Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System (Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up)

  • For the study of Hericium erinaceum as a useful functional foods and materials, liquid cultivation under two different bioreactors(air-lift fermenter and jar fermenter) which was not studied systematically until now, was conducted as a method of mass cultivation for H. erinaceum. A batch cultivation in an air-lift fermenter and a jar fermenter was examined for enhancing the productivity because of small amounts of mycelial weight and slow growth in case of a liquid culture for H. erinaceum. We found that air lift fermenter system was more effective than jar fermenter for mycelial production of H. erinaceum, and mycelial morphology was a critical factor of the growth. By scale-up and cultivation based on morphological analysis, the conditions for mass production with 30 L and 500 L jar fermenter was 200 and 150 rpm of agitation speed at 1 vvm of aeration rate, respectively, and mycelial dry weight under these conditions was enhanced to about $13{\sim}14g/L$.

Phylogenetic Relationships and Cultural Characteristics among Inonotus obliquus Strains Collected in Korea (국내 수집 차가버섯 균주의 배양특성과 유전적 유연관계 분석)

  • Fruiting bodies of Inonotus obliquus were collected from the trunk of Betula ermani at 1,100 m of Mt. Odae. Diameter range of the trees at breast height (DBH) was $10{\sim}50$ cm and size range of the sclerotia was $8{\times}5{\sim}20{\times}16cm$. Relationships between the examined strains and Inonotus obliquus strain registered in National Center for Biotechnology Information (NCBI) were very near. And all of 10 strains except strains registered in NCBI showed high homologous characteristics by neighbour joining analysis of ITS sequence. Mycelial growth showed a big difference among strains. Mycelial growth of KFRI 744 was fastest and KFRI 739 was slowest. Difference of mycelial growth between KFRI 735 and 738 was slight, but the difference of mycelial growth between KFRI 744 and 739 was almost twice. Also weight reduction rate among strains showed some difference. KFRI 744 was highest and KFRI 741 was lowest. Vegetative incompatibilities were observed in all mycelial pairings except for KFRI 740-741 and KFRI 742-743 combinations.

Antifungal Activity of Lichen-forming Fungi against Colletotrichum acutatum on Hot Pepper

  • Antifungal activity of Korean and Chinese lichen-forming fungi (LFF) was evaluated against plant pathogenic fungus of Colletotrichum acutatum, causal agent of anthracnose on hot pepper. This is the first attempt to evaluate antifungal activity of LFF, instead of lichen thalli, against C. acutatum. Total 100 LFF were isolated from the lichens with discharged spore method or tissue culture method. Among the 100 isolates, 8 LFF showed more than 50% of inhibition rates of mycelial growth of the target pathogen. Especially, Lecanora argentata was highly effective in inhibition of mycelial growth of C. accutatum at the rate of 68%. Antifungal activity of other LFF was in the order of Cetrelia japonica (61.4%), Ramalina conduplicans (59.5%), Umbilicaria esculenta (59.5%), Ramalina litoralis (56.7%), Cetrelia braunsiana (56.5%), Nephromopsis pallescensn (56.1%), and Parmelia simplicior (53.8%). Among the tested LFF, 61 isolates of LFF exhibited moderate antifungal activity against the target pathogen at the inhibition rates from 30 to 50%. Antifungal activity of the LFF against C. acutatum was variable at the species level rather than genus level of LFF. This study suggests that LFF can be served as a promising bioresource to develop novel biofungicides.

