The Effects of Paraquat on Arachidonate Lipoxygenase Metabolism (Paraquat가 Arachidonate Lipoxygenase의 대사에 미치는 영향)

  • Using mixed incubation of cultured endothelial cells, cultured fibroblasts, neutrophils activated with PMA and paraquat, the production of superoxide anion, $H_2O$$_2$and lipoxygenase metabolites (5-HETE and 12-HETE) of arachidonate was estimated. The results was as follows: 1. Neutrophils activated with PMA was produced superoxide anion,$H_2O_2$and lipoxygenase metabolites (5-HETE and 12-HETE) of arachidonate. 2. Fibroblasts did not alter the production of superoxide anion and $H_2O_2$ by neutrophils, it was markedly reduced by mixed incubation with endothelial cells. 3. Mixed incubation with endothelial cells significantly augmented the production of 5 or 12-HETEs, but fibroblasts did not. 4. Using mixed incubation of endothelial cells, fibroblasts, neutrophils and paraquat (50ug/m1 and 100ug/m1), the production of superoxide anion, $H_2O_2$and HETEs was significantly increased on both cells at low concentration (50ug/m1), but markedly reduced at high concentration (1001g/m1). 5. Paraquat showed concentration dependent effects on arachidonate lipoxygenase metabolism.

2-Undecanone derived from Pseudomonas aeruginosa modulates the neutrophil activity

  • Pseudomonas aeruginosa (P. aeruginosa) is a well-known Gramnegative opportunistic pathogen. Neutrophils play key roles in mediating host defense against P. aeruginosa infection. In this study, we identified a metabolite derived from P. aeruginosa that regulates neutrophil activities. Using gas chromatography-mass spectrometry, a markedly increased level of 2-undecanone was identified in the peritoneal fluid of P. aeruginosa-infected mice. 2-Undecanone elicited the activation of neutrophils in a Gαi-phospholipase C pathway. However, 2-undecanone strongly inhibited responses to lipopolysaccharide and bactericidal activity of neutrophils against P. aeruginosa by inducing apoptosis. Our results demonstrate that 2-undecanone from P. aeruginosa limits the innate defense activity of neutrophils, suggesting that the production of inhibitory metabolites is a strategy of P. aeruginosa for escaping the host immune system.

Anti-apoptotic Effects of House Dust Mite, S100A8 and S100A9 on Spontaneous Apoptosis of Neutrophils in Coculture with Immune Cells and in the Presence of T Helper Cytokines

  • House dust mite (HDM) as a major allergen and damage-associated molecular pattern (DAMP) such as S100A8 and S100A9 trigger the pathogenesis and severity of allergic disease such as asthma. Regulation of neutrophil apoptosis is an important immune response and its dysregulation is involved in pathogenesis of allergic diseases. In this study, we examined the effects of HDM, S100A8 and S100A9 on spontaneous apoptosis of normal neutrophils. We considered the importance of the difference between in vitro and in vivo results and developed a new in vitro system consisting of a combination of immune cells and T helper (Th) cytokines. Extract of Dermatophagoides pteronyssinus (DP), S100A8, and S100A9 inhibited neutrophil apoptosis in culture of neutrophils alone without other leukocytes. DP and S100A8 more strongly suppressed neutrophil apoptosis in combinations of neutrophils, eosinophils, lymphocytes or monocytes than in a culture of neutrophils alone. Anti-apoptotic effect of S100A9 in the mixture of immune cells was similar to that in neutrophils. DP, S100A8, and S100A9 blocked neutrophil apoptosis, regardless of pretreatment with a T helper (Th) 1 cytokine (IFN-$\gamma$), Th2 cytokines (IL-4 and IL-10), a Th9 cytokine (IL-9), a Th17 cytokine (IL-17), a Treg-producing cytokine (TGF-$\beta$). These findings may enable elucidation of allergy pathogenesis due to HDM and DAMP.

The Inhibitory Effect of Eupatilin on Helicobacter Pylori-Induced Release of Leukotriene $D_4$ in the Human Neutrophils and Gastric Mucosal Cells

  • In this report, the inhibitory action of eupatilin was investgated by using leukotriene $D_4$ in the human neutrophils and Kato III cells (Gastric adenoma cells as a substitute for gastric mucosal cells) stimulated by Helicobacter pylori. Leukotriene $D_4$ ($LTD_4$) was released from both neutrophils and Kato III cells when these cells were incubated with H. pylori. The release of $LTD_4$ increased time-dependently and the maximum release of $LTD_4$ was $2.3{\sim}2.5$ pmol. But in the presence of eupatilin, the release of $LTD_4$ from these cells was inhibited in a dose-dependent manner. In the neutrophils, the release of $LTD_4$ was suppressed to 70% and 50% of the control levels when neutrophils was incubated with 0.01 and 0.1 mM of eupatilin. In the Kato III cells, the release of $LTD_4$ was suppressed to 59% and 27% of the control levels by adding 0.01 and 0.1 mM of eupatilin. We estimated the intracellular $Ca^{2+}$ levels when Kato III cells and neutrophils were stimulated by H. pylori using $^{45}Ca$. But the suppressive effect of eupatilin on $Ca^{2+}$ influx into these cells was not significant. We also obtained the results that H. pylori induced $Ca^{2+}$ influx into these cells by confocal microscopy, however there was no differences in the dose level of eupatilin. These results were confirmed by 1H Nuclear Magnetic Resonance(NMR) spectroscopy. The NMR patterns of eupatilin in the absence of $Ca^{2+}$ was changed compare with when $Ca^{2+}$ was present, but its effect was not strong.

