Reduction Effect of Air Cleaner on Particulate Matters and Biological Agents in a Swine Facility (공기정화기 적용에 따른 돈사 작업장내 입자상 물질 및 생물학상 물질 저감 효과에 관한 연구)

  • Objectives: This on-site study was performed to evaluate the reduction efficiency of an air cleaner on particulate matters and biological agents in a swine facility. Materials and Methods: Particulate matter was measured using a real-time monitoring recorder and biological agents were sampled with a one-stage impactor and then analyzed based on the microbial culture method. An experimental process for the reduction effect on airborne pollutants through air cleaner operation consisted of three conditions: no treatment, wet scrapper by water spray and wet scrapper by disinfectant spray. Results: Geometric mean levels of particulate matter(TSP, $PM_{10}$, $PM_{2.5}$ and $PM_1$) were presented at $1,608{\mu}g/m^3$, $1,373.8{\mu}g/m^3$, $401.8{\mu}g/m^3$ and $144.5{\mu}g/m^3$ for no treatment; $1,503{\mu}g/m^3$, $1,017{\mu}g/m^3$, $159.4{\mu}g/m^3$ and $69.8{\mu}g/m^3$ for wet scrapper by water spray; and $1,222.17{\mu}g/m^3$, $477.17{\mu}g/m^3$, $33.2{\mu}g/m^3$ and $11.1{\mu}g/m^3$ for wet scrapper by disinfectant spray, respectively. In the case of biological agents, the geometric averaged concentrations of total airborne bacteria and fungi were as follows: $45,371cfu/m^3$ and $13,474cfu/m^3$ for no treatment, $43,286cfu/m^3$ and $8,610cfu/m^3$ for wet scrapper by water spray, and $2,440cfu/m^3$ and 1,867 cfu/ for wet scrapper by disinfectant spray, respectively. Regardless of particulate matter and biological agent, the highest concentrations were found for no treatment, while the lowest concentrations were found with wet scrapper by disinfectant spray. Conclusions: Based on the results obtained from this on-site evaluation, there was a significant reduction effect on particulate matter and biological agents through the application of an air cleaner in this study.

Biogenic fabrication and characterization of silver nanoparticles using aqueous-ethanolic extract of lichen (Usnea longissima) and their antimicrobial activity

  • Background: Biogenic fabrication of silver nanoparticles from naturally occurring biomaterials provides an alternative, eco-friendly and cost-effective means of obtaining nanoparticles. It is a favourite pursuit of all scientists and has gained popularity because it prevents the environment from pollution. Our main objective to take up this project is to fabricate silver nanoparticles from lichen, Usnea longissima and explore their properties. In the present study, we report a benign method of biosynthesis of silver nanoparticles from aqueous-ethanolic extract of Usnea longissima and their characterization by ultraviolet-visible (UV-vis), Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM) and scanning electron microscopy (SEM) analyses. Silver nanoparticles thus obtained were tested for antimicrobial activity against gram positive bacteria and gram negative bacteria. Results: Formation of silver nanoparticles was confirmed by the appearance of an absorption band at 400 nm in the UV-vis spectrum of the colloidal solution containing both the nanoparticles and U. longissima extract. Poly(ethylene glycol) coated silver nanoparticles showed additional absorption peaks at 424 and 450 nm. FTIR spectrum showed the involvement of amines, usnic acids, phenols, aldehydes and ketones in the reduction of silver ions to silver nanoparticles. Morphological studies showed three types of nanoparticles with an abundance of spherical shaped silver nanoparticles of 9.40-11.23 nm. Their average hydrodynamic diameter is 437.1 nm. Results of in vitro antibacterial activity of silver nanoparticles against Staphylococcus aureus, Streptococcus mutans, Streptococcus pyrogenes, Streptococcus viridans, Corynebacterium xerosis, Corynebacterium diphtheriae (gram positive bacteria) and Escherichia coli, Klebsiella pneuomoniae and Pseudomonas aeruginosa (gram negative bacteria) showed that it was effective against tested bacterial strains. However, S. mutans, C. diphtheriae and P. aeruginosa were resistant to silver nanoparticles. Conclusion: Lichens are rarely exploited for the fabrication of silver nanoparticles. In the present work the lichen acts as reducing as well as capping agent. They can therefore, be used to synthesize metal nanoparticles and their size may be controlled by monitoring the concentration of extract and metal ions. Since they are antibacterial they may be used for the treatment of bacterial infections in man and animal. They can also be used in purification of water, in soaps and medicine. Their sustained release may be achieved by coating them with a suitable polymer. Silver nanoparticles fabricated from edible U. longissima are free from toxic chemicals and therefore they can be safely used in medicine and medical devices. These silver nanoparticles were stable for weeks therefore they can be stored for longer duration of time without decomposition.

