Exposure assessment of Polynuclear aromatichydrocarbons(PAHs) for heat treat workers using Metalworking fluids (금속가공유 취급 근로자의 다핵방향족탄화수소 노출 평가)

  • The purpose of this study was to assess the ambient level of Polynuclear aromatic hydrocarbons(PAHs), oil mist of the metalworking fluids(MWFs), especially in heat treat industry and non heat treat industry. And we assessed the relationship of the pattern of exposed PAHs with oil mist during survey day. The study population of heat treat industries contained 98 workers, non-heat treat industry contained 40 workers. Personal samples were taken for ambient monitoring of PAHs and oil mist. PAHs was to analyze the relationship of airborne oil mist. The geometric mean of airborne total PAHs was $3.44{\mu}g/m^3$ in heat treat industry and $0.13{\mu}g/m^3$ in non heat treat industry, Pyrene and Benzo(a)pyrene was detected from heat treat industry and Naphthalene in heat treat industry was significantly detected higher than in non heat treat industry. The geometric mean of airborne oil mist was $0.19{\mu}g/m^3$ in heat treat industry and $0.70{\mu}g/m^3$ in non-heat treat industry. The correlation between oil mist and total PAHs was shown below, total PAHs in workers of non heat treat industry and heat treat industry were not significantly correlated with oil mist

Growth, secondary metabolite production and antioxidant enzyme response of Morinda citrifolia adventitious root as affected by auxin and cytokinin

  • Morinda citrifolia adventitious roots were cultured in shake flasks using Murashige and Skoog medium with different types and concentrations of auxin and cytokinin. Root (fresh weight and dry weight) accumulation was enhanced at 5 $mg\;l^{-1}$ indole butyric acid (IBA) and at 7 and 9 $mg\;l^{-1}$ naphthalene acetic acid (NAA). On the other hand, 9 $mg\;l^{-1}$ NAA decreased the anthraquinone, phenolic and flavonoid contents more severely than 9 $mg\;l^{-1}$ IBA. When adventitious roots were treated with kinetin (0.1, 0.3 and 0.5 $mg\;l^{-1}$) and thidiazuron (TDZ; 0.1, 0.3 and 0.5 $mg\;l^{-1}$) in combination with 5 $mg\;l^{-1}$ IBA, fresh weight and dry weight decreased but secondary metabolite content increased. The secondary metabolite content (including 1,1-diphenyl-2-picrylhydrazyl activity) increased more in TDZ-treated than in kinetin-treated roots. Antioxidative enzymes such as catalase (CAT) and guaiacol peroxidase (G-POD), which play important roles in plant defense, also increased. A strong decrease in ascorbate peroxidase activity resulted in a high accumulation of hydrogen peroxide. This indicates that adventitious roots can grow under stress conditions with induced CAT and G-POD activities and higher accumulations of secondary metabolites. These results suggest that 5 $mg\;l^{-1}$ IBA supplementation is useful for growth and secondary metabolite production in adventitious roots of M. citrifolia.

  • Mycolic acid-containing gram-positive bacteria, so called nocardioform actinomycetes, have become a great interest to environmental microbiologists due to their metabolic versatility, multidegradative capacity and potential for bioremediation of priority pollutants. For example, Rhodococcus rhodochrous N75 was able to metabolize 4-methy1catechol via a modified $\beta$-ketoadipate pathway whereby 4-methylmuconolactone methyl isomerase catalyzes the conversion of 4-methylmuconolactone to 3-methylmuconolactone in order to circumvent the accumulation of the 'dead-end' metabolite, 4-methylmuconolactone. R. rhodochrous N75 has also shown the ability to transform a range of alkyl-substituted catechols to the corresponding muconolactones. A novel 3-methylmuconolactone-CoAsynthetase was found to be involved in the degradation of 3-methylmuconolactone, which is not mediated in a manner analogous to the classical $\beta$-ketoadipate pathway but activated by the addition of CoA prior to hydrolysis of lactone ring, suggesting that the degradative pathway for methylaromatic compounds by gram-positive bacteria diverges from that of proteobacteria. Mycobacterium sp. Strain PYR-l isolated from oil-contaminated soil was capable of mineralizing various polyaromatic hydrocarbons (PAHs), such as naphthalene, phenanthrene, pyrene, fluoranthrene, 1-nitropyrene, and 6-nitrochrysene. The pathways for degradation of PAHs by this organism have been elucidated through the isolation and characterization of chemical intermediates. 2-D gel electrophoresis of PAH-induced proteins enabled the cloning of the dioxygenase system containing a dehydrogenase, the dioxygenase small ($\beta$)-subunit, and the dioxygenase large ($\alpha$)-subunit. Phylogenetic analysis showed that the large a subunit did not cluster with most of the known sequences except for three newly described a subunits of dioxygenases from Rhodococcus spp. and Nocardioides spp. 2-D gel analysis also showed that catalase-peroxidase, which was induced with pyrene, plays a role in the PAH metabolism. The survival and performance of these bacteria raised the possibility that they can be excellent candidates for bioremediation purposes.

