  • This study was carried out of to determine effects of high voltage pulsed galvanic stimulation on the soleus. target muscle of the sciatic nerve, of white rat two condition. The coditions included normal, and denervated muscle. The histochemical, ultrastructural observations were made. The following results were obtains. 1. The histochemical observations found the inflammatory cells between muscle bundle and muscle fiber since 1-week control group. In addition, nucleus located in the muscle fiber was frequently observed. 2. The experimental group showed a similar phenomenom to the normal muscles in terms of glycogen granules in the 1-week group, where as fiber were not distinguishable in4-weeks group which indicated that the degenerative changes had occured. 3. The NADH-TR reaction showed that the red muscle slightly increased in the 2-weeks group, and the distinguished was impossible the red fiber 4. The ultrastructures of the muscles in both groups were severely bend, and a number of vacuoles were observed due to the destruction of mitochondria..

Efficacy of Alkali-treated Sugarcane Fiber for Improving Physicochemical and Textural Properties of Meat Emulsions with Different Fat Levels

  • The objective of this study was to evaluate the efficacy of alkaline-treated sugarcane bagasse fiber on physicochemical and textural properties of meat emulsion with different fat levels. Crude sugarcane bagasse fiber (CSF) was treated with calcium hydroxide ($Ca(OH_2)$) to obtain alkaline-treated sugarcane bagasse fiber (ASF). The two types of sugarcane bagasse fiber (CSF and ASF) were incorporated at 2% levels in pork meat emulsions prepared with 5%, 10% and 20% fat levels. Alkaline-treatment markedly increased acid detergent fiber content (p=0.002), but significantly decreased protein, fat, ash and other carbohydrate contents. ASF exhibited significantly higher water-binding capacity, but lower oil-binding and emulsifying capacities than CSF. Meat emulsions formulated with 10% fat and 2% sugarcane bagasse fiber had equivalent cooking loss and textural properties to control meat emulsion (20% fat without sugarcane bagasse fiber). The two types of sugarcane bagasse fiber had similar impacts on proximate composition, cooking yield and texture of meat emulsion at the same fat level, respectively (p>0.05). Our results confirm that sugarcane bagasse fiber could be a functional food ingredient for improving physicochemical and textural properties of meat emulsion, at 2% addition level. Further, the altered functional properties of alkaline-treated sugarcane bagasse fiber had no impacts on physicochemical and textural properties of meat emulsions, regardless of fat level at 5%, 10% and 20%.

  • The musculotendon model is presented to show the declines in muscle force and shortening velocity during muscle fatigue due to the repeated functional electrical stimulation (FES). It consists of the nonlinear activation and contraction dynamics including physiological concepts of muscle fatigue. The activation dynamics represents $Ca^{2+}$ binding and unbinding mechanism with troponins of cross-bridges in sarcoplasm. It has the constant binding rate or activation time constant and two step nonlinear unbinding rate or inactivation time constant. The contraction dynamics is the modified Hill type model to represent muscle force - length and muscle force - velocity relations. A muscle fatigue profile as a function of the intracellular acidification, pH is applied into the contraction dynamics to represent the force decline. The computer simulation shows that muscle force and shortening velocity decline in stimulation time. And we validate the model. The model can predicts the proper muscle force without changing its parameters even when existing the estimation errors of the optimal fiber length. The change in the estimate of the optimal fiber length has an effect only on muscle time constant in transient period not on the tetanic force in the steady-state and relaxation periods.

Correlation of Marbling Characteristics with Meat Quality and Histochemical Characteristics in Longissimus Thoracis Muscle from Hanwoo Steers

  • The objective of this study was to investigate the correlations of marbling characteristics, including marbling score, intramuscular fat (IMF) content, and fleck traits, with meat quality traits and histochemical characteristics of the longissimus thoracis muscle from Hanwoo steers. Marbling fleck characteristics, especially area, number, and fineness (F) index, measured by computerized image analysis were strongly correlated with marbling score and IMF content (p<0.05). However, coarseness (C) index and F/C ratio were somewhat limited relationships with marbling score. In contrast, the IMF content and the number of smaller white flecks increased with increasing lightness value (p<0.05). Moreover, beef with higher marbling scores showed lower cooking loss and Warner-Bratzler shear force value compared to beef with lower marbling scores (p<0.05). Regarding the muscle bundle traits, as number of bundle increased, number of marbling flecks increased (p<0.05), although most marbling characteristics did not have significant correlation with muscle fiber or bundle characteristics.

