장내 세균총과 위장관 질환 (Gut Microbiome and Gastrointestinal Diseases)

  • 본 연구에서는 최근에 연구되어 온 장내 세균총이 특히 주요 장질환의 발병에 있어서, 어떠한 역할을 하는지 보고된 여러 문헌들을 중심으로 연구 결과들을 요약 하였다. 예를 들자면, 면역체계를 매개하여 병이 악화되는 여러 질환에서 정상일 때와 달리 바뀐 장내 세균총을 통해 악화된다고 보고된 바 있다. 장내 세균총의 역할에 대해 많이 연구된 장질환 중에서, 많이 연구된 3개의 질환은 과민성 장 증후군, 염증성 장질환, 대장암이다. 그러나, 사람의 장내에 존재하는 세균총은 몸에 이로우며, 비타민 A 합성, 단사슬지방산의 생산, 담즙산 대사과정과 같은 장내 생리적 기능 매개를 통해 장내 항상성을 유지한다고 알려져 있다. 이와 같이, 장내에 존재하는 이로운 세균 군집과 해로운 세균 군집의 균형은 장내 건강에 주요한 영향을 미친다. 장질환을 포함한 여러 질환의 발병 및 진행에서 장내 세균총의 변화가 주요 원인으로 추측되고 있는 실정이다. 현재까지 보고된 많은 연구 결과에도 불구하고, 어떤 장내 세균총 구성이 몸에 가장 이로운 지학계에서도 의견이 분분한 상태이다. 본 논문에서는, 주요 장질환으로 알려진 과민성 장 증후군, 염증성 장질환, 대장암과 장내 세균총과의 관계에 대해 연구한 논문들에 대해 연결 지어 요약하였다. 마지막으로, 장내 세균총을 매개로 악화되는 장질환을 완화하며, 장내 건강을 지키기 위한 수단으로 천연물을 이용한 치료 전략을 제시하고자 한다.

한강의 Microflora에 관한 연구 (제6보) -남한강의 식물성플랑크톤에 대한 분류와 한강중심수역의 수질오탁판정- (A Stduy on the Microflora of the Han River -Taxonomy of Phytoplankton for the South Han River and Estimation of Water Pollution Levels on the Central Area of the Han River-)

  • In order to utilize for the prevention and preservation of the Han River from the environmental pollution the present studies were carried out to clarify the microflora and estimation of the water pollution levels of the Han River. In addition to the above regional and seasonal fluctuation of the phytoplankton was also examined. Samples of phytoplankton were collected from 6 stations in the South Han River during the period from December, 1971 to October, 1972. The results obtained during the present studies are as follows: 1. The phytoplankton samples collected from 6 stations, Yeoju, Hajapo-ri, Yangpyeong, Daruraegi, Giduwon and Paldang were identified and classified by Engler's classification system(1954). It resulted in 2 phylum, 2 classes, 7 orders, 10 families, 29 genera, 137 species, 1 sub-species, 49 varieties, 6 forma and 2 variety-forma. The total numbers of phytoplankton identified were 195 species, of which 7 families, 27 species, 26 varieties, 4 forma and 2 variety-forma are new to Korea, that of 54 species are first described in Korea. 2. In lower area of the Han River it is found 53 species from Paldang and in middle area it is found 114 species from Giduwon, 95 species from Daruraegi, 66 species from Yangpyeong, 71 species from Hajapori and 81 species from Yeoju. In standpoint of seasonal fluctuation of phytoplankton, the total numbers of the plankton is more abundant in summer than in winter season and it shows bimodal pattern. 3. As compared with previous data which obtained from 30 stations covering estuary to upper area, both South and North Han River, during the period from 1965-1972 it is shown that 10 species of the South Han River and 11 species of the North Han River are found throughout all seasons. Among the above species two are common in both area. In the other hand it is found that 9 species in spring season and 6 species in fall season in the South Han River and 10 species in spring, 23 species in summer, 4 species in fall and 15 species in winter season in the North Han River shows their seasonal fluctuation in this area. Among the seasonal occurrence of phytoplankton 10 species were consider to be indicator for the estimation of biological water pollution levels. 4. According to Fjerdingstad's water pollution level system (1963) the total numbers of 1, 230 species which have been collected from the Han River since 1965 includes 27 species of phytoplankton as indicator; 3 species of blue-green algae, 20 species of diatom, and 4 species of green-algae. 5. With 27 indicator species new estimation of water pollution level system was arranged for water pollution in the Han River. 6. The lower part of the central area of the Han River indicates mesosaprobic. In central area of the Han River shows mesosaprobic and oligosaprobic, but predominant in mesosaporobic. And it is indicated that mesosaprobic, oligosaprobic, and polysaprobic factors mixed up in the North Han River. Compare with their water pollution level in the South and North Han River, with author's new system, it is estimated that North Han River is more polluted than South Han River. 7. The reason why North Han River is more polluted suggested that the selfpurification action was limited by their circulation speed. The rapid speed of water in the North Han River is mainly caused by their topography and water-drainage from waterpower plant. In conclusion the central area of the Han River consist of mesosaprobe zone, as a part with oligosaprobe zone. But the presence of polysaprobe zone in the North Han River gives us many problems in future for the nationa development programme and natural conservation in this area.

