Muscle strips and muscle cells from cat stomach were used to investigate whether spontaneously formed cyclic nucleotides were involved in the inhibition of gastric smooth muscle contraction. A phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX), increased the levels of both cyclic GMP (cGMP) and cyclic AMP (cAMP) in resting state cells, while decreasing acetylcholine-induced muscle contraction. Under the influence of IBMX, SQ22536, an adenylyl cyclase inhibitor and methylene blue, a guanylyl cyclase inhibitor completely blocked increases in cAMP and cGMP respectively, without any effect on contraction. However, the combination of SQ22536 and methylene blue completely blocked increases in both cAMP and cGMP levels and stimulated contractions markedly even in the presence of IBMX. Muscle contraction inhibitors such as isoprenaline, vasoactive intestinal polypeptide and sodium nitroprusside also appeared to increase cyclic nucleotide levels which decreased contraction. Which nucleotide increased the most was dependent on the agonist used. Therefore, irrespective of the cyclic nucleotide class, the spontaneous formation of cyclic nucleotides should be considered in evaluating the mechanism of gastric smooth muscle relaxation.
Treatments for asthma are largely pharmaceutical, with some therapies also utilising alternative breathing techniques. The objective of both medical and alternative methods is to relax contracted airway smooth muscle (ASM). In normal subjects, tidal breathing- and deep inspiration-oscillations are believed to have a bronchodilatory effect. Similarly, application of length oscillations to isolated, contracted ASM also elicits muscle relaxation. As a means of investigating more-effective alternative treatment methods for contracted airways, we analyse the combined effects of bronchodilators and length oscillations on isolated, contracted ASM. The contractile state of the muscle tissue prior to treatment is of primary interest. Thereafter, the effect of applying a combination of small superimposed length oscillations with tidal breathing-like oscillations to ASM is studied alone and in combination with a common bronchodilator, isoproterenol (ISO). This work suggests that relaxation of isolated, contracted ASM following application of combined oscillations and ISO is larger than treatments of either combined oscillations or ISO alone. Further, the observed oscillation-associated relaxation is found to be amplitude- rather than frequency-dependent. This study gives additional insight into the role of oscillations and bronchodilators on contracted airways.
Objectives : These present study was designed to investigate the relaxation effects of Alpiniae Oxyphyllae Fructus(AOF) on isolated corpus cavernosum smooth muscle.Methods : Rabbit corpus cavernous tissues were prepared in strip. Then relaxation responses of AOF at 0.01-3 ㎎/㎖ in contracted strips induced by phenylephrine(PE) were measured. To evaluate mechanisms, indomethacin(IM) tetraethylammonium chloride(TEA), Nω-nitro-L-arginine(L-NNA), methylene blue(MB) were treated before AOF extract(0.1-3 ㎎/㎖) infused into precontracted strips induced by PE. And 1 mM Ca2+was infused into precontracted strips after pretreatment of AOF extract(3 ㎎/㎖) in Ca2+-free krebs-ringer solution. NO concentration was measured by Griess reagent system. Ratio of smooth muscles to collagen fibers and eNOS positive reaction were measured by histocheminal and immunohistochemical process.Results : The cavernous strips were significantly relaxed by AOF extract 0.1, 0.3, 1, 3 ㎎/㎖ and the pretreatment with IM 10 μM,L-NNA 100 μM, MB 10 μM inhibited relaxation of AOF compared to non-pretreatment, but the pretreatment with TEA 100 μM didn't affect relaxation of AOF. In a Ca2+-free solution, pretreatment with AOF reduced increase on contraction of strips by Ca2+supply than non-pretreatment. On HUVEC, NO concentration was increased. On corpus cavernosum of penis in Spontaneous Hypertensive Rat, ratio of smooth muscles to collagen fibers and eNOS positive reaction in AOF group were increased compared to PE groupConclusions : Taken this results, we can suggest that AOF extract exerts a relaxation effects on rabbit corpus cavernosum smooth muscle in part by suppressing influx of extracellular Ca2+throughout prostacyclin, the NO-cGMP system.
