Antibodies against Nitric Oxide Damaged Poly L-Tyrosine and 3-Nitrotyrosine Levels in Systemic Lupus Erythematosus

  • Alterations in the amino acid structure or sequence can generate neo-epitopes from self-proteins causing autoaggressive immune attack. Reactive nitrogen species are an important factor that induces post-translational modification of proteins by cellular reduction and oxidation mechanism; cysteinyl-nitrosylation or tyrosine nitration leading to potentially pathogenic pathways. It was thought of interest to investigate the immunogenicity of nitrated poly L-tyrosine vis-$\`{a}$-vis its possible role in the induction of antibodies in systemic lupus erythematosus (SLE). Commercially available poly L-tyrosine was exposed to nitrating species and the damage was monitored by UV spectroscopy and alkaline gel electrophoresis. The results indicated the formation of 3-nitrotyrosine. Nitrated poly L-tyrosine induced higher titre antibodies as compared to the native form. Nitrated poly L-tyrosine was recognized by the autoantibodies present in the sera of patients suffering from SLE by enzyme immunoassays and band shift assay. The possible role of nitrated self-proteins has been discussed in the production of circulating anti-DNA antibodies in SLE.

Antioxidative Properties of Sachil-Tang Extract

  • Sachil-Tang (SCT) has been traditionally used as a prescription of spasm of the esophagus by stress, pectoralgia and oppressive feeling of the chest in Oriental medicine. This study was carried out to investigate the antioxidant activities of the ethanol extract of SCT and its inhibitory effect on intracellular oxidation and vascular cell adhesion molecule-1 expression in human umbilical vein endothelial cells (HUVECs) using various methods. The SCT extract showed a strong inhibitory effect on free radical generating model systems, including DPPH radical, superoxide anions, hydroxyl radical, peroxynirite and nitric oxide. Besides, the SCT extract exhibited a strong inhibitory effect on lipid peroxidation in rat liver homogenate induced by $FeCl_2$-ascorbic acid, and protected plasmid DNA against the strand breakage in a Fenton's reaction system. The SCT extract also inhibited copper-mediated oxidation of human low-density lipoprotein (LDL), and repressed relative electrophoretic mobility of LDL. Furthermore, the SCT extract protected intracellular oxidation induced by various free radical generators and inhibited expression of vascular cell adhesion molecule-1 (VCAM-1) in HUVECs. These results suggest that SCT can be an effective natural antioxidant and a possible medicine of atherosclerosis.

Effects of KanghwalSokdantang(KS) on LDL Oxidation in Macrophage Cell (대식세포의 oxLDL 생성에 미치는 강활속단탕의 영향)

  • Objective : As a link in chain of research to confirm the oriental medical prescription which has the anti-atherosclerosis effects, this research evaluated the effects on the macrophage-related factors by using KanghwalSokdantang(KS). Methods : In order to perform this research, we have evaluated the effects on the oxLDL formation from the macrophages, the nitric oxide formation, and the oxidation of macrophages. Thus, with this evaluation, we have investigated the applicapability on the artherosclerosis. Results : KanghwalSokdantang has showed a noticeable reduction of protein oxidation in the process of oxLDL formation, has remarkably restrained phospholipid peroxidation, an index to estimated the phospholipid oxidation and reduction that are formed in the process of macrophage's oxLDL formation, and has increased the nitrite concentration noticeably in the LDL-dealing macrophages. By increasing the survival rate of macrophages, KanghwalSokdantang has restrained the cellular damages. KanghwalSokdantang is ineffective on the LDH outflow from damaged cells. $1{\mu}g/ml$ KanghwalSokdantang sample has increased acid phosphatase activity remarkably. Conclusion : KanghwalSokdantang has the possibility to be used in the prevention and treatment of atherosclerosis, which is formed by the oxLDL formation of macrophages.