Effects of Soluble Dietary Fiber on Skeletal Muscle GLUT4 Protein Contents in SHRSP Fed a High-Fat Diet (고지방식유래의 뇌졸중 유발 고혈압 흰쥐(SHRSP)에 있어서 수용성 식이 섬유의 급여가 근섬유 형태에 따른 GLUT4 발현에 미치는 영향)

  • The Purpose of this study is to investigate the effect of soluble dietary fiber psyllium on insulin sensitivity and skeletal muscle glucose transporter 4(GLUT4) protein expression in stroke-prone hypertensive rats(SHRSP) fed a high-fat diet containing 5% of psyllium or cellulose from five to nine weeks of age. Obtained results were as follows : (1) In the psyllium diet group fasting plasma glucose level was significantly reduced and glucose levels upon oral glucose tolerance test were significantly lower than cellulose diet group at 30 min(p<0.05) and 60 min(p<0.01) (2) Skeletal muscle GLUT4 contents were significantly increased in the soleus(slow twitch) and extensor digitorum longus(fast twitch) muscle of psyllium diet group. (3) However there was no difference in insulin levels in the fasting and oral glucose tolerance test. These results indicated that psyllium diet improves peripheral insulin sensitivity but not insulin secretion. In conclusion our present finding suggest that soluble fiber diet is effective to increase insulin sensitivity in SHRSP. From these results it was suggested that soluble dietary fiber supplementation effectively prevents insulin resistance.

  • Fiber type-specific programs controlled by the transcription factor MEF2 dictate muscle functionality. Here, we show that HDAC4, a potent MEF2 inhibitor, is predominantly localized to the nuclei in fast/glycolytic fibers in contrast to the sarcoplasm in slow/oxidative fibers. The cytoplasmic localization is associated with HDAC4 hyper-phosphorylation in slow/oxidative-fibers. Genetic reprogramming of fast/glycolytic fibers to oxidative fibers by active CaMKII or calcineurin leads to increased HDAC4 phosphorylation, HDAC4 nuclear export, and an increase in markers associated with oxidative fibers. Indeed, HDAC4 represses the MEF2-dependent, PGC-$1{\alpha}$-mediated oxidative metabolic gene program. Thus differential phosphorylation and localization of HDAC4 contributes to establishing fiber type-specific transcriptional programs.

An Immunohistochemical Study of Effects of Therapeutic Ultrasound on the Expression of VEGF and Substance-P in Muscle Contusion Injury (근타박상시 치료용 초음파가 혈관내피성장인자와 Substance-P 발현에 미치는 효과에 대한 면역조직화학적 연구)

  • Therapeutic angiogenesis is the controlled induction or stimulation of new blood vessel formation to reduce unfavourable tissue effects caused by local hypoxia and to enhance tissue repair. Therapeutic ultrasound can be considered as a physical agent to deliver therapeutic angiogenesis. The purpose of this study was to evaluate the effect of therapeutic ultrasound after muscle contusion injury by observed immunoreactivity of vascular endothelial growth factor(VEGF) that plays an important role in angiogenesis and substance-P in pain transmission. Ultrasound irradiation(1MHz, $1W/cm^2$, continuous mode, treatment time 5 min) was applied through water submersion technique to 1 limb daily by kept off 5cm from muscle belly of gastrocnemius. The result of this study were as follows. 1. In morphological observation, there were no significant changes excepts of 7 days. At 7 days, granular tissue viewed abundantly in control group. In other groups, general feature were increased interspace of muscle fiber; centronucleated muscle fiber; collapsed of muscle and nerve tissue; appeared inflammatory cell. 2. The VEGF was expressed in interspace of muscle fiber. Especially, at 7 days in experimental group, VEGF was showed in connective tissue surrounding gastrocnemius muscle. 3. The VEGF was higher expressed in experimental group at 2 and 3 days, but in control group at 7 days. These data suggest therapeutic ultrasound enhanced production of VEGF in the early day relatively, therefore stimulated angiogenesis in the skeletal muscle induced contusion injury. Also therapeutic ultrasound may stimulate pain relief by diminish of substance-P in dorsal horn of spinal cord.

