Development of GOCHUJANG for Controlling V. parahaemolyticus with Green Tea and Natural Products (녹차 및 천연산물로부터 장염 비브리오균(Vibrio. parahaemolyticus)의 제어를 위한 고추장의 개발)

  • Vibriocidal GOCHUJANG was manufactured using extract of natural products and green tea. Antioxidative capacity of natural products and combination of natural products was measured with Oxidation-Reduction Potential (ORP) system value. Inhibitory ability of vibriocidal GOCHUJANG was compared with Inhibition Zone Diameter (IZD) values. Phellodendri cortex, Schizandrae fructus, Theae folium, Scutellaria baicalensis george and Acanthodanacis cortex and combination of these natural products were showed higher antioxidant capacity and inhibitory effect against V. para-haemolyticus. Two kinds of combinations, A combination was composed of Phellodendri cortex, Schizandrae fructus, Theae folium, Scutellaria baicalensis george, and B combination was consisted of Acanthodanacis cortex and A combination, were presented higher inhibition ratio with IZD values as $1.57\pm0.0051$ and $1.56\pm0.0071$, respectively. Vibriocidal GOCHUJANG was prepared based on A and B combination with their higher IZD Value as $1.76\pm0.0103$ and $1.79\pm0.0103$, respectively. When vinegar was added to A and B based GOCHUJANG, their IZD value was more higher as $2.15\pm0.0071$ and $2.44\pm0.0086$, respectively B combination based GOCHUJANG was exhibited higher inhibitory effect than A based GOCHUJANG. Combination of natural products was more important process for increasing vibriocidal effect. But addition of soybean malt or chitosan or a garlic into A or B combined GOCHUJANG, no different effect was exhibited.

  • Acid mine drainage occurs when sulfide minerals are exposed to an oxidizing environment. The objective of this study was to inhibit the oxidation of pyrite by applying various coating agent such as $KH_2PO_4$, MgO and $KMnO_4$ over its surface as an oxidation inhibitors. Experiments were conducted for 8 days to test the feasibility of oxidation inhibitors. The optimal condition of coating agent for standard pyrite and IK mine was the combination of 0.01M $KH_2PO_4$, 0.01M NaOAc and 0.01M NaClO. Otherwise, for YD mine the combination of 0.01M $KMnO_4$, 0.01M NaOAc and 0.01M NaClO. The $SO_4^{2-}$ reduction efficiency of pyrite, IK and YD mine samples was 70, 92 and 84%, respectively. For 8 days, no significant increase of $SO_4^{2-}$ from pyrite sample coated with inhibitor was observed. The pH of solution remains in between 4 to 6 for the reaction conditions.

Modulation of Bacteria with a Combination of Natural Products in KIMCHI Fermentation

  • Three kinds of bacteria that influence Kimchi fermentation, Lactobacillus plantarium for acidity, Leuconostoc mesenteroides for ripening Kimchi, and Pichia membranifaciens for decreasing Kimchi quality, were regulated by natural products including Theae folium, Taraxacum coreanum, Brassica juncea, Astragali radix, Gynostemma pentaphyllum, Camellia japonica, Agaricus blazei, and Cordyceps militaris. The common prescription combined T. folium, T. coreanum and C. militaris and simultaneously regulated these 3 bacteria as follows: the growth of L. plantarium and P. membranifaciens were inhibited and L. mesenteroides was promoted. The most effective mixing ratio was T. folium: T. coreanum: C. militaris = 3:2:1. With this new prescription, deep flavor, extended preservation, and a special taste are expected in the Kimchi due to these natural products.

  • Antioxidative capacity of natural products and combination of natural products was measured with ORP system value. Inhibitory ability of vibriocidal GOCHUJANG was compared with IZD values, inhibited clear zone was divided by diameter of disk. Phellodendri cortex, Schizandrae fructus, Theae folium, Scutellaria baicalensis george and Acanthodanacis cortex and Combination of these natural products were showed higher antioxidant capacity and controlling ability against V. parahaemolyticus. Two kinds of combinations, A combination was composed of Phellodendri cortex, Schizandrae fructus, Theae folium, Scutellaria baicalensis george, and B combination was consist Acanthodanacis cortex plus A combination, were presented higher inhibition ratio with IZD values as 1.57 and 1.56, respectively. Vibriocidal GOCHUJANG was prepared based on A and B combination with their higher IZD value as 1.67 and 1.79, respectively. and, when vineger was added to A and B based GOCHUJANG their IZD value was more higher as a 2.15 and 2.44, respectively. B combination based GOCHUJANG was exhibited higher controlling effect than A based GOCHUJANG. Combination of natural products was more important precedure for increasing vibriocidal effect.

  • The combined effect of berberine isolated from the bark of Phellodendron amurense and some antibiotics was evaluated in order to measure the antibiotic activities. In this study, in the presence of streptomycin, tetracycline, cephradine as antibiotics, Staphylococcus aureus, Escherichia coli, Shigella sonnei as microorganisms were grown in an Automated Microbiology System. In case of Staphylococus aureus, combination with berberine and cephardine resulted in the strongest synergistic activity and in case of Escherichia coli and Shigella sonnei, combination with berberine and streptomycin resulted in the strongest synergistic activity. The combination with berberine and antibiotics increased the antibiotic activities, thereby showing a synergistic action.