인공 및 천연제올라이트의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향

  • 본 실험은 석탄재를 이용하여 합성한 인공제올라이트의 사료적 가치를 구명하기 위하여 천연 제올라이트와 동일한 수준으로 첨가 ·급여하여 육계의 생산성 및 장내 미생물, ND항체가등에 미치는 영향을 5주간 고찰하였다. 전기 3주 동안 사료내 조단백질은 21.5%, ME 3,100kcal/kg으로 하였고 후기에는 조단백질 19%, ME 3,100kcal/kg 수준으로 급여하였다. 처리구는 대조구(무처리구), 인공제올라이트와 천연제올라이트를 각각 1.5, 3.0%를 육계용 사료에 급여하였으며, 처리구당 4반복, 반복당 16수로 전체 320수를 공시하였다. 조사항목으로 증체량, 사료섭취량, 사료요구율은 주간별로 측정하였으며, 장내 미생물, 혈중 콜레스테를 및 ND 항체가는 실험종료시에 측정하였고 영양소 이용율은 실험 종료후 대사실험을 통하여 측정하였다. 증체량은 NZ 처리구에서 증가하는 경향을 보였으나 대조구와 통계적인 차이가 없었고 AZ 3.0% 처리구는 대조구에 비하여 현저하게 감소하였다(P<0.05). 사료섭취량은 처리구간에 차이가 없었으며 사료요구율은 AZ 3.0% 처리구에 비하여 NZ 3.0%가 현저하게 개선되었다(P<0.05). 제올라이트가 육계의 장내 미생물에 미치는 영향은 대조구와 차이가 없었으며 혈중 총콜레스테롤은 NZ 처리구가 감소하였다(P<0.05). 또한 NZ 처리구는 조섬유 소화율이 다른 처리구에 비하여 현저하게 증가하였다(P<0.05). 본 실험의 결과 인공제올라이트는 1.5% 급여구에서 대조구와 비슷한 결과를 얻었으므로 사료내 첨가수준은 1.5%로 사료된다.

  • 본 실험은 석탄재를 이용하여 합성한 인공제올라이트 (AZ)의 사료적 가치를 구명하기 위하여 천연제올라이트(NZ)와 동일한 수준으로 첨가 . 급여하여 육계의 생산성, 장내 미생물 및 혈중 콜레스테롤 등에 미치는 영향을 5주간 고찰하였다. 전기 3주 동안 사료내 조단백질은 21.5%, ME 3,100 kal/kg으로 하였고, 후기에는 조단백질 19%, ME 3,100kcal /kg 수준으로 급여하였다. 처리구는 대조구(제올라이트 0%), AZ와NZ를 각각 1.5,3.0% 씩 육계용 사료에 배합하였으며, 처리구당 4반복, 반복당 16수로 전체 320수를 공시하였다. 조사항목으로 증체량, 사료섭취량, 사료요구율은 주간별로 측정하였다. 장내 미생물, 혈중 콜레스테롤 및 뉴켓슬병 백신에 대한 항체가는 실험종료시에 측정하였다. 증체량은 NZ 처리구에서 증가하는 경향을 보였으나 대조구와 통계적인 차이가 없었고, AZ 3.0%처리구는 대조구에 비하여 현저하게 감소하였다(P<0.05). 사료섭취 량은 처리구간에 차이가 없었으며 사료요구율은 AZ 3.0% 처리구에 비하여 NZ 3.0%가 현저하게 개선되었다(P<0.05).제올라이트가 육계의 장내 미생물에 미치는 영향은 대조구와 차이가 없었으며 혈중 총콜레스테롤은 NZ 처리구에서 현저하게 감소되었고 뉴켓슬병백신에 대한 항체가는 제올라이트 처리구가 현저하게 증가하였다(P<0.05). 본 실험의 결과, AZ 1.5% 급여수준은 육계의 생산성에서 대조구와 차이가 없었으므로 육계 사료에 이용될 수 있는 수준으로 사료된다.

