Ultrastructural Aspects of Mixed Infections with Turnip mosaic virus (TuMV- ACl8 and -C5) and Ribgrass mosaic virus (RMV-CA1) in Oriental Cabbage

  • Mixed infections of two economically important viruses, Turnip mosaic virus(TuMV) in the family Potyviridae and Ribgrass mosaic virus(RMV) in the genus Tobamo-virus, were studied ultrastructurally on oriental cabbage. TuMV-ACl8 (alpine isolate in Korea) induced chlorotic spots on inoculated leaves of both ‘SSD63’ inbred line known as susceptible to TuMV, and ‘Tambok’ commercial cultivar, known as resistant to the virus, in the early stages of infection. TuMV-C5 (Taiwan isolate) caused severe mosaic and malformation on the upper leaves of ‘SSD63’, and necrotic spots in both inoculated and upper leaves of ‘Tambok’. RMV-CA1 (oriental cabbage isolate from alpine in Korea) induced vein chlorosis, leaf malformation, and midrib necrotic streak in the upper leaves of both ‘SSD63’ and ‘Tambok’. Both oriental cabbages infected with a combination of TuMV-ACl8 and RMV-CA1 showed synergistic symptoms of severe yellowing, severe mosaic, and necrotic spot or vein necrosis on their leaves. A combination of TuMV-C5 and RMV-CA1 produced synergistic symptoms only in ‘SSD63’. In ‘Tambok’ infected with the combination of TuMV-C5 and RMV-CA1, the number of necrotic spots on the inoculated leaves was one half lesser than that on singly infected with TuMV-C5. A few necrotic spots progressed systemically. In cells infected with a combination of TuMV-ACl8 and RMV-CA1 or TuMV-C5 and RMV-CA1, the particles of the two viruses made nonagon-like rings(NLR); one TuMV particle was surrounded loosely by nine RMV particles. Two unrelated viruses of TuMV and RMV were compacted in the central part of the spiral aggregates(SA) that was induced strikingly in cells by the mixed infections. The SA showed NLR in its center of the cross-sectioned side. Many particles of RMV of Tobamovirus were closely associated with Potyvirus-characteristic cylindrical inclusions. The SAs in the mixed infections were formed easily by the Potyvirus of TuMV-ACl8 or -C5 isolates.

Chinese yam necrotic mosaic virus Isolated from Chinese Yam in Korea (한국산 장마(Dioscorea oppasita cv. Jang-Ma)에서 분리한 Chinese yam necrotic mosaic virus)

  • Chinese yam (Dioscorea opposita cv. Jang-Ma) plants showing necrotic mosaic symptom were collected from their growing fields in Andong, Euisong, Gunwi and Daegu, Korea. Direct negative stainning method by electron microscope showed filamentous particles of about 660 nm in length. Immunosorbent electron microscopy (ISEM) revealed filamentous particles of 660nm decorated with antiserum of Chinese yam necrotic mosaic virus (ChYNMV). The virues purified partially were used to isolate viral RNA as template for RT-PCR to amplify about 1.2 kbp of 3'-terminal region (coat protein, 3'-UTR) with ChYNMV specific and oligo-dT primers. Amino acids sequences of amplified CP genes revealed that the viruses shared 97.9% similarity with ChYNMV (AB044386) wh ich was known as the member of Macluravirus. So the viruses from Chinese yam (D. opposita cv. Jang-Ma) plants were identified as ChYNMV. Comparing the CP amion acid sequences of ChYNMV strains with other macluraviruses such as Cardamon mosaic virus (CdMV), Narcissus latent virus (NLV) and Maclura mosaic virus (MacMV) revealed that N-terminal was the most varialbe region and conserved regions were present within the genus Macluravirus.

First Report of Impatiens Necrotic Spot Virus on Plantago asiatica Cultivated in Open Fields (노지재배 질경이(Plantago asiatica)에서 봉선화괴저반점바이러스병 발생 국내 첫 보고)

  • In August 2020, necrotic ringspots on leaves were observed on 20 from 143 Plantago asiatica plants in open fields in Eumseong, Chungcheongbuk-do. Eight symptomatic Plantago asiatica plants were subjected to investigation on viral infection by reverse transcription and polymerase chain reaction. Impatiens necrotic spot virus, tomato spotted wilt virus and cucumber mosaic virus were detected from the symptomatic plants. Two impatiens necrotic spot virus (INSV) isolates ('INSV-plantain kr1' and '-plantain kr5') were sequenced and analyzed by comparing L genomic segment, nucleoprotein (N) gene and non-structural protein S (NSs) gene sequences. The nucleotide sequence of 'INSV-plantain kr1' isolate (MW114834) was most closely related to that of a 'Phalaenopsis' isolate (GQ336991) from China in the L genomic segment. 'INSV-plantain kr1' and '-plantain kr5' isolates shared the highest identities with those from 'Pepe' isolate (LC384872) and 'J' isolate (AB109100) in the NSs gene, respectively, and with that from 'YSMi-SH' isolate (FN400773) in the N gene. Phylogenetic analysis based on L genomic segment grouped the INSV-plantain kr1 isolate together with isolates from Korea (LC384870), China (GU112505, GQ336991), and Italy (DQ425094). This is the first report on INSV in P. asiatica from Korea.

