between tree density and basal area and the C and N status of litterfall components, but C and N status was not correlated with mean diameter at breast height in the pine wilt disease stands. Needle-litter C and N concentrations were linearly related (P < 0.05) to basal area in pine wilt disease stands. Needle-litter C concentration decreased with a decrease in damage intensity due to pine wilt disease, whereas litter N concentration increased with an increase of basal area in pine wilt disease stands. The linear regression equations developed for litterfall C and N inputs were significant (P < 0.05), with basal area accounting for 50-86% of the variation, except for cone and flower litter. The results indicated that the incidence of pine wilt disease could impact the quality and quantity of C and N in litterfall of pine stands suffering from pine wilt disease.

  • Production of poly-$\beta$-hydroxybutyrate (PHB) in fed-batch fermentation was studied. Utilization of carbon for PHB biosynthesis was investigated by using feeding solutions with different ratios of carbon to nitrogen (C/N). It was observed that at a high C/N ratio carbon source was more preferably utilized for PHB accumulation while its consumption for cellular metabolism appeared to be more favored at a low C/N value. A high cell concentration (184 g/l) was achieved when ammonium hydroxide solution was fed to control the pH, which was also utilized as the sole nitrogen source. For the mass production of PHB, two-stage fed-batch operations were carried out where PHB accumulation was observed to be stimulated by switching the ammonium feeding mode to the nitrogen limiting condition. A large amount of PHB (108 g/l) was obtained with cellular content of 80% within 50 hrs of operation.

  • Ordinary biological methods of evaluating protein quality are subjected to various sources of error, such as preliminary feeding, body weight of the animals, length of feeding period, endogenous nitrogen metabolism, etc. (1). Wide variations in the reported data may be partly due to this fact. The author postulated that by applying labelled protein these shortcomings may be avoided and that a balance study using nitrogen-15 would give most reliable data, uninfluenced by other factors which are unavoidable in ordinary balance studies using unlabelled nitrogen (2). However, the most important prerequisite for testing this assumption is to obtain labelled protein. As it has been known long since that chlorella contains a large amount of protein and can be easily cultured in quantity (3, 4), it was thought that labelled protein might be obtained by adding $N^{15}$ into the culture media. In this study it has been tried to find out, therefore, whether it is possible to obtain labelled protein by incorporating $KN^{15}\;O_3$ into the culture fuid and at what stage of growth the incorporation of the label into chlorella occurs.

A Resistance Property Against High Velocity Impact on Oxynitride Glasses (질화유리의 고속충돌 저항물성)

  • Several oxynitride glasses were fabricated by means of adding $Si_3N_4$ powders as nitrogen source to Ca-Al-Si-O-N (CAS) and Mg-Al-Si-O-N (MAS) glass powders, and heat-treated in graphite crucible at 1600$^{\circ}C$ for 1 h. The physical and mechanical properties as well as impact resistance were generally increased and compared with each other. The impact resistance properties of those manufactured glasses were evaluated by DOP (depth of penetration) method which is a way to analyze armor materials. There were two means to be used herein; the copper jet impacted at hyper velocity by exploding K2l5 warhead and tungsten heavy alloy (WHA) impact bar at high velocity by firing in 30 mm solid propellent gun. The impact resistance properties against copper jet were increased and then decreased with increasing nitrogen content, while those against WHA bar were not changed apparently with nitrogen content.

Performance of a submerged membrane bioreactor for wastewater mimicking fish meal processing effluent

  • The objective of this work was to analyze organic matter removal, nitrification, biomass growth and membrane fouling in a submerged flat-sheet membrane bioreactor, fed with synthetic wastewater, of similar composition to the effluents generated in a fish meal industry. After biomass acclimatization with saline conditions of 12 gNaCl/L and COD/N ratio of 15 in the bioreactor, results showed that the organic matter removal was higher than 90%, for all organic loading rates (0.8, 1, 1.33 and $2gCOD/L{\cdot}d$) and nitrogen loading rates (0.053, 0.067, 0.089 and $0.133gN/L{\cdot}d$) tested during the study. However, nitrification was only carried out with the lowest OLR ($0.8gCOD/L{\cdot}d$) and NLR ($0.053gN/L{\cdot}d$). An excessive concentration of organic matter in the wastewater appears as a limiting factor to this process' operating conditions, where nitrification values of 65% were reached, including nitrogen assimilation to produce biomass. The analysis of membrane fouling showed that the bio-cake formation at the membrane surface is the most impacting mechanism responsible of this phenomenon and it was demonstrated that organic and nitrogen loading rates variations affected membrane fouling rate.