The oral microbiome of implant-abutment screw holes compared with the peri-implant sulcus and natural supragingival plaque in healthy individuals

  • Purpose: An implant-supported prosthesis consists of an implant fixture, an abutment, an internal screw that connects the abutment to the implant fixture, and the upper prosthesis. Numerous studies have investigated the microorganisms present on the implant surface, surrounding tissues, and the subgingival microflora associated with peri-implantitis. However, there is limited information regarding the microbiome within the internal screw space. In this study, microbial samples were collected from the supragingival surfaces of natural teeth, the peri-implant sulcus, and the implant-abutment screw hole, in order to characterize the microbiome of the internal screw space in healthy subjects. Methods: Samples were obtained from the supragingival region of natural teeth, the peri-implant sulcus, and the implant screw hole in 20 healthy subjects. DNA was extracted, and the V3-V4 region of the 16S ribosomal RNA was sequenced for microbiome analysis. Alpha diversity, beta diversity, linear discriminant analysis effect size (LEfSe), and network analysis were employed to compare the characteristics of the microbiomes. Results: We observed significant differences in beta diversity among the samples. Upon analyzing the significant taxa using LEfSe, the microbial composition of the implant-abutment screw hole's microbiome was found to be similar to that of the other sampling sites' microbiomes. Moreover, the microbiome network analysis revealed a unique network complexity in samples obtained from the implant screw hole compared to those from the other sampling sites. Conclusions: The bacterial composition of the biofilm collected from the implant-abutment screw hole exhibited significant differences compared to the supra-structure of the implant. Therefore, long-term monitoring and management of not only the peri-implant tissue but also the implant screw are necessary.

식품내의 미생물 분리를 위한 dryfilm 방법의 평가연구

  • Dryfilm method by using 3M Petrifilm$^{TM}$ has been examined to replace conventional agar method for isolation of microorganisms from foods. The objectives of the present study were to evaluate suitability of dryfilm method as a microbial isolation method and to determine the effect of antimicrobial agent on dryfilm for isolation of microorganisms from foods. Five different foods, milk, ground beef, fishery surimi, Takju and wheat flour were used to isolate the natural microflora in foods and the inoculated Escheri chia coli. Standard method agar (SMA, Difco) and Petrifilm$^{TM}$ aerobic count (PAC, 3M) were used to isolate total microorganisms from foods. Violet red bile agar (VRBA), brilliant green lactose bile (BGLB) broth and Petrifilm$^{TM}$ coliform count (PCC, 3M) were used to isolate coliforms from foods. E. coli broth (EC broth) and Petrifilm$^{TM}$ E. coli count (PEC, 3M) were used to isolate E. coli from foods. Acidified potato dextrose agar (APDA) and Petrifilm$^{TM}$ yeast & mold count (PYMC, 3M) were used to isolate yeasts and molds from foods. Total aerobic plate counts isolated from five different foods by SMA and PAC (3M) were riot significantly different each other at P<0.05 level and were highly correlated each other ($\geq$0.96). Mugwort extract as an antimicrobial agent did not affect microbial enumeratiion of Dryfilm. Significantly higher number of coliform colonies were formed on VRBA than PCC (3M) from ground beef, but they were not significantly different in coliform colonies from milk samples. PCC (3M) and BGLB were not significantly different for enumeration of coliforms in milk and beef samples. Significantly higher number of E. coli were isolated by EC broth than PEC from ground beef, but these were not significontly different for enumeration of E. coli from milk. Yeast and mold counts isolated from Takju and wheat flour by APDA and PYMC (3M) were not significantly different at P<0.05 level. These data indicate that dryfilm method by using 3M Petrifilm$^{TM}$ can be successively used as an alternative to conventional agar method for enumeration of microorganisms in various foods.

Scientific Analysis of Formulation Theory of Chungpesagan-tang; The purgative Action of Chungpesagan-tang

  • To analyze scientifically the fundamental formulation theory and drug interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke, water extracts of herbal medicines with Rhei Rhizoma were administered into mice and the purgative action was measured. Water extracts of Raphani Semen or Cimicifugae Rhizoma with Rhei Rhizoma had more potent purgative activity than Rhei Rhizoma alone, although Raphani Semen alone displayed no purgative action. However, Platycodi Radix and Puerariae Radix inhibited the purgative action of Rhei Rhizoma. When Rhei Rhizoma with Raphani Semen or Cimicifugae Radix was extracted with water, sennoside A content in the water extract was increased. However, Puerariae Radix and Platycodi Radix decreased the content of sennoside A in the extracted water. The transforming rate of sennoside A of Rhei Rhizoma was inhibited by Puerariae Radix and Platycodi Radix. The transforming rate of sennoside A of Chungpesagan-tang to rheinan-throne by human intestinal microflora and its purgative activity were similar to those of Rhei Rhizoma. Finally we thought that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

