Influence of oxytetracycline on the fate of Nitrogen species in a recirculating aquaculture system

  • Common aquaculture practices include the use of certain pharmaceuticals such as antibiotics in avoiding diseases and promoting a healthier growth of the culture. The aim of this study is to monitor and assess the influence of different low oxytetracycline concentrations on the transformation of nitrogen compounds under aeration condition in a lab-scale recirculating aquaculture system (RAS). Over $1mg\;L^{-1}$ dose of oxytetracycline to aquaculture had induced ammonia($NH_4-N$), nitrate($NO_3-N$), soluble COD accumulation in RAS. In addition, nitrous oxide ($N_2O$) emission from RAS was significantly reduced during the oxytetracycline dose periods. After ceasing the dose of oxytetracycline, ammonia oxidation and nitrous oxide re-emission were observed. This observation indicated that low concentrations of oxytetracycline could affect the nitrogen species in RAS. Also, the emission mechanisms of $N_2O$ may not be only dependent on nitrification process but also dependent on denitrification process in our RAS system.

Nitroglycerin Biodegradation under Denitrification Conditions and Corresponding Microbial Community Shifts upon Acclimation (탈질조건에서 nitroglycerin의 생물학적 분해 동역학 및 미생물 군집 변화)

  • Biodegradation of an explosive compound, glyceryl trinitrate (GTN), was studied with a denitrifying microbial culture grown in a sequencing batch reactor and a GTN acclimated denitrifying culture. The GTN acclimated culture, which were fed on GTN for 1 month, degraded GTN regioselectively via denitration on C1 position as compared to C2 position denitration by denitrifying culture that has never been exposed to GTN. Accumulation of two isomeric glyceryl dinitrates (GDNs) in both culture medium suggests that GDN denitration is the rate-limiting step in GTN biodegradation. The first order GTN degradation rate normalized to cell concentration of the acclimated culture was calculated to be 0.045 (${\pm}0.002$) L/g-hr. Increasing concentration of electron acceptor(nitrate) resulted in discouraged GTN degradation. According to microbial community analysis, prolonged GTN exposure resulted in 25% increase in the genus level of the GTN acclimated culture with the disappearance of two dominating denitrifying microbial species of Methyloversatilis universalis and Hyphomicrobium zavarzinii in the denitrifying culture.

Growth and Yield of Strawberry $(Fragaria\;\times\;ananassa\;Duchesne)$ 'Nyoho' and Salt Accumulation in PE Film House Soil as Affected by Fertilization Program (시비방법이 무기염의 토양집적과 딸기 생육 및 수량에 미치는 영향)

  • This research was conducted to determine the effect of fertilization programs on crop growth, nutrient uptake and yield of strawberry and salt accumulation in PE film house soil. To achieve these, experiments were conducted through two years with various fertilization program such as traditional fertilization(A), recommended fertilization of Rural development administration (B), Tochigi prefectural experiment station (C) and Nonsan strawberry experiment station (D), and control (E). In 1st year, statistical differences in growth characteristics were not observed among treatments except (E). Yield per 10 was the highest in (D) followed by (A), (B), (C) and (E) and there was the statistical difference between (D) and (E). In the 2nd year experiment, growth characteristics and yield showed similar trends to those of 1st year experiment. In the results of soil analysis of 2nd year, the soil pH of all treatments were in the acceptable range, while electrical conductivity of (A) and (D) were 2.36 and 2.19 %$dS{\cdot}m^{-1}$, respectively. After finishing of strawberry cultivation, nitrate concentration in soil solution of (A) and (D) were 74.6 and 65.0%$mg{\cdot}L^{-1}$ and Na in those were 3.71 and 3.53 $cmol+kg^{-1}$, respectively. Above results indicated that (A) and (D) were good fertilization program for strawberry cultivation, but those also resulted in accumulation of $NO_{3-}N$ and Na in PE house soil.

Assessment of Environmentally Sound Function on the Increasing of Soil Fertility by Korean Organic Farming (한국 토착유기농업의 토양비옥도 증진책에 대한 환경보전적 기능 평가)

