Asterias amurensis (starfish) is a marine organism that is harmful to the fishing industry, but is also a potential source of functional materials. The present study was conducted to analyze the profiles of fatty acids extracted from A. amurensis tissues and their anti-inflammatory effects on RAW264.7 macrophage cells. In different tissues, the component ratios of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids differed; particularly, polyunsaturated fatty acids such as dihomo-gamma-linolenic acid (20:3n-6) and eicosapentaenoic acid (20:5n-3) were considerably different. In lipopolysaccharide-stimulated RAW264.7 cells, fatty acids from A. amurensis skin, gonads, and digestive glands exhibited anti-inflammatory activities by reducing nitric oxide production and inducing nitric oxide synthase gene expression. Asterias amurensis fatty acids effectively suppressed the expression of inflammatory cytokines such as tumor necrosis $factor-{\alpha}$, interleukin-$1{\beta}$, and interleukin-6 in lipopolysaccharide-stimulated cells. Cyclooxygenase-2 and prostaglandin $E_2$, which are critical inflammation biomarkers, were also significantly suppressed. Furthermore, A. amurensis fatty acids reduced the phosphorylation of nuclear $factor-{\kappa}B$ p-65, p38, extracellular signal-related kinase 1/2, and c-Jun N-terminal kinase, indicating that these fatty acids ameliorated inflammation through the nuclear $factor-{\kappa}B$ and mitogen-activated protein kinase pathways. These results provide insight into the anti-inflammatory mechanism of A. amurensis fatty acids on immune cells and suggest that the species is a potential source of anti-inflammatory molecules.
This study was observed the effect of n6 and n3 polyunsaturated fatty acids of dietary corn oil and fish oil which was supplemented with similar levels of tocopherol in high fat diet on the levels of tocopherol, malondialdehyde ( MDA) productions of plasma and tissues of rats. Also RBC hemolysis, superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) activities In liver were determined. Male Sprague Dawley rats were fed high fat (40%Cal) diet which was different only In fatty acid composition for 6 weeks. Dietary (arts were beef tallow(BT) as a source of saturated fatty acid, corn oil(CO) for n6 linoleic acid (LA) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) + docosahexaenoic acid (DHA). Plasma and liver tocopherol levels were lowered by n3 PUy4 but there was no difference in malondialdehyde(MDA) level by different dietary PUFA. However, MDA content of RBC and hemolysis were increased by n3 PUFA. MDA content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in liver were increased in more unsaturated dietary fat groups. Especially, SOD activity was increased in proportion to the degree of fat unsaturation.

This study was designed to investigate fatty acid composition of salt-fermented seafoods in Chonnam area. The seven samples were analyzed for fatty acid composition. The contents of lipid was highest in salt-fer-mented anchovies and followed by salt-fermented small mullets, salt-fermented oysters, salt-fermented sea-arrows and salt-fermented small shrimps. The ratio of fatty acid composition depended on the samples but the major fatty acids were $C_{l6:0}$, $C_{20:5}$, $C_{l6:1}$, $C_{22:6}$, and $C_{l8:1}$ among all samples. The composition of n-3 polyunsaturated fatty acids was highest in salt-fermented sea-arrows (39.11%) and followed by salt-fermented small shrimps, salt-fermented oysters, salt-fermented anchovies and salt-fermented small mullets. In n-3 polyunsaturated fatty acids, $C_{20:5}$ was relatively abundant in salt-fermented oysters (17.71%) and salt-fermented sea-arrows (16.38%), and $C_{22:6}$ in salt-fermented sea-arrows (22.22%) and salt-fermented small shrimps (15.13~21.50%). However, $C_{22:6}$ was very little in salt-fermented small mullets.l mullets.
