Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro-N-nitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30℃ and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.

The ubiquinone and G+C contents of the bioflocculant-producing fungus, a new Aspergillus strain, were detennined using high-perfonnance liquid chromatography. The internal transcribed spacers 1 and 2 (ITS1 and ITS2), and the 5.8S ribosomal DNA (rDNA) of the strain were amplified and sequenced. The strain contained ubiquinone-l0($H_2$)as a major quinone and the G+C content was 49 mol%. A phylogenetic analysis of the ITS regions indicated that the strain belonged to the genus Aspergillus according to its previously classified morphological characteristics. Based on a sequence homology search, the strain was most closely related to Petromyces muricatus (anamorph, A. muricatus; accession number, AJ005674). The sequence of a new Aspergillus strain in ITS1 and ITS2, and 5.8S rDNA showed 97% homology to P. muricatus. Therefore, the strain is believed to be a new bioflocculant-producing Aspergillus strain.
A new and a unique Brachionus rotifer was found in Hwajinpo coastal lagoon in Gangwon Province, South Korea. This Brachionus certainly originated from the wild rather than from aquaculture stations because Hwajinpo coastal lagoon has been under rigorous control as a military protected area and therefore could not have been contaminated by aquaculture stations. The new strain was identified as Brachionus rotundiformis based upon its morphological characteristics. The parthenogenetic female of this new rotifer strain typically shows characters similar to those of B. rotundiformis, such as the pot shape of the body, rounded dorsal plate compared with flattened ventral plate, elliptical mictic egg, four frontal spines, six pointed occipital spines, non-nodal foot, two toes, trophi typical of the Brachionus genus with five uncus plates resembling comb teeth, one wide symmetrical manubrium and ramus, and no stiffened spine as is seen in freshwater Brachionus rotifers. Moreover, its lorica was rather small in size compared with other common rotifer strains that serve as live-food organisms (Guam, Thai, and Bali strains). This new and unique Korean brackish rotifer, a B. rotundiformis strain, was therefore named the National Fisheries Research and Development Institute (NFRDI) $N^{\circ}1$ rotifer strain.
This paper presents a new uniaxial material model for rate-sensitive analysis addressing both the transient and steady-state responses. The new model adopts visco-plastic theory for the rate-sensitive response, and employs a three-parameter representation of the overstress as a function of the strain-rate. The third parameter is introduced in the new model to control its transient response characteristics, and to provide flexibility in fitting test data on the variation of overstress with strain-rate. Since the governing visco-plastic differential equation cannot be integrated analytically due to its inherent nonlinearity, a new single-step numerical integration procedure is proposed, which leads to high levels of accuracy almost independent of the size of the integration time-step. The new model is implemented within the nonlinear analysis program ADAPTIC, which is used to provide several verification examples and comparison with other experimental and numerical results. The companion paper extends the three-parameter model to trilinear static stress-strain relationships for steel and concrete, and presents application examples of the proposed models.
The companion paper presents a new three-parameter model for the uniaxial rate-sensitive material response, which is based on a bilinear static stress-strain relationship with kinematic strain-hardening. This paper extends the proposed model to trilinear static stress-strain relationships for steel and concrete, and discusses the implementation of the new models within an incremental-iterative solution procedure. For steel, the three-parameter rate-function is employed with a trilinear static stress-strain relationship, which allows the utilisation of different levels of rate-sensitivity for the plastic plateau and strain-hardening ranges. For concrete, on the other hand, two trilinear stress-strain relationships are used for tension and compression, where rate-sensitivity is accounted for in the strain-softening range. Both models have been implemented within the nonlinear analysis program ADAPTIC, which is used herein to provide verification for the models, and to demonstrate their applicability to the rate-sensitive analysis of steel and reinforced concrete structures.