축산업 분야에서의 박테리오신의 산업적 이용 및 향후 전망 (Perspectives for the Industrial Use of Bacteriocin in Dairy and Meat Industry)

  • 최근까지 박테리오신 생산균주의 분리, 분자생물학적 메커니즘, 정제, 구조 및 작용기작, 산업적인 적용 등의 논문 등이 보고되고 있다. 또한 50개국 이상에서 치즈, 통조림식품 등에서 식품보존제로서 승인되어 있다. 박테리오신의 여러 장점으로 인해 현재 추진되고 있는 축산업 분야의 HACCP의 범위인, 사육장에서부터 식품제조에 이르기까지 항생물질, 인공적인 방부제나 식품첨가물이 적게 들어가거나 첨가되지 않은 자연식품을 선호하고 있는 실정에 적합하다. 향후 축산업에서 항생제 대체방안의 하나로서 박테리오신의 사용은 확대되리라 기대된다. 경제적인 가격 경쟁력을 갖기 위해, 대량생산에 대한 연구가 이루어져야 하며, 적절한 제형으로 보다 구체적인 적용실험이 수행되어 산업화를 앞당겨야 한다.

리그난의 화학적 특성, 인체 건강에 미치는 영향 및 리그난의 우유 기능 강화에 관한 연구 고찰 (Chemical Properties of Lignans, Their Effects on Human Health, and the Enhancement of Milk Function of Lignans)

  • This review discusses the characteristics of major lignans and related studies and provides a basis for future studies. Lignans are present in various food products consumed daily, such as flaxseed and other seeds, vegetables, fruits, and beverages including coffee, tea, and wine. Lignans are natural phytoestrogens with a structure similar to that of secoisolariciresinol (Seco), mataireinol (Mat), pinoresinol (Pin), medioresinol (Med), lariciresinol (Lari), and syringaresinol, which is then converted to enterodiol (END) and enterolactone (ENL), which are mammalian lignans and the primary biologically active enterolignans, by the intestinal microflora. The associations between lignans and a decreased risk of cardiovascular disease are promising; however, they are not yet well-established, probably owing to low lignan intake in habitual Western diets. Nonetheless, these associations were more prominent at the higher doses in interventional studies. Many studies on humans and animals have reported the benefits of lignan consumption in protecting against CVD and metabolic syndrome by reducing lipid and glucose concentrations. END and ENL reportedly exert protective effects including phytoestrogenic, antioxidant, anti-inflammatory, and anticancer effects through various mechanisms. Moreover, lignans reportedly exert beneficial effects in breast, colon, and prostate cancer and osteoporosis have reported that. However, future studies are required to confirm the association between lignan and disease.

Antibiosis and Bacteriocin Production of Lactic Acid Bacteria Isolated from Kimchi

  • In order to elucidate roles of lactic acid bacteria(LAB) for the antibiosis occurring in th fermenting environment of Kimchi, 2.052 strains of LAB were isolated from Kimchi. Fifty tow strains which showed antagonistic effect against 4 indicator strains were finally selected and investigated. Based upon responses to protease treatment, antibiosis of the 52 strains of LAB were classified into 3 types. Type A antibiosis resulted from action of antibiotic-like substances which were not affected by protease treatment and which had broad action spectra against even natural inhabitants of Kimchi. Type B antibiosis was due to bacteriocin-like substances which were very sensitive to treatment of protease and more effective against foreign bacteria than original inhabitant microflora. Type C antibiosis was owing to proteinaceous compounds which were activated or induced by the presence of protease and then exerted antibacterial activities. Therefore, lactic acid bacteria appeared to contribute to antibiosis of Kimchi by the concerted action of these three different types of antibacterial compounds. As one of model system for type B bacteriocin, the antagonistic compound produced by LAB31-9 as well as th producer strain itself was further charaacterized. Strain LAB31-9 was identified as L. casei. Bacteriocin produced by LAB31-9 was proteinaceous and stable over wide range of pH and to various solvents, but very labile to heat treatment. Its mode of action was bactericidal. Based upon these data, bacteriocin produced by LAB31-9 was named as 'caseicin K319'. Genetic determinant for the bacteriocin production of LAB31-9 was located in the chromosome.
