The Role of Neuropeptide Y in the Central Regulation of Grass Intake in Sheep

  • The physiological role of brain neuropeptide Y (NPY) in the central regulation of grass intake in sheep was investigated through a continuous intracerebroventricular (ICV) infusion of NPY at a dose of $5{\mu}g/0.2ml/hr$ for 98.5 hours from day 1 to day 5. Sheep (n=5) were fed for 2 hours once a day, and water and 0.5 M NaCl solution were given ad libitum. Feed intake during ICV NPY infusion increased significantly compared to that during ICV artificial cerebrospinal fluid (CSF) infusion. Water and NaCl intake during ICV NPY infusion remained unchanged. Mean arterial blood pressure (MAP) and plasma osmolality during ICV NPY infusion were not significantly different from those during ICV CSF infusion. On the other hand, plasma glucose concentration during ICV NPY infusion increased significantly compared to that during ICV CSF infusion. The results suggest that brain NPY acts as a hunger factor in brain mechanisms controlling feeding to increase grass intake in sheep.

  • Dorsal root ganglion(DRG) cells are primary sensory neurons which contain some biologically active neuropeptides which play a role as neurotransmitters or neuromodulators. This study was performed to observe normal distribution of calcitonin gene-related peptide (CGRP) and substance P (SP) immunoreactive cells and colocalization of CGRP and SP in a single DRG cell of the lumbar DRGs($L_1{\sim}L_6$) in the Wistar Kyoto(WKY) rat by immunohistochemistry. About 55.8% of DRG cells contained CGRP-immunoreactivity, while about 12.7% of DRG cells showed SP-immunoreactivity. There was no significant difference in percentage of each neuropeptied-immunoreactive cells between each neuropeptide-immunoreactive cells between each levels of DRGs ($L_1{\sim}L_6$) (p>0.01). In size distribution, CGRP-immunoreactive cells were identified below $1,500{\mu}m^2$; SP-immunoreactive cells below $600{\mu}m^2$. In serial sections, about 86.7% of the SP immunoreactive cells contained CGRP immunoreactivity.

전침자극이 흰쥐 대뇌피질의 NADPH-diaphorase와 NPY에 미치는 영향 (Effects of Electroacupuncture by Different Frequencies and Treatment-times on NADPH-diaphorase and Neuropeptide Y in Sprague-Dawley Rat Cerebral Cortex)

  • The present study was designed to investigate the effects of various electroacupuncture(EA) stimulation on NADPH-diaphorase and Neuropeptide Y(NPY) in the cerebral cortex of Sprague-Dawley Rats. We evaluated the changes of NADPH-d-positive neurons using a histochemical method and the changes of NPY-positive neurons using a double labeling immunohistochemical method. The staining intensities of NADPH-d-positive neuron and NPY-positive neurons were assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. As to frequency, the optical density of NADPH-d-positive neurons of 2Hz-1 time EA group was significantly higher than that of 100Hz-1 time EA group in M1, Cg, and that of 100Hz-3 times EA group was significantly higher than that of 2Hz-3 times EA group in Vi, Au, Cg and Ins. As to treatment-time, the optical density of NADPH-d-positive neurons of 2Hz-1 time EA group was higher than 2Hz-3 times EA group in Vi, Au, Cg and Ins. And in Vi 100Hz-1 time EA group and in M1 100Hz-3 times EA group was more efficient. As to frequency, the optical density of NPY-positive neurons of 2Hz-1 time was significantly higher in Vi, and that of 100Hz-1 EA group was higher in M1. And that of 2Hz-3 times EA group was higher than 100Hz-3 times EA group in Cg. As to treatment-time, the optical density of NPY-positive neurons of 2Hz-1 time EA group was significantly higer than 2Hz-3 times EA in S1, Vi, Au. And that of 100Hz-1 time EA group was significantly higher than that of 100Hz-3 time EA in Cg. The present results demonstrated that EA changes the activity in NO and NPY system in the cerebral cortex of SHR and EA stimulation, like frequency and treatment-time, is of importance for this effect.

