Exosome-derived microRNA-29c Induces Apoptosis of BIU-87 Cells by Down Regulating BCL-2 and MCL-1

  • Background: Aberrant expression of the microRNA-29 family is associated with tumorigenesis and cancer progression. As transport carriers, tumor-derived exosomes are released into the extracellular space and regulate multiple functions of target cells. Thus, we assessed the possibility that exosomes could transport microRNA-29c as a carrier and correlations between microRNA-29c and apoptosis of bladder cancer cells. Materials and Methods: A total of 28 cancer and adjacent tissues were examined by immunohistochemistry to detect BCL-2 and MCL-1 expression. Disease was Ta-T1 in 12 patients, T2-T4 in 16, grade 1 in 8, 2 in 8 and 3 in 12. The expression of microRNA-29c in cancer tissues was detected by quantitative reverse transcriptase PCR (QRT-PCR). An adenovirus containing microRNA-29c was used to infect the BIU-87 human bladder cancer cell line. MicroRNA-29c in exosomes was measured by QRT-PCR. After BIU-87 cells were induced by exosomes-derived microRNA-29c, QRT-PCR was used to detect the level of microRNA-29c. Apoptosis was examined by flow cytometry and BCL-2 and MCL-1 mRNA expressions were assessed by reverse transcription-polymerase chain reaction. Western blotting was used to determine the protein expression of BCL-2 and MCL-1. Results: The expressions of BCL-2 and MCL-1 protein were remarkably increased in bladder carcinoma (p<0.05), but was found mainly in the basal and suprabasal layers in adjacent tissues. The expression of microRNA-29c in cancer tissues was negatively correlated with the BCL-2 and MCL-1. The expression level of microRNA-29c in exosomes and BIU-87 cells from the experiment group was higher than that in control groups (p<0.05). Exosome-derived microRNA-29c induced apoptosis (p<0.01). Although only BCL-2 was reduced at the mRNA level, both BCL-2 and MCL-1 were reduced at the protein level. Conclusions: Human bladder cancer cells infected by microRNA-29c adenovirus can transport microRNA-29c via exosomes. Moreover, exosome-derived microRNA29c induces apoptosis in bladder cancer cells by down-regulating BCL-2 and MCL-1.

  • This study was carried out with an objective to prepare the framework of conformity for the 21st century modern fashion, which is interactive with mankind, based on a position that the 21st century contemporary fashion with its amalgamating trend of diversified artistic forms may not be elucidated on a standpoint centering on one certain modality. The hybrid fashion trend of historical modality which provides freedom and satisfaction in creation of an individuals expressive power in expressing true desire of mans inside through man-centered thought of the times has made multifarious motives of the past and present fragmentary. It thus extracts inner divisions and the concept of consolidation through three-dimensional form. Modern fashion is newly interpreted by the material and details of high technologies. and is harmonized by mix and match with various expressions. By having it, it is being expressed together with many types of dresses and ornaments. The fact that regional elements could be expressed as if they were interrelated without each being deprived of originality may be explained with an interpretation on pluralism. The hybrid fashion design trend of cultural aspect, which manifests in the background of scientific technological culture as it transcends the societal-cultural boundaries based on the de-centralization theory of Frederic Jameson, has supplemented the imperfect meanings through conversional correlation with other forms and internal program changes. The middle stratal and polyhedral characteristics are seen as each cultural element is dismantled and reassembled by application of 'multiple-time point expression'. Design forming method is not to bring destruction from outside, but to embrace instability and chaos through radical dismantling of the inside. and to pursue diversity and openness. Thus. it is implemented by an approach that takes the role of design process. In communication of discontinuity, continuity was dismantled through forms of mixing, overlap, perversion, insertion and coincidence.

Expression patterns of TRα and CRABPII genes in Chinese cashmere goat skin during prenatal development

  • Background: The physiologic characteristics of the cashmere trait and many of the differentially expressed genes relevant to hair cycling have been extensively studied, whereas genes involved in the prenatal development of hair follicles have been poorly investigated in cashmere goats. The aim of this study, therefore, was to quantify the time-course changes in the expressions of $TR{\alpha}$ and CRABPII genes in the fetal skin of Chinese cashmere goats at the multiple embryonic days (E70, E75, E80, E90, E100, E120 and E130) using real-time quantitative PCR (RT-qPCR). Results: RT-qPCR showed that $TR{\alpha}$ was expressed at E70 with relatively high level and then slightly decreased (E75, E80, and E90). The highest expression of $TR{\alpha}$ mRNA was revealed at E130 (P > 0.05). The expression pattern of CRABPII mRNA showed an 'up-down-up' trend, which revealed a significantly highest expression at E75 (P < 0.05) and was down-regulated during E80 to E120 (P < 0.05) and mildly increased at E130, subsequently. Conclusion: This study demonstrated that $TR{\alpha}$ and CRABPII genes expressed in different levels during prenatal development of cashmere. The present study will be helpful to provide the comprehensive understanding of $TR{\alpha}$ and CRABPII genes expressions during cashmere formation and lay the ground for further studies on their roles in regulation of cashmere growth in goats.

