Rice is the most important cereal crop not only in supplying the basic staple food for more than half of the world's population but also as a model plant for functional genomic studies of monocotyledons. Although rice transformation method using A. tumefaciens has already been widely used to generate transgenic plants, the transformation rate is still low in most Korean elite cultivars. We made several modifications of the standard protocol especially in the co-cultivation step to improve the efficiency of the rice transformation. The co-culture medium was modified by the addition of three antioxidant compounds (10.5 mg/L L-cysteine, 1 mM sodium thiosulfate, 1 mM dithiothreitol) and of Agrobacterium growth-inhibiting agent (5 mg/L silver nitrate). Co-cultivation temperature ($23.5^{\circ}C$ for 1 day, $26.5^{\circ}C$ for 6 days) and duration (7 days) were also changed. The plasmid of pMJC-GB-GUS carrying the GUS reporter gene and the bar gene as the selectable marker was used to evaluate the efficiency of the transformation. After co-cultivation, a high level of GUS gene expression was observed in calli treated with the modified method. It is likely that those newly added compounds helped to minimize the damage due to oxidative bursts during plant cell-Agrobacterium interaction and to prevent necrosis of rice cells. And the transformation rate under the modified method was also remarkably increased approximately 8-fold in Heungnambyeo and 2-fold in Ilmibyeo as compared to the corresponding standard method. Furthermore, we could produce the transgenic plants stably from Ilpumbyeo which is a high-quality rice but its transformation rate is extremely low. Transformation and the copy number of transgenes were confirmed by PCR, bar strip and Southern blot analysis. The improved method would attribute reducing the effort and the time required to produce a large number of transgenic rice plants.

The effects of macronutrients $(NO_3^-,\; NH_4^+\;and\;PO_4^{3-})$ on cell growth and triterpenoids production in Centella asiatica cell suspension cultures were analyzed using the BoxBehnken response surface model experimental design. In screening and optimization experiments, $PO_4^{3-}$ as a single factor significantly influenced cell growth where increasing the phosphate level from 0.1 to 2.4 or 2.6 mM, elevated cell growth from 3.9 to $14\~16g/L$. The optimum values predicted from the response surface model are 5.05mM $NH_4^+$, 15.0mM $NO_3^-$ and 2.6mM $PO_4^{3-}$, yielding 16.0g/L cell dry weight with $99\%$ fitness to the experimental data. While the $NH_4^+-NO_3^-$ interaction influenced cell growth positively in the optimization experiment, $NH_4^+$ and $NO_3^-$ as single factors; and interactions of $NO_3^--PO_4^{3-},\;NH_4^+-PO_4^{3-}$ and $NH_4^+-NO_3^-$ were all negative in the screening experiment. Cell growth and the final pH level were positively affected by $PO_4^{3-}$, but negatively affected by $NH_4^+\;and\;NH_4^+-PO_4^{3-}$ interactions. The different effects of factors and their interactions on cell growth and final pH are influenced by a broad or narrow range of macronutrient concentrations. The productions of triterpenoids however were lower than 4mg/g cell dry weight.
The positive matrix factorization (PMF2) and multilinear engine (ME2) models have been shown to be powerful environmental analysis techniques and have been successfully applied to the assessment of ambient particulate matter (PM) source contributions. Because these models are difficult to apply practically, the US EPA developed a more user-friendly version of the PMF. The initial version of the EPA PMF model does not provide any rotational capabilities; for this reason, the model was upgraded to include rotational functions in the EPA PMF ver. 2.0. In this study, PMF and EPA PMF modeling identified ten particulate matter sources including secondary sulfate I, vehicle gasoline, secondary sulfate II, secondary nitrate, secondary sulfate III, incinerators, aged sea salt, airborne soil particles, oil combustion, and diesel emissions. All of the source profiles determined by the two models showed excellent agreement. The calculated average concentrations of $PM_{2.5}$ were consistent between the PMF2 and EPA PMF ($17.94{\pm}0.30{\mu}g/m^3$ and $17.94{\pm}0.30\;{\mu}g/m^3$, respectively). Also, each set of estimated source contributions of the PMF2 and EPA PMF showed good agreement. The results from the new EPA PMF version applying rotational functions were consistent with those of PMF2. Therefore, the updated version of EPA PMF with rotational capabilities will provide more reasonable solutions compared with those of PMF2 and can be more widely applied to air quality management.

