This experiment was conducted to obtain the basic information on new- and latent-bud formation, and stem vestige arrangement on the rhizome of Panax ginseng C.A. Meyer. Latent buds emerged from meristematic region between shoot and root of the embryo, and new buds for the next year were distributed both at the bottom portion of the stem and the rhizome. In the new buds, organs such as leaf, stem, and flower bud were already completely differentiated, while the latent bud had an undifferentiated meristematic tissue arranged linearly in a vertical line, indicating that each year new- and latent-buds are formed successively. This result suggests that the number of stem vestige may be used for the determination of ginseng age. Bud sport (bud mutation) is a valuable source for existing new genotypes in mulberry (Morus spp.) as well as critical materials for studying the molecular mechanisms underlying essential traits. Thus, identification, collection, characterization, and conservation of such natural variants are prerequisites for enhancing the mulberry genetic resource in the germplasm. In this context, we identified and characterized three bud sports (VBS-1, VBS-2, and VBS-3) of a popular mulberry cultivar, Victory-1 (V-1). These bud sports are morphologically, anatomically, and genetically more distinct from their mother plant, Victory-1. Moreover, these bud sports display lower growth and yield potential. Furthermore, these showed remarkably lower 2C DNA contents of 0.74 pg (VBS-1), 0.78 pg (VBS-2), and 0.76 pg (VBS-3), when compared to their mother plant V-1 (2C = 0.81 pg). On the other hand, molecular characterization between the bud sports and their mother plant revealed the existence of genetic variation due to the natural bud mutation that occurred in the mulberry cultivar Victory-1.
The spatial landmark protein Bud8 plays a crucial role in bipolar budding in the budding yeast Saccharomyces cerevisiae. The unconventional yeast Yarrowia lipolytica can also bud in a bipolar pattern, but is evolutionarily distant from S. cerevisiae. It encodes the protein YALI0F12738p, which shares the highest amino acid sequence homology with S. cerevisiae Bud8, sharing a conserved transmembrane domain at the C-terminus. Therefore, we named it YlBud8. Deletion of YlBud8 in Y. lipolytica causes cellular separation defects, resulting in budded cells remaining linked with one another as cell chains or multiple buds from a single cell, which suggests that YlBud8 may play an important role in cell separation, which is distinct from the function of Bud8 in S. cerevisiae. We also show that the YlBud8-GFP fusion protein is located at the cell membrane and enriched in the bud cortex, which would be consistent with a role in the regulation of cell separation. The coiled-coil domain at the N-terminus of YlBud8 is important to the correct localization and function of YlBud8, as truncated proteins that do not contain the coiled-coil domain cannot rescue the defects observed in $Ylbud8{\Delta}$. This finding suggests that a new signaling pathway controlled by YlBud8 via regulation of cell separation may exist in Y. lipolytica.
1t is well known that the stem is filed with shoot units in the rice plant and each internode bears several crown roots. But it has not yet been ascertained that what controls the differential elongation of the crown roots in the same internode. Thereupon, author had been carried out this experiment to ascertain what controls the elongation of the crown roots in the same internode, especially on the conception of sink-source by leaf-cutting method. Generally, one shoot unit has two important sinks: one axillary bud (tiller) and several crown roots. When we removed the axillary bud, namely shoot unit has one sink: several crown roots, the crown roots formed near the midvein (source) were longer than the crown roots born near the axillary bud. And when the shoot unit has two sinks: one axillary bud and several crown roots, the other way, the crown roots formed at the prophyll unit of the tiller were longest, and the crown roots formed near the midvein were shortest and the crown roots born the near the tiller showed interim length. Juding from the present results, we can suppose that, when shoot unit has two sinks, axillary bud is superior sink than the crown roots. So that axillary bud grows faster than crown roots and tiller becomes a new source. Therefore the crown roots which formed at the new source and the crown roots born the near the new source are longer than others.
Characteristics of vegetative spur leaf, spur bud, fruiting supr leaf and flowering in relation to canopy position was investigated on order to promote better spur bud and flower bud differentiation on Citrus junos. There was no difference in vegetative spur leaf characteristics among directions. Vegetative spur leaf number and leaf area showed no difference between top and bottom position. There was no difference in spur bud size among directions, but spur bud at top was larger than that at bottom. Specific leaf weight of vegetative spur was mostly influenced by light interception, and leaf dry weight per spur average leaf dry weight, spur bud length and diameter had also a very high correlation with light interception rate. Shading and GA treatment in spur bud and flower bud differentiation was ineffective than natural light. Fruit thinning enhanced fruit quality as well as flower bud differentiation through an increase of leaf number per fruit, over 40 in Citrus junos.
