Antiviral Effects of Natural Products on the Inhibition of Hepatitis B Virus DNA Replication in 2.2.15 Cell Culture System

  • Evaluation of plant extracts that might inhibit hepatitis B virus (HBV) replication was performed to find potent anti-HBV agents. Eighty-five species of plants from forty-three families were tested for their anti-HBV activities using HBV-producing HepG2-derived 2.2.15 cells. The anti-HBV activity of plant extracts was measured by slot blot hybridization technique and cytotoxicity was determined by crystal violet staining procedure. All plants were extracted with methanol and the extracts were partitioned into n-hexane, ethyl acetate and aqueous layer. The ethyl acetate fractions of Rhus verniciflua $(stem:\;EC_{50},\;8.2{\mu}g/ml;\;CC_{50},\;9.4{\mu}g/ml)$, Gastrodia elata $(root:\;EC_{50},\;17.7{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, Raphanus sativus $(seeds:\;EC_{50},\;17.3{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, and Angelica gigas $(root:\;EC_{50},\;8.3{\mu}g/ml;\;CC_{50},\;15.6{\mu}g/ml)$ revealed the anti-HBV activity in 2.2.15 cell culture system and these fractions are under the process of further sequential fractionation by column chromatography to find the active principles against HBV.

Tyrosinase-inhibitory and Radical Scavenging Activities from the Seeds of Coix lachryma-jobi L. var. ma-yuen [Roman] Stapf (율무의 tyrosinase 저해활성과 radical 소거효과)

  • The seeds of Coix lachryma-jobi L. var. ma-yuen(yullmoo) were investigated for the free radical scavenging and mushroom tyrosinase inhibitory activities. Brown yullmoo, which contains bran in the seed, has also been evaluated for comparison. After partitioning of the total methanol extracts of yullmoo and brown yullmoo, each organic layer exhibited mild or potent dose-dependent inhibition(about 20-80%) on 1,1-diphenyl picryl hydrazyl(DPPH) free radical with the concentration of $300\;{\mu}g/ml$ to $800\;{\mu}g/ml$. n-Butanol fraction of yullmoo showed the most potent scavenging effect on DPPH free radical. $IC_{50}$ of n-butanol fraction was $423\;{\mu}g/ml$, about six to seven fold higher concentration than standard BHT at the same inhibition rate. As for the tyrosinase inhibitory activity, all of the fractions including the methanol extract exhibited the similar activities even after partitioning. The chloroform fraction of brown yullmoo was found to show the most potent inhibitory activity with an $IC_{50}$ of $321.5\;{\mu}g/ml$.

Suppressive Effects of Crude Extracts of Bacillus sp. CT16 and Neobacillus sp. JC05 against Egg Hatch of Meloidogyne incognita (근권세균 Bacillus sp. CT16과 Neobacillus sp. JC05의 배양액 추출물에 의한 뿌리혹 선충의 알 부화 억제 효과)

  • Root-knot disease caused by Meloidogyne incognita is major soil pathogen and cause severe economic damages to vegetable crops. In this study, we selected rhizobacteria for biocontrol of the root-knot nematode, M. incognita, and identified; performed bioassay of the bacterial extracts in cucumber seedlings. The crude extracts of strains CT16 and JC05 out of 180 strains inhibited egg hatching and increased juvenile mortality in vitro assay; based on 16S rRNA sequences analysis, the two strains were identified as Bacillus sp. CT16, and Neobacillus sp. JC05. After extracting the bacterial supernatants by using various organic solvents, n-butanol and n-hexane extracts of strain CT16 and n-butanol extract of strain JC05 showed inhibitory activity of egg hatching depending on concentrations. Subsequently, n-butanol extracts of two strains significantly suppressed formation of egg masses in cucumber seedling. Therefore, these results indicated that strains CT16 and JC05 could be used as potential biocontrol agents against M. incognita.