  • Background: A physically active lifestyle and regular exercise training incurs many health benefits. One recently recognized benefit of regular moderate exercise is stress reduction and immune enhancement. Thus, a physical stress such as exercise may act at any number of points in the complex sequence of events collectively termed the immune response. Although exercise causes many propound changes in parameters of immune function, the nature and magnitude of such changes rely on several factors including the immune parameters of interest; type, intensity, and duration of exercise; fitness level or exercise history of the subject; environmental factors such as ambient temperature and humidity. Methods: This study was undertaken to investigate the effect of different type of exercise on superoxide dismutase (SOD), neutrophils, and T lymphocytes of Sprague-Dawley rats. Sprague-Dawley rats were randomly divided into three groups; a non-Trained group (NTG, n=6), a swim-Trained group (STG), and a treadmill-Trained group (TTG). The exercise regimen was designed in a treadmill (5 times/5 days/week) during 8-weeks for TTG, and swim training (5 times/5 days/week) during 8-weeks for STG, and the volume of exercise training was the same in both groups. Results: 8 weeks of regular swim and treadmill training significantly increased liver SOD concentration however, muscle SOD concentration was not statistically significant. In the level of neutrophils, TTG and STG showed significant difference, compared to NTG. TTG was the highest level of neutrophils. In the level of immune cell counts, there was significant difference among TTG, STG, and NTG both in the spleen and thymus. Conculsion: In conclusion, it can be stated that eight weeks swim and treadmill exercise training has beneficial effect in improving immune response and antioxidant defence capacity by augmenting immune cells and SOD activities of SD rats.

Increase in phagocytic activity of peripheal neutrophil and lysozyme activity of blood serum in Korea catfish (Silurus asotus) intraperitoneally injecteded with $\beta$-glucan ($\beta$-glucan을 접종한 한국산 메기(Silurus asotus)의 호중구와 리소자임 활성 증강)

  • To examine the effectiveness of $\beta$-glucan in non-specific defence mecanisms of Korean catfish(Silurus asotus), the activities of peripheral neutrophils and serum lysozyme were evaluated after once or twice $\beta$-glucan injections. The number of peripheral neutrophils increased after the injection. The increased number in neutrophils in twice $\beta$-glucan injections maintained longer than in single glucan injecton. But the neutrophils did not showed any remarkable changes in PAS-positive reaction. Peripheral neutrophils from fish injected glucan had enhanced phagocytic activities but there was no significant difference in phagocytic index for the glucan and control groups. An elevation of lysozome activity was also observed in the fish received twice glucan but neither single glucan or physiological saline injection could not evoked an increase of lysozyme activity. These results suggest that $\beta$-glucan can enhance the resistance of Korean catfish against to bacteria infection through the activation of peripheral neutrophils and lysozyme.

Chemotactic Effect of Leukotactin-1/CCL15 on Human Neutrophils

  • Leukotactin-l (Lkn-l )/CCL15 has been known as a potent chemoattractant of leukocytes. However, the precise function of Lkn-l in human neutrophils has not been explained well. In the present study, we investigated the contribution of Lkn-1 in chemotactic activity of human neutrophils. Both CCR1 and CCR3 mRNA expressions are strongly expressed in human neutrophils but CCR2 protein expression was uniquely detected on the cell surface. Lkn-l binding to CCR1 and CCR3 induced chemotactic activity of neutrophils. Chemotactic index of Lkn-l was comparable to that of IL-8. $MIP-1{\alpha}/CCL3$ binding to CCR1 and CCR5 has no effect on neutrophil migration. Cell migration, in response to Lkn-l, was blocked by pertussis toxin (Ptx), a $G_o/G_i$ protein inhibitor, and U73122, a phospholipase C(PLC) inhibitor but not by protein kinase C inhibitor such as rottlerin, and Ro-31-8425. Taken together, our results demonstrate that Lkn-l transduces the chemotaxis signal through $G_o/G_i$ protein and PLC. This finding provides the molecular mechanism by which Lkn-l may contribute to neutrophil movement into the site of inflammation.

