Effect of $GA_3$ and ABA on Peroxidase and Catalase Activities and Isoperoxidase Patterns in Mung Bean Seedling (녹두의 발아과정 중 $GA_3$ 및 ABA의 처리가 Peroxidase, Catalase 활성변화와 Isoperoxidase Pattern에 미치는 영향)

  • The changes in peroxidase and catalase activities, and isoperoxidase patterns in different parts of mung bean seedling caused by the treatment with plant growth substances, $GA_3$ and ABA, were examined. As germination proceeded, the activity of peroxidase in all part except hypocotyl was increased, while that of catalase decreased. The separate application of $GA_3$ and ABA increased the activity of peroxidase which was more influenced by $GA_3$ than by ABA only in cotyledon, while that of catalase was more affected by ABA than by $GA_3$. Electrophoretic study revealed that the number of isoperoxidase was increased continuously in all parts during development. A greater influence was exerted on the intensity of isozyme than the number of isozyme by the hormonal treatment.

  • Effects of the plant growth regulator (P. G. R.)on the reaction of proteinase, $\gamma-amylase$ and acid phosphatase were investigated, and also were the conditions of production of P. G. R. by Stroptomyces sp. 445. The P. G. R. had no effect on the act ivities of such enzymes in mung bean seedling. But in germinating seed previously treated with P. G. R. it effected the activity of protease in cotyledon. In the conditions of production of P. G. R., the maxim, activity was appeared in shaking cutlure at $30^{\circ}C$ for 5 days, and by the addition of peptone or casein hydrolysate as nitrogen source, soluble starch as carbon source, and sulfur as metal ion.

  • The changes in peroxidase activity and its isozyme pattern in the different parts of mung bean sprout were investigated; The enzyme activity in cotyledon and root showed a tendency to increase at an early stage and then decreased gradually as germination continued. However, the crude homogenate of epicotyl and hypocotyl showed a continuous decline in the enzyme activity. In particular, the enzyme activity of the root was $1.5{\sim}3.5$ times higher than that of other parts. Gel electrophoresis of the crude homogenate revealed that the number of isozyme in every part of the mung bean sprout increase during germination up to 6th days. Two isozymes from root were partially purified by ammonium sulfate fractionation, gel filtration by Sephadex G-75 and DEAE cellulose column chromatography. One of the isozymes (A) was purified 16-fold by the present procedure, but the purity of the other isosyme (B) was not increased , significantly. Isozyme A was the most active at $65^{\circ}C$ and isozyme B at $70^{\circ}C$, while both isozyme (A, B) have a optimal pH of 5.6. The Km values of isozyme A and B for 0-dianisidine as a hydrogen donor determined to be 0.071 mM and 0.052mM, respectively, and those for isozyme A and B using $H_2O_2$ as a hydrogen acceptor were 0.28mM and 0.23mM, respectively.

The Effects of ultrasonic irradition on germination of mung bean (녹두 발아에 미치는 초음파 조사의 영향)

  • The experiment was carried out to examine on effect of ultrasonication on the growith of a mungbean while a mungbean is germinated and to measure amino acid content of a mungbean during its germination. Mungbean seeds soaked are divided into three groups; the first group was control group, untreated and naturally germinated. the second was ultrasonic treated group(UTG I), irradiated by ultrasonication for 8, 16 and 24min respectively immediately after soaking. the third was ultrasonic treated group(UTG II), irradiated by ultrasonication after 48hr germinating. The results were as foolows; Fresh weight, moisture, length of mungbean increased as its germination proceeds. while dry weight decreased. Ultrasonication tends to inhibit its germination of mungbean and results in the decrease in weight, length and moisture. Specially ultrasonication had a marked effect on UTG II. The longer the time of ultrasonication was, the more was the growth inhibited. In the control groups, the content of amino acid increased until 72 hours, but after that, it decreased. of the amino acid determined, glutamic acid decreased marekedly in the cotyledons, While the opposite was the case with aspartic acid in the hypocotyls. Comparing with the control groups, UTG I, the content of amino acid decreased. As the ultrasonic treaed time was increased, the concentration of amino acid in hypocotyl cotyledon tended to decreased. UTG II, the content of amino acid was lower than control groups, but higher than UTG I, and the effects of ultrasonication were similar to UTG I.