First Report f Pink Mold Rot on Matured Fruit of Cucumis melo Caused by Trichothecium roseum (Pers.) Link ex Gray in Korea (Trichothecium roseum에 의한 시설재배 멜론성과의 분홍빛열매썩음병(신칭) 발생)

  • A severe pink mold rot on matured melon fruits occurred under a glass tunnel cultivation in Chinju at Kyeongasngnam-do Agricultural Research and Extention Services on May of 1998. Basal portion of the fruits toward blossom end was preferably infected and colonized by the fungus. The causal fungus consistently isolated from the lesions was identified as Trichothecium roseum based on following mycological characteristics. Conidia were hyaline or brightly colored, 2-celled, ovoid or elipsoid, characteristically held together zi-zag chains. Conidiophore was long, slender, simple, septate, bearing conidia-meristem arthrospores-apically, singly when young and successively by slight growth of conidiphore apex. Optimum temperature for mycelial growth and conidial germination was recorded at 20~$25^{\circ}C$. However, over 48% of the fungal conidia were germinated at 15$^{\circ}C$ and mycelial growth was only slightly slower than $25^{\circ}C$. The temperature profiles and high humidity available during the cultivation were considered favorable for the pathogen as showing 22% infection rate on matured melon fruits cv. Saros. This is the first report of pink mold rot of melon caused by T. roseum in Korea.

Fundamental Cultural Characteristics of Chlorociboria spp. Native to Korea (한국에 자생하는 녹청균류의 기초 배양 특성)

  • Members of Chlorociboria are soft-rot ascomycetes that produce blue-green pigment. We investigated the growth characteristics of two Korean species of Chlorociboria, eight strains of Chlorociboria aeruginascens and Chlorociboria poutoensis, under various culture conditions (solid media, temperature, pH) and screened them for extracellular enzyme activity. Although the growth rate was slow, all tested strains of Chlorociboria spp. grew well on potato dextrose agar (PDA; 16.3~42.6 mm after 60 days) or Sabouraud dextrose agar (SDA), but not on malt extract agar (MEA). Compared with C. aeruginascens strains, C. poutoensis strains exhibited higher expression of blue-green pigments on both PDA and SDA media. The optimal temperature for mycelial growth was $20{\sim}25^{\circ}C$, and mycelial growth was lower at $30^{\circ}C$ than at $10^{\circ}C$. All strains tended to have increased mycelial growth as the incubation temperature increased in the range of 10 to $20^{\circ}C$. The optimal pH of potato dextrose broth (PDB) for mycelial growth varied according to the strain under static culture conditions. Maximum biomass production was obtained at pH 6.0 for NIFoS 579 ($114.3{\pm}5.1mg/60days$), but it maintained a stable pigment expression under a broad pH spectrum. The activities of both cellulase and laccase were observed in all tested strains of Chlorociboria spp. Enzyme activities of NIFoS 579 were remarkably higher than those of the other strains. From these results, we suggest that C. poutoensis NIFoS 579 is a potential candidate for use as a source of natural blue-green dye.

Synthesis of Pyto-patch as Silver Nanoparticle Product and Antimicrobial Activity (은나노 제품인 Pyto-patch의 제조공정 및 Pyto-patch의 고추 탄저병 방제효과)

  • The commercial product "pyto-patch" prepared as nano sized silver particle less than 5 nm, has effective antifungal activity against Collectotrichum gloeosporioides, Botrytis cinerea, Sclerotinia sclerotiorum in vitro. As a fungal growth inhibiton mechanism, it can reduce spore germination rate and mycelial growth. As s promising fungicide, Phyto-patch can control anthracnose effectively. The spore of C. gloeosporioides dipped in 5 ppm phyto-patch dilute suppressed germination rate to 13.2%, and mycelial growth stopped for 15 days at 10 ppm. The spore postinoculated on 10 ppm phyto-path smeared PDA surface could not germinate for 3 days and prohibit pathogen infection effectively. In field test, the anthracnose development of 4 ppm phyto-patch treated plot was less than 7% after 21 days compaired to 40% of it in untreated plot. In heavy rainfall season, pepper anthracnose effectivly contrrolled by regular 10 ppm phytopatch spraying every 7 days. The diseased pepper fruit decreased to 5.8% compaired to 94.6% in untreated plot. During drying period, the diseased pepper fruit havested in phyto-patch treated plot was 24.2%, but pepper fruit havested in untreated plot destroyed to 100% within 3 days. The nano silver particle coated on multching textile prevented late blight of pepper effectively and disease occurance delayed about month.