Synthesis of CNFs(Carbon Nanofibers)/DAAQ electrode for Supercapacitor (슈퍼커패시티용 DAAQ/CNFs 전극의 제조)

  • A new type electric double layer capacitor (EDLC) was constructed by using carbon nanofibers (CNFs) and DAAQ(1,5-diaminoanthraquinone) electrode. Carbonaceous materials are found in variety forms such as graphite, diamond, carbon fibers etc. While all the carbon nanofibers include impurities such as amorphous carbon, nanoparticles, catalytic metals and incompletely grown carbons. We have eliminated of Ni particles and some carbonaceous particles in nitric acid. Nitric acid treated CNFs could be covered with very thin DAAQ oligomer from the results of CV and TG analyses and SEM images. A crystalline supramolecular oligomer of 1,5-diaminoanthraquinone(DAAQ) was successfully prepared as a thin film by electrochemical oxidation from an acidic non-aqueous medium. DAAQ oligomer film exhibited a specific capacity as 45-50 Ah/kg in 4M $H_2SO_4$. Its electrochemical characteristics were investigated by cyclic voltammetry. And compared with different electrolyte of aqueous type. During ultrasonic irradiation CNFs was to disperse in 0.1M $H_2SO_4$. As a result, CNFs coated by DAAQ composite electrode showed relatively good electrochemical behaviors.

Change of Hydroponic Components by Plasma Treatment (플라즈마 처리에 의한 양액 성분 변화)

  • The influence of plasma discharge on the nutrient components ($NO_3$-N, $NH_4$-N, $PO_4$-P, K, Ca, and Mg) and water quality [pH, ORP (oxidation-reduction potential) and electric conductivity] of hydroponic water were investigated. It was observed that the $NH_4$-N, $PO_4$-P, K, Ca, and Mg were kept at the constant concentrations for plasma discharging of 90 min. On the other hand, $NO_3$-N concentration was increased with proceeding of the plasma discharge. The increase of $NO_3$-N concentration was considered with the fact that nitric acid was created from nitrogen among supplying air for the insulation of inside of dielectric barrier. ORP and electric conductivity was increased with plasma discharging time. However, pH was decrease with what because of the generation of the nitric acid. When adjusting the hydroponic ingredients, pH and conductivity must to be considered because of the change of pH and conductivitiy with the discharging.

  • The dynamics of gas release at thermal oxidation of extraction systems on the basis of diamides of dicarbonic acids in fluorinated sulphones with 14 mol/L $HNO_3$ was investigated. The effect of preirradiation of the mixtures with accelerated electrons on the kinetics of their thermolysis was determined. The mixtures were heated in an autoclave at temperatures of 170 and $200^{\circ}C$ and irradiated using an electron accelerator to absorbed doses of 0.1, 0.5, and 1.0 MGy. It has been shown that no conditions for autocatalytic oxidation at thermolysis of extraction mixtures irradiated up to a dose of 1 MGy were developed.

Supercritical Water Oxidation of Anionic Exchange Resin (초임계수 산화를 이용한 음이온교환수지 분해)

  • The characteristics of supercritical water oxidation have been studied to decompose the waste anionic exchange resins which were produced from a power plant. The waste resins from a power plant were mixture of anionic and cationic exchange resins. The waste anionic exchange resins had been separated from the waste resins using a solid-liquid fluidized bed. It was confirmed that the cationic exchange resins were not included in the separated anionic exchange resins by the elemental and thermogravimetric analysis. A slurry of anionic exchange resins which could be fed continuously to a supercritical water oxidation apparatus by a high pressure pump was prepared using a wet ball mill. Although the COD of liquid effluent had been reduced more than 99.9% at 25.0 MPa and $500^{\circ}C$ within 2 min, the total nitrogen content was reduced only 41%. The addition of nitric acid to the slurry could reduce the total nitrogen content in treated water. The central composite design as a statistical desist of experiments had been applied to optimize the conditions of decomposing anionic resin slurry by means of the COD and total nitrogen contents in treated waters as the key process output variables. The COD values of treated waters had been reduced sufficiently to $99.9{\sim}100%$ af the reaction conditions of $500{\sim}540^{\circ}C$, 25.0 MPa within 2 min. The effects of temperature and nitric acid concentration on COD were not significant. However, the effect of nitric acid concentration on the total nitrogen was found to be significant. The regression equation for the total nitrogen had been obtained with nitric acid concentration and the coefficient of determination($r^2$) was 95.8%.

