제주산 감귤과즙을 이용하여 감귤식초를 제조할 목적으로 식초산 발효균 CV1과 CV2를 분리하고 식초산 생성능력을 산업적으로 이용되고 있는 균주와 비교 검정하고 165 rDNA 염기서열을 분석하고 동정하였다. 분리균은 8일간의 발효에 의하여 산도 $5.5\%$ 이상의 식초산을 생성하였으며, 감귤식초 발효에는 사과식초나 감식초 발효에 이용되고 있는 균주 보다 더욱 적합한 것으로 판정되었다. 분리한 균주는 Gluconacetobacter hanenii(CV2) 및 G. hanenii의 변이주(CV1)로 동정되었다.
줄알락명나방과 미끌애꽃노린재의 효율적인 대량생산을 위하여 온도 $23{\pm}2^{\circ}C$, 습도 $70{\pm}5%$, 광조건 16L : 8D의 실내사육실에서 증식 최적화 조건을 탐색하였다. 줄알락명 나방 생산효율은 $20{\ell}$ 사육용기에서 28.6배, $3{\ell}$ 사육용기에서 6.4배였다. 미끌애꽃노린재는 $5{\ell}$ 사육용기에 성충 1,500, 10 cm 채란식물을 10개 접종했을때 효율이 16.3배였다. 줄알락명나방은 모든 사료배합 실험구에서 알 수확량은 큰 차이가 없었으나, 소맥피(43%), 쌀겨(43%), 이스트(14%)로 조성된 먹이사료에서 알 수확량(5.3 g/통)이 가장 많았고, 혼합한 사료도 비용면에서 경제적이었다.
Microbial wound infection prolonged the hospitalization and increase the cost for wound management. Silver is commonly used as antimicrobial wound dressing. However, it causes several adverse side effects. Hence, this study was aimed to evaluate the antimicrobial efficiency of Swietenia macrophylla seed extract on clinical wound pathogens. Besides, the bioactive constituents of the seed extract were also determined. S. macrophylla seeds were extracted with methanol by maceration method. The seed extract inhibited 5 test bacteria and 1 yeast on disc diffusion assay. The antibacterial activity was broad spectrum, as the extract inhibited both Gram positive and Gram negative bacteria. On kill curve analysis, the antibacterial activity of the seed extract was concentration-dependent, the increase of extract concentration resulted in more reduction of bacterial growth. The extract also caused 99.9% growth reduction of Bacillus subtilis relative to control. A total of 21 compounds were detected in gas chromatography- mass spectrometry analysis. The predominant compounds present in the extract were oleic acid (18.56%) and linoleic acid (17.72%). In conclusion, the methanolic extract of S. macrophylla seeds exhibited significant antimicrobial activity on clinical wound pathogens. Further investigations should be conducted to purify other bioactive compounds from the seeds of S. macrophylla.
More and more available fungal genome sequence data reveal a large amount of secondary metabolite (SM) biosynthetic 'dark matter' to be discovered. Heterogeneous expression is one of the most effective approaches to exploit these novel natural products, but it is limited by having to clone entire biosynthetic gene clusters (BGCs) without errors. So far, few effective technologies have been developed to manipulate the specific large DNA fragments in filamentous fungi. Here, we developed a fungal BGC-capturing system based on CRISPR/Cas9 cleavage in vitro. In our system, Cas9 protein was purified and CRISPR guide sequences in combination with in vivo yeast assembly were rationally designed. Using targeted cleavages of plasmid DNAs with linear (8.5 kb) or circular (8.5 kb and 28 kb) states, we were able to cleave the plasmids precisely, demonstrating the high efficiency of this system. Furthermore, we successfully captured the entire Nrc gene cluster from the genomic DNA of Neosartorya fischeri. Our results provide an easy and efficient approach to manipulate fungal genomic DNA based on the in vitro application of Cas9 endonuclease. Our methodology will lay a foundation for capturing entire groups of BGCs in filamentous fungi and accelerate fungal SMs mining.