Automatic Titration for KMnO4 Consumption Test of Tap Water Using Personal Computer Camera (PC 카메라를 이용한 수돗물의 과망간산칼륨소비량 적정 자동화)

  • An automatic titration system using a PC-camera with a color filter on its lens was used in the $KMnO_4$ consumption test of tap water and distilled water in relation to blank tests. The very faint pink color of titration end point could be effectively detected by using a yellow cellophane paper as a color filter. The average hue value (Havg) of 192 pixels in the image of the sample solution being titrated was computed and followed up at regular time intervals during titration in order to detect the titration end point. The Havg decrease of 2 degrees from the average of first 10 Havgs was regarded as reaching the end point. The volume of 0.01N $KMnO_4$ consumed by a tap water sample was $0.728{\pm}0.022ml$ in manual titration and $0.735{\pm}0.013ml$ in automatic titration (p=0.580). The volume of 0.01N $KMnO_4$ consumed by a distilled water sample was $0.383{\pm}0.015ml$ in manual titration and $0.367{\pm}0.015ml$ in automatic titration (p=0.252). The high p-values for t-test suggested that there were good agreements between manual and automatic titration data and the automatic method proposed in this article was considered to effectively replace the manual titration.

Lead Effects among Secondary Lead Smelter Workers with Blood Lead Levels below $80{\mu}g/100\;ml$

  • A subgroup or workers from a secondary lead smelter was defined to include those with blood lead levels not exceeding $80{\mu}g/100\;ml$ and with no pest history of elevated blood lead. Central nervous system symptoms (tiredness, sleeplessness, irritability, headache) were reported by 55% of the group and muscle and joint pain by 39%. Zinc proteporphyrin (ZPP) levels were elevated in 71% or cases. Low hemoglobin levels (less than 14 gm/l00 ml) were round in more than a third of the workers. While BUN and creatinine were mostly in the normal range, there - nevertheless n correlation between ZPP and both BUN and creatinine. Reduced nerve-conduction velocities were present in 25% or the group; this was not significantly different from findings in Ii control group. The data indicate that n blood level of $80{\mu}g/100\;ml$ is an inappropriate biological guide in the prevention or lead disease.

Studies on Tolerance Level Against Antheraea mylitta Cytoplasmic Polyhedrosis Virus (AmCPV) in Different Eco-races of Tropical Tasar Silkworm, Antheraea Mylitta Drury

  • Pathogenic infection in tasar silkworm, Antheraea mylitta Drury is common and there is a breed specific response regarding tolerance. Hence, the tolerance level of three eco-races of tasar silkworm viz. Daba, Sarihan and Raily to Antheraea mylitta cytoplasmic polyhedrosis virus (AmCPV) infection was tested. The survival of Daba, Sarihan and Raily eco-races was significantly different (p<0.05) when challenged with the same concentration ($1{\times}10^5$ polyhedra/ml) of AmCPV. Daba eco-race was more tolerant to the AmCPV infection having higher survival (65.7%) and $LC_{50}$ values (1000893.17 polyhedra/ml) of AmCPV followed by Sarihan eco-race (50.7% survival and $LC_{50}$ value of AmCPV 187203.6168 polyhedra/ml) and Raily eco-race (24.3% survival and $LC_{50}$ value of AmCPV 5176.37 polyhedra/ml). The tolerance level of Daba and Sarihan eco-races against AmCPV was more than two times than Raily eco-race.

A Comparison of Hydromorphone-Bupivacaine and Fentanyl-Bupivacaine in Patient Controlled Epidural Analgesia after Thoracotomy (개흉술 후 경막외 통증자가조절을 설시한 환자에서 Hydromorphone-Bupivacaine과 Fentanyl-Bupivacaine 투여의 비교)

  • Background: Hydromorphone has an intermediate lipid solubility range that falls between morphine and fentanyl. Lipophilic activity during opioid epidural administration is important in relation to both the side effects and analgesic efficacy. The purpose of this study was to compare epidural hydromorphone and fentanyl when concomitantly infused with bupivacaine in patients undergoing a thoracotomy. Methods: Seventy-seven thoracotomy patients, with patient-controlled epidural analgesia (PCEA), were blindly allocated into two groups [group F (n = 34); 0.1% bupivacaine and fentanyl $5{\mu}g/ml$, group H (n = 34); 0.1% bupivacaine and hydromorphone $16{\mu}g/ml$)]. The basal PCEA rate and demand dose were 4 ml/hr and 3 ml, respectively. The visual analogue scale (VAS) for pain, and pruritus, sedation and nausea were measured at 6, 12 and 24 hours after the operation. Results: There were no significant differences in the VAS pain scores and the incidences of pruritus, nausea and sedation between the two groups. The total infused volume after 24 hours was lower in H compared to that of F group (P < 0.05). Conclusions: We conclude that epidural hydromorphone or fentanyl administration has a similar analgesic efficacy and shows similar incidences of side effects, when concomitantly infused with bupivacaine, in the management of acute pain following a thoracotomy.