  • In order to get some basic data on environmental friendly function by Korean organic farming, the chemical characteristics of soil were determined on 100 farm cultivating site in Paldang watershed area of Great Seoul. The EC and content of $NO_3-N$ and Av. $P_2O_5$ in topsoil(0~30cm) showed $2.30dS\;m^{-1}$, $82mg\;kg^{-1}$, $918mg\;kg^{-1}$ in the soil cultivated chinese cabbage. $2.29dS\;m^{-1}$, $86mg\;kg^{-1}$, $954mg\;kg^{-1}$ in the soil of lettuce, $1.83dS\;m^{-1}$, $66mg\;kg^{-1}$, $1114mg\;kg^{-1}$ in the soil of crown daisy. These salt accumulation(EC) and the high concentration of mineral content in topsoil such as nitrate and phosphate showed the soils of organic farming were contaminated by practice of organic farming for the maintenance strategy of soil fertility. The $NO_3-N$ and Av. $P_2O_5$ in the subsoil(30~60cm) showed $75mg\;kg^{-1}$ and $641mg\;kg^{-1}$, $72mg\;kg^{-1}$ and $466mg\;kg^{-1}$, $42mg\;kg^{-1}$ and $873mg\;kg^{-1}$ in soil cultivated chinese cabbage, lettuce and crown daisy respectively. It indicates eventually the high concentration of nitrate and phosphate in topsoil caused penetration to subsoil, and the high concentration of mineral contents in subsoil indicate the potential risk of leaching of ground water by Korean organic farming. The positive correlation at 1% between EC and $NO_3-N$, $K_2O$, T-C, $P_2O_5$ and T-N show the salt accumulation in the both soil depth of Korean organic farming were caused by minerals such as $NO_3-N$, $K_2O$, T-C, $P_2O_5$ and T-N by overuse of organic fertilizer.

Dry Matter, Nitrogen Distribution and Organic Reserves Accumulation as Affected by Nitrate Supply Level in Alfalfa (Medicago sativa L.) (질산태 질소의 공급수준에 따른 알팔파의 건물, 질소의 분포 및 저장영양소의 축적)

  • To investigate the effects of the exogenous N $O_3$$^{[-10]}$ supply level on the nitrate assimilation and growth during the vegetative growth stage, and on the accumulation of organic reserves during the successive regrowth period, dry matter (DM), the amount of nitrogenous compounds, total N and starch in alfalfa plants grown hydroponically with 0.2, 1.0 and 3.0 mM KN $O_3$ was estimated, respectively, during vegetative growth period and two cycle of regrowth. When compared with DMs and N contents in various N compounds in the organs grown with 1.0 mM N $O_3$$^{[-10]}$ , N starvation symptoms were found in 0.2 mM and a depressive effect was observed in 3.0 mM after 10 weeks of vegetative growth. Total starch content in root system gown with 0.2, 1.0 and 3.0 mM N $O_3$$^{[-10]}$ during the first regrowth was 50.96, 15.47 and 6.37 mg plant$^{-1}$, respectively. Starch was contained mainly in taproots. The starch content was not significantly changed by 24 days of the second regrowth with 1.0 mM N $H_4$N0$_3$. Total nitrogen content in root system grown with 0.2, 1.0 and 3.0 mM N $O_3$$^{[-10]}$ during the first regrowth was 6.66, 8.43 and 11.09 mg plant$^{-1}$ , respectively. Nitrogen was contained mainly in lateral roots; 80% (in 0.2 mM), 74% (1.0 mM) and 76% (3.0 mM) of total nitrogen in root system. Total N content in root system at the end of the second regrowth also closely affected by the N $O_3$$^{[-10]}$ supply level during the first regrowth. These results suggest that the level of N $O_3$$^{[-10]}$ may strongly influence the accumulation of organic reserves in root system, and that the initial level of organic reserves for the successive regrowth was one of the determinants for shoot regrowth.

Survey on the Green house Flower Soil Chemicophysical Properties and Amount of Fertilizers and Soil Amendment Applications (시설화훼(施設花卉) 재배지(栽培地) 토양(土壤)의 이화학성(理化學性)과 화학비료(化學肥料) 및 토양개량제(土壤改良制) 시용량(施用量) 조사(調査))

  • This study was conducted to obtain basic information for soil improvement in flower crop cultivating greenhouse soil through survey on the chemical and physical properties of greenhouse soils. Total of 85 Flowcultivating farms were surveyed and analysis was done on the soil characteristics, amounts of chemical fertilizer and soil amendmentuse. The result are as follows: In soil properties of flower cultivating greenhousees, silt clay loam was 51%and 68% of the surveyed soils had good drainage condition. Ground water table was over 90-120cm which was optimum range for flower cultivation. Flower cultivating farms had problem with accumulation of fertility. Nitrate nitrogen was accumulated in Gypsophila paniculate farms and available phosphorus, and exchangeable postassium were significantly higher in greenhouse soils about 2 times than in open field soil. Application amount of chemical feltilizers in greenhouses were nitrate 211,phosphorus 135, and potassium 132kg/ha, respectively. Amount of organic matter used in greenhouse were high in order of cattle manure> compost> organic fertilizer> poultry manure> swine manure and their application amounts were69, 103, 32, 20, and 43 MT/ha, respectively.

