Experimental Studies on Activity of the Cultivated Mycelia of Phellinus linteus (상황(桑黃) 배양균사체의 활성에 관한 연구(I))

  • Phellinus linteus was examined for its anticancer activity using an animal model. Water extract of Phellinus linteus was prepared from artificially cultivated mycelia. Neither toxicity nor abnormal changes of hematological parameters were observed in the rat given orally with high doses of drug extract for 15 days. ICR mice were transplanted with Sarcoma-180 tumor cells intraperitoneally and drug extract was daily given to the mice from 1 day after tumer transplantation for 3 weeks. Administration of drug extract significantly prolonged the survival duration of Sarcoma 180-transplanted mice. For the better understanding of the anticancer activity, we have examined the effect of the drug extract administration on various killer cell functions, such as natural killer(NK) cells, cytotoxic T-lymphocytes (CTL) and macrophages which have been known to be main effector cells in immune responses against tumors. The results from the 4 hr $^{51}Cr-release$ assay have shown that the drug extract augments mouse NK cell activity but neither CTL nor macrophages. It is possible, then, that the anticancer activity of the Phellinus linteus may be associated with augmentation of NK cell function in the cancerated hosts.

Effects of Quercetin on the Immune Responses in Mice (Quercetin이 마우스의 면역반응에 미치는 영향)

  • Effects of quercetin on the specific and non-specific immune responses were studied in vivo. Quercetin at a dose of 2.5, 5, 10, 20 and 40 mg/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune responses were evaluated by humoral and cellular immune reponses and non-specific immune response. The results of this study were summarized as followings; 1. Quercetin significantly decreased the body weight, and introduced the atrophy of liver, spleen and thymus gland dose-dependently, but increased the numbers of white blood cell. 2. Querectin significantly depressed the hemagglutination titer, Arthus reaction and hemolytic plaque forming cell. 3. Quercetin significantly depressed the delayed type hypersensitivity and rosette forming cell. 4. Quercetin at a dose of 2.5, 5 and 40 mg/kg significantly depressed phagocytic activity. 5. Quercetin at a dose of 10 and 20 mg/kg significantly increased natural killer cell activity.

  • The objective of the study is to estimate the potential of gintonin, as an immune enhancing agent through natural killer cell (NK cell) activity in cyclophosphamide (CY)-induced immunosuppressive animals. Accumulated results reveals that, gintonin attenuated CY-induced immunosuppression and it might modulate NK cell activity to boost the immunity.

Improved Anti-Cancer Effect of Curcumin on Breast Cancer Cells by Increasing the Activity of Natural Killer Cells

  • Curcumin is known to possess various biological functions, including anti-inflammatory, anti-oxidative, and anti-cancer activities. Natural killer (NK) cells are large lymphocytes that directly kill cancer cells. However, many aggressive cancers, including breast cancer, were reported to escape the successful killing of NK cells in a tumor microenvironment. In this study, we investigated the anti-cancer effect of curcumin in coculture of human breast carcinoma MDA-MB-231 and NK (NK-92) cells. We found that curcumin had an immune-stimulatory effect on NK-92 by increasing the surface expression of the $CD16^+$ and $CD56^{dim}$ population of NK-92. We confirmed that the cytotoxic effect of NK-92 on MDA-MB-231 was significantly enhanced in the presence of curcumin, which was highly associated with the activation of Stat4 and Stat5 proteins in NK-92. Finally, this improved anticancer effect of curcumin was correlated with decreased expression of pErk and PI3K in MDA-MB-231.

A STUDY ON THE CHANGE OF T-LYMPOCYTE AND NATURAL KILLER CELL IN H & N CANCERS (두경부악성 종양 환자의 T-lymphocyte 및 Natural Killer Cell에 대한 연구)

  • 악성종양의 발생과 진행에 면역 방어기능이 중요한 역할을 하리라는 가설은 모두가 공감하는 사실이다. 이중 T-lymphocyte와 Natural killer cell (이하 NK cell이라함)은 종양 면역학에 특히 중요한 임파구로 이런 임파구의 혈액분포양상은 면역방어기능을 짐작할 수 있는 간접적인 자료가 될 수 있다. 저자들은 치료전 두경부 악성종양환자에서 혈액을 채취하여 T-lymphocyte와NK cell의 분포양상을 검사하고, 방사선치료 환자에서는 NK cell activity를 측정하였기에 다음과 같은 결과를 보고하는 바이다. 1) 두경부 악성 종양 환자군에서 CD3+ cell은 감소하고 NK cell은 증가하며 CD4/CD8 비율은 변화가 없었다. 2) 병변이 진행되면서 CD3+ cell과 CD4+ cell은 감소하고 NK cell은 증가하였으며 CD4/CD8 비율의 변화는 없었다. 3) 방사선치료에 의해 CD3+ cell과 CD4+ cell, CD4/CD8 비율은 감소하였고, NK cell과CD8+cell은 증가하였다. 4) 방사선치료에 의한 CD4/CD8 비율의 감소와, CD8+ cell의 증가는 NK cell의 증가에 의한 것이라 추정되고, NK cell을 제외하면 CB4/CD8 비율의 변화는 없었다. 5) 방사선치료 환자에서 NK cell activity는 증가하였고, 이런 증가가 T-lymphocyte기능의 감소를 보상해 주고 있었다.