The hydrolysis characteristics of each fatty acid composing the fish oil by means of the lipase from Candida cylindracea was investigated. The saturated fatty acids, C14:0, C16:0 and C18:0, and the unsaturated fatty acids with one double bond, C16:1, C18:1(n-7), C18:(n-9), C20:1 and C22:1 were more easily hydrolyzed than the $\omega$-3 polyunsaturated fatty acids. when the number of carbon of the $\omega$-3 fatty acids was same but that of double bond was different, the hydrolysis of the $\omega$-3 fatty acids having lower number of double bond was more rapidly carried out. When the degree of polyunsaturation was same but the number of carbon was different, the lipase acted more rapidly upon the $\omega$-3 fatty acids with lower number of carbon. Docosahexaenoic acid(DHA) was most highly concentrated in the glyceride mixture after 120hr hydrolysis among the various $\omega$-3 polyunsaturated fatty acids. The weight percentage of DHA to the $\omega$-3 fatty acids in the fish oil was 31.87% but that in the glyceride mixture after 120hr hydrolysis was 51.89%.
The differential effects of various fatty acids such as n-3 and n-6 types or degrees of unsaturation on the CYP2E1 induction and the production of lipid peroxidation (LPO) were investigated. The CYP2E1-transduced human hepatoma HepG2 cells (E47) were cultured in RPMI 1640 media containing different concentrations of various fatty acids up to 48 h incubation compared to 04 cells and CYP2E1-null cells. Treated fatty acids were linoleic acid (LA:n-6, C18:2), arachidonic acid (AA:n-6, C20:4) and docosahexaenoic acid (DHA:n-3, C22:6). The cell survival rate was decreased corresponding to the degree of unsaturation (LA>AA $\cong$DHA) and to LPO production in E47 and 04 cells. The four or five unsaturation degree of fatty acids, AA and DHA, caused time- and dose-dependent cell death in E47 cells but not as much as in C34 (without CYP2E1), suggesting an important role of CYP2E1 in the DHA mediated damage. In the levels of lipid peroxides (LPO), AA also elevated LPO by 3- and 5- fold compared to DHA or LA treated E47 cells. However, AA did not increase LPO until 48 h incubation in C34 cells. In conclusion, the polyunsaturated fatty acids induced CYP2E1 induction might be changed by the elevated levels of lipid peroxide (LPO) and oxidative stress through the connection of CYP2E1 and degrees of unsaturated fatty acids.
Since the n-6 polyunsaturated fatty acid, docosatetraenoic acid (22:4n-6), is a major functional constituent of avian spermatozoa, the effects of two dietary oils rich in fatty acids which are metabolic precursors of 22:4n-6 on the fatty acid profiles of testicular lipids were investigated during a 39 week period of supplementation from 21 to 60 weeks of age. The effects on liver lipids were determined for comparison. Dietary supplementation of male chickens with Arasco Oil, which provides a large amount of arachidonic acid (20:4n-6), increased the proportion of 20:4n-6 in liver phospholipid by almost 2.5-fold. Although liver phospholipid normally contains very little 22:4n-6, this proportion was significantly increased as a result of Arasco feeding, indicating that the conversion of 20:4n-6 to 22:4n-6 was occurring. The phospholipid of the testis contains much higher proportions of 20:4n-6 and particularly of 22:4n-6 than the liver; supplementation with Arasco Oil significantly increased the proportions of both these polyunsaturates in testis phospholipid but the magnitude of this effect was much lower than that which occurred in the liver. Dietary supplementation with Evening Primrose Oil which contains ${\gamma}-linolenic $ acid (18:3n-6) resulted in significant increases in the proportions of 20:4n-6 and 22:4n-6 in liver phospholipid, although the extent of this increase was less than that produced by the Arasco Oil. By contrast, the feeding of Evening Primrose Oil did not alter the fatty acid composition of phospholipid in the testis. The findings raise the possibility that dietary supplementation with Arasco Oil may modulate the fatty acid profile of avian spermatozoa in a way which could potentially be beneficial for fertility. Moreover, the weights of the testes were almost doubled as a result of supplementation with Arasco Oil or Evening Primrose Oil.
We analyzed the compositions of proximate and total fatty acids in the reproductive organs of male (testis, vas deferens, seminal vesicle, and spermatophore sac) and female (ovary, oviduct, oviducal gland, and nidamental gland) common squid. The protein contents were 12.1-22.7 and 13.2-19.4% for males and females, respectively, and the lipid contents were 1.0-2.7 and 2.1-8.0%, respectively. The prominent fatty acids in all reproductive organs were 22:6n-3, 20:5n-3, 16:0, 20:1n-9, 18:0, 18:1n-9, and 20:4n-6. Females had 28.6-32.0% of 22:6n-3 and 11.3-22.6% of 20:5n-3, while males had 20.8-26.8% of 22:6n-3 and 14.4-28.7% of 20:5n-3. These results indicate that the reproductive organs of both male and female squid are potential sources of n-3 polyunsaturated fatty acid.