A Study on Nitridation and Oxidation Reaction of Titanium Powder (금속티타늄분말의 질화반응과 산화반응에 관한 연구)

  • The nitridation kinetics of titanium powder were studied by isothermal and non-isothermal (dynamic) methods in high purity nitrogen under I atm pressure. For the comparison with nitridation, the oxidation kinetics of titanium powder were also studied in dry oxygen at I atm pressure. An automatic recording electrobalance was used to measure the weight gain as a function of time and temperature. For the reaction with nitrogen, the nitride was formed at over $700^{\circ}C$. The reaction with nitrogen followed the parabolic rate law, and the activation energy was calculated to be 31 kcal/mol in the isothermal method (above $900^{\circ}C$). The non-stoichiometric TiNx has been synthesized by the nitridation at a proper temperature and time, followed by the homogenizing treatment above $1100^{\circ}C$. In comparison with the stoichiometric $TiN_{1.0}$ and the non-stoichiometric TiNx ($TiN_{0.5}$ and $TiN_{0.65}$), the hot oxidation characteristics of the former is superior to that of the latter. However, both non-stoichiometric nitrides make little difference in the hot oxidation characteristics.

Study of the Nitrogen-Beam Irradiation Effects on ALD-ZnO Films (ALD로 성장된 ZnO박막에 대한 질소이온 조사효과)

  • ZnO, a wurtzite lattice structure, has attracted much attention as a promising material for light-emitting diodes (LEDs) due to highly efficient UV emission resulting from its large band gap of 3.37 eV, large exciton binding energy of 60 meV, and low power threshold for optical pumping at room temperature. For the realization of LEDs, both n-type ZnO and p-type ZnO are required. Now, n-type ZnO for practical applications is available; however, p-type ZnO still has many drawbacks. In this study, ZnO films were grown on glass substrates by using atomic layer deposition (ALD) and the ZnO films were irradiated by nitrogen ion beams (20 keV, $10^{13}{\sim}10^{15}ions/cm^2$). The effects of nitrogen-beam irradiation on the ZnO structure as well as the electrical property were investigated by using fieldemission scanning electron microscopy (FESEM) and Hall-effect measurement.

Growth and Gibberellins level of Two Rice Cultivars as Influenced by Different Nitrogen Containing Compounds

  • Seedlings of two rice cultivars i.e. cv. Daesanbyeo and cv. Dongjinbyeo were analyzed for growth and endogenous gibberellins (GAs) in response to nitrogen nutrition applied in the forms of $KNO_3$, $(NH_4)_2SO_4$ and $NH_4NO_3$. All the growth parameters showed an increase in N applied treatments and their magnitudes of increase were different depending on different nitrogen fertilizer forms. The endogenous GAs contents were increased with N application but differentially affected by various N-forms in both rice cultivars. In cv. Daesanbyeo, maximum amount of bioactive $GA_1$ was recorded for $(NH_4)_2SO_4$, while maximum amount of bioactive $GA_1$ in cv. Dongjinbyeo was observed in $NH_4NO_3$ applied treatments. In both rice cultivars, $KNO_3$ applied rice plants contained least $GA_1$ contents. Also, $GA_{19}$ was the most abundant GA found in rice seedlings whereas $GA_4$ was absent at seedling stage. Our study indicated that different rice cultivars showed different responses for the same fertilizer depending upon the response potential of each cultivar and the pertinent physiological responses to changes of endogenous GAs in rice cultivars, which were comparatively lesser in magnitude.
