Evaluation of the Antioxidant Activity of Cooked Gomchwi (Ligularia fischeri) Using the Myoglobin Methods

  • This study evaluated the influence of cooking methods (blanching, pan-frying, and microwaving) on the antioxidant activity of Ligularia fischeri (LF) using myoglobin methods against five different reactive oxygen species (ROS) (i.e., 2,2-diphenyl-1-picrylhydrazyl (DPPH), hypochlorite ions, hydroxyl radicals, peroxyl radicals, and peroxynitrite ions). With respect to DPPH scavenging activity, the antioxidant activities of blanched LF and pan-fried LF were significantly higher (P<0.05) than that of fresh LF for all cooking times. Pan-fried LF and microwaved LF showed higher antioxidant activities against hydroxyl radicals and peroxyl radicals than uncooked LF, while the protective effect of blanched LF was low, except for the at the 3 min cooking time point. Microwaved LF showed high antioxidant activity against all ROS at the 2 min cooking time point. Interestingly, LF that had been microwaved for 2 min and 3 min had high antioxidant values, whereas blanched LF and pan-fried LF had low antioxidant values. The activities of cooked LF against five different ROS were characterized comprehensively by 5-axe cobweb charts.

삼지구엽초의 부위별 추출물에 따른 항산화 활성 (Antioxidative Effects of Extracts from Different Parts of Epimedium koreanum Nakai)

  • 본 연구에서는 삼지구엽초의 뿌리, 줄기, 잎의 부위별 항산화 활성을 알아보기 위하여 각 부위를 70% 에탄올 용액을 이용하여 얻은 추출물의 총페놀 함량, 총플라보노이드 함량, DPPH 라디칼 소거 활성, ABTS 라디칼 소거 활성, 미오글로빈 보호 효과, FRAP를 추출 농도별(1, 2, 10, 20 mg/mL)로 측정하였다. 부위별 총페놀 함량의 경우 뿌리(2.468 mg), 잎(2.307 mg), 줄기(2.277 mg) 순으로 함량이 높았으며, 총플라보노이드 함량은 뿌리(2.071 mg), 잎(1.875 mg), 줄기(0.616 mg)로 순으로 측정되었다. DPPH 라디칼 소거활성, 미오글로빈 보호 효과 그리고 FRAP 실험에서는 농도 의존적으로 항산화 활성을 보였으며 플라보노이드 및 페놀성 화합물의 함량이 적은 줄기에 비하여 잎과 뿌리의 추출물의 활성이 좋았고 ABTS 라디칼 소거 활성의 경우 뿌리 추출물의 활성이 가장 높았다. 이는 총페놀 함량과 총플라보노이드의 함량이 항산화 활성에 상관관계가 있음을 확인할 수 있었다. 삼지구엽초는 주로 잎 부분이 식용으로 쓰이고 있으며 최근 지상부만을 제한적으로 식품원료로 허용하고 있는데 본 연구를 통하여 지상부 외에 지하부에서도 항산화 활성이 높은 것으로 확인되었다.

Comparative Study of the Protective Effects of Citral, Thymoquinone, and Silymarin on Methotrexate-induced Cardiotoxicity in Rats

  • Objectives: Methotrexate (MTX), an immunosuppressant and anti-cancer medication, can harm the heart. The goal of the current investigation was to assess the cardiotoxicity caused by MTX and the potential cardioprotective properties of silymarin, citral, and thymoquinone as antioxidants. Methods: Forty-eight rats were divided into six groups, which included control, MTX, cosolvent, citral, thymoquinone, and silymarin groups. At the end of the study, the rats were anesthetized (ketamine and xylazine) and killed using CO2. Their blood samples were collected to measure the enzymatic activities of creatine kinase-myoglobin binding (CK-MB), creatine phosphokinase (CPK), and lactate dehydrogenase (LDH). Also, the heart tissue was sampled to determine the antioxidant capacity and examine the histopathology. Results: The findings revealed that the activity of CPK, CK-MB, and LDH enzymes significantly reduced in the thymoquinone treatment group compared to the MTX group (p < 0.05). On the other hand, total antioxidant capacity was significantly increased in the thymoquinone group compared to the MTX group (p < 0.05). The pathological modifications (i.e. severe congestion, edema fluid, the presence of inflammatory cells around the blood vessels, mild to moderate hemorrhaging between cardiac muscle fibers) were seen in the MTX group. The treatment groups, particularly thymoquinone, did not experience any appreciable pathological changes. Conclusion: The thymoquinone was found to have the strongest protective effect against the heart damage caused by MTX.