This study was designed to examine the mechanism of penile erection in adult bull by analyzing the responses of bovine proximal retractor penile muscle strips(BRP) to electtical field stimulation(EFS), exogenous nitric oxide(NO), NO synthesis precursor(L-arginine), NO synthase inhibitors(L-NAME, L-NMMA), guanylate cyclase inhibitor(methylene blue) and nonspecific potassium channel blocker(tetraethylammonium, TEA) treatments. Isometric tension of BRP was measured using physiograph. Results were summarized as follows: 1. EFS of nonadrenergic noncholinrgic(NANC) nerve in BRP produced frequency-dependent inhibitory responses to the contraction induced by co-treatment of epinephrine, guanethidine and atropine. The inhibitory responses to EFS were blocked by tetrodotoxin(TTX, $1{\mu}M$). 2. Treatment of L-NAME ($10,\;20{\mu}M$) inhibited the relaxation to EFS whereas L-NMMA ($100{\mu}M$) had no effect. 3. Treatment of NO($20,\;40{\mu}M$; as an acidified solution of $NaNO_2$) induced concentration-dependent relaxation whereas preincubation of TTX($1{\mu}M$) and L-NAME($20{\mu}M$) had no effect on the relaxation response. 4. L-arginine treatment(10mM) blocked the inhibitory effect of L-NAME($20{\mu}M$). 5. Pretreatment of methylene blue($40{\mu}M$) reduced the NANC-induced relaxation of BRP. 6. Tetraethylammonium(TEA, 80mM) reduced NANC relaxation. These results suggest that NO may act as a NANC neurotransmitter in BRP and the effects might be mediated by cGMP and potassium channel.

Purpose : The purpose of this study was to investigate the effect of applying intensity on the muscle activity and muscle hardness of the upper trapezius muscle in adult males when TENS (transcutaneous electrical nerve stimulation) was applied. In addition, this study intends to prepare the scientific basic data of TENS for the purpose of relaxation based on the experimental results. Methods : Eighty-seven healthy adult males participated in the experiment, and they were randomly divided into an experimental group and a control group. All subjects in this study were healthy subjects without musculoskeletal or nervous system damage. All subjects were subjects who voluntarily consented to the purpose and method of the experiment. All subjects were provided with a load by typing for 20 minutes, and muscle activity and muscle hardness of the upper trapezius muscle were measured immediately. Afterwards, TENS was given to each groups for 15 minutes, and the experimental group received stimulation at the motor threshold level, and the control group received a placebo stimulation. After stimulation, muscle activity and muscle hardness of the upper trapezius muscle were measured in the same method. The measured data were compared between groups through an independent t-test and dependent t-test. The statistical significance level was set at .05. Results : The application of TENS statistically significantly decreased the muscle activity and muscle hardness of the trapezius muscle in the experimental group, and the results showed a significant difference from the control group. Conclusion : Application of TENS significantly decreased the muscle activity and muscle stiffness of the upper trapezius muscle. The application of TENS of applying intensity that induces muscle contraction may induce relaxation by reducing the muscle activity and muscle hardness of the trapezius muscle.

Deep blocking of consciousness alone does not prevent a reaction to severe stimuli, and copious amounts of pain medication do not guarantee unconsciousness. Therefore, anesthesia must satisfy both: the loss of consciousness as well as muscle relaxation. Muscle relaxants improve the intra-bronchial intubation, surgical field of vision, and operating conditions, while simultaneously reducing the dose of inhalation or intravenous anesthesia. Muscle relaxants are also very important for breathing management during controlled mechanical ventilation during surgery. Excessive dosage of such muscle relaxants may therefore affect neurological examinations during surgery, but an insufficient dosage will result in movement of the patient during the procedure. Hence, muscle relaxation anesthesia depth and neurophysiological monitoring during surgery are closely related. Using excessive muscle relaxants is disadvantageous, since neurophysiological examinations during surgery could be hindered, and eliminating the effects of complete muscle relaxation after surgery is challenging. In the operation of neurophysiological monitoring during the operation, the anesthesiologist administers muscle relaxant based on what standard, it is hoped that the examination will be performed more smoothly by examining the trends in the world as well as domestic and global trends in maintaining muscle relaxant.
The present study was aimed at investigating possible transmitter mechanisms in the endothelial cell layer in regulating the tone of the vascular smooth muscle. The thoracic aorta was isolated from the anesthetized male white rabbits and its helical strips were prepared. Electrical field stimulation was delivered to platinum wire electrodes positioned parallel to the vessel segment preconstricted with phenylephrine [3.5x10-6 mol/L at a distance of 1.5-2.0 mm. The electrical stimulation [70 V, 5 msec, 0.5-200 Hz caused either relaxation only [34% or a biphasic response [prolonged relaxation following a weak and transient contraction, 66% . The relaxation response was frequency- dependent, and at 200 Hz a complete relaxation was noted. Mechanical rubbing of the endothelial layer abolished or greatly attenuated the relaxation. The relaxation was also markedly attenuated in the presence of NG-nitro- L-arginine methyl ester [10-3mol/L or procaine hydrochloride [3.5x10-4mol/L . Tetrodotoxin,guanethidine, atropine or indomethacin failed to block or enhance the relaxation response to electrical field stimulation. It is concluded that the vascular endothelium in the aorta contains diffusible substances that regulates the function of the smooth muscle layer, in which relaxation is more prominent than contraction. Their release by the electrical stimualtion in vitro may not involve classic neuronal transmitter release mechanisms or metabolism of arachidonic acids by cyclooxygenase. The release of the relaxing agents may require an increase in cytosolic calcium level. The chemical nature of the relaxant may be, to a large extent, nitric oxide.

