Expression of Mouse $\alpha-Amylase$ Gene in Methylotrophic Yeast Pichia pastoris

  • The expression of the mouse $\alpha-amylase$ gene in the methylotrophic yeast, P pastoris was investigated. The mouse $\alpha-amylase$ gene was inserted into the multi-cloning site of a Pichi a expression vector, pPIC9, yielding a new expression vector pME624. The plasmid pME624 was digested with SalI or BglII, and was introduced into P. pastoris strain GSl15 by the PEG1000 method. Fifty-three transformants were obtained by the transplacement of pME624 digested with SaiII or BglII into the HIS4locus $(38\;of\;Mut^+\;clone)$ or into the AOX1 locus $(15\;of\;Mut^s\;clone)$. Southern blot was carried out in 11 transformants, which showed that the mouse $\alpha-amylase$ gene was integrated into the Pichia chromosome. When the second screening was performed in shaker culture, transformant G2 showed the highest $\alpha-amylase$ activity, 290 units/ml after 3-day culture, among 53 transformants. When this expression level of the mouse $\alpha-amylase$ gene is compared with that in recombinant Saccharomyces cerevisiae harboring a plasmid encoding the same mouse $\alpha-amylase$ gene, the specific enzyme activity is eight fold higher than that of the recombinant S. cerevisiae.

항투명대 항체의 특성에 관한 연구 II. 항투명대 항체가 돼지난자와 생쥐난자의 체외수정에 미치는 영향 (Studies on the Characteristics of Anti-Zona Antibody II. Effect of Anti-Zona Antibody on Fertilization of Porcine and Mouse Eggs In Vitro)

  • These experiments were undertaken as a basic study to develop immunocontraceptive vaccine and to understand the role of zona pellucidae in early fertilization process by investigating the effect of monoclonal and polyclonal antibody to porcine zona pellucidae and polyclonal antibody to mouse zona pellucidae on the fertilization of porcine and mouse eggs in vitro. The results obtained in these experiments were summarized as follows : 1. Treatment of porcine and mouse eggs with undiluted anti-zona serum produced intense precipitation layer on the poricne and mouse zonae, respectively, thus resulting in the total inhibition of sperm adherence on surface of zona. 2. In vitro fertilization of eggs pre-treated with 0.3∼10% of various antibodies was examined, and resulting in that 5 and 10% of rabbit polyclonal antibodies to porcine zona inhibited completely both in vitro fertilization and polyspermy of porcine eggs while monoclonal to porcine zona and rabbit polyclonal antibody to mouse zona did not inhibit in vitro fertilization but monoclonal antibody reduced the rate of polyspermy compared to that of control group. Almost the same results were obtained in the study on the effect of anti-zona serum on in vitro fertilization of mouse eggs.

Shikonin ameliorates salivary gland damage and inflammation in a mouse model of Sjögren's syndrome by modulating MAPK signaling pathway

  • Sjögren syndrome (SS) is a systemic inflammatory autoimmune disease that involves exocrine glands. Shikonin is extracted from comfrey, which is conventionally used as an anti-tumor, antibacterial, and antiviral drug in China. However, the application of Shikonin in SS remains unreported. This study aimed to verify the potential functions of Shikonin in SS progression. Firstly, non-obese diabetic mice were used as the SS mouse model, with C57BL/6 mice serving as the healthy control. It was demonstrated that the salivary gland damage and inflammation were aggravated in the SS mouse model. Shikonin improved salivary gland function decline and injury in the SS mouse model. Moreover, Shikonin reduced inflammatory cytokines and immune infiltration in the SS mouse model. Further experiments discovered that Shikonin attenuated the MAPK signaling pathway in the SS mouse model. Lastly, inhibition of the MAPK signaling pathway combined with Shikonin treatment further alleviated the symptoms of SS. In conclusion, Shikonin ameliorated salivary gland damage and inflammation in a mouse model of SS by modulating the MAPK signaling pathway. Our findings indicate that Shikonin may be a useful drug for SS treatment.

TC1 (C8orf4) is involved in ERK1/2 pathway-regulated G1- to S-phase transition

  • Although previous studies have implicated a role for TC1 (C8orf4) in cancer cell proliferation, the molecular mechanism of its action is still largely unclear. In this study, we showed, for the first time, that the mRNA levels of TC1 were upregulated by mitogens (FBS/thrombin) and at least partially, through the ERK1/2 signaling pathway. Interestingly, the over-expression of TC1 promoted the $G_1$- to S-phase transition of the cell cycle, which was delayed by the deficiency of ERK1/2 signaling in fibroblast cells. Furthermore, the luciferase reporter assay indicated that the over-expression of TC1 significantly increased Cyclin D1 promoter-driven luciferase activity. Taken together, our findings revealed that TC1 was involved in the mitogen-activated ERK1/2 signaling pathway and positively regulated $G_1$- to S-phase transition of the cell cycle. Our results may provide a novel mechanism of the role of TC1 in the regulation of cell proliferation.

