Effect of Different Amount of Dietary n-3 PUFA on Colon Carcinogenesis in DMH-treated Rats (쥐에서 식이에 첨가한 n-3 PUFA함량에 따라 대장 암화과정에 미치는 영향)

  • The objective of the study was to observe the effect of n-3 PUFA on cell proliferation and apoptosis by determining mRNA and protein of COX-2 and eicosanoid product and the mRNA and protein of Bu and Bcl-2 related to apoptosis in colon carcinogenesis of 1,2- dimethylhydrazine (DMH)-treated rats. Ninety male Sprague Dawley rats weighing about 170g were divided into 3 groups, control and n-3 PUFA supplemented groups (FO group: 6.2 mmoles n-3 PUFA; 2FO group: 12.4 mmoles n-3 PUFA) and fed experimental diet for 14 weeks. All rats were intramuscularly injected with DMH 15 mg/kg twice a week for 6 weeks to deliver total dose of 180 mg/kg body weight. Compared with the control group, 6.2 mmoles n-3 PUFA significantly reduced the levels of mRNA and protein expression of COX-2 and 2-series eicosanoids ($TXB_{2}$ and $PGE_{2}$) and decreased cell proliferation in colonic mucosa. However, high levels of n-3 PUFA supplementation significantly increased the levels of mRNA and protein expression of COX-2, TXB2 and PGE2. and increased cell proliferation which was similar level to that of control group. Compared with the control group, n-3 PUFA, regardless of the amount, significantly increased apoptotic index in colonic mucosa. Western blot and RT-PCR analyses showed that the levels of mRNA and protein expression of Bax were significantly increased by 6.2 mmoles n-3 PUFA, but decreased by 12.4 mmoles n-3 PUFA. The analyses also showed the levels of mRNA and protein expression of Bcl-2 were significantly reduced by 6.2 mmoles n-3 PUFA, but increased by 12.4 mmoles n-3 PUFA. The ratio of Bcl-2/Bax in mRNA and protein was significantly reduced by 6.2 mmoles n-3 PUFA but increased by 12.4 mmoles n-3 PUFA. Overall, these results indicate that n-3 PUFA could be effective in preventing colon carcinogenesis by reducing cell proliferation with lower level of COX-2 and 2-series eicosanoid, and increasing apoptosis by inducing pro-apoptotic gene, Bax and inhibiting anti-apoptotic gene, Bcl-2 in the colonic mucosa of DMH-treated rats. However, high level of n-3 PUFA supplementation could stimulate colon carcinogenesis by increasing cell proliferation and inhibiting apoptosis. (Korean J Nutrition 38(10): 807$\sim$816,2005)

Effects of Fish Oil and Some Seed Oils on Fatty Acid Compositions of Liver and Brain Tissue in Rats (어유 및 식물 종자유의 급이가 흰쥐가 간장, 뇌조직의 지방산 조성에 미치는 영향)

  • This study was designed to observe the effects of the fish oil and some seed oils on the improvement of the fatty acid compositions of liver and brain tissue in rats. In order to induce the hypertriglyceridemia in the rats of the Sprague-Dawley, 12% coconut oil and 3% each of olive oil, lard, fish oil, perilla oil, corn oil, red pepper seed oil and evening primrose oil were administered to the rats for 4 weeks. In the fatty acid composition of liver lipid, n-3 PUFA contents were most in the fish oil and perilla oil groups of phospholipid fraction, and n-6 PUFA contents were most in the corn oil, red pepper seed oil and evening primrose oil groups of triglyceride fractions. Fatty acid composition of liver lipid fractions were influenced from the fatty acid composition of the test lipids. In the fatty acid composition of brain phospholipid, n-3 PUFA contents (8.8~17.2%) were most in the fish oil group, and n-6 PUFA (34.6~38.2%), though it contains high percentage, showed little difference between groups.

Effect of n-3 Polyunsaturated Fatty Acids on Glucose Uptake of Soleus Muscle in NIDDM Diabetic Rats (NIDDM 당뇨병 흰쥐에서 n-3 다가불포화지방산이 가자미근의 Glucose Uptake에 미치는 영향)