Total Phenolic Contents, Radical Scavenging Capacities and Inhibitory Effects on Lipid Peroxidation and LDL Oxidation of Prunus persica Branch

  • This study was undertaken to elucidate the antioxidant activity of the ethanol (EEPB) and water (WEPB) extracts of Prunus persica branches. The extracts contained a high phenolic content and revealed a potent hydrogen donating activity in DPPH scavenging assay. Compared to $\alpha$-tocopherol, EEPB (p < 0.001) and WEPB (p < 0.05) significantly inhibited $FeCl_2$-ascorbic acid-induced lipid peroxidation, and also exhibited potent antiradical activities against hydroxyl radical, superoxide anion, nitric oxide and peroxynitrite. In copper- and AAPH-mediated human low-density lipoprotein (LDL) oxidation systems, the extracts demonstrated a strong antioxidant function by metal chelating, rather than direct scavenging, action. Furthermore, EEPB at 5 ${\mu}g/mL$ concentration showed 80.77% inhibition of the electrophoretic mobility of LDL, compared to 77.69% for ascorbic acid and 76.92% for BHT. These results suggest that PB branch extracts may protect against oxidative stress-induced diseases.

Facile Separation of Zinc Oxalate to Oxalate and its Conversion to Glycolic Acid via Electrochemical Reduction (ZnC2O4의 Oxalate로의 효과적 분리 및 이의 전기화학적 환원을 통한 글리콜산으로의 전환)

  • Oxalic acid has been traditionally obtained via the oxidation of carbohydrates using nitric acid and catalysts. However, this process produces a variety of nitrogen oxides during oxidation and requires a separation process due to its various intermediates. These products and additional steps increase the harmfulness and complexity of the process. Recently, the electrochemical reduction of carbon dioxide into oxalic acid has been suggested as an environmentally friendly and efficient technology for the production of oxalic acid. In this electrochemical conversion system, zinc oxalate (ZnC2O4) is obtained by the reaction of Zn2+ ions produced by Zn oxidation and oxalate ions produced by CO2 reduction. ZnC2O4 can then be converted to form oxalic acid, but this requires the use of a strong acid and heat. In this study, a system was proposed that can easily convert ZnC2O4 to oxalic acid without the use of a strong acid while also allowing for easy separation. In addition, this proposed system can also further convert the products into glycolic acid which is a high-value-added chemical. ZnC2O4 was effectively separated into Zn(OH)2 powder and oxalate solution through a chemical treatment and a vacuum filtration process. Then the Zn(OH)2 and oxalate were electrochemically converted to zinc and glycolic acid, respectively.

Equilibrium Removal of Pb (II) Ions from Aqueous Solution onto Oxidized-KOH-Activated Carbons

  • In the present study, the removal of Pb (II) ions on oxidized activated carbons (ACs) was investigated. ACs were derived from activation of indigenous cotton stalks waste with potassium hydroxide (KOH) in two-stage process. The KOH-ACs were subjected to liquid-phase oxidation with hot $HNO_3$ and one untreated sample was included for comparison. The obtained carbons were characterized by Fourier transform infrared (FTIR), slurry pH and $N_2$-adsorption at 77 K, respectively. Adsorption capacity of Pb (II) ions on the resultant carbons was determined by batch equilibrium experiments. The experimental results indicated that the oxidation with nitric acid was associated with a significant increase in mass of yield as well as a remarkable reduction in internal porosity as compared to the untreated carbon. The AC-800N revealed higher adsorption capacity than that of AC-800, although the former sample exhibited low surface area and micropore volume. It was observed that the adsorption capacity enhancement attributed to pore widening, the generation of oxygen functional groups and potassium containing compounds leading to cation-exchange on the carbon surface. These results show that the oxidized carbons represented prospective adsorbents for enhancing the removal of heavy metals from wastewater.