  • A toxoplasmic uveitis case was reported on the focus of impairment of pathological findings and serological antibody titers after chemotherapy. A chief complaint of a 60-year-old male was a decreased and blurred vision in his right eye for 2 weeks after experiencing tremendous stress and fatigue. A steroid therapy for 3 weeks was not effective and the retinal lesion became necrotic. Anti-Toxoplasma gondii antibody titer was checked to be a strong positive by both ELISA and indirect latex agglutination assay (lLA). He was treated with Fansidar F for 8 weeks. His vision improved as the necrotic lesion healed by scarring, but the antibody titers still remained very high without any signs of negative conversion. It is suggested to be a recurrent case of the past asymptomatic infection by presumed immune suppression caused by excessive stress.

Effects of Air Pollution on Pinus thunbergii in Yocheon Industrial Comples (麗川工業 地의 大氣汚染이 곰솔에 미치는 影響)

  • To examine the effects of air pollution on Pinus thunbergii growing around Yocheon Industrial Complex, evaluation of visible injury based on Injury Index and chemical analysis of water-soluble sulfur and fluorine contents in needles were carried out on 30 sampling sites. Histological responses of needles were also investigated on the necrotic needle tissues. Not only Injury Index but water-soluble sulfur and fluorine contents were higher at all sites than those at control site, and showed significant correlationship among them. Especially, they were rather higher in the vicinity of air pollutants sources. Water-soluble sulfur contents were positively correlated with average atomospheric $SO_3$ concentrations. Microscopic examination of necrotic needle tissues showed that mesophyll cells were collapsed with hypertrophy and collapse of transfusion cells.

  • Surgery, Chung-Ang Unviersity College of Medicine, Yong-San Hospital, Seoul, Korea Apoptosis is considered to be a programmed and controlled mode of cell death, whereas necrosis has long been described as uncontrolled and accidental cell death resulting from extremely harsh conditions. In the following review, we will discuss the features and physiological meanings as well as recent advances in the elucidation of the signaling pathways of both apoptotic cell death and programmed necrotic cell death.

First Report of a Foliar Nematode Aphelenchoides fragariae (Aphelenchidae) on Stachys riederi var. japonica, a Medicinal Plant, in Korea

  • Leaf blight symptoms and abnormal defoliation were found on Stachys riederi var. japonica, a medicinal plant, grown at Hongcheon, Gangwon province, Korea in 2007. A foliar nematode, Aphelenchoides fragariae was isolated from infected leaf tissues. The symptoms observed on host plant were leaf blotching, discoloration and browning of tissues, tan-colored interveinal necrotic lesions and large dead patches of necrotic tissues. This is the first report of A. fragariae occurring on S. riederi var. japonica.

  • Sputum smears of 116 cases in 55 pulmonary tuberculosis patients with the demonstration of acid-fast bacilli in sputum were retrospectively reviewed to investigate the cytologic diagnosis of pulmonary tuberculosis in sputum. Epithelioid cells, multinucleated giant cells of Langhans or nonspecific type, or necrotic materials of caseation or nonspecific nature were found in 40% of the cases, but in 60% of the cases, only nonspecific findings including squamous metaplasia were found. Only in 6% of the cases, the cytologic diagnosis of tuberculosis was possible with the findings of epithelioid cells and muitlnucleated giant cells in a necrotic background.

  • Clinically, three holstein dairy cattle showed the inappetence and decrease of milk production with severe lameness and pain. Grossly, strawberry-like proliferation of the skin between the heel bulbs were observed 4${\times}$5${\times}$1.5cm in size. Histopathologically, severe hyperplasia of epidermis, parakeratotic hyperkeratosis and multifocal ulceration of cornified cell layer, acanthosis, spongiosis of prickle cell layer and irregular arrangement of basal cell layer were seen. And focally observed the lesions of necrotic vasculitis, infiltration of inflammatory cells, congestion and hemorrhage. Spirochete bacteria were found in necrotic and outer proliferated epidermal cells by Levaditi's silver stain. Clinical and pathological findings were consistent with dermatitis verrucosa.
