대황으로부터 세균성 질염 치료를 위한 항균성 물질의 분리 및 특성 (Isolation and Characterization of an Antibacterial Substance from Rheum palmatum for Treatment of Bacterial Vaginosis)

  • Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.

  • Coix lacryma-jobi (Hanjeli) is known to posses anti-microbial properties. Therefore, phytochemical compounds of C. lacryma-jobi have been studied to produce novel antimicrobial agents as treatments against antibiotic-resistant bacteria.The objective of this study was to determine the phytochemical composition and antibacterial activity of the C. lacryma-jobi oil against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. The phytochemical composition of the oil was determined via gas chromatography mass spectrophotometry (GC-MS). Moreover, agar disk and agar well diffusion were employed to screen the antibacterial activity of the oil. An agar well diffusion test was implemented to determinate MIC's (minimum inhibitory concentrations). Dodecanoic acid, tetradecanoic acid, 2,3-dihydroxypropylester, 1,3-dioctanoin, N-methoxy-N-methyl-3,4-dihydro-2H-thiopyran6-carboxamide, propanamide, 5-Amino-1-(quinolin-8-yl)-1,2,3-triazole-4-carboxamide, and pyridine were identified in the C. lacryma-jobi oil. The MIC value of the oil was 0.031 g/L and the MBC of the oil was 0.125 g/L effective in all test bacteria. Dodecanoic acid displayed inhibitory activity against gram-positive and gram-negative bacteria. Therefore, our research demonstrated C. lacryma-jobi (Hanjeli) oil exhibited antibacterial activity against E. coli, S. aureus, and B. subtilis. These research suggest that C. lacryma-jobi root oil could be used for medicinal purposes; however clinical and in vivo tests must be performed to evaluate its potential as an antibacterial agent.

Antibacterial Effect of Amentoflavone and Its Synergistic Effect with Antibiotics

  • Selaginella tamariscina is a traditional herb used in medicine. Phytochemical amentoflavone, a biflavonoid class of flavonoids, was isolated from the plant of Selaginella tamariscina. In this study, the antibacterial effects and combination effects of amentoflavone and conventional antibiotics such as ampicillin, cefotaxime, and chloramphenicol were investigated. These results showed that amentoflavone had a considerable antibacterial effect and synergistic interaction with antibiotics against various bacterial strains (fractional inhibitory concentration index ${\leq}$ 0.5), except for Streptococcus mutans. To study the mechanism(s) involved in the synergistic activities between amentoflavone and antibiotics, we detected hydroxyl radical formation using 3'-(p-hydroxyphenyl) fluorescein and measured the $NAD^+/NADH$ ratio by $NAD^+$ cycling assay. The results indicated that the formation of hydroxyl radical would be a cause of the synergistic effect and that this oxidative stress originated from a transient NADH depletion. This study suggests that amentoflavone synergizes with antibiotics and has potential as a therapeutic agent for antimicrobial chemotherapy.

천연 항균물질이 첨가된 소금의 식중독 세균에 대한 항균효과 (Antibacterial Effects of Salt with Natural Antimicrobial Substances against Foodborne Pathogens)

  • Salt is a common seasoning agent used in various processed foods, especially in kimchi and salted seafood (jeotgal). This study was conducted to investigate the efficacy of salt with antimicrobial substances (acetic acid, garlic extract, carvacrol, nisin, thymol, and their combination (acetic acid+nisin+thymol)) on improvement of antibacterial effects of salt against foodborne pathogens. Salt (10%) was prepared using six different types of 0.2% natural antimicrobial substances. The antibacterial effect of salt combined with natural antimicrobial substances was evaluated against foodborne pathogens using the broth micro-dilution method and growth curve plotted using absorbance measurements. For the five foodborne pathogens, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of salt without antimicrobial substances as control were in the range of 24~>50,000 ㎍/mL and >50,000 ㎍/mL, respectively. Salt with nisin, thymol, or garlic extract showed strong inhibitory effects and their MIC against L. monocytogenes were 49, 12,500, and 24 ㎍/mL, respectively. In particular, salt with nisin showed inhibitory activities against Gram-positive bacteria. However, all the antimicrobial substances were less effective against Gram-negative bacteria such as E. coli O157:H7 and S. Typhimurium than Gram-positive bacteria. These results could be used for the development of salt with natural antimicrobial substances especially targeted against L. monocytogenes. This would enable the lowering of saline concentration while improving the storability of food.