Isolation and Characterization of Growth Stimulating Thermophilic Fungi on Oyster Mushroom from Oyster Mushroom (Pleurotus ostreatus) Compost (느타리버섯 배지로부터 느타리버섯 균사의 성장을 돕는 고온성 곰팡이의 분리 및 동정)

  • Some of thermophilic fungi which has growth-promoting effect on Pleurotus ostreatus were isolated from compost during high temperature fermentation process. The temperature optima of 7 isolated thermophilic fungi were $50^{\circ}C$ on PDA media. Isolated strains S-1 and S-2 have the best mycelial growing rate, so these isolates were expected as excellent thermophilic fungi for high temperature composting and mycelial growing of oyster mushroom. In liquid culture, the optimal pH of thermophilic fungi observed variously, pH 7.0-10.0 but most of thermophilic fungi grow well in pH 8.0-pH 9.0 and the final pH of media after cultured was done pH 5.5-6.0. In liquid culture of thermophilic fungi on the optimal condition, S-2 have the best mycelial growing rate. The growing rate of thermophilic fungi S-1, S-2, S-5, and S-10 on lignocellulosic substrates was good but Humicola grisea var. thermoidea, well know thermophilic fungi which has growth-promoting effect on Agaricus bisporus, was poor and which was well grown on PDA at $50^{\circ}C$, pH 7.0. Isolated strain S-1 was identified as Trichophyton sp. and other 6 strains were identified as Sepedonium sp. by morphological characteristics.

  • Erinacine which is produced from Hericium erinaceus mycelia is known to have a stimulating activity of nerve growth factor(NGF) synthesis. Thus, this work was concentrated on the maximum production of Hericium erinaceus mycelia. In order to maximize cell growth, the cultivation was performed with varying the agitation rate and applying the additional medium. As the results, the mycelium concentration was 24.2g/L.

Development of Effective Screening Method for Resistance of oyster mushroom to Trichoderma disease in vitro (느타리버섯 푸른곰팡이병 저항성 실내검정 방법의 개발)

  • Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.

Impact of Rhizosphere Competence of Biocontrol Agents upon Diseases Suppression and Plant Growth Promotion

  • Root colonization of biocontrol agents via seed treatment was investigated and a compatible combination, Gliocladium virens G872B and Pseudomonas putida Pf3, in colonizing cucumber rhizosphere was confirmed through the study. Much higher number of fungal and bacterial propagules were detected when two isolates were inoculated together. The presence of Pf3 in root system was greatly helpful to G872B to colonize at root tip. The mechanism of this phenomenon is partially elucidated through the results of in vitro experiments and the observations of scanning electron and fluorescence microscope. Addition of Pf3 cells resulted earlier germination of G872B conidia and increased mycelial growth. And the more number of germinated conidia on seed coat, the more vigorous hypal streching and sporulation on the root surface were observed in coinoculated treatment. The propagules of G872B on the cucumber root when they were challenged against the pathogenic Fusarium oxysporum, were even higher than that of G872B treated alone, and the magnitude of such a difference was getting grater toward the root ip and the population of F. oxysporum on the root was reduced by seed inoculation of G872B. The rhizosphere competence was obviously reflected to disease suppression and plant growth promotion that induced by the given isolate. Green house experiments revealed that the combined treatment provided long-term disease suppression with greater rate and the larger amount of fruit yield than single treatments. Through this study the low temperature growing Pseudomonas fluorescens M45 and MC07 were evaluated to apply them to the winter crops in field or plastic film house. In vitro tests reveal that M45 and MC07 inhibited the mycelial growth of Pythium ultimum, Rhizoctona solani and Phytophthora capsici and enhanced growth of cucumber cotyledon in MS agar. This effect was more pronounced when the bacteria were incubated at 14$^{\circ}C$ than at 27$^{\circ}C$. And disease suppression and plant growth promotion in green house were also superior at low temperature condition. Seed treatment of M45 or soil treatment of MC07 brought successful control of damping-off and enhanced seedling growth of cucumber. The combined treatment of two isolates was more effective than any single treatment.