The purpose of this study was to identify $PM_{2.5}$ sources and to estimate their contributions to the border of Yongin-Suwon area, based on the analysis of the $PM_{2.5}$ mass concentration and the associated inorganic elements, ions and carbon components. The contribution of $PM_{2.5}$ sources were estimated by using a positive matrix factorization (PMF) model to identify air emission sources. For this study, $PM_{2.5}$ samples were collected from May, 2007 to April, 2008. The inorganic elements were analyzed by an ICP-AES. The ionic components in $PM_{2.5}$ were analyzed by an Ie. The carbon components were also analyzed by DRI/OGC analyzer. After performing PMF modeling, a total of 12 sources were identified and their contributions were quantitatively estimated. The contributions from each emission source were as follows: 11.3% from oil combustion source, 3.4% from bus/highway source, 5.8% from diesel vehicle source, 4.7% from gasoline vehicle source, 8.8% from biomass burning source, 15.1 % from secondary sulfate, 5.2% from secondary nitrate source, 13.4% from industrial related source, 4.1% from Cl-rich source, 19.6% from soil related source, 1.0% from aged sea salt, and 7.4% from coal combustion source, respectively. This study provides basic information on the major sources affecting air quality, and then it will help to effectively control $PM_{2.5}$ in this study area.

Water content of $PM_{2.5}$ (particles in the atmosphere with a diameter of less than or equal to a nominal $2.5{\mu}m$) was estimated by using a gas/aerosol equilibrium model, SCAPE2, for the particles collected at Seoul and Gosan, Korea. From measured and analyzed characteristics of the particles, the largest difference between Seoul and Gosan is the proportions of total ammonia (t-$NH_3$=gas phase $NH_3$+particle phase ${NH_4}^+$), total nitric acid (t-$HNO_3$=gas phase $HNO_3$+particle phase ${NO_3}^-$) and sulfuric acid ($H_2SO_4$). Even though both sites have sufficient t-$NH_3$ to neutralize acidic species such as $H_2SO_4$, t-$HNO_3$, and t-HCl (total chloric acid=gas phase HCl+particle phase $Cl^-$), equivalent fraction of t-$NH_3$ and t-$HNO_3$ are higher at Seoul and $H_2SO_4$ is higher at Gosan. Based on the modeling result, it is identified that the $PM_{2.5}$ at Seoul is more hygroscopic than Gosan if the meteorological conditions are the same. To reduce water content of $PM_{2.5}$, and thus, mass concentration, control measures for ammonia and nitrate reduction are needed for Seoul, and inter-governmental cooperation is required for Gosan.

Soil aquifer treatment is a water reuse technology that secondary or tertiary treated wastewater is infiltrated into the aquifer in which physical and biochemical reactions occur. Major consideration in SAT is the removal and transport of DOC and nitrogen species. In this study, reaction mechanism in SAT was examined considering nitrification, denitrification and organic oxidation. In addition, SAT modeling system was developed as the reaction mechanism was applied to groundwater flow and transport model. In verification of the reaction module by 1-dimensional unsaturated soil column test, the experimental data of all of the species, ammonium, nitrate, DOC and DO, were well matched with the simulation results. In sensitivity analysis, ammonium partition coefficient, dissolved oxygen inhibition constant and biomass decay rate affect ammonium, DOC and DO concentration of effluent, respectively.
The objectives of the study were to evaluate seasonal patterns of epilimnetic water quality, and determine interannual eutrophication patterns at the dam site of Yong-dam Reservoir using long-term data during 2002~2009. Ionic dilutions, based on specific conductivity, occurred in the summer period in response to the intense monsoon rain and inflow, and suspended solid analysis indicated that the reservoir was clear except for the monsoon. Seasonality of nitrogen contents varied depending on the types of nitrogen and responded to ionic dilution; Ammonia-nitrogen ($NH_4$-N) peaked at dry season but nitrate-nitrogen ($NO_3$-N) peaked in the monsoon when the ionic dilution occurred. The maxima of $NO_3$-N seemed to be related with external summer N-loading from the watershed and active nitrogen fixation of bluregreens in the summer. $NO_3$-N was major determinant (>50%) of the total nitrogen pool and relative proportion of $NH_4$-N was minor. Long-term annual $NO_3$-N and TDN showed continuous increasing trends from 2004 to 2009, whereas TP and TDP showed decreasing trends along with chlorophyll-a (CHL) values. Empirical model analysis of log-transformed nutrients and N : P ratios on the CHL showed that the reservoir CHL had a stronger linear function with TP ($R^2$=0.89, p<0.001) than TN ($R^2$=0.35, p=0.120). Overall results suggest that eutrophication progress, based on TP and CHL, is slow down over the study period and this was mainly due to reduced phosphorns, which is considered as primary nutrient by the empirical model.