Anti-tuberculosis effects of frankincense through immune responses of Mycobacterium tuberculosis-infected macrophages (결핵균이 감염된 대식세포의 면역반응을 통한 유향(Frankincense)의 항결핵효과)

  • Frankincense has been used as a traditional medicine for treating rheumatoid arthritis, dermatitis, and muscle pain. In this study, the anti-tuberculosis effects of Frankincense were evaluated in immune responses of macrophages. Frankincense methanol extract was not cytotoxic to the host. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction assay using human macrophage (THP-1) cells did not show cytotoxic effects or morphological changes with treatments of 31.3, 62.5, and 125 ㎍/mL Frankincense methanol extract (FRM). Inhibitory effects of Frankincense methanol extract on the growth of Mycobacterium tuberculosis in human macrophages were investigated. The immune response was measured by monitoring the levels of TNF-α and IL-1β in THP-1 cells with or without M. tuberculosis infection under Frankincense methanol extract treatment. Inflammatory cytokine levels and M. tuberculosis numbers were reduced in THP-1 cells treated with Frankincense methanol extract. Therefore, Frankincense methanol extract could be used as a potential anti-tuberculosis agent.

Determinations of Toltrazuril and Toltrazuril Sulfone Levels in Olive Flounder Paralichthys olivaceus Samples Using Liquid Chromatography-Electrospray Ionization Tandem Mass Spectrometry (LC-MS/MS를 이용한 넙치(Paralichthys olivaceus)시료의 톨트라주릴 및 톨트라주릴 설폰 분석)

  • Several studies investigating the prevention and treatment of external parasites in farmed olive flounder Paralichthys olivaceus have found that the anticoccidial agent toltrazuril sulfone is an effective antiparasitic. Prior to undertaking a full-scale study, we developed analytical methods to detect the levels of toltrazuril and toltrazuril sulfone in farmed flounder samples using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-MS/MS). This analysis showed that LC-MS/MS changed the mobile phase and collision energy of toltrazuril and toltrazuril sulfone. This was validated using established conditions. Sample pre-treatment for this process involved extraction with dichloromethane and purification by liquid-liquid extraction in formic acid, acetonitrile, and h-hexane, followed by determination of all compounds by LC-MS/MS. Separation was achieved within 10 min by gradient elution using a Capcell Pak C18 ($3.0{\mu}m$, $100{\times}2.0mm$) analytical column (Shiseido UG 120V) with a mixture of 0.1% (v/v) formic acid and acetonitrile. Multiple reaction monitoring was used for selective detection of toltrazuril and toltrazuril sulfone. This method yields satisfactory results for linearity, precision, and limits of quantification. Therefore, the method established in our study will serve as a basis for further research on parasite control by toltrazuril and toltrazuril sulfone.

Isolation Characteristics of causative agent of Streptococcosis and Serotype Changes of Streptococcus parauberis from olive flounder (Paralichthys olivaceus) in Jeju (제주지역 넙치(Paralichthys olivaceus) 연쇄구균병 원인체의 분리특성과 Streptococcus parauberis의 혈청형 변화)

  • During the period from 2003 to 2020, a total of 470 Streptococcus species were isolated from farmed flounder in Jeju. Out of 470 isolates, 92 (19.6%) isolates and 378 isolates (80.4%) were identified as Streptococcus iniae and S. parauberis, respectively by multiplex PCR assay. During that period, the percentage of S. iniae decreased from 56.9% in 2003 to 0.0% in 2020 whereas that of S. parauberis increased from 43.1% in 2003 to 100% in 2020. In the PCR assay for serotyping, the isolated S. parauberis showed 3 subserotypes, Ia (34.9%), Ib/Ic (46.3%) and II (18.8%). In 2003 and 2004, serotype II was dominant at 59.1% and 50.0% of isolation rates, however between 2005 and 2009, subserotype Ib/Ic was dominant (57.6%, 86.0%, 84.6%, 57.9%, and 83.3%). After 2010, except for 2015, subserotype Ia was the most dominant one. In the last 3 years (2018 to 2020), subserotype Ia was most abundant (70%), followed by subserotype Ib/Ic (16-30%). Serotype II was not isolated in 2018, but in 2019 and 2020, it showed an increased tendency to 3.4% and 16.7%, respectively. It is believed that continuous monitoring is necessary for research on counter-measures against streptococcosis of flounder.

