Optimization of Culture Conditions for the Production of Pyrimidine Nucleotide N-Ribosidase from Pseudomonas oleovorans (Pseudomonas oleovorans의 pyrimidine nucleotide N-ribosidase의 생성 최적조건)

  • Pyrimidine nucleotide N-ribosidase (pyrimidine 5'-nucleotide phosphoribo (deoxyribo) hydrolase/pyrimidine 5'-nucleoude nucleosidase, EC 3.2.2.10) directly catalyzes pyrimidine 5'-nucleotide to pyrimidine base and ribose (deoxyribo) 5-phosphate. In order to clarify the best nutritional conditions for the growth and the pyrimidine nucleotide N-ribosidase production of Pseudomonas oleovorans ATCC 8062 the effects of various nutrients such as different carbon and nitrogen sources were studied. For the both the growth and the enzyme production, 2% fumarate, 1.5% peptone, 5% corn steep liquor (CSL) and 1% ammonium chloride were excellent carbon and nitrogen sources, respectively. Optimum pH, temperature, and cultivation time for the enzyme production were 7.0, $28^{\circ}C$, and 48 h, respectively. The pyrimidine nucleotide N-ribosidase of P. oleovorans ATCC 8062 was not induced by UMP and its derivatives, and was constitutive enzyme.

Optimal Application Rate of Mixed Expeller Cake and Rice Straw and Impacts on Physical Properties of Soil in Organic Cultivation of Tomato (토마토 유기재배에서 혼합유박과 볏짚의 적정시용량 및 토양 물리성에 미치는 영향)

  • BACKGROUND: In this study, 5 different treatments such as non-treatment, mixed expeller cake 1.0 N (standard nitrogen fertilizer), rice straw, rice straw+mixed expeller cake 0.5 N, rice straw+mixed expeller cake 1.0 N were performed over 4 cropping seasons over 2 years in order to identify the optimal application rate of mixture of rice straw and mixed expeller cake, organic source in organic cultivation of tomatoes. METHODS AND RESULTS: There was no difference in all treatments in case of 200 mg/kg in the nitrate nitrogen content in soil prior to the first cropping season test under the criteria for nitrogen nutrient based on yield of crops, cultivation without fertilizers seems possible. But in the second cropping season, no treatment and rice straw showed the reduction of yield and in the third cropping season, rice-straw+mixed expeller cake 0.5 N treatment showed the significant difference. The content of nitrate nitrogen in soil prior to cropping seasons was evaluated in 160 mg/kg and standard fertilization such as mixed expeller cake, source of nitrogen, are needed due to the deficiency of nitrogen. In terms of application of organic resources, rice straw showed the effects of improvements on physical properties of soil such as bulk density, cation exchange capacity and humus contents, but the mixed expeller cake did not show any significant differences in improvements on physical properties of soil. CONCLUSION(s): Fertilizer management in organic cultivation of tomatoes is thought to produce the reliable quantity of crops as well as keep the high quality of soils by using the optimal application rate of mixed expeller cake according to the contents of nitrate nitrogen in soil and rice straw which improves the physical properties of soil.

Effects of Split Nitrogen Application on Agronomic Characteristics, Forage Yield, and Chemical Composition of Kenaf in Jeju (제주지역에서 질소분시 횟수에 따른 양마의 생육특성, 사료수량 및 조성분 변화)

  • Evegradies 71' kenaf (Hibiscus cannabinus L.) was grown at 160,000 plants per ha in a volcanic ash soil in Jeju island to determine the optimum frequency of split N application fer forage production. Nitrogen fertilizer was applied with 240 kg/ha, and frequencies of the split N applications were 1, 2, 3, 4 and 5 times. Plant height averaged across two harvests increased from 187 to 201cm as N was split-applied from one to three applications and then decreased to 201cm in five applications. This pattern held for the number of branches and leaves per plant stem diameter and weight of plant per plant. Fresh forage yield increased from 91.8 to 114.2 MT/ha. dry matter yield from 12.70 to 16.6 MT/ha, crude protein yield from 1.75 to 2.48MT/ha, and total digestible nutrients (TDN) yield from 5.39 to 7.63 MT/ha as N was split-applied from one to four applications, and then decreased to 106.6, 15.0, 2.32, and 7.22 MT/ha in five applications, respectively As N was split-applied from one to five applications, crude protein content increased from 13.8 to 15.4%. ether extract content from 4.1 to 5.9%, and TDN content from 42.4 to 48.1%. but crude fiber decreased from 38.2 to 37.1% and crude ash content from 11.5 to 8.6%. Nitrogen free extract content was about 19.0% regardless of cutting height.

Characteristics of Organics and Nitrogen Removal with the Recycle Ratio in Anoxic / Oxic Packed Bed Process (충전탑형 무산소/호기 공정에서 반송비에 따른 유기물 및 질소 제거 특성)

  • This study was focused on the investigation of the characteristics of organics and nitrogen removal with the recycle ratio in anoxic/oxic(A/O) packed bed process that consisted of the anoxic reactor and the aerobic reactor. As increasing the recycle ratio by 0.5, 1.0, 2.0, the COD removal efficiency increased by 94.0%, 98.5%, 98.8% respectively. The aerobic reactor showed the perfect nitrification efficiency by 98.5%, 99.2%, 98.0% respectively. The T-N removal efficiency with the recycle ratio, increased by 56%, 67%, 70% respectively. As increasing the recycle ratio by 0.5, 1.0, 2.0, T-P removal efficiency decreased by 62.1%, 57.4%, 51.3% respectively. The process by releasing the stored phosphorus in the anoxic reactor and uptaking the excess phosphorus in the aerobic reactor, occurred well comparatively when recycle ratio is 0.5. But this process did not occur when the recycle ratio is 1.0 and 2.0. And optimum pH of nitrification was about 6~7 and alkalinity decreased as nitrification rate increased. As increasing the recycle ratio in the anoxic reactor, DO concentration and ORP increased.