  • Fluidity retention of concrete used high range water reducing AE agent(HWAE) is varied according to main component, dosage amount and dosing method of HWAE. The type and substitution ratio of mineral admixture also have influence on the fluidity retention of concrete used HWAE. In this study, for the purpose of improving the fluidity retention in concrete used HWAE. 3 types of HWAE and ground granulated blast furnace slag(SG) are used in cement paste, mortar and concrete varing dosage amount and dosing time of HWAE and substitution ratio of SG respectively. According to using the HWAE of naphthalene sulfonates and SG, the fluidity retention of mortar and concrete is improved remarkably. And after 30 min, dosing method of HWAE is very effective in improving the fluidity retension and strength of concrete regardless of type of HWAE.

Bioassays of Polycyclic Aromatic Hydrocarbons Using cyp1a1-Luciferase Reporter Gene Expression System in Mouse Liver Hepa 1 Cells

  • Recent industrial society has human widely exposed to PAHs (polynuclear aromatic hydrocarbons) that are comming from the incomplete combustion of organic material as wider spread environmental contaminants. Biological activities of PAHs are not known although PAHs are considered as carcinogens. Our laboratory have been studied the effect of PAHs in the mouse liver hepa 1 cells. In this study, we examined the mouse liver hepa-l cells as a new bioassay system to evaluate bioactivity of PAHs. We have selected 13 PAHs to examine bioassay using cyp1a1-luciferase reporter gene expression system where cyp1a1 1.6 Kb 5flanking region DNA was cloned in front of luciferase reporter gene and this plasmid was transfected into hepa 1 cells transiently. This cells then used for the study to observe the effect of PAHs. We demonstrated that PAHs induced the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase (EROD) activities in a concentration-dependant manner. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD. Acenaphthene, anthracene, fluorine, naphthalene, pyrene, phenanthrene, carbazole were weak responders to cyp1a1 promoter activity stimulation and EROD induction in hepa 1 cells and these chemicals seemed to respond less to EROD than cyp1a1 promoter activity. Benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, chrysene, and dibenzo(a,h)anthracene showed strong response to cyp1a1 promoter activity stimulation and also EROD induction in hepa 1cells. Results of dose response study suggested that four strong responding PAHs, such as benzo(a)anthracene benzo(k)fluoranthene, chrysene, and dibenzo(a, h)anthracene might be mediated through arylhydrocarbon receptor system in hepa1 cells.

  • Polycyclic aromatic hydrocarbons (PAMs) are ubiquitous contaminants in marine environments. PAMs enter estuarine and nearshore marine environment via several routes such as combustion of fossil fuels, domestic and industrial effluents and oil spills. PAHs have been the focus of numerous studies in the world because they are potentially carcinogenic, mutagenic, and teratogenic to aquatic organisms and humans from consuming contaminated food. However, one can hardly find any available data on PAM content in marine organisms in Korea. The present study was carried out in order to determine PAM content in oysters from the intertidal and subtidal zones of Chinhae Bay, which is located in near urban communities and an industrial complex, and the bay is considered to be a major repositories of PAHs. 16 PAHs were analyzed by High Performance Liquid Chromatography (HPLC) with uv/vis and fluorescence detectors in oysters: they are naphthalene (NPTHL), acenaphthylene (ANCPL), acenaphthylene (ACNPN), fluorene (FLURN), phenanthrene (PHEN), anthracene (ANTHR), fluoranthene (FLRTH), pyrene (PYR), benzo(a)anthracene (BaA), chrysene (CHRY), benzo(b)fluoranthene (BbF), benzo(k)fluoranthene (BkF), benzo(a)pyrene (BaP), dibenz(a,h)anthracene(DahA), benzo(g,h,i)peryne (BghiP) and indeno(1,2,3,-cd)pyrene (I123cdP). The PAM contents in oysters from the intertidal and subtidal zones of Chinhae Bay ranged from < 0.1 to 992.0 ug/kg (mean $69.8\pm9.8$ ug/kg).

Characteristics of Atmospheric Concentrations of Volatile Organic Compounds at a Heavy-Traffic Site in a Large Urban Area (대도시 교통밀집지역 도로변 대기 중 휘발성유기화합물의 농도분포 특성)