To improve new propagation method, I try to propagate wild polygonatum odoratum roots which formate adventitious bud without bud. The obtained results of size of roots and adventitious bud to formate for multiple propagation of wild polygonatum odoratum as the follow. One year grown wild polygonatum odoratum root in 5cm diameter formate 66.5% adventitious buds. One year grown wild polygonatum odoratum root in 15cm in diameter germinate 113% adventitious bud. Two years grown root in 15cm diameter formate adventitious bud three times more then one year root. Wild polygonatum odoratum in 15cm diameter without any bud in experiment plot makes 4 times of propagation. Sand and Perlite media propagaton plot was increased 3.1∼3.3times more than common culture medium. The best result of mass propagation wild polygonatum odoratum is 10mm diameter root and 5cm cut in length to plant sand media plot.
Growth hadit and demograph in a mature population of polygonatum and polygonatum humile in kanghwa island were studied for two growing seasons. Shoots of two species emerged early spring from the growing apices of the underdground rhizomes which had persisted for up to 1~5 year.after flowering, the ramets produced two rhizome buds at the shoot base. Bacause of the apical dominance in the rhizome system, a new rhizome was developed from only one bud, an actual bud,and the other latent buds were suppressed sothat remained dormant. The latent dud produced a new rhizome only when the actual dud was severed by the herbivores or by the physical obstacles. Therefore, the ramet number is not increased by the new rhizome from the latent bud. however, new ramets dould sometimes grow from latent buds which had been produced more than a year ago. Production of these ramets was main means increasing the ramet numbew and widening the potential zone of exploitation. Changes in size class of each ramet were noyiceable after a tear in small size-classes. Small ramets replaced themselves with larger-sized ramet, while large ramets with similar-sized or smallar-sized ramets. ramet numbers were average 0.82and 1.14 times of those fromthe previous year inp. involucratum and p. humile, though there was between-site variation. Almost all the ramets in the quadrats were alive during the growing season. when the entire rhizome systems were excavater next spring, there were many rhizomesegments without shoots, especially in p. involucratum. therefore, the drcrease of ramet number in p. involucratum in probably due to the climatic factors of winter.
This study was carried out to obtain the basic information for the shortening of emergence period of ginseng by treatment of growth regulators. Seedlings that removed and non-removed bud sac were treated at 10, 50 and 100ppm of GA$_3$, Kinetin and 2,4-D in early December, and investigated the characteristics of new bud emergence and growth vigor in 2-year-old ginseng. GA treatment showed the most desirable effects in shortening of emergence period of new bud, and elevating its emergence rate with increasing of the GA concentration. In addition, GA treatment especially accelerated the growth of stem and petiole length and early finished the growth of aerial parts of ginseng. On the other hand, root weights were mainly increased by formation of a lot fine roots in GA 50, 100ppm plots.
Gibberellic acid (GA) was applied to field-grown 3-year-old North American ginseng (Panax quinqueiolius L.) between 1 and 4 times, before and during bloom in 1999. Applications of both GA$_3$ and GA$\sub$4+7/ four times (x4) to the developing inflorescences increased maximum pedicel length, and seed head diameter and height. Treatment with GA$\sub$4+7/ increased mean and total root fresh weight linearly, whereas those treated with GA$_3$ did not show similar increases. Both GA$_3$ and GA$\sub$4+7/ at 50, 100 and 200 mg L$\^$-1/ (x4) increased the incidence of breaking of dormancy of perennating buds with GA$_3$ being twice as effective as GA$\sub$4+7/. Both GA$_3$ and GA$\sub$4+7/ treatments resulted in an increased number of new bud initials forming per root, with the number of new initials per root increased two-fold by the GA$_3$ sprays compared to GA$\sub$4+7/.

Yacon [Samallanthus sonchifolius (Poepp. & Endl.) H. Robinson], a member of Compositae plants, has sweet taste and crisp texture. Unlike other Andean root crops such as potato and sweet potato, the cultivation area of yacon has increased recently, since it is known to have large content of fructooligosaccharides (FOS). Since there are no yacon varieties bred in Korea, we have been trying to create new genetic resources using gamma-ray. The optimal gamma-ray dosage for mutation breeding in yacon was investigated. Crown bud and green bud of yacon were exposed to doses of gamma rays from 20 Gy to 80 Gy, and subsequently planted in a greenhouse. After 50 days of sowing, the survival rates and growth decreased rapidly at doses above 40 Gy, while all of crown bud individuals died above 60 Gy. The median lethal dose (LD50) of crown bud and green bud was 22.4 and 36.6 Gy, and the median reduction doses (RD50) for plant height, fresh weights, and tuberous root weight were 20-40 Gy, respectively. A dose of 20-40 Gy was found to be optimal for mutation breeding in yacon. Considering the growth factors, the optimum doses were determined to be within the range of 20-40 Gy for the selection of useful mutant lines. M2-M3 mutant lines were obtained from 20-60 Gy gamma-ray-irradiated M1 plants through clonal propagation. These mutant lines will be used for the development of a new variety of yacon plant with high FOS and no crack tuberous root.
