Neural Growth Factor Stimulates Proliferation of Spinal Cord Derived-Neural Precursor/Stem Cells

  • Objective : Recently, regenerative therapies have been used in clinical trials (heart, cartilage, skeletal). We don't make use of these treatments to spinal cord injury (SCI) patients yet, but regenerative therapies are rising interest in recent study about SCI. Neural precursor/stem cell (NPSC) proliferation is a significant event in functional recovery of the central nervous system (CNS). However, brain NPSCs and spinal cord NPSCs (SC-NPSCs) have many differences including gene expression and proliferation. The purpose of this study was to investigate the influence of neural growth factor (NGF) on the proliferation of SC-NPSCs. Methods : NPSCs ($2{\times}10^4$) were suspended in $100{\mu}L$ of neurobasal medium containing NGF-7S (Sigma-Aldrich) and cultured in a 96-well plate for 12 days. NPSC proliferation was analyzed five times for either concentration of NGF (0.02 and 2 ng/mL). Sixteen rats after SCI were randomly allocated into two groups. In group 1 (SCI-vehicle group, n=8), animals received 1.0 mL of the saline vehicle solution. In group 2 (SCI-NGF group, n=8), the animals received single doses of NGF (Sigma-Aldrich). A dose of 0.02 ng/mL of NGF or normal saline as a vehicle control was intra-thecally injected daily at 24 hour intervals for 7 days. For Immunohistochemistry analysis, rats were sacrificed after one week and the spinal cords were obtained. Results : The elevation of cell proliferation with 0.02 ng/mL NGF was significant (p<0.05) but was not significant for 2 ng/mL NGF. The optical density was increased in the NGF 0.02 ng/mL group compared to the control group and NGF 2 ng/mL groups. The density of nestin in the SCI-NGF group was significantly increased over the SCI-vehicle group (p<0.05). High power microscopy revealed that the density of nestin in the SCI-NGF group was significantly increased over the SCI-vehicle group. Conclusion : SC-NPSC proliferation is an important pathway in the functional recovery of SCI. NGF enhances SC-NPSC proliferation in vitro and in vivo. NGF may be a useful option for treatment of SCI patients pending further studies to verify the clinical applicability.

  • 본 논문에서는 셀룰러 기반 무선 인지망에서 스펙트럼 인지(CR)기술을 이용하여 모바일 사용자의 핸드오버 호의 손실확률을 줄이는 방법을 제안한다. 제안한 방법에서는 모바일이 방문할 셀을 Ziv-Lempel 알고리듬을 이용하여 예측하고, 방문할 셀에 할당된 채널이 부족할 때는 CR기술에 기초한 스펙트럼 홀 자원을 예측하여 모바일 사용자를 지원한다. 스펙트럼 홀 자원의 크기는 신경망기법으로 예측하며, 예측된 스펙트럼 홀 자원은 핸드오버 호가 초기 발생 호 보다 우선하여 사용할 수 있게 한다. 시뮬레이션을 통하여 셀룰러 이동 통신망에 CR기술을 사용함으로써 모바일 사용자의 핸드오버 호 손실확률을 줄일 수 있음을 보인다.

Classification of Leukemia Disease in Peripheral Blood Cell Images Using Convolutional Neural Network

  • Classification is widely used in medical images to categorize patients and non-patients. However, conventional classification requires a complex procedure, including some rigid steps such as pre-processing, segmentation, feature extraction, detection, and classification. In this paper, we propose a novel convolutional neural network (CNN), called LeukemiaNet, to specifically classify two different types of leukemia, including acute lymphoblastic leukemia (ALL) and acute myeloid leukemia (AML), and non-cancerous patients. To extend the limited dataset, a PCA color augmentation process is utilized before images are input into the LeukemiaNet. This augmentation method enhances the accuracy of our proposed CNN architecture from 96.9% to 97.2% for distinguishing ALL, AML, and normal cell images.

Accurate Representation of Light-intensity Information by the Neural Activities of Independently Firing Retinal Ganglion Cells

  • For successful restoration of visual function by a visual neural prosthesis such as retinal implant, electrical stimulation should evoke neural responses so that the informat.ion on visual input is properly represented. A stimulation strategy, which means a method for generating stimulation waveforms based on visual input, should be developed for this purpose. We proposed to use the decoding of visual input from retinal ganglion cell (RGC) responses for the evaluation of stimulus encoding strategy. This is based on the assumption that reliable encoding of visual information in RGC responses is required to enable successful visual perception. The main purpose of this study was to determine the influence of inter-dependence among stimulated RGCs activities on decoding accuracy. Light intensity variations were decoded from multiunit RGC spike trains using an optimal linear filter. More accurate decoding was possible when different types of RGCs were used together as input. Decoding accuracy was enhanced with independently firing RGCs compared to synchronously firing RGCs. This implies that stimulation of independently-firing RGCs and RGCs of different types may be beneficial for visual function restoration by retinal prosthesis.

In Vitro Biocompatibility Test of Multi-layered Plasmonic Substrates with Flint Glasses and Adhesion Films

  • Since in vitro neural recording and imaging applications based on a surface plasmon resonance (SPR) technique have expanded dramatically in recent years, cytotoxicity assessment to ensure the biosafety and biocompatibility for those applications is crucial. Here, we report the cytotoxicity of the SPR substrate incorporating a flint glass whose refractive index is larger than that of a conventional crown glass. A high refractive index glass substrate is essential in neural signal detection due to the advantages such as high sensitivity and wide dynamic range. From experimental data using primary hippocampal neurons, it is found that a lead-based flint glass is not appropriate as a neural recording template although the neuron cells are not directly attached to the toxic glass. We also demonstrate that the adhesion layer between the glass substrate and the gold film plays an important role in achieving the substrate stability and the cell viability.

