구기자 추출물 제조 시 백삼 및 홍삼 첨가에 의한 항산화활성 및 안지오텐신 전환효소에 대한 저해활성 효과 (Antioxidative Activity and Inhibition of Angiotensin Converting Enzyme by Lycii fructus Extracts Prepared by Adding White Ginseng and Red Ginseng)

  • Background: To enhance the taste and physiological characteristics of Lycii fructus (Gugija) extracts, we investigated the changes in the physiological characteristics of Gugija extracts caused by adding white ginseng (WG) and red ginseng (RG) Methods and Results: Gugija extracts, including 10G10, 10GW-G8 : 2, -G6 : 4, -G4 : 6, -G2 : 8, and -G0 (mixtures made by replacing 0, 20, 40, 60, 80, and 100% of Gugija with WG), as well as 10G10, 10GR-G8 : 2, -G6 : 4, -G4 : 6, -G2 : 8, and -G0 (mixture made by replacing 0, 20, 40, 60, 80, and 100% of Gugija with RG) were extracted with water at 10 times the respective mixture's volume. The antioxidant activities of Gugija extracts were investigated by assessing their 1,1-diphenyl-2-picrydrazyl (DPPH) and 2,2'-azinobis(3ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, ferric reducing antioxidant potential (FRAP) activity, nitrite scavenging activity, and angiotensin converting enzyme (ACE) inhibitory activity. As the amount of WG added increased, the DPPH, and, ABTS radical scavenging activity, and FRAP activity of the Gugija extract decreased. The half maximal inhibitory concentration ($IC_{50}$) value of 10G10, 10GW-G6 : 4, 10GR-G6 : 4, and 10GR-G0 for DPPH radical scavenging activity were $25.50{\pm}1.04$, $52.06{\pm}1.46$, $16.87{\pm}1.24$, and $9.50{\pm}0.16{\mu}{\ell}/m{\ell}$, respectively. On the other hand, the physiological activity of Gugija extract increased with the addition of increasing amounts of RG. However, ACE inhibitory activity was the highest ($50.25{\pm}2.58%$) in the Gugija 10-fold extract without any added RG. Conclusions: From the above results, we suggest that adding RG to Gugija extracts increase their antioxidant, FRAP, and nitrite scavenging activities.

추출방법에 따른 양파껍질 추출물의 품질특성 및 항산화활성 비교 (Quality characteristics and antioxidant activity of onion peel extracts by extraction methods)

  • 추출방법에 따른 양파껍질 추출물의 품질 특성 및 생리 활성 특성을 비교하였다. 환류냉각추출의 수율이 13.21%로 가장 높았고, 폴리페놀과 플라보노이드 함량은 가압가열추출이 가장 높게 나타났다. DPPH 라디칼 소거능 및 ABTS 라디칼 소거능 또한 가압가열추출이 가장 높게 측정되었다. ${\alpha}$-Glucosidase 저해 효과는 80.24%로 환류냉각추출이 가장 높은 저해 효과를 나타내었고, ACE 저해 효과는 27.05%로 저온고압추출이 가장 높았다. 아질산염 소거활성은 가압가열추출이 33.97%로 가장 높았고 다음으로 환류 냉각추출이 32.47%로 높은 활성을 나타내었다. Ferrous ion chelating 효과에서는 저온고압추출이 54.73%로 가장 높은 활성을 나타내었다. 이러한 결과를 종합해 볼 때 가압가열 추출이 소재 활용가치가 높을 것으로 사료되며 천연 항산화제 및 기능성 증진을 위한 소재로 이용 가능할 것으로 판단된다.

일당귀 에탄올 추출물의 항산화 효과 (Antioxidative Effect of Angelica acutiloba Kitagawa Ethanol Extract)