Xylophagous termites are capable of degrading lignocellulose by symbiotic gut microorganisms along with the host's indigenous enzymes. Therefore, the termite gut might be a potential niche to obtain natural yeasts with celluloytic, xylanolytic and ethanologenic traits required for bioethanol production from lignocellulosic biomass. In this study, we cultured 79 yeasts from three different termites viz. Coptotermes heimi, Odontotermes javanicus and Odontotermes obesus. After suitable screening methods, we identified 53 yeasts, which belonged to 10 genera and 16 different species of both ascomycetous and basidiomycetous yeasts. Most yeasts in the present study represent their first-ever isolation from the termite gut. Representative strains of identified yeasts were evaluated for their cellulolytic, xylanolytic, and ethanologenic abilities. None of the isolates showed cellulase activity; 22 showed xylanolytic activity, while six produced substantial quantities of ethanol. Among xylanolytic cultures, Pseudozyma hubeiensis STAG 1.7 and Hannaella pagnoccae STAG 1.14 produced 1.31 and 1.17 IU of xylanase. Among ethanologenic yeasts, the strains belonging to genera Candida and Kodamaea produced high amount of ethanol. Overall, highest ethanol level of 4.42 g/L was produced by Candida tropicalis TS32 using 1% glucose, which increased up to 22.92 g/L at 35 ℃, pH 4.5 with 5% glucose. Fermentation of rice straw hydrolysate gave 8.95 g/l of ethanol with a yield of 0.42 g/g using the strain TS32. Our study highlights the gut of wood-feeding termites as a potential source of diverse yeasts that would be useful in the production of xylanase and bioethanol.
에리스리톨은 4탄당의 당알코올에 해당되며 포도당을 원료로 효모에 의해 생산되는 발효 감미료제로 버섯, 포도주, 과실류, 청주, 간장 등의 발효식품에 함유되어 있는 천연당이다. 에리스리톨과 다른 감미제에서 Streptococcus mutans (S. mutans)의 glucosyl transferase (GTF)와 fructosyl transferase (FTF)의 유전자 발현 양상을 확인하여 치아우식 예방을 위한 제품을 생산하거나 활용 시 올바른 정보와 기초자료를 제공하고자 시행하였다. 연구결과 에리스리톨은 치아우식에 관여하는 S. mutans의 생육을 억제하는 것으로 나타났으며 자당(설탕) 대체 감미제로서 우식예방 효과가 우수한 것으로 확인이 되었다. 특히 세포외다당류 합성에 관련된 gtf B, gtf C, gtf D, ftf 의 발현이 낮게 나타나 세포외부에 다당류의 합성을 억제하여 치면세균막 형성과 치면에 세균의 부착률을 감소시킬 수 있을 것으로 생각된다. 따라서 Streptococci에 의한 우식기전에 에리스리톨이 효과적으로 항우식 감미제로서 사용될 수 있다고 생각된다.
Plant biomass, or lignocellulose, is one of the most abundant natural resources on earth. Lignocellulosic biomass, such as agricultural and forestry residue, serves as a renewable feedstock for microbial cell factories due to its low price and abundant availability. However, the recalcitrance of lignocellulosic biomass requires a pretreatment process prior to microbial fermentation, from which fermentable sugars including xylose and glucose are generated along with various inhibitory compounds. The presence of furan derivatives, such as 5-hydroxymethyl-2-furaldehyde and 2-furaldehyde (furfural), hampers the microbial conversion of lignocellulosic biomass into value-added commodities. In this study, furfural tolerance was improved by investigating the detoxification mechanism in non-model yeast. The genes encoding aldehyde dehydrogenases were overexpressed to enhance furfural tolerance and resulted in improving cell growth and lipid production that can be converted into biofuel. Taken together, this approach contributes to the understanding of the reducing toxicity mechanism of furfural by the aldehyde dehydrogenases and provides a promising strategy that the use of microorganism as an industrial workhorse to treat efficiently lignocellulosic biomass as sustainable plant derivatives.
구리(Cu)는 저렴한 비용으로 용이하게 도입이 가능하여 다양한 소재 표면에 살균 코팅제로 쓰이고 있다. 자연적 산화 반응이 구리의 효능을 손상시키지 않아 장기간 노출 조건에서도 항균 성능을 유지할 수 있다. 더 나아가 구리 화합물은 그람 음성균 및 그람 양성균 뿐만 아니라, 병원성 효모, 외피 보유 및 외피 미보유 타입의 바이러스에 대해 모두 폭넓은 살균 효과를 보인다. 구리 코팅 표면의 접촉 살균은 구리의 침투로 단백질 변성을 일으키고 세포막 손상으로 뉴클레오티드 및 세포질 등의 내용물이 용출되게 한다. 또한 구리 산화환원 활성에 의한 활성 산소종 생성으로 효소작용을 억제하고 DNA를 파괴하여 세포를 영구적으로 손상시킨다. 구리는 안정한 금속 성질 때문에 나노입자, 이온, 복합물, 합금 등의 여러 형태로 쓰이고 있으며 코팅 방법이 다양하다. 본 총설에서는 구리 이온과 구리 산화물의 대표적인 표면 도입 방법을 살펴보고 구리 산화수에 따른 항균·항바이러스 특성을 다루고자 한다.