  • The purpose of this study was to investigate the effects of n-3 polyunsaturated fatty acids(PUFA) on glucose and lipids metabolism in high-fat diet rate. Rats were randomly assigned to normal, high-fat with n-3 PUFA and high-fat dietary groups. Experiments were carried out after 5 weeks feeding with prescriptive diets following 7 hrs fasting. Body weight gains tended to be higher in high-fat fed rats than normal. Blood glucose was increased (p<0.05) by high-fat diet compared with normal diet, and decreaseed (p<0.05) to normal level by n-3 PUFA. Plasma insulin level was significcantly higher (p<0.01) in high-fat diet rats than that of normal-diet rats, and also decreased (p<0.01) by n-3 PUFA. Glucose up take of soleus muscle in vitro was decreased markedly in high-fat fed rats than normal diet rats at 0, 1, 10, and 100nM insulin concentration. Therefore insulin sensitivity and responsiveness were decreased by high-fat diet. Omega-3 PUFA made a recover(p<0.01) insulin sensitivity to almost normal level, and improved (p<0.05) insulin responsiveness in some extent. In conclusion, the results suggest that metabolic disorder of glucose and insulin resistance of skeletal muscle are caused by high-fat diet and n-3 PUFA can ameliorate metabolic disorder and insulin resistance.

Effects of dietary omega-3 polyunsaturated fatty acids on growth and immune response of weanling pigs

  • The recognition that omega-3 polyunsaturated fatty acids (n-3 PUFA) possess potent anti-inflammatory properties in human models has prompted studies investigating their efficacy for animal growth and immunity. This study examined the effect of feeding an n-3 PUFA-enriched diet on growth and immune response of weanling piglets. Newly weaned pigs (averaging $27{\pm}2$ days of age and $8.1{\pm}0.7kg$ of body weight) were assigned randomly to receive a control (3% vegetable oil, n = 20) or n-3 PUFA-supplemented (3% marine n-3 PUFA, n = 20) diet for 28 day after weaning. Female pigs consuming the n-3 PUFA-enriched diet were lighter at week 4 post-weaning than those fed the vegetable oil supplement. Weanling pigs gained more weight, consumed more feed and had better growth to feed ratios between days 14 and 28 than between days 0 and 14 post-weaning. Plasma insulin-like growth factor I (IGF-I) decreased between days 0 ($87.2{\pm}17.0ng/mL$) and 14 ($68.3{\pm}21.1ng/mL$) after weaning and then increased again by day 28 ($155.2{\pm}20.9ng/mL$). In piglets consuming the vegetable oil-enriched diet, plasma tumor necrosis factor alpha (TNF-${\alpha}$) increased from $37.6{\pm}14.5$ to $102.9{\pm}16.6pg/mL$ between days 0 and 14 post-weaning and remained high through day 28 ($99.0{\pm}17.2pg/mL$). The TNF-${\alpha}$ increase detected in the piglets fed vegetable oil was not observed in the piglets fed n-3 PUFA. Results indicate that weaning induces considerable immune stress in piglets and that this stress can be mitigated by dietary supplementation of n-3 PUFA.

Association of Fatty Acid Intake and Dyslipidemia in Korean Adults: Korea National Health and Nutrition Survey, 1998-2007 (한국 성인의 지방산 섭취와 이상 지혈증과의 관련성 -1998~2007 국민건강영양조사 자료에 근거하여-)

  • The purpose of this study was to examine the association between fatty acid intake and dyslipidemia in Korean adults, using data from the Korean National Health and Nutrition Examination Survey (KNHANES)I, II, III, and IV. After excluding subjects who had missing data for the dietary intakes, anthropometric measurments and laboratory tests, a total of 15,804 subjects aged 20~64 y (6,838 men and 8,966 women) from KNHANES 1998~2007 were included. All data were analyzed using SPSS Statistics software (version 18.0). Associations between fatty acid intakes and dyslipidemia were analyzed by logistic regression analysis after adjusting for age, BMI, energy intake, marital status, job, education level, alcohol intake, and smoking status. Fatty acid intakes were significantly higher in men than in women. The risk of hypertriglyceridemia was significantly and negatively associated with intakes of TFA, SFA, MUFA, PUFA, EPA, DHA, n-3 PUFA, and n-6 PUFA in both men and women. The risk of hypercholesterolemia was not significantly associated with any fatty acid intakes in either sex. The risk of high LDL cholesterolaemia was significantly and positively associated with intakes of TFA, MUFA, PUFA, LNA, n-3 PUFA, and n-6 PUFA in men however, there was no significant association with any fatty acids in women. The risk of low HDL cholesterolaemia was significantly and positively associated with intakes of TFA, MUFA, PUFA, LNA, n-3 PUFA, and n-6 PUFA in men, and was significantly and negatively associated with intakes of MUFA, PUFA, LNA, and n-3 PUFA in women. These results show that dyslipidemia is significantly associated with individual intake of fatty acids, suggesting that fatty acid type may be related to the risk of dyslipidemia.