  • 본 연구는 일당귀의 생리활성 및 기능성을 검토하기 위하여 in vitro에서 일당귀 에탄올 추출물과 용매별 분획물에 대한 DPPH radical 소거능, 항산화 지수, 아질산염 소거능, linoleic acid에 대한 항산화 효과, 지질과산화 억제 효과를 측정하여 일당귀의 항산화력을 측정하였다. 일당귀 에탄올 추출물을 n-hexane, chloroform, ethylacerate, n-butanol, water로 계통 분획하여 DPPH radical에 대한 자유기 소거능을 측정한 결과 n-hexane 분획이 $39{\mu}g/ml$로 가장 강한 항산화 활성을 나타내었다. 또한 Rancimat로 측정한 항산화 지수도 분획 중 n-hexane 분획이 1.73으로 가장 높게 나타났으며, 아질산염 소거능에서도 모든 pH에서 n-hexane 분획이 가장 높게 나타났으며, pH 변화에 따른 각 분획의 아질산염 소거 작용은 pH 1.2에서 가장 우수하였다. 지질과산화물 생성 억제 효과는 n-hexane 분획이 87.7%로 가장 높았으나, 양성대조군으로 사용한 BHT의 억제율보다는 낮았다. Linoleic acid에 대한 항산화 효과를 과산화물가로 측정한 결과도 n-hexane의 항산화력이 가장 우수하였고 ethylaceta, chloroform, n-butanol, water 순이었다. 이상의 결과 일당귀 추출물은 항산화 효과를 나타내는 물질을 함유하고 있는 것으로 판단되며, 이 원인물질에 대한 연구가 더 진행되어야 할 것이며, 일당귀 추출물의 in vitro 에서 항산화효능의 우수함을 통해 일당귀의 천연항산화제로서 개발 및 활용성을 기대해 본다.

미숙사과 용매 추출물의 항산화성 (Antioxidant Properties of Unripened Apple Extracts)

  • The purpose of this study was to investigate the antioxidant properties of unripened apple extracts. The amount of total flavonoids in the peel and 5/30 samples were 2.7times and 5.0 times higher than the flesh and 6/30 samples, respectively. The degree of angintensin I converting enzyme (ACE) inhibition activity in hot water extracts was higher than the other extracts. In the 5/30 sample, a $0.1\%$ concentration of unripened apple powder showed almost the same electron-donating ability with a $0.5\%$ concentration of the 6/30 sample. More than $90\%$ of the electron-donating ability was observed from the peel extracts regardless of solvents used for extraction. The nitrite-scavenging effects of acetone and methanol extracts of the 5/30 sample were the strongest at pH 1.2, and especially the $80\%$ methanol extracts exhibited a powerful scavenging effect of more than $90\%$ at pH 3.0.

Antioxidant Activities of Methanol Extracts from Root Parts of Korean Salad Plants

  • Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

Antioxidant Activity of Saururus chinensis Pretreated by Mechanochemical Technology - Nitrite Scavenging and Electron Donating Ability -

  • Saururus chinensis is used as a raw material of an anti-rheumatic, poultice, sedative and stomachic. The tea of dried Saururus chinensis leaves has a function of relaxing pains in back and breast and of treating stomach ailments. The plant has the functions of anti-inflammatory, depurative, diuretic, febrifuge and refrigerant. The extraction of functional material is carried out by methanol and ethanol solvents. Mechanochemical grinding is applied as a pretreatment process before extraction to enhance the extraction efficiency by increasing the surface area of the materials while changing the chemical properties of the materials. Extraction of functional materials from the Saururus chinensis after grinding as a pretreatment using the mechanochemical technology was accomplished in this study to investigate the effect of grinding on the antioxidant activities of the extract. Saururus chinensis was ground by the planetary ball mill and the morphology was analyzed by SEM. Mechanochemical pretreatment increased the yields of functional materials from 5.9 g to 6.4, 7.0, and 8.1 g after grinding of 30 minutes, 1 hour and 2 hours, respectively. Nitrite scavenging ability increased from 53.0-71.0% to 61.0-79.0% for the methanol extraction in the solution of pH 1.2. Also, Electron donating abilities were increased from 7.90-33.51% to 11.97-38.51% for the methanol extraction. The extract concentration for the half inhibition of DPPH radicals was reduced from $278{\mu}g/mL$ in the original sample to $263{\mu}g/mL$ in the sample after grinding for the methanol extraction.

마늘 추출물이 Nitrosamine 생성반응에 미치는 영향 (Inhibitory Effects of Garlic Extracts on the Nitrosation)

  • 마늘 추출물의 아질산염 소거능과 항산화 효과 및 nitrosamine 생성 억제 작용에 대해 조사한 결과는 다음과 같다. 아질산염 소거작용은 마늘의 물 추출물과 에탄올 추출물에서 모두 pH가 낮을수록 높게 나타났고, pH가 높을수록 소거작용이 떨어졌으며, 특히 에탄올 추출물 첨가시 pH 1.2에서 89.6%의 뛰어난 소거효과를 나타냈다. 이들 마늘 추출물은 전자공여작용에도 높은 호과를 나타내었는데, 에탄올 추출물이 물 추출물 보다 높았으며, linoleic acid의 항산화 효과도 에탄을 추출물이 우수한 것으로 나타났다. Nitrosation 반응에서는 에탄올 추출물 첨가시 pH 1.2 및 3.0에서 80% 이상, 그리고 pH 6에서는 29.9%의 억제 효과가 있는 것으로 나타나 반응 pH에 따라 억제효과의 차이가 컸다.

