고련피 추출물의 항암활성 (Anti-cancer Activities of Extract from the Bark of Melia azedarach L. var. japonica Makino)

  • In the present study, the anti-cancer activity of 80% ethanol extracts from 120 kinds of medicinal herbs and native plants were investigated. Among them, the barks of Melia azedarach L. var. japonica Makino showed the highest cytotoxicity in HCT-15 human colon cancer cell. With this result, we carried out hollow fiber (HF) assay and anti-metastasis study to confirm the anti-cancer effects of M. azedarach var. japonica. In MTT assay, M. azedarach var. japonica.inhibited the proliferation of HCT-15 cells in dose-dependent manner. HF assay was carried out using A549 human adenocarcinoma cell, HCT-15 and SK-Hep1 human liver cancer cell via intraperitoneal (IP) and subcutaneous (SC) site. As a results, SK-Hep1 implanted in IP site showed the highest cytotoxicity. The result from metastatic model using B16/BL6 mouse corresponded to that of HF assay. These results suggest that the ethanol extract from M. azedarach var. japonica. might have a potent anti-cancer activity and advanced study is needed for the development of novel natural anti-cancer drug.

Process Balance of Natural Indigo Production based on Traditional Niram Method

  • In this work, the natural indigo production process from Polygonum tinctorium was balanced based on the traditional Niram method in Korea. A standard procedure was determined considering the conditions of indican extraction from plant material, the amount of alkali for precipitation, storage of extract, etc. The effect of experimental conditions on the yield of crude dye was investigated. The contents of indigo and indirubin of the crude dyes were analyzed by HPLC. Increase of the amount of crude dye was observed within 1-2.5 days of extraction time. Longer extraction beyond 2.5 days resulted in a slight decrease in the amount of crude dye. There was no consistency in terms of indigo content depending on extraction pH. We found that the storage of extract or harvested plants affected adversely to dye yield and dye quality. Based on the lab scale extraction, large scale extraction was performed for 2-2.5 days in water and 2.0-2.5 g/L of $Ca(OH)_2$ was applied for precipitation of indigo dye. We obtained natural indigo dye containing about 15% of pure indigo in scale-up production using whole plant except root.

Iridoid Compounds from the Whole Plant of Galium verum var. asiaticum

  • One new iridoid glycoside, 10-p-dihydrocoumaroyl-6-${\alpha}$-hydroxygeniposide (1), and six known iridoid glycoside derivatives, 10-p-dihydrocoumaroyl deacetylasperuloside (2), asperulosidic acid methylester (3), asperuloside (4), asperulosidic acid (5), deacetylasperuloside (6), and scandoside (7) were isolated from the methanolic extract of the whole plant of Galium verum var. asiaticum Nakai (Rubiaceae) through repeated column chromatography. Their chemical structures were characterized by spectroscopic analysis. This is the first report of the characterization of compounds 1 - 7 from this plant.

식물성 천연 추출액을 첨가한 고추장의 특성 (Studies on the Properties of kochujang by Addition of Natural Plant Extracts)

  • 식물성 천연 추출액을 첨가하여 60일간 숙성시킨 고추장과 일반 고추장의 유리 아미노산 분석 결과, cysteine, isoleucine, tyrosine, lysine을 제외한 나머지 유리 아미노산들의 함량이 일반 고추장에 비해 식물성 천연 추출액을 첨가한 고추장에서 높게 나타났고, 식물성 천연 추출액을 첨가한 고추장에서 감칠맛의 주요 성분인 gutamic acid 함량이 높게 증가함을 나타내었다. 한편 glutamic acid 외에도 aspartic acid, proline, valine, histidine등의 함량이 식물성 추출액을 첨가한 고추장에서 높았다. 식물성 천연 추출액을 첨가한 고추장의 관능적 특성에 미치는 색, 향, 구수한 맛, 매운맛, 단맛, 짠맛, 전체적인 기호도를 조사한 결과, 일반 고추장에 비해 향, 구수한 맛, 단맛에서 높은 값을 보였고, 색이나 매운 맛, 짠맛은 거의 비슷한 값을 나타내었다. 전체적인 기호도에서 일반 고추장에 비해 기호도가 높은 것을 볼 수 있었다. 따라서 고추장에 식물성 천연 추출액을 첨가하여 숙성시키는 것이 고추장의 구수한 맛, 향, 단맛을 향상시킬 뿐만 아니라 천연물의 한방적인 효능이 있을 것으로 생각된다.

  • Chemical investigation of the hot hexane extract of the stem bark of Gordonia dassanayakei afforded a new oleanane triterpenoid, $11{\alpha},13{\beta}-dihydroxyolean-3,12-dione$ (1) and two other oleanane triterpenoids, $3{\beta}-acetoxy-11{\alpha},13{\beta}-dihydroxyolean-12-one\;(2)\;and\;3{\beta},11{\alpha}-diacetoxy-13{\beta}-hydroxyolean-12-one\;(3)\;and\;a\;hopane\;6{\alpha},22-diol$ (4), which are new to the plant.