Effect of Biphenyl dimethyl Dicarboxylate on the Cellular and Nospecific Immunosuppressions by Ketoconazole in Mice

  • The effect of biphenyl dimethyl dicarboxylate (PMC) on the cellular and nonspecific immunosuppressions by ketoconazole (KCZ) was investigated in ICR mice. PMC at a dose of 6 mg/kg was administered orally to mice daily for 14 consecutive days. KCZ was suspended in RPMI 1640 medium and orally administered at 160 mg/kg/day 2 hrs after the administration of PMC. Immune responses of the delayed-type hypersensitively (DTH) reaction to sheep red blood cells (SRBC), phagocytic activity and natural killer (NK) cell activity were evaluated. DTH reaction to SRBC was enhanced to normal level by the combination of PMC and KCZ, as compared with treatment of KCZ alone. In the combination of PMC and KCZ, as compared with treatment of KCZ alone, there were also significant increases in activities of natural killer (NK) cells and phagocytes along with circulating leukocytes. These findings indicate that PMC shows a significant restoration from the immunotoixc status induced by KCZ.

  • To clarify the activating effects of Buthus martensi Karsch on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated. Buthus martensi Karsch increased the number of both antibody producing cells(anti-IgM and anti-IgG producing plaque forming cells, PFC) and phagocytic activity of peritoneal macrophage. Futhermore, these phenomena were significantly increased with aging in mice. Buthus martensi Karsch also increased natural killer cell activity concerning to cancer immunology. These results suggest that Buthus martensi Karsch markedly increases the reduced activity in the elderly and activates the immune response in senescence mice.

Effects of the Pilose Antler on the Experimental Hepatocarcino- genesis and the Natural Killer Cell Activity in Rats (랫드의 실험적 간암발생과 자연살해세포의 활성에 미치는 녹용의 효과)

  • This study was performed to investigate the modifying effect of the general (GPA) and the fermented pilose antler (FPA) on experimental hepatocarcinogenesis and Natural Killer cell activity in rats. Specific pathogen free, 5-week male Sprague-Dawley rats were divided into four groups. To induce hepatocarcinogenesis, diethylnitrosamine (DEN, 200 mg/kg, i.p.) was used as a tumor initiator and was given in a single dose at experimental onset. All rats were given a partial hepatectomy (PH) at 3 weeks after experimental onset. Sodium phenobarbital (PB, 0.05% in diet), GPA (0.075% in diet) and FPA (0. 075% in diet) were given from 2 to 8 weeks. Group I of the initiation control group was only given DEN. As initiation-promotion group, Group II was given DEN and then PB. Group III and IV were given DEN-PB-GPA and DEN-PB-FPA, respectively. In hematological analysis, as compared with Group I. the number of white blood cells were significantly increased in the GPA (p<0.01) and the FPA treated group (p<0.05), respectively. Natural killer (NK) cell activity by flow cytometer (FCM) analysis was higher in group of treated with the GPA (35%) than that of the FPA (27.5%), but not significant. Result of the immunohistochemical staining of the glutathione S-transferase placental form (GST-p) indicated that the number of and area of the pre-neoplastic lesions was not significantly changed in Group III and IV compared Group II, respectively. In conclusion, the GPA and the FPA treatment significantly increased the number od WBC in peripheral blood, but the enhancing NK activity and the modifying effect on the experimental hepatocarcinogenesis were not observed.

Natural killer cell activity in Naegzeria fowleri infected mice (Naegleria fowleri 자연살세포의 활성에 미치는 영향)

  • The natural killer (NK) cell activity of splenocytes and recycling capacity of NK cells were observed by combining the $^{51}Cr-release$ cytotoxicity assay and single cell cytotoxicity assay against YAC-1. The ICR mice were infected intranasally with Naegleria fewleri, that is a pathogenic free-living amoeba. The mice infected with $1{\times}10^5$ trophosoites showed mortality rate of 76.7% and mean survival time of 12. 9 days. The cytotoxic activity of NK cells in infected mice was significantly higher than that of non-infected mice during the period between 12 hours and day 3 after infection, and highest on day 1. The target-binding capacity of NK cells in infected mice was not different from that of non-infected ones. Maximal killing potential and maximal recycling capacity were remarkably increased in infected mice as compared with the control. The results obtained in this observation indicated that elevated NK cell activity in mice infected with N. fowieri was not due to target-binding capacity of NK cells but due to the increased activity of NK cells and increased recycling capacity of individual NK cells.

Enhancement of Anticancer Activities of Kimchi by Manipulating Ingredients

  • To enhance the antitumor activity of Chinese cabbage kimchi, four kinds of kimchi, which ere differently prepared in kinds and levels of sub-ingredients, were fermented at 15$^{\circ}C$ for 1 day and then at 5$^{\circ}C$ up to pH 4.3. The solid tumor formation, hepatic glutathione S-transferase activity and glutathione contents in the liver, and natural killer (NK) cell activity of spleen were determined from the sarcoma-180 cell injected Balb/c mice that were treated with methanol extracts of the kimchi samples. Kimchi IV, prepared with organically cultivated Chinese cabbage, red pepper powder, garlic, Chinese pepper powder mustard leaf and heat processed salt (Gueun salt), reduced the tumor formation by 39.3% compared to the sarcoma-180 cell treated group, resulting in the smallest tumor weight. Methanol extracts of the kimchi III and kimchi IV recovered the activities of hepatic glutathione S-transferase(GST) that was decreased by the transplantation of the sarcoma-180 cells to th mice. The injections of methanol extracts of kimchi II and kimchi IV increased glutathione contents in sarcoma-180 cells treated mice. The methanol extract of kimchi IV increased the natural killer (NK) cell activity of spleen lymphocytes a more effectively (p<0.05) than those the other kimchi samples. These results suggest that the anticancer activities of kimchi can be increased by changing the kinds and levels of sub-ingredients.