FDA-Test 생사판정법이 초급속 동결된 Mouse 수정란의 배양과 이식후 착상에 미치는 영향 I. FDA 첨가 수준이 초급속 동결된 생쥐상실배의 배양과 이식에 미치는 영향 (Effects of FDA-Test on the Survival and Conception Rate in Vitrified Mouse Embryos I. Effects of Addition Levels of FDA(Florescein Diacetate) on Survival and Conception Rate in Vitrified Mouse Morulae)

  • This study was carried out to test the effects of fluorescein diacetate(FDA 0${mu}ell$/ml, 0.5${mu}ell$/ml, 5${mu}ell$/ml, 10${mu}ell$/ml or 0${mu}ell$/ml, 5${mu}ell$/ml in PBS) treated before culture on the survival of vitrified mouse morulae in vitrification solution(20% glycerol+glycerol+10% ethylene glycol+30% ficoll+10% sucrose). The results were summarized as follows; 1. The survival rate of FDA-tested fresh mouse morulae after 24 hours culture was over 96%((P4.8) in the control or treatment groups with various levels of FDA. Because the rate of mouse morulae which developed to hatched blastocysts was higher with the various levels of FDA treatment(67%) than control(50%), it was considered that toxicity of FDA did not affect the survival of mouse morulae. 2. When mouse morulae were FDA-tested in FDA 0(control), 0.5, 5, and 10${mu}ell$/ml treatment after vitrification, the development rate to expanded blastocyst were 66, 82, 64 and 76%, and FDA scores were P4.2(84%), P4.7(94%), P4.2(84%) and P3.9(78%), respectively. There were no significant differences between control and FDA treatments, but there were significant difference between 0.5${mu}ell$/ml)94%) and 10${mu}ell$/ml(78%) treatment(P<0.01). 3. The survival rates of cultured mouse morulae according to FDA-scores(P0=non-fluorescence; P1~P5=according to their fluorescence) after vitrification were P5;92%, P4;67%, P3;42% and P2.P0;0%, respectively. 4. Implantation rates of morulae stage embryos cultured into early blastocysts and implanted into uterine hornes vitrification were 14 and 11% embryos treated control(0${mu}ell$/ml) and FDA 5${mu}ell$/ml and the normal fetus development was 2% embryos for both treatments. Results of this percent study indicated that toxicity of FDA does not affect not only the survival of fresh and vitrified mouse morulae but also the development rate and implantation of fetus after transfer as well. The development rates of mouse morulae with the FDA score of P5, P4 and P3 were 92, 67 and 42%, respectively, it was considered that FDA-test was fit for the judge of survival.

  • 해바라기 화분립이 mouse의 간장에서 cholesterol 대사에 어떤 영향을 미치는가를 조사하기 위해서 해바라기 화분립으로부터 추출한 지질을 column chromatography, thin-layer chromatography 및 gas-liquid chromatography를 이용해서 분석했다. 또한 mouse에 화분립을 투여하였을 때 간장에서의 cholesterol치에 미치는 영향에 대해서도 조사하였다. 1. 화분립의 지질은 neutral lipid 84.10%, glycolipid 10.50%, phospholipid 5.40%를 함유했다. 2. 중성지방질의 조성은 triglyceride가 71.20%로 가장 많고, sterol ester, diglyceride, monoglyceride, free fatty acid, sterol의 순이다. 3. neutral lipid, glycolipid, phospholip의 주요지방산은 linoleic acid 28.48${\sim}$33.70%, palmitic acid 12.90${\sim}$47.50%, oleic acid 11.20${\sim}$12.20% 이다. 그러나 phospholipid는 다른 지질보다 palmitic acid를 더 많이 함유하고 있다. 4. 해바라기 화분립 투여 mouse의 체중은 대조군 보다 현저하게 증가했다. 5. 화분립을 투여한 mouse의 간장지질대사가 대조군보다 활발함을 알 수 있었다. 6. 화분립 투여 mouse의 간 cholesterol치가 초기에는 증가했으나, 말기에는 대조군과 비슷한 수준으로 감소했다.

연속 촬영 이미지를 이용한 Mouse의 운동 능력 평가 시스템 (A New Locomotor Evaluation System for Mouses Based on Continuous Shooting Images)

  • 본 논문에서는 연속 촬영 이미지를 이용한 실험용 쥐의 운동 능력 평가 시스탬을 제안한다. 의학이나 수의학 분야에서는 인류의 양질의 삶을 위해 mouse를 이용한 동물실험을 실시하고 있다. 특히 실험용 쥐(mouse)를 이용한 실험 중에는 인위적인 부상(injury)을 만들고, 다양한 처치를 통하여 그 부상에서 회복되는 정도를 측정하는 실험을 많이 하고 있다. 이러한 실험에서 그 동안의 전통적인 운동량 측정 방법은 사람이 직접 관찰하는 방법으로 이루어지고 있었다. 제안한 시스템은 실험용 쥐에 대한 움직임을 정해진 단위 시간동안 연속 촬영을 실시하고, 이 연속 촬영된 이미지에서 mouse의 외곽선을 point cloud로 추출한다. 추출된 point cloud를 이용하여 mouse의 몸통에 대한 내부 외곽선(interior contour line)을 다시 추출한 후, 이를 이용하여 그 중심점을 구하고, 이들 중심점들을 연결하여 거리를 계산하여 누적함으로써 mouse가 단위 시간동안 이동한 총 거리를 자동 계산하도록 하는 시스템을 설계하고 구현한다.

  • Several types of IFNs were tested for their ability to suppress TCDD-inducible P-450 dependent mixed function oxidase (MFO) system in mouse primary hepatocytes. Mouse IFN-gamma (IFN-G) markedly suppressed EROD activity when added at the same time as TCDD (10 nM). The antagonism of EROD activity by IFN-G exhibited both a dose-(10-100 U/ml) and time-depedence. In contrast, mouse IFN-A/B was only moderately suppressive and only at high concentrations (500 U/ml). Rat IFN-G was even more selective than mouse, wherase human IFN-G had no activity.

  • [6]-Gingerol, a major pungent ingredient of ginger (Zingiber officinale Roscoe, Zingiberaceae) has a wide array of pharmacologic effects. Our previous studies have demonstrated that [6]-gingerol inhibits mouse skin tumor promotion and anchorage-independent growth of cultured mouse epidermal cells stimulated with epidermal growth factor. In this study, we have investigated the molecular mechanisms underlying anti-tumor promoting effects of [6]-gingerol on mouse skin carcinogenesis. (omitted)