탱자 열매 추출액의 항산화 및 지질저하효과 (Antihyperlipidemic and Antioxidant Effects of Poncirus trifoliata)

  • 탱자 추출액이 비만을 유도한 흰쥐의 지질강하 및 항산화효과에 미치는 영향을 검토했다. 혈장유리지방산 및 triglyceride 량은 대조군과 비교하여 탱자 처리군이 유의하게 낮은 값을 보였다. 그러나 정상군의 수준까지 나타내지는 않았다. 총콜레스테롤 및 LDL-cho.농도는 탱자 처리군 모두가 대조군보다 현저하게 하락한 값을 나타내었다. HDL-cho.은 대조군을 비롯하여 전 처리군에서 유의한 차이를 나타내지 않았다. 간장내 총콜레스테롤과 triglyceride량은 정자 처리군이 대조군 보다 현저하게 낮은 값을 보였다. 혈장 및 간장의 TBARS 농도는 탱자처리량이 증가함에 따라 감소하였다. GSH-Px, SOD 및 CAT 활성치는 탱자 처리군에서 높은 활성치를 보였다. 그러나 SOD의 활성치는 대조군과 유의한 차이를 나타내지는 않았다.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

부처손(Selaginella tamariscina) 추출물의 리파아제 저해 활성 및 지질 축적 억제 효과 (The Effect of Selaginella tamariscina on Inhibition of Pancreatic Lipase and Lipid Accumulation)

  • The objective of this study was to evaluate novel usability as natural anti-obesity supplement of Selaginella tamariscina extract. The total phenol contents and total flavonoid contents were $60.29{\pm}3.11GAE\;mg/g$ and $14.90{\pm}0.34QE\;mg/g$, respectively. To evaluate anti-obesity activity of Selaginella tamariscina extract, pancreatic lipase inhibition activity as well as its inhibition effect of lipid accumulation in adipocytes were conducted by Oil Red O staining and lipolysis assay. The result of pancreatic lipase inhibition activity of S. tamariscina extract showed a wide range between 40 and 73% dose dependently. While the incubation of 3T3-L1 cells with S. tamariscina extract inhibited differentiation of preadipocytes and reduced lipid accumulation, the level of released free glycerol into culturing medium was increased in multiple concentrations. These results showed that S. tamariscina extract inhibit adipogenesis and pancreatic lipase activity. Thus, S. tamariscina extract can be a candidate for regulating lipid accumulation in obesity.

Anti-Melanogenic Activities of Ranunculus chinensis Bunge via ERK1/2-Mediated MITF Downregulation

  • Research on whitening materials using natural alternatives is actively being conducted. The aim of this study was to investigate the in vitro inhibitory effects of Ranunculus chinensis Bunge (RCB) on melanogenesis and associated enzymes, such as tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 in B16F10 murine melanoma cells. We found that RCB extract significantly attenuated melanin synthesis and reduced the activity of intracellular tyrosinase, a rate-limiting melanogenic enzyme. Western blot analysis showed that RCB extract decreased the protein expression of tyrosinase and TRP-1. In addition, it significantly decreased the expression of microphthalmia-associated transcription factor (MITF), a key regulator of melanogenesis. Extracellular signal-regulated kinase (ERK) activation has been reported to be involved in the inhibition of melanogenesis. Thus, we investigated whether the hypopigmentary effects of RCB extract were related to the activation of ERK. RCB extract induced ERK phosphorylation in a dose-dependent manner. Furthermore, it markedly inhibited body pigmentation in a zebrafish model. Our results suggest that RCB extract inhibits melanogenesis by activating ERK pathway-mediated suppression of MITF and its downstream target genes, including tyrosinase. Therefore, RCB extract can be used as a whitening agent in the development of functional cosmetics.

Methanol Extract of Cinnamomum cassia Represses Cellular Proliferation and Gli-mediated Transcription in PANC-1 Human Pancreatic Cancer Cells

  • Twenty five methanolic plant extracts were investigated to determine the anticancer activity against sonic hedgehog (shh)/Gli signaling pathway dependent cancer, PANC-1 human pancreatic cancer cells, through three screening programs. All extracts were inspected their inhibitory properties on sonic hedgehog-conditioned medium (shh-CM) induced alkaline phosphatase (ALP) activity in C3H10T1/2 mouse mesenchymal stem cells to examine whether the plant extracts affect the shh/Gli signaling pathway. Next, plant extracts were screened the ability to suppress the cell proliferation of PANC-1 human pancreatic cancer cells. Finally, active plant extracts from the two screening systems were evaluated for the suppressive effect on Gli-mediated transcriptional activity in PANC-1 cells. Among active plants, Cinnamomum cassia suppressed Gli-mediated transcriptional activity leading to the down-regulated expression of Gli-target genes such as Gli-1 and Patched-1 (Ptch-1). This study provides the consideration for the important role of natural products in drug discovery process as well as the basis for the further analysis of active plant and potential identification of novel bioactive compounds as inhibitors of Gli and therapeutic candidates against shh/Gli signaling pathway dependent cancers.