Assessments of Dissolved Rare Earth Elements and Anthropogenic Gadolinium Concentrations in Different Processes of Wastewater Treatment Plant in Busan, Korea (부산 하수처리장에서 공정별 용존 희토류 원소의 농도 및 인위적 기원 가돌리늄의 배출량 평가)

  • Gadolinium, commonly used as a contrast agent for magnetic resonance imaging (MRI), is discharged into aquatic environments without removal after treatment in wastewater treatment plants (WWTPs) because of its high stability. In this study, we collected water samples from Suyeong WWTP, Busan, to investigate the dissolved rare earth element (REE) removal capacity of each wastewater treatment process and to evaluate the discharge of anthropogenic Gd (Gdanth) from effluents. As wastewater passed through each stage of treatment, the concentrations of light REEs (La-Eu) decreased, whereas those of heavy REEs (Tb-Lu) were relatively consistent. Negative Sm anomalies (<1) were observed in several samples, indicating that Sm can be removed by adsorption onto particles or phosphate during the biological removal process. Positive Gd anomalies (149±50, n=9) were observed in all samples. The ratios of Gdanth concentrations to measured Gd concentrations in all wastewater treatment processes were higher than 97%. This indicates that Gdanth was discharged to the Suyeong River without removal during the wastewater treatment process. Considering the daily treatment capacity in each process, the total flux of Gdanth was estimated to be 259 mmol/day. Our results suggest that mid- and/or long-term monitoring of Gd is needed because Gdanth is continuously discharged into Suyeong Bay through WWTPs.

Balancing Bleeding Risk and Thromboembolic Complications in Elderly Chronic Subdural Hematoma Patients Undergoing Burr Hole Trephination : A Multicenter Retrospective Cohort Study and Literature Review

  • Objective : Chronic subdural hematoma (CSDH) patients using antithrombotic agents (AT) at high risk for cardiovascular disease are increasing. The authors aimed to analyze the factors influencing outcome by targeting patients using AT and to establish a desirable treatment strategy. Methods : A retrospective analysis was performed on data from 462 patients who underwent burr hole trephination (BHT) surgery for CSDH at five hospitals from March 2010 to June 2021. Outcomes included incidence of postoperative acute bleeding, recurrence rate, and morbidity or mortality rate. Patients were divided into the following four groups based on their history of AT use : no AT. Only antiplatelet agents (AP), only anticoagulants (AC), both of AP and AC. In addition, a concurrent literature review was conducted alongside our cohort study. Results : Of 462 patients, 119 (119/462, 25.76%) were using AT. AP prescription did not significantly delay surgery (p=0.318), but AC prescription led to a significant increase in the time interval from admission to operation (p=0.048). After BHT, AP or AC intake significantly increased the period required for an in-dwelling drain (p=0.026 and p=0.037). The use of AC was significantly related to acute bleeding (p=0.044), while the use of AP was not (p=0.808). Use of AP or AC had no significant effect on CSDH recurrence (p=0.517 and p=1.000) or reoperation (p=0.924 and p=1.000). Morbidity was not statistically correlated with use of either AP or AC (p=0.795 and p=0.557, respectively), and there was no significant correlation with mortality for use of these medications (p=0.470 and p=1.000). Conclusion : Elderly CSDH patients may benefit from maintenance of AT therapy during BHT due to reduced thromboembolic risk. However, the use of AC necessitates individualized due to potential postoperative bleeding. Careful post-operative monitoring could mitigate prognosis and recurrence impacts.

A Study on Control Possibility of Ambrosia trifida L., an Invasive Alien Plant by the Feeding of Ophraella communa LeSage (돼지풀잎벌레의 섭식에 의한 생태계교란 식물인 단풍잎돼지풀의 제어 가능성 연구)