Antibacterial Activity of Acanthoic acid Isolated from Acanthopanax koreanum against Oral and Skin Microfloras

  • The (-)-pimara-9 (11), 15-dien-19-oic acid, acanthoic acid was extracted from the roots of Acanthopanax koreanum using bioassay-guided isolation of a MeOH extract. Acanthoic acid was assayed against Streptococcus mutans and Staphylococcus epidermidis causing dental caries and opportunistic pathogen. The minimum inhibitory concentration (MIC) of acanthoic acid against Streptococcus mutans and Staphylococcus epidermidis was 2 and 4 ${\mu}g/mL$, respectively, which was much lower than those of other natural antimicrobial agents such as 8 ${\mu}g/mL$ of tanshinone IIA. Acanthoic acid also significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus and Streptococcus sanguis, and Streptococcus grodenii in the MIC range of 4${\sim}$32 ${\mu}g/mL$. Our findings suggest that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries and skin infections.

애완견의 털에 부착되어 있는 세균의 분리 및 황금의 살균효과 (Identification of Bacterial Strains Adhered to Dog Hair and Antibacterial Effects of Scutellaria baicalensis Gorgi)

  • 애완견의 털에 부착되어 있는 세균을 분리 동정한 후, 이 분리 세균들에 대한 항세균효과가 있는 생약재 추출물을 탐색하였다. 애완견의 털에서 76개의 세균을 분리한 결과, Staphylococcus속이 41균주로 가장 많았으며 그 다음으로 Micrococcus속이 21균 주, Enterococcus속이 8균주, Bacillus속이 3균주, Exiguobacterium속 of 2균주, Shigella속과 Zoogloea속이 각각 1균주씩으로 동정되었다. 이러한 결과는 개의 털에 존재하는 세균들이 사람에게 감염을 일으킬 수 있는 원인이 될 수 있음을 시사한다. 분리균주들에 대한 항생제 저항성과 생약재 추출물의 항세균 활성은 디스크 확산법으로 확인하였다. 황금의 추출물이 애완견의 털로부터 분리한 세균들에 대해 높은 항세균 활성을 나타내어 항생제 저항성이 있는 세균들에 대한 새로운 항미생물제로 사용학 수 있을 것으로 기대된다.

해양미세조류 에탄올 추출물의 항균활성에 관한 연구 (Antibacterial activity of ethanol extracts from marine micro-algae)

  • Mixed A, Chlorella vulgaris, Nannochloropsis oculata, Chaetoceros calcitrans 등 4종류의 미세조류로부터 99.9% 에탄올로 추출한 성분의 항균활성을 검증하기 위해 그람음성세균 2종(E. coli, S. Typhimurium)과 그람양성세균 2종(S. aureus, B. cereus)에 대하여 paper disk diffusion assay, MIC, MBC를 측정하였다. 4종류의 미세조류 추출물은 공시균주에 대하여 0.62-1.66 mg/mL의 MIC를 나타내었으며, C. vulgaris 추출물이 4종의 공시균주에 대한 MIC가 0.62 mg/mL로 가장 강한 항균활성을 나타내었으며, MBC 또한 C. vulgaris 에탄올 추출물이 E. coli에 대해 2.50 mg/mL, S. aureus, B. cereus와 S. Typhimurium에 대해서는 4.16-5.00 mg/mL로 가장 강한 항균활성을 나타내었다. 항균활성이 가장 강한 C. vulgaris 에탄올 추출물의 성분을 분석한 결과, 함량이 가장 많은 성분은 9,12-octadecadienoic acid (linoelaidic acid, peak 5, 7)로 38.8%이었으며, 그 다음이 2-Hexadecen-1-o1, 3,7,11,15-tetramethyl (phytol, peak 2,6)로 30.0%이었다. Octadecadienoic acid는 일명 linoelaidic acid로 탄소수 18개의 고도불포화지방산이며, 두 번째로 함량이 많은 Phytol은 클로로필을 구성하는 불포화 제1급 알코올의 일종으로서(엽록소의 구성 성분) 병원성 대장균 및 황색포도상구균에 항균활성을 나타낸다는 보고로 볼 때, 이 2가지 성분이 항균활성의 주성분인 것으로 사료된다.