  • The release of IEEE 802.15.4e specification significantly develops IEEE 802.15.4. The most inspiring improvement is the enhancement for medium access control (MAC) sublayer. To study the performance of IEEE 802.15.4e MAC, in this paper we first present an overview of IEEE 802.15.4e and introduce three MAC mechanisms in IEEE 802.15.4e. And the major concern here is the Time Slotted Channel Hopping (TSCH) mode that provides deterministic access and increases network capacity. Then a detailed analytical Markov chain model for TSCH carrier sense multiple access with collision avoidance (CSMA-CA) is presented. Expressions which cover most of the crucial issues in performance analysis such as the packet loss rate, energy consumption, normalized throughput, and average access delay are presented. Finally the performance evaluation for the TSCH mode is given and we make a comprehensive comparison with unslotted CSMA-CA in non-beacon enabled mode of IEEE 802.15.4. It can validate IEEE 802.15.4e network can provide low energy consumption, deterministic access and increase network capacity.

Spectrum Hole Utilization in Cognitive Two-way Relaying Networks

  • This paper investigates the spectrum hole utilization of cooperative schemes for the two-way relaying model in order to improve the utilization efficiency of limited spectrum holes in cognitive radio networks with imperfect spectrum sensing. We propose two specific bidirectional secondary data transmission (BSDT) schemes with two-step and three-step two-way relaying models, i.e., two-BSDT and three-BSDT schemes, where the spectrum sensing and the secondary data transmission are jointly designed. In the proposed cooperative schemes, the best two-way relay channel between two secondary users is selected from a group of secondary users serving as cognitive relays and assists the bi-directional communication between the two secondary users without a direct link. The closed-form asymptotic expressions for outage probabilities of the two schemes are derived with a primary user protection constraint over Rayleigh fading channels. Based on the derived outage probabilities, the spectrum hole utilization is calculated to evaluate the percentage of spectrum holes used by the two secondary users for their successful information exchange without channel outage. Numerical results show that the spectrum hole utilization depends on the spectrum sensing overhead and the channel gain from a primary user to secondary users. Additionally, we compare the spectrum hole utilization of the two schemes as the varying of secondary signal to noise ratio, the number of cognitive relays, and symmetric and asymmetric channels.

  • Emotion affects many parts of human life such as learning ability, behavior and judgment. It is important to understand human nature. Emotion can only be inferred from facial expressions or gestures, what it actually is. In particular, emotion is difficult to classify not only because individuals feel differently about emotion but also because visually induced emotion does not sustain during whole testing period. To solve the problem, we acquired bio-signals and extracted features from those signals, which offer objective information about emotion stimulus. The emotion pattern classifier was composed of unsupervised learning algorithm with hidden nodes and feature vectors. Restricted Boltzmann machine (RBM) based on probability estimation was used in the unsupervised learning and maps emotion features to transformed dimensions. The emotion was characterized by non-linear classifiers with hidden nodes of a multi layer neural network, named deep belief network (DBN). The accuracy of DBN (about 94 %) was better than that of back-propagation neural network (about 40 %). The DBN showed good performance as the emotion pattern classifier.

  • In this paper, we describe a case of using robot technology in child studies to examine children's judgement and reasoning of the life phenomenon on boundary objects. In order to control the effects of the appearance of the robot, which children observe or interact directly with, on the children's judgement and reasoning of the life phenomenon, we developed a robot similar to human. Unit experimental scenarios representing biological and psychological properties were implemented based on control of robot's motion, speech, and facial expressions. Experimenters could combine these multiple unit scenarios in a cascade to implement various scenarios of the human-robot interaction. Considering that the experimenters are researchers of child studies, there was a need to develop a remote operation console that can be easily used by non-experts in the robot field. Using the developed robot platform, researchers of child studies could implement various scenarios by manipulating the biological and psychological properties of the robot based on their research hypothesis. As a result, we could clearly see the effects of robot's properties on children's understanding about boundary object like robots.

