This study was carried out to elucidate effective restoration measures for natural forest with recycling native tree un site from November 2001 to October 2002 to obtain a basic information for revegetation measure, eight experimental treatment was done and the length of stump, root-ball size of stump, antisepsis treatment of trunk cut, Planting season and contents of organic matter in soil were effective on regrowth of root-stock. Thirteen tree species including Quercus acutissima among twenty tree species showed outstanding sprout and survival rate(over 90 percent), Planting in November and combinated planting with 5 trees and 9 shrubs of root-stock per 100$m^2$ plot showed a good growth. And 10 percent of organic matter plot showed a good crown coverage.
Glucoamylase was inactivated with 1-ethyl-2-(dimethylaminopropyl)carbodiimide (EDC) at pH 5.0. Time course of inactivation of glucoamylase was at least biphasic. From the results of the titration of SH groups with Ellman's reagent and hydroxylamine treatment at pH 7.0, it was concluded that the crucial sites of modification were carboxyl groups of glucoamylase. The CD spectrum of EDC-modified glucoamylase suggested that the gross conformation of the native enzyme was retained. The inactivation of glucoamylase was reduced remarkably in the presence of maltose. The logarithm of the half-life of the inactivation of glucoamylase by EDC was a linear function of log[EDC] in each stage indicating that one carboxyl group among the modified ones was crucial for inactivation of glucoamylase. The change in the binding affinity due to modification was determined by using an affinity column. It indicates that the carboxyl group of glucoamylase seems to play a role in both, the catalysis and substrate binding in the first stage, but in the second stage the binding affinity is recovered almost up to that of native enzyme.
Background: Revegetating landfills can be a challenging task as the reclaimed soils are typically dry and have low nutrient content. Therefore, selecting suitable plant species is important for initial revegetation. The issue with current practices (in Korea) is that alien plant species have been typically selected for revegetation projects. In this context, this study selects and tests a set of native plant species for landfill revegetation, accompanied by the assessment of the landfill soils. Results: The soil of the landfill (landfill location) was in a very poor condition with high bulk density and low organic matter and nutrient contents. Among 10 tested species, only Brassica campestris showed high coverage and a sufficient number of individuals in study quadrats sown with seeds. Results suggest that plant species with heavy seeds are the only ones that can adapt to the environment of a typical landfill due to the site's aridity and low nutrient content. The reason is due to such species' superior wind resistance and the capacity to provide sufficient energy for the initial growth of the plants for survival in such landfill environment. Conclusions: This study recommends selecting plant species (1) with arid-adapted features and (2) whose seed weight is sufficiently heavy for survival at landfills or areas with a similar condition for future revegetation.

Harringtonine and homoharringtonine known as anti-cancer agents were isolated from Korean native plumyew(Cephalotaxus koreana) using column chromatography(CHCl3:MeOH=19:1, Rf=0.28). The structure of the mixture of two compounds was characterized by 1H-NMR. Comparison of our spectra of harringtonine and homoharringtonine with previously reported ones indicated that the two are identical. The contents of harringtonine and homoharringtonine in the needles, stems, and roots of Korean native plumyew were determined by high performance liquid chromatography(HPLC). The contents of both compounds varied with the site of location and the part of plant. The content of harringtonine was higher in needles and roots than in stems, whereas the content of homoharringtonlne was lower than harringtonine. Homoharringtonine contents in needles at Mt. Palgong, Mt. Dukyu, Mt. Baekyang, Mt. Jiri, and Namhae were higher than in stems and roots. But homoharringtonine contents in needles al Mt. Jokye and Jindo were lower than in stems and roots.
Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri which possessed the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewaters and mine wastewaters polluted with various heavy metals. The binding sites of heavy metal in the cells were investigated by chemical modification of functional groups the cell walls. To determine the binding sites of heavy metal in the cells, electrochemical charge of amine and carboxyl groups in the cell walls of heavy metal-tolerant microorganisms were chemically modified. Chemical modifications of amine groups did not affect the heavy metal uptake as compared to native cell walls. In contrast, modifications of carboxyl groups drastically decreased heavy metal uptake as compared to native cell walls, and electron microscopy confirmed that the form and structure of the heavy metal uptake were different from those of native cell walls. The results suggested that the carboxyl groups were the major sites of heavy metal uptake in the heavy metal-tolerant microorganism cell.
The structural basis of Iysine specificity of Achromobacter protease I (API) was investigated by means of site-directed mutagenesis. The precursor protein in which Glu190, one of the two candidates for determining Iysine specificity, was substituted by glutamine, aspartic acid or leucine was processed autocatalytically to attaln full pretense activity with lysine specificity. The substitution of the other candidate, Asp225, for asparagine or leucine produced no mature active forms of pro-API. The precursor protein of the mutant D225E slowly matured autocatalytically. The lysylendopeptidase activity of the mature D225E was 0.25% of that of native API, and this reduced activity is mainly due to a decrease in the affinity of the enzyme for lysine. These results suggest that Asp225 plays a critical rol in restricted substrate specificity as a lysylendopeptidase. However, D225E exhibited no measurable activity for synthetic ornithine substrate. Since the hydroxyl group of Ser194 in this mutant retained essentially the same reactivity to DFP or PMSF as that in native API, it can be noted that a methylene unit longer side chain of residue 225 is not compensated by a methylene unit shorter side chain at subsite P1 in the bound substrate.
It is important for genetic resources to collect and identify in native medium-leaf type zoysiagrasses species distributed in Korea. This study was conducted to investigate morphological variation and characteristics of native medium-leaf type zoysiagrasses from coastal, island and inland regions in Korea. Among them, 75 collected lines was confirmed to have various morphological variations, accessions were classified into 2 main based group coastal and inland regions by morphological characteristics and site environment. Group I included Z. sinica type, this group showed 3.7 mm in leaf width, 29 in number of seed per spikelet and 5.0 mm in seed length. Group II included Z. japonica type, this group showed 4.4 mm in leaf width, 42 in number of seed per spikelet and 3.5 mm in seed length. There is a need for additional research on growth characteristics and the molecular level for the introgressive hybridization between species which confirmed that cross-pollination is possible due to protogyny. The individuals showing variations should be preserved as valuable genetic resources for the expansion of variations in zoysiagrasses, and the results of this investigation on the genetic resources collected will be highly valuable in breeding high quality turfgrass.

This study is aimed at establishing the categories and items for ecological assessment and evaluation of the environmental friendliness of golf courses in the capital region of Korea. The categories and items for the assessment have been derived based on the existing literature and interviews with golf experts. This study covers 32 golf courses in the capital region of Korea that are available in terms of data and on-site surveys. In order to conduct a comprehensive assessment of the environmental friendliness of the golf courses, the assessment area was divided into 4 categories that include a total of 14 sub-categories. The 4 categories encompass 1) location, 2) topography, 3) vegetation, and 4) construction. As its sub-categories, the location category includes current land use and zoning in the National Land Use Management Law. Topography has 2 sub-categories in the damage ratio of existing topography, gradient, cut area, and slope height. The assessment of vegetation is largely based on site surveys in the categories of preservation of the existing vegetation, the use of natural resources and existing trees, the component ratio of native tree species, the multi-layered structure of vegetation, and the utilization of water purification plants. In the aspect of construction, afforestation on tile slopes and the utilization of existing surface soil were evaluated. The examination of comparative analysis among the 10 items as a ratio measure showed that the scores were low in the sub-categories of current land we, the use of existing trees, and the multi-layered structure of vegetation. However, the rating results were satisfactory in the 2 sub-categories including cut area, and the utilization of native tree species. Those proved to be contributing factors in the ecological health of the golf courses. According to correlation analysis of the 10 items to the overall ecological rating of each golf course, the sizes of the 32 golf courses were mainly affected by the damage ratio of existing topography, gradient, preservation of vegetation and slope height. This study has the initiative to conduct an ecological assessment of golf courses in the country based on site surveys. The study results revealed that location factors such as current land use, damage ratio of topography and gradient and topographical factors were the main factors affecting the environmental friendliness of golf courses. This indicates indicating the significance of these factors in the future construction practices of golf courses. Furthermore, this study raises the need for follow-up studies to establish more detailed assessment criteria and to develop assessment techniques for areas such as slope afforestation and water purification plants that need a qualitative approach.