Effects of Chicken Egg White Derivatives on the Changes of Blood Cells and the Neutrophil Phagocytosis of Rats Administered with Cyclophosphamide (Cyclophosphamide투여 랫드의 혈구세포 변화 밀 호중구 탐식능에 있어서 계난백유래물질의 효과)

  • As an antitumor agents cyclophosphamide (CPA) is frequently used in animal clinic. Important adverse effects of its administration are leukopenia, thrombocytopenia and anemia. We investigated the effects of chicken egg white derivatives (EWD and EF-203) on the changes of blood cells and the neutrophil phagocytosis of rats administered with CPA. Rats were administered CPA peritoneally at dose of 50 mgag once a day far 3 days plus either EWD or EF-203 orally at dose of 200 mg/kg once a day far 3 days. Thereafterl the changes of blood cells by automatic blood cell counter and the phagocytosis of neutrophils by flow cytometry were examined far 7 days. There was no change in RBC values regardless of administration of either EWD or EF-203 throughout experimental period. But rats receiving CPA plus either EWD or EF-203 showed a significant higher PCV values than those of CPA alone (p<0.01). The numbers of peripheral blood platelets and WBC and the differential count of neutrophils in the ra% receiving CPA plus either EWD or EF-203 were significantly higher (p<0.05 to 0.01) than those of CPA alone. Moreover, these rats showed significanly enhanced phagocytoses of neutrophils when compared to rats with CPA alone (p<0.01). These result suggested that chicken egg white derivatives including EWD and EF-2% have immunomodulatory effects in regard to the increase of platelets, WBC, differential count of neutrophils, PCV, and the enhancement of phagocytic activity of neutrophils in immunosuppressed rats by CPA. Thus, co-adminstration of chicken egg white derivatives will be able to reduce the side effects in the animals treated with antitumor agents.

Lipid Peroxidation, $NF-_{\kappa}B$ Activation and Cytokine Production in Neutrophil-Stimulated Pancreatic Acinar Cells

  • Reactive oxygen species (ROS), generated by infiltrating neutrophils, are considered as an important regulator in the pathogenesis and deveolpment of pancreatitis. The present study aims to investigate whether neutrophils primed by $4{\beta}-phorbol\;12{\beta}-myristate\;13{\alpha}-acetate$ (PMA) affect the productions $H_2O_2$ and lipid peroxide (LPO), $NF-_{\kappa}B$ activation and cytokine production in pancreatic acinar cells, and whether these alterations were inhibited by an antioxidant, N-acetylcysteine (NAC) and superoxide dismutase (SOD). $H_2O_2$ (ferrithiocyanate method), LPO (as thiobarbiturate reactive substances), and cytokines $(IL-l{\bata},\; IL-6,\;TNF-{\alpha};\;enzyme-linked\;immunosorbent\;assay)$ and $NF-_{\kappa}B$ activation (electrophoretic mobility shift assay) were analyzed in acinar cells treated with or without PMA-primed neutrophils in the absence or presence of NAC (10 mM) or SOD (300 U/ml). As a result, the productions of H2O2, LPO and $TNF-{\alpha}$ were increased with the ratio of PMA-primed neutrophils to acinar cells while the productions of LPO, $IL-l{\beta},\;IL-6\;and\;TNF-{\alpha}$ were increased with time. PMA-primed neutrophils resulted in the activation of $NF-_{\kappa}B.$ Both NAC and SOD inhibited neutrophil-induced alterations in acinar cells. In conclusion, ROS, generated by neutrophils, activates $NF-_{\kappa}B,$ resulting in upregulation of inflammatary cytokines in acinar cells. Antioxidants might be clinically useful antiinflammatory agents by inhibiting oxidant-mediated activation of $NF-_{\kappa}B$ and decreasing cytokine production.

Der p 1 Inhibits Spontaneous Neutrophil Apoptosis by Cytokine Secretion of Normal and Allergic Lymphocytes (Der p 1에 의한 정상인과 알레르기 환자의 림프구에서 사이토카인 분비를 통한 자발적인 호중구 세포고사 억제)

  • Dermatophagoides pteronissinus (DP) is one of the house dust mites (HDM) associated with allergic diseases. The cysteine protease Der p 1 from DP is a powerful allergen. The pathogenic mechanism of allergy is involved in cytokine secretion of lymphocytes and spontaneous apoptosis of neutrophils. In this study, we investigated whether Der p 1 induces cytokine secretion of lymphocytes and if the release of cytokines is associated with regulation of neutrophil apoptosis. In normal and allergic subjects, Der p 1 increased IL-6, IL-8, MCP-1, and GM-CSF release in a time-dependent course. Supernatants collected from normal and allergic neutrophils after Der p 1 stimulation suppressed the apoptosis of normal and allergic neutrophils, although Der p 1 alone has no effect on neutrophils. Der p 1 suppressed neutrophil apoptosis in coculture of normal neutrophils with normal lymphocytes. Der p 1 more strongly suppressed apoptosis of allergic neutrophils cocultured with allergic lymphocytes than normal neutrophils cocultured with normal lymphocytes. In summary, Der p 1 increases the secretion of cytokines, which has anti-apoptotic effects on neutrophils of normal and allergic subjects. These results will contribute to elucidate the pathogenic mechanism of allergic diseases.