Lipid levels and fatty acid compositions of 20 species of subtropical fish caught off the coast of Jeju Island, Korea were determined. The fish were categorized according to total lipid (TL) content as follows; lean fish ($TL=1.19{\pm}0.32%$, n=10 species), low-fat fish ($TL=3.02{\pm}0.44%$, n=6 species) and medium-fat fish ($TL=6.87{\pm}1.23%$, n=4 species). The prominent fatty acids were saturated fatty acids (SFAs), at 16:0 and 18:0 monounsaturated fatty acids (MUFAs), at 18:1n-9 and 16:1n-7, and polyunsaturated fatty acids (PUFAs), at 22:6n-3, 20:5n-3 and/or 20:4n-6. Across all species, the percentages of PUFAs and SFAs were $38.0{\pm}7.89%$ and $34.3{\pm}3.08%$, respectively, with no significant difference between both groups (P<0.05), but showed higher proportions than that of MUFA group ($27.8{\pm}7.23%$) (P<0.05). The proportion of n-3 PUFAs was $27.1{\pm}8.68%$ across all fish species, similar to levels in other fishes from Korea. These results suggest that subtropical fish from Jeju Island are a good source of n-3 PUFAs for humans.
The edible portions of saury were cooked by pan frying, deep fat frying, steaming and microwave cooking to compare the compositions of fatty acids in cooked fish with those in raw fish. The total lipid in saury studied varied by each fish but ranged from 21.87% to 30.63%. The distribution of each fatty acid (average) was in order; $C_{22:1}\;(21.75%)>C_{20:1}\;(15.81%)>C_{16:0}\;(11.75%)>C_{22:6}\;(DHA,\;10.61%)>C_{14:0}\;(7.04%)>C_{20:5}\;(EPA,\;5.13%)$. The average ratio of PUFA (polyunsaturated fatty acids)/SFA (saturated fatty acids) in saury fillet was 1.07 and that of n-3/n-6 was 9.15, which showed saury might be a good source for PUFA including n-3 fatty acids. Comparing the effects of cooking on EPA and DHA, two of the most important n-3 PUFA, the decreasing rate of EPA were 6.98% (pan frying), 26.93% (deep fat frying), 1.16% (steaming) and 15.12% (microwave cooking), respectively. The decreasing rate of DHA ware 4.30% (pan frying), 15.99% (deep fat frying), 3.26% (steaming) and 9.56% (microwave cooking). This study showed that both EPA and DHA were reduced by steaming least, and pan frying, microwave cooking, deep fat frying most, in order.
To investigate the effect of dietary docosahexaenoic acid(DHA) and environmental enrichment on brain fatty acid composition and acetylcholinesterase(AChE) activity, two groups of was fed isocaloric diets containing 10 or 12% dietary lipids for 7 weeks. A third group was fed 10% (w/w) dietary lipids with supplemented 2% DHA-rich fish oil. Each diet group was housed either in a stainless steel cage individually or in a large enriched cage with toys where 7 rats were kept together. The fatty acid composition of plasma and brain was significantly affected by dietary lipid composition but not by environmental enrichment. Fish oil supplementation significanlty decreased plasma levels of monounsaturated fatty acids(MUFA) and increased polyunsaturated fatty acids(PUFA). Fish oil supplemented groups also maintained lower plasma n-6 fatty acids and higher n-3 fatty acids levels than unsupplemented groups. The fish oil supplementation significantly decreased arachidonic acid and increased eicosapentaenic, docosapentaenoic acids, and DHA in brain fatty acid composition. In addition, brain DHA level in supplemented groups tended higher than the unsupplemented. Brain, AChE activity significantly increased by the environmental enrichment but not by the fish oil supplementation. These finding suggest that the 2% fish oil (0.57% DHA & 0.31% EPA, per diet weigth) supplementation is enough to accumulate n-3 fatty acids and to change the n-6 n-3 ratio in brain and environmental enrichment might promote the learning ability.