Fatty Acid Composition of 72 Species of Korean Fish

  • Fatty acid compositions of seventy-two species of Korean fish muscle, 59 species of sea water fish and 13 species of fresh water fish, were studied. Polyunsaturated fatty acid (PUFA) was the richest fatty acid group in all fish samples, accounting for $38.0\pm10.3\%$ of total fatty acids. Monounsaturated fatty acids $(MUFA,\;31.4\pm9.67\%)$ and saturated fatty acids $(SFA, 30.5\pm3.81\%)$ showed a similar level. There was a positive correlation between the total lipid content and MUFA (r=0.7788, p<0.001) and a negative correlation between the total lipid content and PUFA (r= -0.7786, p<0.001) while there was no correlation between the total lipid content and SFA. The proportion of n-3 PUFA and n-6 PUFA was $29.7\pm8.73\%$ and $6,48\pm3.70\%$, respectively, in all fish samples. The n-3 PUFA was rich in sea water fish while n-6 PUFA was rich in fresh water fish. The migratory fish contained the highest level of the n-3 PUFA $(1.82\pm1.01g/100g\;muscle)$, followed by the fresh water fish $(1.09\pm1.04g/100g\;muscle)$, the reef fish $(0.90\pm0.60g/100g\;muscle)$ and the demersal fish $(0.77\pm0.38g/100g\;muscle)$. There was a positive correlation between the total lipid and n-3 PUFA content, $y=0.2083\times+0.05 (r=0.9352,\;p<0.0010)$.

Fatty Acid Composition in Blood Plasma and Follicular Liquid in Cows Supplemented with Linseed or Canola Grains

  • This study was carried out to evaluate the fatty acid composition in Nellore cows supplemented with either linseed (n-3) or canola grains (n-6 and n-9). Fifteen Nellore cows, aged five years and bodyweight 550 kg${\pm}$48 kg, were randomly distributed to the following treatments: CON (control), LIN (linseed) and CAN (canola grains). The cows were fed for 80 days. The concentrations of C18:0, C18:2 n-6 and C20:3 n-6 fatty acid were higher (p<0.10) in CON blood plasma in comparison to follicular liquid. Likewise, PUFA, n-6 contents, PUFA:SFA and n-6:n-3 ratios were higher (p<0.10) in blood plasma. On the other hand, C18:1 n-9, C22:5 n-3, MUFA and n-3 contents were lower (p<0.10) in blood plasma. C18:0, C18:2 n-6, C18:3 n-3, C22:5 n-3, PUFA, n-6, n-3 contents and PUFA:SFA ratio were higher (p<0.10) in LIN blood plasma than in the follicular liquid. Nevertheless, C14:0, C16:0, C16:1 n-7, PUFA, C16:0, C18:1 n-9 and MUFA contents were lower (p<0.10) in LIN blood plasma. On treatment CAN, the C18:0 and SFA contents, and n-6:n-3 ratios were higher (p<0.10) in blood plasma. However, C20:3 n-6, C22:5 n-3, PUFA and n-3 contents were lower (p<0.10) in blood plasma. C16:0, C18:0, PUFA, SFA contents and PUFA:SFA ratio did not differ (p>0.10) among the treatments. C14:0, C16:1 n-7, C18:2 n-6 and n-6 contents were higher (p<0.10) for CON and CAN than LIN. C17:1 n-7, C20:4 n-6 and C 22:0 contents were higher (p<0.10) for CAN than CON and LIN. C18:1 n-9, C18:3 n-3, MUFA and n-3 contents were higher (p<0.10) for LIN and CAN than CON. C20:3 n-6 content and n-6:n-3 ratio were higher (p<0.10) for CON than LIN and CAN. C22:5 n-3 content were higher (p<0.10) for CON and LIN than CAN. The concentrations of fatty acids in blood plasma and follicular liquid were not correlated for any fatty acid, independent of the treatment studied. Canola grain added to the diet of Nellore cows resulted in increased concentrations of fatty acids n-6 and n-3 in follicular liquid.