  • This study was carried out to evaluate the temporal (daily, weekly, and seasonal) variations of volatile organic compounds (VOCs) concentrations at a road-side site in a heavy-traffic central area of Metropolitan Taegu. Ambient air sampling was undertaken continuously for 14 consecutive days in each of four seasons from the spring of 1999 to the winter of 2000. The VOC samples were collected using adsorbent tubes, and were determined by thermal desorption coupled with GC/MS analysis. A total of 10 aromatic VOCs of environmental concern were determined, including benzene, toluene, ethylbenzene, m+p-xylenes, styrene, o-xylene, 1,3,5-trimethylbenzene, 1,2,4-trimethylbenzene, and naphthalene. Among 10 target VOCs, the most abundant compounds appeared to be toluene (1.5 ∼ 102 ppb) and xylenes (0.1 ∼ 114 ppb), while benzene levels were in the range of 0.3 ∼6 ppb. It was found that the general trends of VOC levels were significantly dependent on traffic conditions at the sampling site since VOC concentrations were at their maximum during rush hours (AM 7∼9 and PM 7 ∼9). However, some VOCs such as toluene, xylenes, and ethylbenzene were likely to be affected by a number of unknown sources other than vehicle exhaust, being attributed to the use of paints, and/or the evaporation of solvents used nearby the sampling site. In some instances, extremely high concentrations were found for these compounds, which can not be explained solely by the impact of vehicle exhaust. The results of this study may be useful for estimating the relative importance of different emission sources in large urban areas. Finally, it was suggested that the median value might be more desirable than the arithmetic mean as a representative value for the VOC data group, since the cumulative probability distribution (n=658) does not follow the normal distribution pattern.

  • A series of water-soluble polyparacyclophanes bearing two diphenylmethane or two diphenyl ether skeletons were investigated to develop useful host compounds by using 1-anilinonaphthalene-6-sulfonate (ANS) and 2-p-toluidinylnaphthalene-6-sulfonate (TNS) as fluorescent hydrophobic substrates in aqueous solution. It was noteworthy that remarkable fluorescent enhancements and blue shifts of ANS and TNS were observed only in the presence of 1,6,20,25-tetraaza[6.1.6.1] paracyclophane (CPM 44) and 1,6,21,27-tetraaza [7.1.7.1] paracyclophane (CPM 55) for diphenylmethane skeleton, and 1,7,21,27-tetraaza-14,34-dioxa [7.1.7.1] paracyclophane (CPE 55) and 1,8,22,29-tetraaza-15,36-dioxa [8.1.8.1] paracyclophane (CPE 66) for diphenyl ether skeleton, comparing with ${\alpha}-\;and\;{\beta}-cyclodextrins$. However, their acyclic analogues such as 4,4'-dimethylaminodiphenylmethane and 4,4'-dimethylaminodiphenyl ether, and paracyclophanes whose cavities were smaller showed only small effects under the same conditions. These facts suggested that hosts and substrates were in an intimate contact which would not occur without larger structures, and thus that guest molecules were strongly incorporated in the hydrophobic cavities of these larger paracyclophanes. The effects of pH on the fluorescent intensity of ANS-CPM 44, ANS-CPM 55, ANS-CPE 55, ANS-CPE 66, TNS-CPM 44, TNS-CPM 55, TNS-CPE 55 and TNS-CPE 66 systems were not significant below pH 2.0, but their fluorescent intensities were markedly reduced with increasing ionic strength.

  • The purpose of the present study was to kinetically investigate the carrier-mediated uptake in the hepatic transport of organic anions, and to simulate the 'in vivo counter-transport' phenomena, using kinetic model which was developed in this study. The condition that the mobility of carrier-ligand complex is greater than that of free carrier is not essential for the occurrence of 'counter-transport' phenomenon. To examine the inhibitory effects on the initial uptake of organic anions by the liver, it is necessary to judge whether the true counter-transport mechanism (trans-stimulation) is working or not. Effects of bromophenol blue (BPB) or bromosulfophthalein (BSP) on the plasma disappearance curves of a 1-anilino-8-naphthalene sulfonate (ANS) were then kinetically analyzed based on a flow model, in which the ligand is eliminated only from the peripheral compartment (liver compartment). Moreover, 'in vivo counter-transport' phenomena were simulated based on the perfusion model which incorporated the carrier-mediated transport and the saturable intracellular binding. The 'in vivo counter-transport' phenomena in the hepatic transport of a organic anions were well demonstrated by incorporating the carrier-mediated process. However, the 'in vivo counter-transport' phenomena may be also explained by the enhancement of back diffusion due to the displacement of intracellular binding. In conclusion, one should be more cautious in interpreting data obtained from so-called 'in vivo counter-transport' experiments.

Effect of soil Venting on Dissolution Potential of Gasoline Components in Contaminated Soil : 2. Modeling Approach (Soil Venting이 오염토양중 가솔린 성분의 용출성에 미치는 영향: 2. 모델링 접근)

  • The effects of soil venting on the leaching potential of residual gasoline were characterized by applying a simple multi-component volatilization/dissolution model based on Raoult's law. The validity of Raoult's law in describing dissolution of gasoline was evaluated separately using both pure gasoline and gasoline contaminated soil. The aqueous concentrations of gasoline components equilibrated with pure gasoline could be described by Raoult's law within one order of magnitude, regardless of the composition of the gasoline. The leaching concentrations from contaminated soil could be well predicted at a relatively high gasoline concentration in soil. However, after 93.5% removal of gasoline by venting, the calculated values were higher than the experimental values by 50∼100%. A model involving multi-component evaporation and dissolution was applied and the results were compared with the experimental values. Possible causes of the discrepancy between the predicted values and experimental values were given.