  • In order to study the influence of culture conditions on cell growth and lipid formation by Mucor sp, various carbon and nitrogen sources initial pH and C./N ratio of medium were investigated. Glucose was found to be suitable carbon source in terms of lipid yield and ${\gamma}$-linolenic acid(GLA) content. When NH4Cl and (NH4)2SO4 were used as nitrogen source lipid content was high(19-21%) but GLA content was low(15-17%) On the other hand when NaNO3 and KNO3 were used lipid content was low(about 13%) but GLA content was high(22-23%). The highest production of lipid was obtained at a C/N ratio of 40 using glucose and (NH4)2SO4 as carbon and nitrogen source respectively. it was found that lipid yield was high at pH4.6 Also this fungus did not grow at 35$^{\circ}C$ and lipid yield was highr at 15$^{\circ}C$ than $25^{\circ}C$.

A Study on Optimum Rate of Cattle Slurry Application for Forage Production using Idled Arable Land. I. The effect of applying time of cattle slurry and fertilization added mineral nitrogen on dry matter yield in Orchardgrass (유휴지에서 조사료 생산을 위한 적정 가축분뇨의 시용에 관한 연구 I. 액상구비의 시용시기와 무기태 질소의 첨가가 Orchardgrass의 건물수량에 미치는 영향)

  • The aim of this experiment was to investigate the effects of applying time of cattle slurry and fertilizing added mineral nitrogen on dry matter yield of orchardgrass and also to estimate proper levels of fertilizing nitrogen when idled arable land rapidly increased these days, which was used for the production of Orchardgrass. The results were as follows. 1. The annual yields of dry matter were produced 5.62~6.67 ton per ha when cattle slurry applied at rates of $30m^3$(average mineral fertilizer equivalent to 120kg) per ha in different dressing times. Those were higher 0.60~1.65 ton or 0.16 ton per ha than the yields from non-fertilizing or phosphrous and potassium fertilizing. 2. Relative yields of annual dry matter from mineral nitrogen fertilization of 120kg per ha were 142~146% in comparsion with control plots(cattle slurry plot or phosphrous and potassium plot). On the other hand, those of cattle slurry application were 112~133% in comparion with non-fertilizing plot. 3. The efficiencies of nitrogen on dry matter yield(kg DM/kg N) were 5.0~13.8 DM kg/N kg when cattle slurry applied to Orchardgrass at rates of $30m^3$(average mineral fertilizer equivalent to 120kg) per ha, and were 21.3~23.1kg DM/kg N at the same level fertilization of mineral nitrogen. 4. On the condition of this study, mineral nitrogen were fertilized after applying cattle slurry at rates of $30m^3$(average mineral fertilizer equivalent to 120kg) per ha in order to minimize chemical fertilizer and increase the potential production of Orchardgrass. At this situation, marginal dry matter yields of 8.48~8.63 ton per ha were obtained as the level of economic even point goint when mineral nitrogen fertilization is between 119.4 and 133.3kg per ha. It was possible to obtain the maximal dry matter yield(8.98 tons/ha) with fertilization of 202.8kg N/ha.

  • Hydrogenated amorphous carbon nitride[a-C:H(N)] films were deposited on p-type Si(100) at room temperature with bias voltage of 200 V by DC saddle-field plasma-enhanced chemical vapor deposition. Effects of the ratio of $N_2$ to $CH_4$($N_2/CH_4$), in the range of 0 and 4 on such properties as optical properties, microstucture, relative fraction of nitrogen and carbon, etc. of the films have been investigated. The thickness of the a-C:H(N) film was abruptly decreased with the addition of nitrogen, but at $N_2/CH_4$>0.5, the thickness of the film gradually decreased with the increase of the $N_2/CH_4$. The ratio of N to C(N/C) of the films was saturated at 0.25 with the increase of $N_2CH_4$. N-H, C≡N bonds of the films increased but C-H bond decreased with the increase of $N_2CH_4$.Optical band gap energy of the film decreased from 2.53 eV at the ratio of $N_2CH_4$=4.

Electronic Transitions and Ligand Field Analysis of mer-[N-(2-aminoethyl)-1,2-ethanediamine](glycylglycinato) chromium (III) Perchlorate (mer-[Cr(dien)(glygly)]$CIO_4$ 의 전자전이와 리간드장 해석)

  • The emission and excitation spectra of mer-[Cr(dien)(glygly)]ClO$_4$(glygly=glycylglycinate;dien=N-(2-aminoethyl)-1,2-ethanediamine) taken at 77 K are reported. The infrared and visible spectra at 298 K are also measured. The twelve electronic bands due to spin-allowed and spin-forbidden transitions are assigned. Using the observed transitions, a ligand field analysis has been performed to determine the bonding property of coordinated atoms in the chromium(III) complex. It is confirmed that the amine nitrogen atoms of the dien and glygly have strong $\sigma$-donor characters, but the peptide nitrogen of glygly has weak $\pi$-donor property toward chromium(III) ion.