Determination of Copper, Lead, Cadmium and Zinc in Water by Square Wave Polarography with Dithizone-Chloroform Extraction Method (디티존-클로로포름 抽出, 短形波폴라로그래피에 의한 물중의 구리, 납, 카드뮴 및 아연의 定量)

  • 500 ml of a sample water was extracted with 10 ml of 0.01 % dithizone-$CHCl_3$three times. When $CHCl_3$ layer was back extracted with 10 ml of 0.1 N HCl containing mercuric ion, the free metal ions come into aqueous layer. The aqueous layer was added with 2 ml of 2 N KCl and was washed with 10 ㎖ of $CHCl_3$two times in order to remove the trace dithizone, and then was recorded square wave polarogram. The concentration of copper, lead and cadmium can be determined up to 3 ppb and that of zinc up to 14 ppb with an error of 10 %.

Bioequivalence Assesment of Tiropramide in Korean Male Volunteers

  • Two formulations of tiropramide {(${\pm}$)${\alpha}$-(benzoylamino)-4-[2-(diethylamino)-ethoxy]-N,N-dipropyl-benzenepropanamide hydrochloride}, an antispasmodic agent, were orally administered to 16 healthy Korean male volunteers by Latin crossover design with the purpose of evaluating bioeqivalence and phamacokinetics of tiropramide. Tiropramide in human plasma was determined by a gas chromatography/nitrogen phosphorus detector. Detection limit of tiropramide was 5 ng/ml. C$\_$max/ values in test and reference formulations were 93.9 ${\pm}$ 54.3 and 96.4 ${\pm}$ 51.6 ng/ml, respectively. AUC$\_$0\longrightarrowlast/ and AUC$\_$0\longrightarrowinf/ were, respectively, 330.7 ${\pm}$ 193.9 and 349.5 ${\pm}$ 205.3 ng.hr/ml for test formulation, 348.9 ${\pm}$ 207.7 and 380.8 ${\pm}$ 239.0 ng.hr/ml for reference formulation. Terminal half-life was 2.3-2.6 hr. Bioavailability differences for C/aub max/ and AUC$\_$0\longrightarrowlast/ were 2.48% and 5.22%, respectively. Minimum detection differences were less than 20% in both C$\_$max/ AUC. Based on this results, two formulations of tiropramide were considered to be bioequivalent

Pharmacokinetic Properties and Antiinflammatory Activity of Ketoprofen Lotion (케토프로펜 로오숀의 약물동력학적 특성과 항염증작용)

  • The pharmacokinetic properties and antiinflammatory activity of 3% ketoprofen lotion (ID-lotion), formulated with poloxamer 407, were evaluated using rats. For the pharmacokinetic study, the lotion, at the dose of 4.5 mg/kg, was applied on the dorsal skin of rats and the drug concentration in plasma was determined using an HPLC method. As references, ketoprofen suspended in saline was administered orally, and E-lotion, which is a 3% ketoprofen lotion in the Japanese market was applied transdermally. Following the transdermal application of ID-lotion and E-lotion, $C_{max}$ were 316 $\pm$22.3 ng/ml and 163 $\pm$ 12.2 ng/ml, respectively, at the same Tma of 2 hours postdose, while $C_{max}$ and $T_{max}$ after oral administration of the drug were 1,030$\pm$89.1 ng/ml and 0.25 hours, respectively. Relative bioavailabilities of ID-lotion and I-lotion were 69.3% and 34.2%, respectively. The antiinflammatory activity of the two 3% ketoprofen lotions was evaluated with carrageeneninduced edema method after 50 mg of the lotions was applied on the paw of rats. ID-lotion showed 67.6% inhibition of the edema formation, while I-lotion showed 34.\\\\`r%. The calculated ED5o after transdermal application of ID-lotion was 2.5 mg/kg, while that after oral administration was 7.0 mg/kg. Based on these results, the relative equiponderal availability of ID-lotion was 296% compared to the oral administration of ketoprofen.n.n.n.