This study examined the effect of Gamma-Amino butyric acid (GABA) and selective GABA receptor related drugs on the electrically stimulated relaxation in the lower esophageal sphincter muscle (LES) of a cat. Tetrodotoxin $(10^{-6}\;M)$ suppressed the electrically stimulated (0.5-5 Hz) relaxation of the LES. However, guanethidine $(10^{-6}\;M)$ and atropine $(10^{-6}\;M)$ had no effect indicating that the relaxations were neurally mediated via the nonadrenergic and noncholinergic (NANC) pathways. NG-nitro-L-arginine methyl ester ($10^{-4}M$, L-NAME) also inhibited the relaxant response but did not completely abolish the electrically stimulated relaxation with 60% inhibition, which suggests the involvement of nitric oxide as an inhibitory transmitter. This study examined the role of GABA, an inhibitory neurotransmitter, on neurally mediated LES relaxation. GABA ($10^{-3}-10^{-5}M$, non selective receptor agonist), muscimol ($10^{-3}-10^{-5}M$, GABA-A agonist), and baclofen ($10^{-3}-10^{-5}M$, GABA-B agonist) had no significant effect on the electrically stimulated relaxation. Moreover, bicuculline ($10^{-5}M$, GABA-A antagonist) and phaclofen ($10^{-5}M$, GABA-B antagonist) had no inhibitory effect on the electrically stimulated relaxation. This suggests that GABA and the GABA receptor are not involved in the electrically stimulated NANC relaxation in the cat LES.
Objective: This study aims to investigate the immediate effects of myofascial release and Duoball assisted self-relaxation (DASR) techniques on pain and muscle tension in patients experiencing chronic cervical pain. Design: A randomized controlled trial. Methods: This study is a randomized controlled experimental study. Eighteen patients with chronic neck pain who met the selection criteria were randomly assigned to myofascial release group and myofascial release group using Duoball. Results: The frequency results for assessment muscle tension showed a decrease of about 10% in the suboccipital muscle, SCM, Pect m, UT, and LS in both the MFR and DASR groups, and the stiffness results showed a decrease in all muscles except the upper trapezius in the MFR group and the DASR group. All were found to decrease by about 10% in the suboccipital muscle, SCM, Pect m, UT, and LS, and the decrement results showed an increase of about 15% in the suboccipital muscle, SCM, Pect m, UT, and LS in both the MFR and DASR groups(p<0.05). Conclusions: In patients experiencing chronic neck pain, application of MFR and duoball assisted self relaxion was shown to be effective on pain and muscle tension. MFR is a non-pharmacological intervention method with few potential side effects and is considered a universal and easily applicable treatment method.

Background: The aim of this study was to determine the impact of applied progressive muscle relaxation training on the levels of depression, anxiety and stress among prostate cancer patients. Materials and Methods: A quasi-experimental study was conducted at the University Malaya Medical Centre (UMMC) and Universiti Kebangsaan Malaysia Medical Centre (UKMMC) over six months. Prostate cancer patients from UMMC received the intervention and patients from UKMMC were taken as controls. The level of depression, anxiety and stress were measured using Depression, Anxiety Stress Scales - 21 (DASS-21). Results: A total of 77 patients from the UMMC and 78 patients from the UKMMC participated. At the end of the study, 90.9% and 87.2% of patients from the UMMC and UKMMC groups completed the study respectively. There were significant improvements in anxiety (p<0.001, partial ${\eta}^2$=0.198) and stress (p<0.001, partial ${\eta}^2$=0.103) at the end of the study in those receiving muscle training. However, there was no improvement in depression (p=0.956). Conclusions: The improvement in anxiety and stress showed the potential of APMRT in the management of prostate cancer patients. Future studies should be carried out over a longer duration to provide stronger evidence for the introduction of relaxation therapy among prostate cancer patients as a coping strategy to improve their anxiety and stress.