Accelerated aging test for the double base propellant was carried out at three different temperatures (60, 70, and $75^{\circ}C$) for over a year. To evaluate the aging characteristics of the double base propellant, the stabilizer contents and thermal decomposition kinetics were analyzed by using high performance liquid chromatography (HPLC) and AKTS-Thermokinetics software. As a result, stabilizer contents in the double base propellant gradually decreased according to the aging temperature and aging duration. The consumption rate of 2-NDPA in the accelerated aged propellants showed that it was two times faster at $75^{\circ}C$ in compared with ther rate at $70^{\circ}C$. These experimental values were simulated by the SB kinetic model, and it was shown that the two-step model with constant reaction orders n1=1 and n2=0 best describes the process of the stabilizer depletion for the double base propellant.

A one-dimensional numerical model was developed to simulate vertical profiles of electron acceptors and their reduced species in benthic sediments. The model accounted for microbial degradation of organic matter and subsequent chemical reactions of interest using stoichiometric relationships. Depending on the dominant electron acceptors utilized by microorganisms, the benthic sediments were assumed to be vertically subdivided into six zones: (1) aerobic respiration, (2) denitrification, (3) manganese reduction, (4) iron reduction, (5) sulfate reduction, and (6) methanogenesis. The utilizations of electron acceptors in the biologically mediated oxidation of organic matter were represented by Monod-type expression. The mass balance equations formulated for the reactive transport of organic matter, electron acceptors, and their corresponding reduced species in the sediments were solved utilizing an iterative multistep numerical method. The ability of model to simulate a freshwater sediments system was tested by comparing simulation results against published data obtained from lake sediments. The simulation results reasonably agreed with field measurements for most species, except for ammonia. This result showed that the C/N ratio (106/16) in the sediments is lower than what the Redfield formula prescribes. Since accurate estimates of vertical profiles of electron acceptors and their reduced species are important to determine the mobility and bioavailability of trace metals in the sediments, the model has potential application to assess the stability of selected trace metals in the sediments.

Groundwater in the Waikatoregion is a valuable resource for agriculture, water supply, forestry and industries. The 434,000 ha study area comprises the upper Waikato River catchment from the outflow of Lake Taupo (New Zealand's largest lake) through to Lake Karapiro (a man-made hydro lake with high recreational value) (Figure 1). Water quality in the area is naturally high. However, there are indications that this quality is deteriorating as a result of land use intensification and deforestation. Compounding this concern for decision makers is the lag time between land use changes and the realisation of effects on groundwater and surface water quality. It is expected that the effects of land use changes have not yet fully manifested, and additional intensification may take decadesto fully develop, further compounding the deterioration. Consequently, Environment Waikato (EW) have proposed a programme of work to develop a groundwater model to assist managing water quality and appropriate policy development within the catchment. One of the most important and critical decisions of any modelling exercise is the choice of the modelling platform to be used. It must not inhibit future decision making and scenario exploration and needs to allow as accurate representation of reality as feasible. With this in mind, EW requested that two modelling platforms, MODFLOW/MT3DMS and FEFLOW, be assessed for their ability to deliver the long-term modelling objectives for this project. The two platforms were compared alongside various selection criteria including complexity of model set-up and development, computational burden, ease and accuracy of representing surface water-groundwater interactions, precision in predictive scenarios and ease with which the model input and output files could be interrogated. This latter criteria is essential for the thorough assessment of predictive uncertainty with third-party software, such as PEST. This paper will focus on the attributes of each modelling platform and the comparison of the two approaches against the key criteria in the selection process. Primarily due to the ease of handling and developing input files and interrogating output files, MODFLOW/MT3DMS was selected as the preferred platform. Other advantages and disadvantages of the two modelling platforms were somewhat balanced. A preliminary regional groundwater numerical model of the study area was subsequently constructed. The model simulates steady state groundwater and surface water flows using MODFLOW and transient contaminant transport with MT3DMS, focussing on nitrate nitrogen (as a conservative solute).