The Effects of Shihosogan-san on Alcohol-induced Muscle Atrophy in Rats (흰쥐의 만성 알콜성 근위축에 시호소간산(柴胡疎肝散)이 미치는 효과)

  • Chronic or acute alcohol abuse often leads to liver injury associated with alcoholic hepatitis, liver fibrosis, cirrhosis, and liver cancer. In addition to the liver, alcohol abuse also induces a variety of other tissue injuries including pancreatitis, cardiomyopathy, neurotoxicity and muscle loss. Chronic skeletal muscle myopathy, independent of peripheral neuropathy, is well recognised in alcoholic patients. Several mechanisms may be involved in the pathogenesis of alcoholic myopathy. Ethanol is a potent inhibitor of muscle protein synthesis. Gastrocnemius and plantaris muscles are Type II fiber-predominant and usually considered representative of the musculature as a whole. Whereas, soleus muscle is Type I fiber predominant. Shihosogan-san is a traditional Korean medicine that is widely employed to treat indigestion and liver diseases. Muscle diseases are often related to liver diseases and conditions. We therefore tested the hypothesis that treatment with Shihosogan-san could ameliorate the ethanol-induced changes in muscle protein synthesis. Young male Sprague-Dawley rats were orally given 25% ethanol (5ml/kg, body weight) daily with Ethanol for 28 days. Normal group was similarly administrated with saline. In Shihosogan-san treated group, rats were orally administrated Shihosogan-san extract, and rats of EtOH group were given with the vehicle only. After 4 week, the morphology of gastrocnemius and plantaris muscles were assessed by hematoxylin and eosin staining. For comparative purposes, liver function was also investigated. The muscles from rats of EtOH group displayed a significant reduction in average cross section area compared to Normal group. Shihosogan-san treated group had increased fiber compared to the EtOH group. Moreover, Shihosogan-san treated group compared with EtOH group showed significantly decreased pro-apoptotic BAX expression and increased anti-apoptotic Bcl-2 expression. In conclusion, Shihosogan-san extract showed ameliorating effects on chronic alcohol toxicity in skeletal muscle.

Laminin as a Key Extracellular Matrix for Proliferation, Differentiation, and Maturation of Porcine Muscle Stem Cell Cultivation

  • Extracellular matrix (ECM) proteins play a crucial role in culturing muscle stem cells (MuSCs). However, there is a lack of extensive research on how each of these proteins influences proliferation and differentiation of MuSCs from livestock animals. Therefore, we investigated the effects of various ECM coatings-collagen, fibronectin, gelatin, and laminin-on the proliferation, differentiation, and maturation of porcine MuSCs. Porcine MuSCs, isolated from 14-day-old Berkshire piglets, were cultured on ECM-coated plates, undergoing three days of proliferation followed by three days of differentiation. MuSCs on laminin showed higher proliferation rate than others (p<0.05). There was no significant difference in the mRNA expression levels of PAX7, MYF5, and MYOD among MuSCs on laminin, collagen, and fibronectin (p>0.05). During the differentiation period, MuSCs cultured on laminin exhibited a significantly higher differentiation rate, resulting in thicker myotubes compared to those on other ECMs (p<0.05). Also, MuSCs on laminin showed higher expression of mRNA related with maturated muscle fiber such as MYH1 and MYH4 corresponding to muscle fiber type IIx and muscle fiber type IIb, respectively, compared with MuSCs on other ECM coatings (p<0.05). In summary, our comparison of ECMs revealed that laminin significantly enhances MuSC proliferation and differentiation, outperforming other ECMs. Specifically, muscle fibers cultured on laminin exhibited a more mature phenotype. These findings underscore laminin's potential to advance in vitro muscle research and cultured meat production, highlighting its role in supporting rapid cell proliferation, higher differentiation rates, and the development of mature muscle fibers.

The effects of aqua-exercise on the muscle atrophy of hind limb in rats

  • This study was peformed to investigate the effects on skeletal muscle recovery with aqua-exercise; swimming to take the muscle endurance for 20 days on two group of white rats which were the low extremity atrophy group(control groups) by fixed for two weeks and aqua-exercise group(experimental groups) after it. The effects was observed with light and electron microcope to measure the morphological changes of muscle fibers. The results obtained were as follow. 1. Light microscope: In the case of control groups, quadriceps fibers had been irregular alignment, decreased muscle width and the irregular alignment nuclear appeared, as it is degenerative muscle fibers. In the case of experimental groups, the fibers had been regular alignment cells and fibers. The nucleus of muscle had been normal characterized by oval shape and fiber sarcomere clearly classified. 2. Electron microscope: In the case of control groups, there were the quadriceps which was Z-line streaming phenomenon induced at the sarcomere and cells nuclear separated from basal membrane. It was not only observed the sarcomere alignment irregularly and mitochondria damaged, but also vacuoles found. In the case of experimental groups, A band, I band, H band had been clearly appeared, classified at the myofibrils of quadriceps, and electronic dense M-line found in sarcomere. There were observed satellite cells and basal laminas that usually to be appeared at the time of mitochondrial development, skeletal muscle fiber regeneration or development. This results suggest that the aqua-exercise assisted to inhibit the degenerative morphological changes of skeletal muscle cells and help to recover from abnormal states. Especially, it is considered to effect on a normal structural formation.