New Azafluorenone Derivative and Antibacterial Activities of Alphonsea cylindrica Barks

  • A phytochemical study of Alphonsea cylindrica King (unreported) has led to the isolation of six alkaloids. The compounds were identified as kinabaline (1; azafluorenone alkaloid), muniranine (2), O-methylmoschatoline (3; oxoaporphine alkaloid), lysicamine (4), atherospermidine (5) and N-methylouregidione (6; 4, 5-dioxoaporphine alkaloid). The structures of the isolated compounds were determined based on the spectroscopic techniques and by comparison with data reported in the literature. Alkaloid 2 was isolated as a new derivative of azafluorenone while alkaloids 1, 3 - 6 were isolated for the first time from Alphonsea species. In addition, alkaloid 3 and 4 showed inhibition zone against Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus cereus in disc diffusion test. The minimum inhibition concentration (MIC) values of lysicamine (4) against S. aureus, B. cereus and P. aeruginosa were found to be smaller than O-methylmoschatoline (3). Therefore, the reported antibacterial activity showed the potential of this plant as natural antibacterial agent and supported the documented traditional use of Alphonsea sp. in the treatment of diarrhea and fever.

광귤 추출물의 Salmonella Typhimurium에 대한 항균 메커니즘 및 샐러드 세척 효과 (Antibacterial Mechanism and Salad Washing Effect of Bitter Orange Extract Against Salmonella Typhimurium)

  • 본 연구에서는 천연 항균제인 광귤 추출물의 항균 활성과 항균 메커니즘을 조사해 즉석섭취식품인 샐러드에서 Salmonella Typhimurium을 제어하기 위한 세척수로써 적용 가능성을 평가하였다. 액체배지희석법으로 S. Typhimurium에 대한 광귤 추출물의 최소 억제 농도(MIC)를 구했다. 그런 다음 다양한 농도(1/16 MIC-2 MIC)에 해당하는 광귤 추출물에 S. Typhimurium을 접종하고 성장곡선을 분석해 대조군과 성장값을 비교하여 항균 활성을 확인하였다. 광귤 추출물을 처리한 후, S. Typhimurium의 세포 내 활성산소종 수준과 막 전위 및 손상도의 변화, 핵산 누출량을 측정하여 광귤 추출물의 항균 메커니즘을 확인하였다. 최종적으로 S. Typhimurium을 인위적으로 접종한 샐러드에 다양한 농도의 광귤 추출물을 다양한 시간 동안 침지 방법으로 항균 처리하여 저감화 효과를 확인했다. S. Typhimurium에 대한 광귤 추출물의 MIC는 195.313 mg/L으로, 1 MIC와 2 MIC의 광귤 추출물은 S. Typhimurium의 성장을 완전히 억제하였다. 광귤 추출물의 처리농도가 높아질수록, 세포 내 ROS 수준과 막 전위, 막 손상도 그리고 핵산 방출량은 증가하였다. 마지막으로, 세척수인 광귤 추출물의 농도가 높고 처리 시간이 길수록 샐러드의 S. Typhimurium의 수가 감소하였다. 따라서 광귤 추출물은 S. Typhimurium를 효과적으로 제어할 수 있음을 입증했다. 광귤 추출물은 차아염소산나트륨과 비교하였을 때 효과적인 항균 활성을 보이며 안전한 샐러드 세척수로 사용될 수 있다. 이는 샐러드와 같은 식품에서 광귤 추출물이 식중독 발생을 미연에 방지하는데 기여할 수 있음을 시사한다.

새송이버섯 추출물이 구강세균에 작용하는 항균효과 (The antibacterial effect of Pleurotus eryngii extracts on oral bacteria)

  • Objectives: Pleurotus eryngii is used both for edible and medicinal purposes, and has a physiological activity. The purpose of this study is to investigate the antibacterial effect of Pleurotus eryngii against six oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Methods: The antibacterial activities of various extracts of Pleurotus eryngii were examined by disc diffusion assay and minimum inhibitory concentration (MIC). The disc diffusion assay was performed by putting a paper disc soaked in extracts on plates inoculated bacterial cultures. The MIC of these extracts was determined by using a broth microdilution assay at a concentration ranging between 0.03 mg/ml to 15.00 mg/ml. The growth inhibition effect of extracts was measured at 600 nm for 24 hrs. Results: The antibacterial activity was confirmed against all six tested bacteria at Pleurotus eryngii ethyl acetate extract by the disc diffusion method. Acetone extract showed the antibacterial activity only against 4 strains containing Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, and Actinomyces viscosus. In ethanol extract, no activity was observed against other strains except Staphylococcus aureus. MIC values of ethyl acetate extract were the same, 7.50 mg/ml in all tested bacteria. Conclusions: Pleurotus eryngii exhibited the antibacterial activity against oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Thus, Pleurotus eryngii may be considered as a natural antibacterial agent for treatment of dental diseases.