Strain NR $15-1^T$ isolated from traditional Korean Nuruk is described as a new species and named as Aspergillus coreanus NR $15-1^T$ sp. novo Strain NR $15-1^T$ grew rapidly to form yellow-green colonies whose surfaces were velvety on Czapek solution agar. Conidial heads were yellow to light and elliptical, whereas the conidiophore was colorless and typically long. In addition, vesicles were from flask-shaped to globose, and sterigmata are uniseriate. Conidia were spherical and deep yellow-green, and their surfaces were lightly roughened. The G+C content of strain NR $15-1^T$ was 51 mol% and strain NR $15-1^T$contained a dihydrogenated ubiquinone with Q9 (94.9%) as a major quinone. The nucleotide sequences of strain NR $15-1^T$ in the two Internal Transcribed Spacers (ITS 1 and 2) and 5.8S rDNA showed highest similarity when compared with that of A. tubingensis and A. phoenicis NRRL $365^T$. However, based on morphological and chemotaxonomic characteristics, this strain was different from A. tubingensis and A. phoenicis NRRL $365^T$. On the basis of the data presented, it is proposed that strain NR $15-1^T$ should be placed in the genus Aspergillus as a new species, Aspergillus coreanus sp. novo Therefore, the type strain of the new species is strain NR $15-1^T$(=KCTC 18075P^T,=KCCM 80006^T$.
Strain 88-GT-161 producing new phthalic acid derivative and basic macrolide antibiotics was identified as being S. melanosporofuciens based on numerical taxonomic data. However, 4 unit characters among 139 units were clearly different from the common properties of 6 strains belonging to cluster No. 32 represented by the name of S. violaceoniger or S. violaceusniger, leading us to designate as a variety of S. melunosporofaciens. This paper describes the taxonomic characteristics of the strain. Isolation and chemical structures, including biological activities of the active compounds produced by this strain will be presented elsewhere.
Group A Streptococcus strain ST4529 is a provisional new ems type which has been recently reported in Malaysia (Jomal, et al. 1999. Energ. Infect . Dis. 5,10-14). This strain was found to be opacity factor (OF) negative with a Tl phenotype. Usually, OF negative strains with T1 phenotypes are associated with acute rheumatic fever. However, strain ST4529 was isolated from the blood of a patient with septicemia. Comparison of the deduced amino acid sequence of the mature hypervariable N-terminus of ST4529 showed only 43% identity with that of M5, the closest matched OF negative strain with a T1 phenotype. Thus, ST4529 most probably encodes a new serospecifically unique M protein which is associated with septicemia rather than pharyngitis infections. The strains with these phenotypes are very important because their sequences should be considered for developing any anti-streptococcal vaccines.

Most metallic materials and alloys show rate independence or negative rate sensitivity in some temperature region when dynamic strain aging occurs. It is generally recognized that negative rate sensitivity is an essential feature of dynamic strain aging that can depend on strain and/or strain rate. The unified viscoplasticity theory based on overstress is applied to reproduce a change of rate sensitivity type that depends on strain or strain rate. This is accomplished through the introduction of a single new term in the growth law of the equilibrium stress, which is a tensor valued state variable of the model. It is also shown that the new term can be used to reproduce a dramatic increase of rate sensitivity in dynamic plasticity.
High deformable concrete (HDC) elements have compressive strength rates equal to conventional concrete and have got a high compressive strain at about 20% to 50%. These types of concrete elements as prefabricated parts have an abundance of applications in the construction industry which is the most used in the construction of tunnels in squeezing grounds, tunnel passwords from fault zones or swelling soils as soft supports. HDC elements after reaching to compressive yield stress, in nonlinear behavior have hardening combined with increasing strain and compressive strength. The main aim of this laboratory and numerical research is to construct concrete elements with the above properties so the compressive stress-strain behavior of different concrete elements with four categories of mix designs have been discussed and finally one of them has been defined as HDC element mix design. Furthermore, two columns with and without implementing of HDC elements have been made and stress-strain curves of them have been investigated experimentally. An analysis model is presented for columns using finite element method adopted by ABAQUS. The results obtained from the ABAQUS finite element method are compared with experimental data. The main comparison is made for stress-strain curve. The stress-strain curves from the finite element method agree well with experimental results. The results show that the dimension of the HDC samples is significant in the stress-strain behavior. The use of the element greatly increases energy absorption and ductility.