Effect of Rice Straw and Woodchip Application on Greenhouse Soil Properties and Vegetable Crops Productivity (볏짚과 파쇄목 시용이 시설하우스 토양 성질과 작물 수량에 미치는 영향)

  • There have been increasing concerns about decreasing crop productivity due to salt accumulation in greenhouse soils. The objective of the study was to investigate the impact of rice straw and woodchip application to a salt accumulated greenhouse soil on crop productivity and soil quality. The application of rice straw (RS) and woodchip (W) increased tomato yield and decreased blossom-end rot, and increased yield of Chinese cabbage compared with standard recommended fertilization ($204-103-122kg\;ha^{-1}\;N-P_2O_5-K_2O$ for tomato and $222-64-110kg\;ha^{-1}\;N-P_2O_5-K_2O$ for Chinese cabbage), while less soil residual nitrate, phosphatephosphorus, and potassium. In addition to the organic material application, fertilization reduction based on soil testing may also contribute to relatively low level of soil residual nutrients. Application of the organic material reduced soil bulk density presumably because of improved soil aggregation and structure, and increased biomass C and dehydrogenase activity. In comparison to rice straw, woodchip application resulted in higher crop yield, less amount of soil residual nitrate and lower soil EC, and greater biomass and dehydrogenase activity. The results obtained in this study indicateshowed that woodchip amendment along with reduced fertilization based on soil testing can be one of essential management practices for salt accumulated greenhouse soils.

Effects of Application Amount of Organic Compound Fertilizer on Lettuce Growth and Soil Chemical properties under Plastic film house (시설재배지에서 유기복합비료 시용량에 따른 상추 생육 및 토양화학성에 미치는 영향)

  • The Project supporting organic fertilizer started in 1999 as a national policy. In farmhouse, over application of mixed organic compound fertilizer(OC) caused salt accumulation in plastic film house soil. To replace inorganic fertilizer with OC fertilizer, this study was investigated the effect of OC application on yield and soil chemical properties for lettuce cultivation in plastic film house. The OC fertilizer was applied at 50(OC50+N50), 100(OC100), and 150(OC150) % level of the basal amount of nitrogen fertilizer in soil testing recommendation. And these were compared to NPK(nitrogen, phosphat, and potash fertilizer) and PK treatment. The yield of lettuce in OC100 was similar to that of NPK treatment. In OC 50, 100 and 150 treatments, pH had a tendency to increase than that of NPK treatment. Nitrate nitrogen(NO3-N) and electrical conductivity(EC) were similar to NPK treatment. These showed that nutrients from OC fertilizer were less likely to accumulate in soil than NPK. Also, use efficiency of nitrogen in OC100 treatment were similar to NPK treatment. These results suggest that OC application as the basal dressing at the 100% level could be best to prevent a nutrient accumulation of soil and to increase the yield and commercial quality for lettuce.

Effects of Mycorrhizal Inoculation on Plant Growth and N Metabolites in Relation to drought-stress Tolerance (Mycorrhiza 접종이 가뭄 스트레스하의 식물성장과 질소 대사산물에 미치는 영향)

  • The effects of arbuscular mycorrhizal (AM) fungus (Glomus intraradices) on plant growth and N metabolic responses were examined in perennial ryegrass plants exposed to drought-stressed or well-watered condition. Mycorrhizal inoculation improved significantly leaf water potential, dry mass and P content. Drought stress increased significantly nitrate concentration in roots where the increase was much less in AM than non-AM. Drought stress decreased the concentration of soluble proteins in non-AM shoots, whereas non-significant decline occurred in AM shoots even under drought condition. The concentrations of ammonia and proline in drought stressed non-AM plants significantly increased, while mycorrhizal inoculation lowered significantly ammonia and proline accumulation. The decrease in leaf dry weight in drought stressed-plants was significantly correlated to the increase in ammonia (p<0.01) and proline concentration (p<0.01). These results suggested that the increased P content and N assimilation by mycorrhizal inoculation may be associated with drought stress tolerance, showing the moderating effects on shoot growth inhibition and ammonia accumulation in drought stressed-plants.

Salt Accumulation and Desalinization of Rainfall Interception Culture Soils of Rubus sp. in Gochang-gun, Jeollabuk-do (복분자 비가림 하우스 토양 중 염류집적 요인과 물리적 제염효과)

  • This study was carried out to investigate the factors of desalinization of the rainfall interception culture soils of Rubus sp. in Gochang-gun, Jeollabuk-do. Soil samples were collected from 85 different sites of the rainfall interception culture soils of Rubus sp. in Gochang-gun, Jeollabuk-do. The electrical conductivity in paste saturation of rainfall interception culture soils ranged from $1.0\sim28.4dS\;m^{-1}$ (average: $4.8dS\;m^{-1}$) and salt affected soil which EC was higher than $4dS\;m^{-1}$, covered nearly 55% of all field surveyed. Salts in rainfall interception culture soils were accumulated by increasing the cultivation period. Electrical conductivity in rainfall interception culture soils was positively correlated with water soluble anions such as chloride ion ($r=0.85^{**}$), nitrate ion ($r=0.94^{**}$), phosphate ion ($r=0.88^{**}$), and sulfate ion ($r=0.84^{**}$), respectively. As a result of desalinization experiments carried out by water management practices, the rinsing method was more effective than leaching method.