Deformation prediction by a feed forward artificial neural network during mouse embryo micromanipulation

  • In this study, a neural network (NN) modeling approach has been used to predict the mechanical and geometrical behaviors of mouse embryo cells. Two NN models have been implemented. In the first NN model dimple depth (w), dimple radius (a) and radius of the semi-circular curved surface of the cell (R) were used as inputs of the model while indentation force (f) was considered as output. In the second NN model, indentation force (f), dimple radius (a) and radius of the semi-circular curved surface of the cell (R) were considered as inputs of the model and dimple depth was predicted as the output of the model. In addition, sensitivity analysis has been carried out to investigate the influence of the significance of input parameters on the mechanical behavior of mouse embryos. Experimental data deduced by Fl$\ddot{u}$ckiger (2004) were collected to obtain training and test data for the NN. The results of these investigations show that the correlation values of the test and training data sets are between 0.9988 and 1.0000, and are in good agreement with the experimental observations.

실영상처리를 위한 셀룰러 신경망 설계 (A Design of a Cellular Neural Network for the Real Image Processing)

  • 셀룰러 신경망(Cellular Neural Network: CNN)은 간단한 처리요소인 셀들의 배열로 이루어져 있으며 각 셀들은 국부적인 연결특성과 공간불변 템플릿 특성을 갖는다. 따라서 CNN은 하드웨어로 구현하는데 매우 적합한 구조를 갖는다. 그러나 CNN 하드웨어 프로세서를 실제의 대형 영상의 화소와 1:1로 매핑하는 것은 불가능하다. 본 논문에서는 소규모의 CNN 셀 블록을 사용하여 대규모의 영상을 블록단위로 처리하는 실용적인 시다중화 영상처리 기법을 적용할 수 있는 파이프라인 입${\cdot}$출력을 갖는 $5{\times}5$ CNN 하드웨어 프로세서를 설계하였다. 그리고 윤곽선 검출과 그림자 검출 실험을 통하여 구현된 CNN 하드웨어 프로세서의 동작을 검증하였다.

소부혈(少府穴) 자침(刺鍼)이 Kainic Acid로 유도(誘導)된 간질(癎疾) 동물(動物) 모델의 해마(海馬) 치상회(齒狀回)에 미치는 영향(影響) (Acupuncture Treatment at HT8 Protects Hippocampal Cells in Dentate Gyrus on Kainic Acid-Induced Epilepsy Mice Model)

  • Objectives : Epilepsy is one of the most common serious brain disorders that affect people of all ages, and it is characterized by recurrent unprovoked seizures. We examined whether acupuncture can reduce both the incidence of seizures and hippocampal cell death in dentate gyrus (DG) using a mouse model of kainic acid (KA)-induced epilepsy. Methods : ICR mice ($20{\sim}25$ g) were given acupuncture once a day at acupoint HT8 (sobu) bilaterally during 2 days before KA injection. After an intracerebroventricular injection of 0.1${\mu}g$ of KA, acupuncture treatment was subsequently administered once more (total 3 times), and the degree of seizure was observed for 20 min. Three hours after injection, we confirmed the neural cell death using cresyl violet staining and silver impregnation staining, and determined the expressions of c-Fos and glutamate decarboxylase (GAD)-67 using immunohistochemistry techniques in the DG. Results : KA induced epileptic seizure, neural cell death, increased c-Fos expression and decreased GAD-67 expression in the DG. Acupuncture treatment at HT8 reduced the severity of the epileptic seizure and inhibited neural cell death from KA. In addition, acupuncture normalized the expressions of c-Fos and GAD-67 in the same areas. Conclusions : These results demonstrated that acupuncture treatment at HT8 may reduce the KA-induced epileptic seizure and neural cell death in the DG possibly by normalizing c-Fos expressions and the gamma-aminobutyric acid neurons.

Isolation of human mesenchymal stem cells from the skin and their neurogenic differentiation in vitro

  • Objectives: This aim of this study was to effectively isolate mesenchymal stem cells (hSMSCs) from human submandibular skin tissues (termed hSMSCs) and evaluate their characteristics. These hSMSCs were then chemically induced to the neuronal lineage and analyzed for their neurogenic characteristics in vitro. Materials and Methods: Submandibular skin tissues were harvested from four adult patients and cultured in stem cell media. Isolated hSMSCs were evaluated for their multipotency and other stem cell characteristics. These cells were differentiated into neuronal cells with a chemical induction protocol. During the neuronal induction of hSMSCs, morphological changes and the expression of neuron-specific proteins (by fluorescence-activated cell sorting [FACS]) were evaluated. Results: The hSMSCs showed plate-adherence, fibroblast-like growth, expression of the stem-cell transcription factors Oct 4 and Nanog, and positive staining for mesenchymal stem cell (MSC) marker proteins (CD29, CD44, CD90, CD105, and vimentin) and a neural precursor marker (nestin). Moreover, the hSMSCs in this study were successfully differentiated into multiple mesenchymal lineages, including osteocytes, adipocytes, and chondrocytes. Neuron-like cell morphology and various neural markers were highly visible six hours after the neuronal induction of hSMSCs, but their neuron-like characteristics disappeared over time (24-48 hrs). Interestingly, when the chemical induction medium was changed to Dulbecco's Modified Eagle Medium (DMEM) supplemented with fetal bovine serum (FBS), the differentiated cells returned to their hSMSC morphology, and their cell number increased. These results indicate that chemically induced neuron-like cells should not be considered true nerve cells. Conclusion: Isolated hSMSCs have MSC characteristics and express a neural precursor marker, suggesting that human skin is a source of stem cells. However, the in vitro chemical neuronal induction of hSMSC does not produce long-lasting nerve cells and more studies are required before their use in nerve-tissue transplants.