Joint Destination-Relay Selection and Antenna Mode Selection in Full-Duplex Relay Network

  • In this paper, a joint destination-relay selection and antenna mode selection scheme for full-duplex (FD) relay network is investigated, which consists of one source node, N FD amplify-and-forward (AF) relays and M destination nodes. Multiple antennas are configured at the source node, and beamforming technique is adopted. Two antennas are employed at each relay, one for receiving and the other for transmitting. Only one antenna is equipped at each destination node. In the proposed scheme, the best destination node is firstly selected according to the direct links between the source node and destination nodes. Then the transmit and receive mode of two antennas at each relay is adaptively selected based on the relaying link condition. Meanwhile, the best relay with the optimal Tx/Rx antenna configuration is selected to forward the signals. To characterize the performance of the proposed scheme, the closed-form expression of the outage probability is derived; meanwhile, the simple asymptotic expressions are also obtained. Our analysis shows that the proposed scheme obtains the benefits of multi-relay diversity and multi-destination diversity. Moreover, extra space diversity in the medium SNR region can be achieved due to the antenna selection at the relay. Finally, Monte-Carlo simulations are provided to consolidate the analytical results, and show the effectiveness of the proposed scheme.

Regulation of RANTES and MCP Expression in Human Nasal Mucosal Fibroblasts (비점막(鼻粘膜) 섬유모세포에서의 RANTES와 MCP의 발현 조절)

  • Background: Fibroblast functions both as a structural element and as a vital immunoregulatory cell. Fibroblasts regulate inflammation through governing of chemokine expression. In order to elucidate the mechanisms by which the expressions of chemokines were regulated, the co-stimulatory effects of Th1 and proinflammatory cytokines were compared using nasal mucosal fibroblasts. Methods: Human nasal mucosa was obtained from surgery for septal deviation and the growth of fibroblasts was established. Fibroblasts from 4th to 6th passage were stimulated with various combinations of cytokines. To inhibit selected signaling pathways, fibroblasts were pretreated with cyclosporin A, wortmannin, staurosporine, and dexamethasone prior to the stimulation with cytokines. The supernatants were collected and chemokines were detected with a sandwich enzyme-linked immunosorbent assay. Results: $TNF-{\alpha}/IFN-{\gamma}$-induced production of RANTES was inhibited by all inhibitors used. MCP-1 was produced constitutively and $TNF-{\alpha}$-induced or $TNF-{\alpha}/IFN-{\gamma}$-induced production of MCP-1 was not inhibited by cyclosporin A or wortmannin, but by stauroporine or dexamethasone. All inhibitors used in this experiment inhibited $TNF-{\alpha}/IFN-{\gamma}$-induced or $IL-1{\beta}/IFN-{\gamma}$-induced production of MCP-2 in nasal mucosal fibroblasts. Although staurosporine or dexamethasone showed strong inhibitory effects, cyclosporin A or wortmannin did not inhibit the production of MCP-3 by $IL-1{\beta}/IFN-{\gamma}$ treatment. Conclusion: Chemokines were strongly induced by stimulation of cytokines in combination and showed different pattern of inhibition by the inhibitors. Therefore, it was assumed that cytokines acted on multiple pathways or on unknown pathways which converged to gene-specific transcription factors.

Differential Expression of $PKD2$-Associated Genes in Autosomal Dominant Polycystic Kidney Disease

  • Autosomal dominant polycystic kidney disease (ADPKD) is characterized by formation of multiple fluid-filled cysts that expand over time and destroy renal architecture. The proteins encoded by the $PKD1$ and $PKD2$ genes, mutations in which account for nearly all cases of ADPKD, may help guard against cystogenesis. Previously developed mouse models of $PKD1$ and $PKD2$ demonstrated an embryonic lethal phenotype and massive cyst formation in the kidney, indicating that $PKD1$ and $PKD2$ probably play important roles during normal renal tubular development. However, their precise role in development and the cellular mechanisms of cyst formation induced by $PKD1$ and $PKD2$ mutations are not fully understood. To address this question, we presently created $Pkd2$ knockout and $PKD2$ transgenic mouse embryo fibroblasts. We used a mouse oligonucleotide microarray to identify messenger RNAs whose expression was altered by the overexpression of the $PKD2$ or knockout of the $Pkd2$. The majority of identified mutations was involved in critical biological processes, such as metabolism, transcription, cell adhesion, cell cycle, and signal transduction. Herein, we confirmed differential expressions of several genes including aquaporin-1, according to different $PKD2$ expression levels in ADPKD mouse models, through microarray analysis. These data may be helpful in $PKD2$-related mechanisms of ADPKD pathogenesis.