While the recent lift of restrictions in greenbelt areas is expected to generate a number of development plans, there are efforts to create various development plans into spatial plans that consider the natural and ecological conditions of development sites. However, these development plans consider Degree of Green Naturality or Degree of Ecology only when designating areas for conservation within development sites. It is true that they don't fully reflect the value of green areas and wetlands as habitats and natural resources. Therefore, this study built an conservation value assessment model that is applicable to sites where development is planned in Korea by reviewing prior case examples md studies and applied the developed model to a case study area. The site where the conservation value assessment model was applied to is an area around Yongsan-ri and Suha-ri, Doam-myon, Pyeongchang-gun, Gangwon province where quality natural resources are located in and wend the site. This is a site for the development of Alpensia Resort where a resort including facilities for the Winter Olympics is planned to be introduced. In order to assess the conservation value of the site for Alpensia Resort, a total of eight items including area, distribution of communities, habitation of species with conservation value, functions of habitats, connectivity of habitats, vegetation layers of forests, age of forests, and ratio of non-native plants were studied through literature review and field surveys. The assessment was made by dividing the site into 95 habitats that are perceived by aerial photographs and each habitat unit was assessed on the eight items in a 3-point scale. Each unit habitat assessed in a 3-point scale was segmented into primary, secondary or tertiary areas based on the conservation value. Habitats assessed as primary were designated as priority (absolute) conservation areas and those assessed as secondary and tertiary were set as secondary conservation areas and tertiary conservation areas, respectively. As a result, each area represented 26.9%, 20% and 3% of total site area. Based on this result, habitat management plans were developed to conserve primary conservation areas, improve secondary conservation areas and restore tertiary conservation areas. In addition, a development plan was developed to create habitats in areas where new habitats are requested in order to build an eco-network in the site and a plan to create eco-corridors was proposed. In developing a land use plan for the site, a development plan that considers conservation areas first should be set up based on the outcome of conservation value assessment. This can be linked to the development of an environment-friendly land use plan as well as easing the establishment of a green area and eco-network. This study will facilitate the implementation of the 'conservation before development' principle, which can prevent reckless development, by assessing conservation value in setting up development plans.

An exhibit entitled 'Revelatory Landscapes' was held at the San Francisco Museum of Modem Art from May 5 to October 14, 2001. For this Museum's rot off-site outdoor exhibition, five outstanding design teams-Kathryn Gustafson, Hargreaves Associates, Hood Design, Tom Leader Studio, and ADOBE LA-created site-specific installations in Berkeley, Oakland, San Jose, and the San Francisco Bay Area. Each project showcased a hybridization of environmental art and landscape design. The main content revealed through revelatory landscapes were the natural, cultural, and historical palimpsest of the sites-particularly as related to the history of minorities such as Native American, African Americans and Latin Americans-as well as the every day life of ordinary people. To represent these ideas, a juxtaposition of the past and the present was broadly applied. Furthermore, the use of dramatic colors, textures, and forms in consideration of materials coupled with the revelation of natural elements such as wind and sunlight accelerated the effect of this juxtaposition. Every project of the Revelatory Landscapes exhibit requires a phenomenological experience to be appreciated. Via the five senses, these experiences cause a synesthetic experience beyond solely the visual. By examining the projects of 'Revelatory landscapes', the threshold for a new blending between environmental art and landscape design as well as new landscape design strategies that overcome the dichotomy between nature and culture will change and evolve.