글루타티온은 간 질환용 치료제 내지는 영양보충제로 시판되고 있다. 이전 실험에서 글루타티온 고함유 효모 Saccharomyces cerevisiae FF-8 균체(FF-8GY) 투여가 사염화탄소 및 알코올성 유발 간독성 개선효과를 나타내었다. 따라서, 알코올성 지방간의 산화 스트레스에 미치는 영향을 조사하기 위하여 시판되고 있는 글루타티온 고함유 효모 추출물(GYE)과 본 실험실에서 분리한 글루타티온 고함유 효모 FF-8GY을 비교실험 하였다. FF-8GY 또는 GYE 투여에 의한 간 조직 중의 산화스트레스 측정지표로서는 과산화지질(TBARS) 및 글루타티온 함량을 측정하였다. TBARS 농도는 간 조직 homogenize 분획에서 알코올 투여 대조군에 비해 FF-8GY 투여 실험군에서 5%수준에서 유의적으로 감소하였으며, GYE 투여에 의해서는 감소경향을 보였다. 간 조직 microsomal 및 mitochondria 분획에서도 같은 경향이 관찰되었다. 이때 간 조직 중의 글루타티온 농도는 정상군 보다 알코올 투여 대조군에서 5%수준에서 유의적으로 감소하였으며, 이러한 감소는 GYE 투여에 의해 더욱 감소되었으나 FF-8GY 투여에 의해서 정상군 수준까지 회복되는 것으로 나타났다. 한편 간조직의 손상정도를 나타내는 또 하나의 지표인 혈청 총단백질 및 알부민 농도는 알코올 처리 대조군에서 낮게 나타난 반면 FF-8GY 투여에 의해 정상군 수준 이상으로 증가하는 것으로 나타났다. 이상의 결과에서 글루타티온 고함유 효모 Saccharomyces cerevisiae FF-8 균체(FF-8GY)의 투여에 따른 알코올성 유발 산화스트레스의 경감 효과는 시판되고 있는 글루타티온 고함유 효모 추출물 보다 더욱 현저한 개선 효과가 있는 것으로 확인되었다.
Alaska pollack caught in the Northern Pacific Ocean and frozen aboard vessel are skipped to the plant and processed into frozen fillets. In the present paper quality changes during thwaing, refreezing and storage at $-20^{\circ}C$ are discussed. Natural, running-water, vacuum and steam thawing were employed as thawing methods. And contact plate, air blast, immersion in dry ice-alcohol solution freezing and storage at $-5^{\circ}C$ were applied to refreeze the thawed fillets. As quality factors content of drip released, salt-extractable protein, VBN, DNA in the drip and pH were determined. In addition, bacteriological tests were also carried out along with the whole process. In thawing of round material, the vacuum thawing was more effective than any other method, resulting in drip, salt-extractable protein $(N\%)$, VBN and DNA as $4.4\%,\;1.82\%,\;16.21mg\%$ and $13.70\;{\mu}g/ml$, respectively. Storage at $-5^{\circ}C$ as refreezing method yielded lower in drip and DNA content but similar to or slightly higher in both salt-extractable protein and VBN, which might postulate that the quality of the frozen fillet depends not largely on the secondary freezing but on the conditions of thawing and primary freezing. It seemed that most of the bacterial flora in thawed fillet came from skin and viscera of fish, worker's hands, utensils and other processing facilities, since sanitary indicative bacteria were not detected in the frozen flesh of round Alaska pollack. Bacterial quality of fillet varied with thawing methods, vacuum thawing appeared more sanitative compared with other methods as natural, running-water, and steam thawing. Bacterial colonies isolated from the thawed fillet were composed of $73.8\%$ Gram negative rod shape, $4.9\%$ Gram positive rod shape, $18.0\%$ cocci, and $3.3\%$ yeast. Decreasing rate of coliform group of the fillet during the storage at $-20^{\circ}C$ for 30 days was more than $70\%$ and that of plate count was less than of coliform group.