마치현 열수 및 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Hot Water and Ethanol Extracts from Portulacae Herba)

  • Objectives : This study aims to provide basic data about Portulacae Herba (PH) extracts as natural antioxidants by considering diverse antioxidant activities of PH depending on solvents. Methods : The samples of PH were pulverized, and A hot water and a 70% EtOH were stir-extracted for two hours three times repeatedly in a water bath with a temperature of 95 degrees and at room temperature respectively to measure 7 kinds of antioxidant activities. Results : There were significant differences in total phenol content, because the total phenol content of the 70% EtOH extract was higher than the hot water extract's, and the total flavonoid content of the 70% EtOH extract($4.40{\mu}g/mg$) was nearly 3.8 times higher than the hot water extract's($1.16{\mu}g/mg$). DPPH and ABTS radical scavenging activities were 70% EtOH extract showed a little higher activity than the hot water extract, and at a concentration of $500{\mu}g/mg$, the highest scavenging activity was found in the 70% EtOH extract, not in the control group. Hydroxyl radical and $Fe^{2+}$ chelatingactivities were slightly higher in hot water extract than in 70% EtOH extract, and increased in a dose-dependent manner. Nitrite scavenging activities increased dose-dependently in the hot water and the 70% EtOH extract, regardless of the pH level, and scavenging activity of the 70% EtOH extract was higher at pH 1.2 than at pH 3.0. Conclusions : In conclusion, it is thought that PH hot water and 70% EtOH extract have antioxidant activities, and can be used as natural antioxidants in future.

국내 야생버섯의 항산화 활성 및 베타글루칸 함량 분석 (Antioxidant activities and β-glucan contents of wild mushrooms in Korea)

  • 국내 자생하는 야생버섯 추출물의 항산화 활성 및 베타글루칸 함량을 알아보기 위하여 버섯추출물에 대한 DPPH 라디컬 소거능, $ABTS^+$ 라디컬 소거능, 총 폴리페놀 함량, 베타글루칸 함량을 분석하였다. 수집 야생버섯 중에서 DPPH 라디컬 소거능과 ABTS 라디컬 소거능이 높은 버섯은 간버섯속(OK1071), 명아주자작나무버섯(OK1090), 등갈색미로버섯(OK1094)이었다. 아질산염 소거능과 총 폴리페놀 함량은 간버섯속(OK1071)이 각각 74.2%와 37.7 mg GAE/g의 값을 나타내며 가장 높았다. 베타글루칸 함량은 조개껍질버섯(OK1040), 구름송편버섯(OK1044), 간버섯속(OK1071)이 각각 47.8%, 43.9%, 41.8%로 높은 수치를 나타냈다. 본 연구결과로부터 야생 버섯들 중에서 항산화 활성 등이 우수한 버섯자원들이 존재하는 것을 확인하였으며, 기능성 소재로서의 활용가능성이 높을 것으로 기대한다. 더 나아가 국내 버섯산업의 확대를 위하여 토종 버섯자원을 이용한 새로운 천연물 유래 생리활성 물질로 활용하기 위한 기초자료로 활용도가 높으리라 기대된다.

Antioxidant Properties and Total Phenolic Contents of Cherry Elaeagnus (Elaeagnus multiflora Thunb.) Leaf Extracts

  • In Korea and China, cherry elaeagnus (Elaeagnus multiflora Thunb.) has been used traditionally to treat cough, diarrhea, itching, and foul sores. Therefore, in this study, the ethanol and water extracts of cherry elaeagnus leaves were examined for their antioxidant activities. The ethanol extract of the cherry elaeagnus leaves contained more phenolics than the water extract. All the cherry elaeagnus leaf extracts had higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability than ascorbic acid at concentrations of $250-1,000\;{\mu}g/mL$. The ethanol extract also showed higher superoxide dismutase (SOD)-like activity compared to the water extract. Furthermore, the SOD-like activity of the ethanol extract amounted to 89% of that of ascorbic acid at a concentration of $500\;{\mu}g/mL$. The nitrite scavenging ability and xanthine oxidase inhibitory (XOI) activity of the ethanol extract were higher than those of the water extract. In particular, the ethanol extract had higher XOI activity than ascorbic acid at a concentration of $1,000\;{\mu}g/mL$.