  • Corticotropin-releasing factor(CRF) and neuropeptide Y(NPY) are known to play important roles in mediating stress responses and stress-related behavior. To elucidate the role of neuropeptides in response to the condition that had paired with traumatic event, we observed the changes of CRF and NPY by immunohistochemistry using a conditioned footshock paradigm. Male Sprague-Dawley rats were placed in a shuttle box and exposed to 20 pairings of a tone(< 70dB, 5sec) followed by a footshock(FS, 0.8mA, 1sec) over 60min. A second group was exposed to the tone-footshock pairings, returned to the homecage for 2days, and then reexposed to the test chamber and 20tones alone for 60min, prior to sacrifice. Control groups were : a) sacrificed without exposure to FS ; b) exposed to the tone-footshock pairings and then sacrificed two days later ; or c) exposed to the chamber and tones alone, returned to the homecage for 2days and then reexposed to the chamber and 20tones over 60min prior to sacrifice. CRF was increased in animals exposed to FS or the aversive condition(context and tone) that had paired to FS in bed nucleus of the stria terminalis (BNST) compared to the control. NPY was increased by FS in amygdala and PVN, but the condition previously associated with FS results in slight increase only in amygdala area. These results suggest that the BNST appears to be the mostly involved neural circuit in response to explicit cues previously paired with footshock. Moreover, this study raise the possibility that increased CRF peptide in the BNST in response to re-exposure to the aversive condition may underlie, in part, the experience of conditioned fear-related anxiety behavior.

The Body Weight-related Differences of Leptin and Neuropeptide Y (NPY) Gene Expression in Pigs

  • To determine if body weight change is directly related to altered leptin and neuropeptide Y (NPY) gene expression, we assessed adipose tissue weight, percent body fat, leptin and NPY mRNA levels and serum leptin concentration in pigs at weights of 1, 20, 40, 60, and 90 kg. The results indicated that the weight of adipose tissues and the percent body fat of pigs significantly increased and correlated with body weight (BW) from 1 to 90 kg (p<0.01). Serum leptin concentrations and leptin mRNA levels in omental adipose tissue (OAT) increased from 1 to 60 kg, and then decreased from 60 to 90 kg. At 60 kg, the serum leptin concentration and leptin mRNA level significantly increased by 33.5% (p<0.01) and 98.2% (p<0.01), respectively, as compared with the levels at 1 kg. At 60 kg, the amount of leptin mRNA in subcutaneous adipose tissue (SAT) was significantly higher than that of 1 and 40 kg animals (p<0.05). NPY gene expression in the hypothalamus also changed with BW and at 60 kg the NPY mRNA level significantly decreased by 54.0% (p< 0.05) as compared with that in 1 kg. Leptin mRNA in OAT was correlated with serum leptin concentrations (r = 0.98, p<0.01), body weight (r = 0.82, p<0.05) and percent body fat (r = 0.81, p<0.05). This is the first report of the developmental expression of leptin in porcine OAT, peritoneal adipose tissue (PAT) and SAT, and proves that the expression of leptin in OAT could reflect the levels of circulating leptin. These results provide some information for nutritional manipulation of leptin secretion which could lead to practical methods of controlling appetite and growth in farm animals, thereby regulating and improving efficiency of lean meat production and meat production quality.

  • Allatotropin is a 13-residue amidated neuropeptide isolated from pharate adult heads of the tobacco hornworm, Manduca serta and strongly stimulates biosynthesis of juvenile hormones in adults, but not larval, lepidopteran corpora allata. From a Bombyx mori midgut cDNA library, a cDNA that encodes a 130-amino-acid polypeptide containing M. sexta allatotropin sequence was isolated. The B. mori allatotropin cDNA consists of 1196 nucleotides. (omitted)