  • To develop an effective management plan for Ambrosia trifida L., an invasive alien plant in Korea, we assessed the potential of Ophraella communa LeSage as a biological control agent. This involved investigating the host specificity of the herbivore Ophraella communa LeSage, its annual travel distance, and the impact of this insect on the fitness of Ambrosia trifida L. We confirmed the host plant preference of Ophraella communa LeSage. The travel distance of this insect was determined by monitoring its appearance in selected Ambrosia trifida L. communities without these insects at distances of 10, 20, 30, and 100 meters, based on the locations where the presence of Ophraella communa LeSage was observed. The growth, reproductive, and physiological responses of Ambrosia trifida L. were measured according to feeding by Ophraella communa LeSage. As a result, Ophraella communa LeSage fed on only three taxa and moved short distances within a radius of 30 m per year from the host. The feeding behavior of the herbivore had a negative impact on the growth, reproductive, and physiological responses of Ambrosia trifida L. And the plant's growth and reproduction improved with increasing distance from the herbivore. Furthermore, the introduction of herbivores was able to control over 90% of Ambrosia trifida L. when the coverage of the Ambrosia trifida L. group was below 50%. However, the effectiveness of the removal decreased when the coverage exceeded 90%. These results are likely to be utilized by Ophraella communa LeSage as an ecological control agent. It is advantageous to introduce them in spring (May) when the coverage is low to maximize the effectiveness of control.

Assessment of Clinical Applicability of a New Plaque Scoring System Using Quantitative Light-Induced Fluorescence-Digital (Quantitative Light-Induced Fluorescence-Digital을 이용한 치면세균막 검사법의 임상적 활용 가능성 평가)

  • The aim of this study was to suggest a convenient method of monitoring the gingival state through plaque detection. Quantitative Light-Induced Fluorescence-Digital (QLF-D), which can assess mature plaque, can be used to assess the oral hygiene status of individuals and to establish an adequate intervention plans for them. The subjects of the study participated in the clinical training at Department of Dental Hygiene, N University. The subjects completed questionnaires on general characteristics and oral hygiene methods. Then, photographs of maxillary and mandibular anterior teeth of the subjects were taken using the QLF-D. After the oral examinations, gingival state was recorded according to the $L{\ddot{o}}e$ & Silness's Gingival Index (GI). In addition, a plaque control record was calculated in percentage using disclosing agent. The analysis of the relation between the plaque control record and the QLF-D scores showed positive correlation (r=0.638, p<0.001), and the analysis of the relation between the QLF-D scores and the gingival bleeding index scores also showed positive correlation (r=0.562, p<0.001). Besides, the study classified the participants into healthy gingiva group and the gingivitis group according to the classification criteria of GI, and when the difference of QLF-D scores between the groups was analyzed, the QLF-D scores were statistically significantly higher (t=-2.785, p=0.007) in the gingivitis group ($1.71{\pm}1.545$) than the healthy gingiva group ($0.74{\pm}1.290$). When the differences in mean values of the QLF-D scores were analyzed against and the gingival bleeding index scores, the QLF-D scores of 0 and 1 belonged to the category of normal gingival state while the QLF-D scores of 2, 3 and 4 belonged to the category of gingivitis state. Therefore, it is deemed that the red fluorescence detected by the QLF-D from the mature supragingival plaque can be used for monitoring the state of gingivitis.

The Effect of Midazolam As Sedative Agent in Bronchoscopy (기관지내시경 검사시 전처치로서 Midazolam의 효과)

  • Background : Bronchoscopy is an important diagnostic and a therapeutic tool in chest medicine. However, most patients feel that a bronchoscopy is an unpleasant procedure, and it is important to sedate the patients appropriately, particularly where repetitive examinations are required. Midazolam is a sedative drug with amnestic qualities and a rapid 2 hour half-life. This study have attempted to determine the safety, appropriate dosage, and the effect of midazolam premedication in patients who underwent a bronchoscopy. Methods : One hundred and eighty consecutive patients undergoing bronchoscopy were enrolled in this study. The patients received a midzolam doses of 0.03 mg/kg, 0.06 mg/kg, or a placebo. An additional dose of lidocaine, the total number of coughs, and the duration of the procedures were recorded with monitoring the the blood pressure, heart rate, and oxygen saturation. The level of satisfaction was assessed by the patient, bronchoscopist, and the nurse. Results : The blood pressure, pulse rates, oxygen saturation, number of coughs, lidocaine dose, and procedure time in the 3 groups were similar. There was a trend for the midazolam 0.03 mg/kg group to satisfy bronchoscopists more than the other two groups. The nurses' acceptability was lower in the midazolam 0.06 mg/kg group than the other groups. The patients' acceptablity was greater in both the midazolam 0.03 mg/kg and 0.06 mg/kg groups than in the control group. Conclusion : Sedation with low doses of intravenous midazolam is a safe technique for fiberoptic bronchoscopy with a low morbidity and high acceptable to patients and bronchoscopists.