  • Two meroterpenoids (1 and 2) along with twelve known compounds (3 - 14) were isolated from the culture broth of a Penicillium sp. fungus collected from Chuja-do, Korea. Based on the results of a combination of spectroscopic analyses, the new compounds, preaustinoids E (1) and F (2), were determined to be epimeric austintype penta-cyclic lactones.

  • P-glycoprotein (P-gp) is a permeability glycoprotein also known as multidrug resistance protein 1 (MDR1). P-gp is an ATP-binding cassette (ABC) transporter that pumps various types of drugs out of cells. These transporters reduce the intracellular concentrations of drugs and disturb drug absorption. The Caco-2 cell permeability assay system is an effective in vitro system that predicts the intestinal absorption of drugs and the functions of enzymes and transporters. Rhodamine-123 (R-123) and digoxin are well-known P-gp substrates that have been used to determine the function of P-gp. Efflux of P-gp substrates by P-gp has been routinely evaluated. To date, a number of herbal medicines have been tested with Caco-2 cell permeability assay system to assess bioavailability. There are growing efforts to find phytochemicals that potentially regulate P-gp function. The Caco-2 cell permeability assay system is a primary strategy to search for candidates of P-gp inhibitors. In this mini review, we have summarized the P-gp modulation by herbal extracts, decoctions or single components from natural products using Caco-2 cell permeability assays. Many natural products are known to regulate P-gp and herbal medicines could be used in combination with conventional drugs to enhance bioavailability.

  • A convex combination of two products with same degree of finitely many finite geometric series with each having even degree does not always have all its zeros on the unit circle. However, in this paper, we show that a polynomial obtained by just adding a finite geometric series multiplied by a large constant to such a convex combination has all its zeros on the unit circle.

The Growth Inhibiting Effect of Enterobacteria, Clostridium pefringens KCTC 5014, by the Combination of Natural Products (Clostridium perfringens KCTC 5014의 성장 억제를 위한 항산화 천연산물의 효과)

  • The growth of C. pefringens was inhibited by the combination of natural products which have antioxidative capacity C. perfringens was controlled very effectively by each or the combination of Paeonia japonica, Atractylodis rhizome alba, Epimedii herba, Acori graminei rhizoma, Schizandraefructus, Lyciifructus, Cinnamomi cortex Sophorae flos Phellodendri cortex, Nelumbo nuclfera gaertner, Asparagi tuber, Mori cortex radicis, Astrafali radix and Coptidis rhizome The combination of Phellodendri cortex, Epimedii herba, Astragali radtx and Glycyrrhizae radix inhibited the growth of C. pefringen by 75% and increased the antioxidative activity by 4.4 times. This combination also eliminated more than 80% of hydroxyl radical. These natural products can be utilized as pharmaceuticals, functional foods, antiaging tea, and multifunctional fermentative beverages.

Natural Products for Cancer-Targeted Therapy: Citrus Flavonoids as Potent Chemopreventive Agents

  • Targeted therapy has been a very promising strategy of drug development research. Many molecular mechanims of diseases have been known to be regulated by abundance of proteins, such as receptors and hormones. Chemoprevention for treatment and prevention of diseases are continuously developed. Pre-clinical and clinical studies in chemoprevention field yielded many valuable data in preventing the onset of disease and suppressing the progress of their growth, making chemoprevention a challenging and a very rational strategy in future researches. Natural products being rich of flavonoids are those fruits belong to the genus citrus. Ethanolic extract of Citrus reticulata and Citrus aurantiifolia peels showed anticarcinogenic, antiproliferative, co-chemotherapeutic and estrogenic effects. Several examples of citrus flavonoids that are potential as chemotherapeutic agents are tangeretin, nobiletin, hesperetin, hesperidin, naringenin, and naringin. Those flavonoids have been shown to possess inhibition activity on certain cancer cells' growth through various mechanisms. Moreover, citrus flavonoids also perform promising effect in combination with several chemotherapeutic agents against the growth of cancer cells. Some mechanisms involved in those activities are through cell cycle modulation, antiangiogenic effect, and apoptosis induction.Previous studies showed that tangeretin suppressed the growth of T47D breast cancer cells by inhibiting ERK phosphorylation. While in combination with tamoxifen, doxorubicin, and 5-FU, respectively, it was proven to be synergist on several cancer cells. Hesperidin and naringenin increased cytotoxicitity of doxorubicin on MCF-7 cells and HeLa cells. Besides, citrus flavonoids also performed estrogenic effect in vivo. One example is hesperidin having the ability to decrease the concentration of serum and hepatic lipid and reduce osteoporosis of ovariectomized rats. Those studies showed the great potential of citrus fruits as natural product to be developed as not only the source of co-chemotherapeutic agents, but also phyto-estrogens. Therefore, further study needs to be conducted to explore the potential of citrus fruits in overcoming cancer.