Effects of dietary n-6:n-3 polyunsaturated fatty acid ratio on growth performance, blood indexes, tissue fatty acid composition and the expression of peroxisome proliferator-activated receptor gamma signaling related genes in finishing pigs

  • Objective: This study investigated the effects of dietary n-6:n-3 polyunsaturated fatty acid (PUFA) ratio on growth performance, blood indexes, tissue fatty acid composition and the gene expression in finishing pigs. Methods: Seventy-two crossbred ([Duroc×Landrace]×Yorkshire) barrows (68.5±1.8 kg) were fed one of four isoenergetic and isonitrogenous diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1, and 8:1. Results: Average daily gain, average daily feed intake and gain-to-feed ratio had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The concentrations of serum triglyceride, total cholesterol and interleukin 6 were linearly increased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio, while that of high-density lipoprotein cholesterol tended to decrease (p = 0.062), and high-density lipoprotein cholesterol:low-density lipoprotein cholesterol ratio and leptin concentration were linearly decreased (p<0.05). The concentration of serum adiponectin had a quadratic response but the measurement was decreased and then increased (quadratic, p<0.05). The proportion of C18:3n-3 was linearly decreased (p<0.05) in the longissimus thoracis (LT) and subcutaneous adipose tissue (SCAT) as dietary n-6:n-3 PUFA ratio increasing, while the proportion of C18:2n-6 and n-6:n-3 PUFA ratio were linearly increased (p<0.05). In addition, the expression levels of peroxisome proliferator-activated receptor gamma (PPARγ) and lipoprotein lipase in the LT and SCAT, and adipocyte fatty acid binding protein and hormone-sensitive lipase (HSL) in the SCAT had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The expression of HSL in the LT was linearly decreased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio. Conclusion: Dietary n-6:n-3 PUFA ratio could regulate lipid and fatty acid metabolism in blood and tissue. Reducing dietary n-6:n-3 PUFA ratio (3:1) could appropriately suppress expression of related genes in PPARγ signaling, and result in improved growth performance and n-3 PUFA deposition in muscle and adipose tissue in finishing pigs.

The Effect of Long Chain n-3 Polyunsaturated Fatty Acids on Development of Collagen-induced Arthritis in Rats (흰쥐에서 다족쇄 n-3 다불포화지방산이 콜라젠-유도 관절염의 발생에 미치는 영향)

  • Background: The treatment of rheumatoid arthritis still depend on conserve therapy in major. Recent studies report that n-3 polyunsaturated fatty acids(PUFA) could modulate the incidence and progress of arthritis. The purpose of this study was to investigate the effects of n-3 PUFA on the development of collagen-induced arthritis in rats. Materials and Methods: Female Louvain rats were used for this experiment. Rats were randomly assigned into either normal (n=8) or collagen-immunized groups, and collagen immunized groups were divided into control(n=8, normal diet) and n-3 PUFA(n=8, 5% n-3 PUFA in diet) groups. One week after feeding n-3 PUFA to rats, they were immunized with type II collagen emulsified in incomplete Freund's adjuvant into tail and back. Development of arthritis was confirmed by x-ray and microscopic examination. Results: Incidence of arthritis at the 5th week after immunization was 38% in control and 0% in n-3 PUFA. Rats with arthritis showed edema in hind paws and inflammation in synovial membrane of the knee joint. Plasma glucose and insulin were not changed by both of immunization and diet. Plasma triglycerides and cholesterol concentrations were decreased by n-3 PUFA. Conclusion: n-3 PUFA may prevent or treat collagen-induced arthritis m rats. Further studies are needed for action mechanism of it.

Proximate and Fatty Acid Compositions of Commercial Domestic and Imported Melania Snails Semisulscospira sp. (시판 국내산 및 수입산 다슬기(Semisulscospira sp.)의 일반성분 및 지방산 조성)

  • The proximate, fatty acid and dimethyl acetal (DMA) compositions of domestic and imported melania snails Semisulcospira sp. being sold commercially were studied. The protein content ranged from 8.09% (China) to 11.3% (North Korea). The lipid content ranged from 0.84 (China) to 2.36% (North Korea). Domestic melania snail had protein (10.5%) and lipid (1.02%) contents intermediate between those of the two imported melania snails. The prominent fatty acids were 16:0 and 18:0 saturated fatty acids, 16:1n-7, 20:1n-11, 18:1n-9, 18:1n-7 and 16:1n-9 monounsaturated fatty acids, and 20:4n-6, 20:5n-3 (eicosapentaenoic acid, EPA), 22:2 non-methylene-interrupted diene (NMID), 18:3n-3, 18:2n-6 and 22:5n-3 (docosapentaenoic acid, DPA) polyunsaturated fatty acids (PUFA). The percentage of n-3 PUFA was higher in domestic (19.6%) and North Korean (imported, 19.5%) snails than in that from China (imported, 16.8%), while the n-6 PUFA level was higher in snails from China (21.2%) and domestic snails (14.7%) than in that from North Korea (9.87%). The total DMA, i.e., 18:0 DMA, 16:0 DMA and 18:1 DMA was 7.30, 7.27, and 4.86% domestic snail and those from China and North Korea, respectively. These results suggest that melania snail is a source of n-3 PUFA, and DMA derived from plasmalogen.
